토마토 꽃과 수정용 벌집으로부터 잿빛곰팡이병 방제 길항균주 선발 (Selection of Biocontrol Agent of Tomato Gray Mold Disease from Flower and Pollinator Hive)

  • 국내에서 생산하는 과채류에서 발생되는 잿빛곰팡이병은 명확한 생물학적 방제 방법으로 제시된 바가 없는 실정이다. 국내에서 사용 및 개발되고 있는 화학적 방제 약제의 경우 약제저항성 병원균의 출현이 지속되고 있는 상황이다. 또한 임의 기생성으로 인해 기주의 여부에 상관 없이 생존이 가능하므로 경종적 방제 또한 어려운 실정이다. 이에 본 연구에서는 토마토 꽃에 정착력이 우수한 길항미생물을 선발하기 위해 시설재배지의 토마토 화기와 벌집 시료로 미생물을 분리하여 방제균을 선발하였다. 총 6개월의 토마토 재배기간 중 꽃에서 1,004개 균주, 벌집에서 925균주를 분리하여, 잿빛곰팡이 병원균을 억제력이 우수한 6균주를 선발하였다. 선발된 6균주는 잿빛곰팡이 병원균에 대한 항균활성이 우수할 뿐 아니라, cellulase와 protease의 활성 또한 우수한 것으로 검증되었다. 선발된 균주는 Paenibacillus polymyxa 로 동정 되었으며, 토마토의 잿빛곰팡이 발병률을 약 75% 감소시키는 효과를 나타내었다.

토마토 재배에서 점무늬병 및 잎곰팡이병 발생 예측 및 방제력 연구 (Forecasting Leaf Mold and Gray Leaf Spot Incidence in Tomato and Fungicide Spray Scheduling)

  • 이번 연구는 토마토잎곰팡이병 및 점무늬병에 대한 방제력을 개발하고, 온도와 잎의 결로시간에 따른 발생정도를 평가하기 위해 수행되었습니다. 첫 번째 병발생 예측 실험에서는 토마토묘에 Fulvia fulva, Stemphylium lycopersici 분생포자를 각각 1 × 104 conidia·mL-1로 접종시키고 10-25℃(F. fulva) 및 10-30℃(S. lycopersici)에서 0-18시간 동안 이슬생육상에 두었다. 살균제를 이용한 방제 연구에서는 잎곰팡이병 방제를 위하여 트리미다졸, 폴리옥신 B, 이미녹타딘 트리스(Belkut)를 처리하였으며 점무늬병 방제를 위하여서는 에트리디아졸 + 티오파네이트(가지란)과 삼염기 황산구리(세빈나)를 처리하였다. 엽결로시간이 9시간 이상에서 잎곰팡이와 점무니병이 발생하였으며 결로시간이 길어질수록 병발생률이 높았다. 잎곰팡이병의 발생률은 20℃와 15℃에서 더 높았고, 점무늬병은 25℃와 20℃에서 발병률이 증가하였다. 포자 접종 14일 후에 잎곰팡이병 및 점무늬병이 발생하였으며 잠복기는 14-15일로 추정되었다. 잎곰팡이 및 점무늬병 포자가 처리된 식물체에 접종 후 0시간부터 240시까지 살균제 처리를 한 결과 살균제 종류와 상관없이 일찍 약제를 처리한 식물체에서 방제가가 높았으며 150시간이 지난 처리에서는 방제가가 급격히 떨어졌다. 이번연구에서 개발된 병예측모델과 방제력은 토마토의 잎곰팡이병과 점무늬병을 적기에 방제하고 효과적으로 관리하는 데 도움이 될 수 있을 것이다.

