Activation of Antioxidant-Response Element (ARE), Mitogen- Activated Protein Kinases (MAPKs) and Caspases by Major Green Tea Polyphenol Components during Cell Survival and Death

  • Green tea polyphenols (GTP) have been demonstrated to suppress tumorigenesis in several chemical-induced animal carcinogenesis models, and predicted as promising chemopreventive agents in human. Recent studies of GTP extracts showed the involvement of mitogen-activated protein kinases (MAPKs) in the regulation of Phase II enzymes gene expression and induction of apoptosis. In the current work we compared the biological actions of five green tea catechins: (1) induction of ARE reporter gene, (2) activation of MAP kinases, (3) cytotoxicity in human hepatoma HepG2-C8 cells, and (4) caspase activation in human cervical squamous carcinoma HeLa cells. For the induction of phase IIgene assay, (-)-epigallocatechin-3-gallate (EGCG) and (-)-epicatechin-3-gallate (ECG) potently induced antioxidant response element (ARE)-mediated luciferase activity, with induction observed at 25 $\mu\textrm{m}$with EGCG. The induction of ARE reporter gene appears to be structurally related to the 3-gallate group. Comparing the activation of MAPK by the five polyphenols, only EGCG showed potent activation of all three MAPKs (ERK, JNK and p38) in a dose- and time-dependent manner, whereas EGC activated ERK and p38. In the concentration range of 25 $\mu\textrm{m}$ to 1 mM, EGCG and ECG strongly suppressed HepG2-ARE-C8 cell-growth. To elucidate the mechanisms of green tea polyphenol-induced apoptosis, we measured the activation of an important cell death protein, caspase-3 induced by EGCG, and found that caspase-3 was activated in a dose- and time-dependent manner. Interestingly, the activation of caspase-3 was a relatively late event (peaked at 16 h), whereas activation of MAPKs was much earlier (peaked at 2 h). It is possible, that at low concentrations of EGCG, activation of MAPK leads to ARE-mediated gene expression including phase II detoxifying enzymes. Whereas at higher concentrations of EGCG, sustained activation of MAPKs such as JNK leads to apoptosis. These mechanisms are currently under investigation in our laboratory. As the most abundant catechin in GTP extract, we found that EGCG potently induced ARE-mediated gene expression, activated MAP kinase pathway, stimulated caspase-3 activity, and induced apoptosis. These mechanisms together with others, may contribute to the overall chemopreventive function of EGCG itself as well as the GTP.

  • In general, transportation sources include both on-road vehicles and off-road equipment. Off-road vehicles have usually used diesel engines, which have the disadvantage of high NOx emission. Common rail direct injection (CRDI) and after-treatment systems have been applied to meet the exhaust gas emission regulations for diesel vehicles. In the present, agricultural machinery has satisfied the Tier 3 emission regulations by using waste gate turbocharger (WGT) and internal exhaust gas recirculation (EGR). In this paper, the combustion and emission characteristics of an EGR system applied to a 56kW off-road vehicle in non-road transient cycle (NRTC) mode have been investigated. The EGR map was made from foundation experiments determining the EGR duty for all engine operating conditions, and then this map was applied to the NRTC mode. Consequently, the NOx emission was reduced by the EGR system, and the Tier 4 interim emission regulations were satisfied by using both the EGR system and an after-treatment system.

Effects of Hot Water and Specific Gravity Treatment on Germination of Legumes and Green Manure Crops (열탕처리와 염수선 처리가 콩과 녹비작물 발아에 미치는 영향)

  • Green manure crops have a variety of benefits, including improving soil fertility, removing soil salinity, enhancing soil aeration and moisture regulation, reducing the use of chemical fertilizers, reducing nitrous oxide emissions, reducing herbicide use through weed-suppressive effects, promoting agricultural ecosystem protection and carbon dioxide absorption capacity. To find ways to increase the germination rate of legumes and green manure crop seeds, which are imported in large quantities every year. It was carried out that two seed priming methods, osmotic priming and heat treatment, and compared their effects. Heat treatment was treated for 10 minutes at 40℃ or 60℃, followed by rinsing with water. Osmotic priming was applied for 30 seconds in a saline solution with a specific gravity of 1.13. Overall, there was no significant difference in the final germination rate, but it was found that osmotic priming and heat treatment affected the germination speed. However, applying heat treatment and osmotic priming simultaneously did not affect the germination characteristics. Therefore, it is suggested that heat treatment and osmotic priming can increase the germination speed of soybean and green manure crops.

