Rapid Detection of Fluorescent DNA on Denaturing Polyacrylamide Gel by Using Gel Scanner (겔스캐너를 이용한 변성아크릴아마이드 겔의 형광 DNA 검출)

  • The denature polyacrylamide gel stain silver nitrate is used for routine nucleic acid detection. More sensitive stains, such as Vistra Green, SYBR Green are available to address a broad range of DNA applications requiring lower detection limits in polyacrylamide gel formats. Gel Scanners, laser-scanning instruments, provide sensitive fluorescence detection of DNA gel stains. We established one step fluorescent impregnation enhanced sensitivity with simple, rapid and low cost. We have applied this fluorescent staining procedure for the routine analysis of DNA profiles generated by SSR amplification.

Study for Energy Transfer from Rhodamine 6G to Malachite Green Using Time Correlated Single Photon Counting Method (시간상관 단일광자 계수법에의한 Rhodamine 6G에서 Malachite Green으로의 에너지 전달 연구)

  • We investigated the nonradiative energy transfer process from Rhodamine 6G to Malachite Green in ethylen glycol solvent using time correlated single photon counting system equipped with a modelocked Ar ' laser. The reduced concentration and critical transfer distance for various acceptor concentration were obtained by using a full-fitting analysis of the fluorescence decay curves. We found that Huber model is more suitable than Forster model and the influence of energy migration through the dipole-dipole interaction becomes more significant for the low acceptor concentrations relative to the donor concentration($5\times 10^4$mol/l).

Output power characteristics of a CW Nd:YVO4/KTP laser pumped by a tunable Ti:Sapphire laser (파장가변 티타늄 사파이어 레이저로 펌핑하는 연속발진 Nd:YVO4/KTP 레이저의 출력 특성)

  • We measured the absorption rate of a Nd:YVO$_4$crystal with a thickness of 1 mm and the output power characteristics of a cw Nd:YVO$_4$/KTP laser with respect to the change of wavelength and the polarizations of a tunable Ti:sapphire pump laser with a linewidth of 0.2 nm. In the case of S-polarization (E┴$\pi$) and P-polarization (E∥$\pi$) of a pump laser, the maximum absorption rate of the crystal was 82% at 809.4 nm and 98% at 808.8 nm, and slope efficiencies for the output power of the Nd:YVO$_4$laser (1064 nm) were 43% and 52%, respectively. The maximum Nd:YYO$_4$laser output power of 516 mW was obtained from the P-polarization pump laser of 1000 mW. As a result of an intracavity frequency-doubling, slope efficiency for the output power of the Nd:YVO$_4$/KTP green laker (532nm) was 23% and the maximum output power of 205 mW with the beam quality (M$^2$) of 1.42 was obtained from the P-polarization pump laser of 1000 mW.

Two Cases of Primary Localized Amyloidosis of Larynx (원발성 국소 후두아밀로이드증 2례)

  • Primary localized laryngeal amyloidosis is an uncommon disorder of unknown cause that occurs in the absence of systemic amyloidosis or associated disease. There is a risk of either missing concomitant systemic amyloidosis or exhaustively investigating for this when it is not present through failure to appreciate the nature of the disease. We present 2 cases of primary localized laryngeal amyloidosis in supraglottic region. Biopsy of the mass of patients revealed findings consistent with amyloidosis, which were Congo red reaction with a apple green birefringence in polarized light fluorescence microscopy. An extensive workup for systemic amyloidosis was negative. All of two cases were treated by vaporization via $CO_2$ LASER using "Swiftlase Flshscan" for creating a wide, shallow char-free treatment site by precisely controlling ablation depth without causing residual thermal damage to tissue. The postoperative recovery of all cases was uneventful with good vocal quality and no aspiration. At the present time, the patients have no evidence of disease, recurrence and complication.

Unequal-path Low-coherence Interferometry Using Femtosecond Pulse Lasers for Surface-profile Metrology (펨토초 레이저를 이용한 형상 측정용 비동일 광경로 저결 맞음 간섭계)

  • We discuss two possibilities of using femtosecond pulse lasers as a new interferometric light source for enhanced precision surface-profile metrology. First, a train of ultra-fast laser pulses yields repeated low temporal coherence, which allows unequal-path scanning interferometry, which is not feasible with white light. Second, the high spatial coherence of femtosecond pulse lasers enables large-sized optics to be tested in nonsymmetric configurations with relatively small-sized reference surfaces. These two advantages are verified experimentally using Fizeau and Twyman-Green type scanning interferometers.

  • In this paper, we propose an BRDF measurement method based on laser light scattering, which is very effective for roughness measurement. The measurement setup has a very simple configuration, which consists of a collimated green laser, a detector. The experimental tests show that the BRDF measurement has difference between good Au Pad and bad Au Pad.

  • Particle accelerator has mainly used in nuclear field only because of the large scale of the facility. However, since laser-plasma particle accelerator which has smaller size and spends less cost developed, the availability of this accelerator is expended to various research fields such as industrial and medical. This paper suggests a visualization system to control the laser-plasma particle accelerator efficiently. This system offers real-time 3D images via convert HDF file comes from plasma data obtained from PIC simulation into OpenGL texture type to analyse and modify plasma data. After that, it stores high-resolution rendering images of the data with external renderer hereafter.

  • We constructed a frequency doubler with a frequency-stabilized Nd:YAG Laser, a nonlinear crystal KTP, and an external resonant cavity. Using type II phase-making method and a resonant cavity, one usually have a serious problem that the second harmonic conversion efficiency is decreased greatly, for under usual circumstances ordinary ray and extraorninary ray cannot be resonated in a cavity simultaneously. With the change of temperature and incident angle of KTP, we found an optimized condition for the phase-matching and double-resonance. As a result we produced 26 mW of green light with 318 mW of fundamental laser power from a cw Nd:YAG laser.

Microstructure-Properties Relationships of Ti-6Al-4V Parts Fabricated by Selective Laser Melting

  • This work investigates the relationships between the static mechanical properties of Ti-6Al-4V manufactured through selective laser melting (SLM) and post-process heat treatments, namely stress relieve, annealing and hot isostatic pressing (HIP). In particular, Ti-6Al-4V parts were fabricated in three different build orientations of X, Z, and $45^{\circ}$ to investigate the multi-directional mechanical properties. The results showed that fully densified Ti-6Al-4V parts with densities of up to 99.5% were obtained with optimized SLM parameters. The microstructure of stress relieved and mill annealed samples was dominated by fine ${\alpha}^{\prime}$ martensitic needles. After HIP treatment, the martensite structure was fully transformed into ${\alpha}$ and ${\beta}$ phases (${\alpha}+{\beta}$ lamellar). Within the realm of tensile properties, the yield and ultimate strength values were found statistically similar with respect to the built orientation for a given heat treatment. However, the ductility was found orientation dependent for the HIP samples, where a lower value was observed for samples built in the X direction.