시설재배에서 Botrytis cinerea에 의한 가지 잿빛곰팡이병 (Gray Mold Rot of Eggplant Caused by Botrytis cineraea in Greenhouse)

  • Botrytis cinerea E12 was isolated from the leaves, flowers and fruits of eggplant in the greenhouse in Halrim, Kimhae and Dejeo, Pusan. The leaves infected with the pathogen were appeared initially brown-color, small gray spots at the edge, and finally fall down. The fruit was showed the symptoms of circular or irregular shapes, followed by sunken. When the symptoms were developed, the conidia formed on the surface with gray color. To determine the pathogenicity of B. cinerea E12 against the eggplants, the conidia were suspended with 30% tomato juice, PDB and sterile water, respectively. The result showed that the conidial suspension with 30% tomato juice was highly effective on the pathogenicity as more than 90%. Moreover, the symptoms caused by inoculum were the same as those of wild-type pathogen.

금형주조한 오스템퍼 회주철의 기계적성질 및 파괴인성 (Mechanical Properties & Fracture Toughness of Austempered Gray Cast Iron(AGI) by Permanent Mould Casting)

  • The mechanical properties and fracture toughness of permanent mold cast austempered gray cast iron(AGI) were compared to those of sand cast AGI. The iron was melted to eutectic composition in order to get better castability especially in permanent mold casting. Specimens prepared for tensile, impact and fracture toughness test were austenitized at $900^{\circ}C$ and austempered at $270^{\circ}C$ and $370^{\circ}C$ for 1 hour. The strength, impact and fracture toughness of permanent mold cast AGI were found to be superior to those of sand cast AGI. The maximum value of 836 MPA in tensile strength, was obtained at the austempering temperature of $270^{\circ}C$. But ductility of AGI was not improved by permanent mold casting.

First Report of Botrytis cinerea as a Postharvest Pathogen of Blueberry in Korea

  • Gray mold of blueberry caused by Botrytis sp. is reported for the first time in Korea. A detailed description of the fungus is given, along with its rDNA internal transcribed spacer sequence. The fungus was identified as Botrytis cinerea based on mycological characteristics and molecular data.

  • 무화과 잿빛곰팡이병은 2010년부터 2011년에 아열대작목을 시범적으로 재배하고 있는 전북 농업기술원 하우스 포장에서 발병하였다. 잿빛곰팡이병은 11월경의 저온상태에서 발병이 심하였으며, 병원균은 갈색의 일부분으로 시작하여 점차 진전되어 후기에는 과실 전체가 잿빛곰팡이로 뒤덮였다. 잿빛곰팡이로 뒤덮인 과실은 갈색 솜털을 덮고 있는 모양이었으며, 육안으로도 곰팡이 포자를 쉽게 관찰할 수 있었다. 병원균에 감염된 과실은 처음에 갈색~암 갈색 수침상으로 부패하기 시작하였고, 잿빛곰팡이균사와 포자가 밀생하였다. 무화과 잿빛곰팡이병의 분생포자는 구형으로 $7.3-14.6{\times}6.8-11.1{\mu}m$이며, 무색으로 단세포이었다. 또한 수지상으로 분지한 분생자경위에 무수히 형성 하였다. 분생자경의 크기는 $15-33{\times}2{\mu}m$이며, 갈색 또는 투명한 색을 띄고 있었다. rDNA ITS 영역의 염기서열 분석결과 무화과 잿빛곰팡이병을 일으키는 병원균은 Botrytis cinerea로 등록된 GenBank accession number HQ171052, EU519210, HQ171053, FN812726, HM849615, EU563120 과 100% 일치하는 것으로 나타났다. 또한 Botrytis spp. 종간 비교에서도 B. cinerea와 같은 clade에 속함을 확인할 수 있었으며, 다른 group과는 57%의 similarity를 보였다. 따라서 무화과 잿빛곰팡이병을 일으키는 병원균을 분리배양하여 균학적 특성과 rDNA ITS영역의 염기서열을 분석한 결과 이미 보고된 B. cinerea와 일치하였다.