  • The purpose of this study was to investigate the consistency of socially responsible consumer attitudes and behaviors toward environment problem. The major findings were as follows; 1. The socially responsible consumer behaviors toward environment problem were classified into resource conservation purchasing & using in green product and recycling behavior by factor analysis. 2. Correlation coefficient of the socially responsible attitudes and behaviors toward environment problem was found to be above. 43. This result implies that the consistency of socially responsible attitudes and behaviors was very high. 3. In the area of total socially responsible consumer attitudes and behaviors toward environment problem the related variable on the consistency of attitude and behavior were altruism perceived power of big business liberalism alienation dogmatism perceived consumer effectiveness faith on the environmental regulation policy experience of environment education and consumer i formation.

  • Needs for high ash loading in printing paper increase as green house gas (GHG) emission regulation becomes more stricter and pressure of lowering paper production cost increases. To meet the needs, a new type of filler was developed. The mixture of ground calcium carbonate (GCC) and calcium oxide was pre-floccuated with polyelectrolyte and further treated with carbon dioxide to form new calcium carbonate between GCCs. The final products were called as hybrid calcium carbonate (HCC), and its properties were compared to the GCC and the pre-flocculated GCC. Results showed increase in tensile, smoothness, opacity, and bulk for HCC.

  • Vitamin K, which is a nutritional factor, play a role in the regulation of bone metabolism. Vitamin K exists naturally in 2 forms, namely, vitamin $K_1$ (phylloquinone) in green plants and vitamin $K_2$ (menaquinone) in animals and bacteria. Vitamin $K_1$ has an effect on bone metabolism. Vitamin $K_2$ is essential for the ${\gamma}$-carboxylation of osteocalcin, a bone matrix protein containing ${\gamma}$-carboxyglutamic acid (Gla) which is synthesized in osteoblast of bone tissues. This paper is to provide the basic information of vitamin K by supplying function and biological activity of vitamin K together with vitamin K producing lactic acid bacteria and it's utilization for the dairy products production.

  • Energy saving in the transport sector in the framework of the annual growth of energy consumption, the degree of negative impact on the environment and the amount of harmful emissions are becoming increasingly important. The article considers the world tendencies of energy consumption in transportation sector and emphasizes its dependency from oil. Also article describes the dynamics of energy use and CO2 emissions from transport of city Almaty. In conclusion authors identify a number of problems in the transport sector, which hinder the implementation of energy efficiency measures and measures to reduce CO2 emissions.

  • In this article we consider the modern trends of global warming, GHG pollutions and discussions of the obligations of developed and developing countries before the UN Global Climate Summit in Paris. The article considers decarbonization as a national strategy, including complex tools for the improvement of energy efficiency, reduction of CO2 and development of emissions trading systems. The author underlines that the Central Asian countries such as Kazakhstan and Turkmenistan, have the largest GHG potential in the region, and for this reason they should be within the framework of the UNFCCC and join the international process on development of the national decarbonization strategies.Thesemeasuresallowthese countries to join the global carbon trade marketing, international financial recourses, and significantly reduce CO2 pollutions in the region.

  • United Nations Framework Convention on Climate Change(UNFCCC), an international treaty to prevent the regulation and global warming, has passed through the General Assembly for 17 times from 1992 to 2011. To create a performance plan specific, such as the Kyoto Protocol, it is to promote the reduction of energy consumption and greenhouse gases worldwide. In South Korea, Korea Infrastructure Safety and Technology Corporation(KISTEC) is working and supporting for developing method of green remodeling policy. Among several solutions, "Remodeling" ensures to increase the energy efficiency of buildings and to reduce the emissions of greenhouse gases, and consequently it could maintain buildings high efficiency. For remodeling projects, it is most important to determine construction plan including remodeling scopes and methods, while the plan has been determined by the engineer's background and experience. Therefore, this study aims at developing a method for selecting optimal skin system which has remarkable influence to energy performance of the buildings.

  • The increased interest in the design of energy efficient ships post IMO regulation on enforcing EEDI has encouraged researchers to reevaluate the numerical methods in predicting important hull design parameters. The prediction of added resistance and stability of ships in the rough sea environment dictates selection of ship hulls. A 3D panel method based on Green function is developed for vessel motion prediction. The effects of parametric instability are also investigated using the Volterra series approach to model the hydrostatic variation due to ship motions. The added resistance is calculated using the near field pressure integration method.