Effect of Chlorine Dioxide on Freshness of 'Maehyang' Strawberries during Export

  • The objective of this study was to assess the effect of precooling and application of gaseous $ClO_2$ on the retention of freshness and quality of 'Maehyang' strawberry fruits intended for export. 'Maehyang' strawberry fruits (Fragaria ${\times}$ ananassa Duch.) were grown in commercial greenhouses and then harvested. Fruits of uniform and medium size at 60% ripeness were selected and assigned to one of four treatment groups: non-treatment (control), precooling only (PO), gaseous $ClO_2$ only (GCO) or precooling combined with gaseous $ClO_2$ (P + C). Weight loss was lowest in the PO treatment and greatest in the GCO treatment after export. Compared to the control and PO treatment groups, strawberry fruits in the GCO treatment group maintained high brightness and high chroma. Six days after shipping, fruits in the P + C treatment group had the highest soluble solids content, even as high as $10.05^{\circ}Brix$; the lowest value was observed in the PO treatment. The incidence rate of gray mold in strawberry fruits was 20% and 17% in the control and the PO treatment, respectively; in the GCO treatment, the incidence rate of gray mold amounted to 10%. No gray mold was observed in the P + C treatment group. These results indicate that gaseous $ClO_2$ treatment combined with precooling (P + C) was effective in maintaining the freshness of 'Maehyang' strawberry fruits intended for export from South Korea to Hong Kong.

Production of Surfactin and Iturin by Bacillus licheniformis N1 Responsible for Plant Disease Control Activity

  • Bacillus licheniformis N1, previously developed as a biofungicide formulation N1E to control gray mold disease of plants, was investigated to study the bacterial traits that may be involved in its biological control activity. Two N1E based formulations, bacterial cell based formulation PN1E and culture supernatant based formulation SN1E, were evaluated for disease control activity against gray mold disease of tomato and strawberry plants. Neither PN1E nor SN1E was as effective as the original formulation N1E. Fractionation of antifungal compounds from the bacterial culture supernatant of B. licheniformis N1 indicated that two different cyclic lipopeptides were responsible for the antimicrobial activity of the N1 strain. These two purified compounds were identified as iturin A and surfactin by HPLC and LCMS. The purified lipopeptides were evaluated for plant disease control activity against seven plant diseases. Crude extracts and purified compounds applied at 500 ${\mu}g/ml$ concentration controlled tomato gray mold, tomato late blight and pepper anthracnose effectively with over 70% disease control value. While iturin showed broad spectrum activity against all tested plant diseases, the control activity by surfactin was limited to tomato gray mold, tomato late blight, and pepper anthracnose. Although antifungal compounds from B. licheniformis N1 exhibited disease control activity, our results suggested that bacterial cells present in the N1E formulation also contribute to the disease control activity together with the antifungal compounds.

Effect of Thymol and Linalool Fumigation on Postharvest Diseases of Table Grapes

  • Several postharvest diseases of table grapes (Vitis vinifera) occur during storage, and gray mold rot is a particularly severe disease because the causal agent, Botrytis cinerea, grows at temperatures as low as $0^{\circ}C$. Other postharvest diseases, such as those caused by Penicillium spp. and Aspergillus spp., also often lead to deterioration in the quality of table grapes after harvest. The use of plant essential oils such as thymol and linalool, to reduce postharvest diseases in several kinds of fruits, including table grapes and oranges, has received much attention in European countries. However, to the best of our knowledge there has been no report of the use of thymol fumigation to control gray mold in table grapes in Korea. Thymol ($30{\mu}g/mL$) and linalool ($120{\mu}g/mL$) significantly inhibited mycelial growth and conidia germination of B. cinerea. The occurrence rate of gray mold rot of B. cinerea and other unknown fungi was significantly reduced by fumigation with $30{\mu}g/mL$ thymol in several table grape cultivars, such as Campbell early, Muscat Bailey A, Sheridan, and Geobong. In this study, fumigation with $30{\mu}g/mL$ thymol, had no influence on the sugar content and hardness of grapes, but reduced fungal infection significantly. This suggests that $30{\mu}g/mL$ thymol could be utilized to reduce deterioration of grapes due to gray mold and other fungal infections during long-term storage.
