Studies on Anti-Inflammatory and Anti-Melanogenic Effect of Grape Fruit Stem Extract (포도송이가지 추출물의 항염증 및 미백효능에 대한 연구)

  • Objectives : The various grape extracts derived from grape pulp, seed and skin, containing various types of polyphenols and flavonoids, have been known to have anti-inflammatory, antioxidant and improve cardiovascular condition as well as sun's damaging effects. However, there have been rare reports of various beneficial effects of grape fruit stem extract (GFSE), one of the waste products of grapes. We investigated anti-inflammatory and melanogenesis inhibitory effects of GFSE. Methods : One-hundred gram of grape fruit stem was extracted with 80% ethanol at room temperature for 3 days. After filtration, the ethanol was removed using vacuum evaporator, then lyophilized to obtain the dry extract which was stored at $-20^{\circ}C$ until used. NO levels were measured by using Greiss reagent. Prostaglandin $E_2$ ($PGE_2$) production was measured by ELISA assay. The expression levels of iNOS, COX-2, TRP-1 and TRP-2 were evaluated by western blot analysis. Results : GFSE reduced the level of nitric oxide and prostaglandin $E_2$ ($PGE_2$) production in a dose-dependent manner, compared to control. Expressions of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) protein were also effectively inhibited by the GFSE. In a tyrosinase inhibitory activity, GFSE significantly reduced the tyrosinase activity and melanin content in a dose dependent manner, compared to control. GFSE also decreased the expression of tyrosinase related protein-1 (TRP-1) and tyrosinase related protein-2 (TRP-2), known as a melanocyte-specific gene product involved in melanin synthesis. Conclusions : Therefore, these results indicated that GFSE had powerful anti-inflammatory and anti-melanogenic effects.

Resveratrol Extraction from Grape Fruit Stem and its Antioxidant Activity (포도 송이가지를 이용한 레스베라트롤의 추출 및 항산화 활성)

  • The extraction conditions for resveratrol production from grape fruit stem, which is a by-product of grape processing, were optimized to develop high-functional grape-based products. Additionally, the bioefficacy of grape fruit stem extract (GFSE) as an antioxidant agent was evaluated. Resveratrol was extracted using various experimental conditions such as extractant type, extractant concentration, raw material-extractant ratio, extraction time and temperature, and the results were analyzed using a statistical program (SPSS). The resveratrol yield was the highest when 80% ethanol with a raw-material-extractant ratio of 1:10 (w/v) was used. In addition, the optimal temperature and time were selected as $60^{\circ}C$ and 90 min, respectively. When the antioxidant activity was analyzed and expressed as DPPH radical scavenging activity and SOD-like activity, the antioxidant activity of GFSE was higher than that of BHT, BHA and L-ascorbic acid. Finally, it was found that GFSE could be used as a raw material for the production of high antioxidant agents.

Effects of Auxins end Cytokinins on Callus Induction from Leaf Blade, Petiole, and Stem Segments of in Vitro-grown 'Sheridan' Grape Shoots

  • To establish an the mass production system of grape anthocyanin pigments through callus and cell suspension culture, the effects of various combinations of auxins and cytokinins on friable callus production were studied. for friable callus production, 2,4-D was superior to other regulators. IAA at 2 mg/L induced callus from stem and petiole while NAA resulted in rooting. Callus induction rate increased with the 2,4-D level, and stem segments were superior to leaf blade or petiole, showing nearly 100% with 1 and 2 mg/L 2,4-D from petiole and stem. Combined treatments of 2,4-D + kinetin and NAA + BA also yielded friable callus from stem segments. In treatments with 1 mg/L 2,4-D + 1 mg/L kinetin and 1 mg/L NAA + 1 mg/L BA, callus induction rate was nearly 100%. The combination effect of 2,4-D and BA on anthocyanin production was not significant.

Evaluating the resistance to crown gall in grape rootstocks.

  • To evaluate the resistance to crown gall in grape rootstocks, cuttings from twenty seven grape rootstocks were inoculated with Agrobacterium vitis Cheonan 493 and size of galls from grapevines was measured in a greenhouse. Tumors were formed in all varieties of grape rootstocks tested in this study and no grape rootstock variety was immune to crown gall. Tumors were found on the stems of all plants tested in '196-17'and '41B' Based on measuring size and weight of galls formedon the stem of grape rootstocks, '779P' was extremely susceptible to crown gall. Some varieties such as 'Gloire', '140R', '101-l4M', '3309C', and '333EM' found to be resistant, while '99R', '1447P', 'Rupestris du lot', '110R', 'Freedom', and '41B'were susceptible and '1103P', '5C', '420A', 'Golia', and '5BB' were moderately susceptible to crown gall.

Grape Pruning Stem Extract (GPSE) Suppresses Allergy and Skin Proliferation Inhibition against UVB Induced Skin Damage (포도전정가지 추출물이 UVB에 유도된 피부 손상에 대한 알레르기와 피부증식 억제효과)

  • This study examined the effects of polyphenols in grape pruning stem extracts (GPSE) using grape stems discarded after harvest. The inhibitory effects on allergy, proliferation, and apoptosis in UVB-induced HR-1 hairless mice were analyzed. The applicability as a material for functional food and functional cosmetics was evaluated. The contents of the active ingredients of GPSE were analyzed by HPLC. After UVB irradiation on the dermis of HR-1 hairless mice, the number of mast cells was determined by toluidine blue staining to confirm that the skin allergic reaction was caused by GPSE. The results were similar to the normal group in the group applied GPSE, and there was no allergic reaction in the GPSE application group and a significant decreased compared to the sun cream control. PCNA immunohistochemical staining of the epidermal proliferation factor had an inhibitory effect on epidermal epithelial cell proliferation in all concentrations of GPSE and serum base mixture as an average of 42% compared to the control group. The mixture of GPSE and serum base suppressed the apoptosis inhibition rate by 27% on average compared to the control. These results confirmed the inhibitory effects GPSE on the allergic, proliferation, and apoptosis activities by with a serum base on UVB-induced skin damage. GPSE is a functional ingredient with potential skin protection effects, and has a high utilization as an ingredient for functional cosmetics.

Optimum Dyeing Condition of Cotton by Fermented Grape By-products with Degraded Protein Mordant (발효 포도부산물의 단백질 분해물 매염제를 활용한 염색 최적조건에 관한 연구)

  • Many of the natural dyes used for natural dyeing are difficult to maintain colorfastness due to their complex structure and specific properties. Therefore, there is a need for developing of color sustainable ability for use as an advanced coloring agent for fabrics, which would eco benign or not. In this study, the natural dye extracted from the waste of grape fruits was used to dye cotton fabric. Thus, the present study aims at extraction of color from grape seeds, skin, and stem through fermentation and then employing the same in dyeing and mordanting of cotton. Dyeing experiments were done under different conditions of fermentation and protein type mordants which were treated before and after dyeing. Experimental fabrics were used with cotton after scouring. Color value of dyed fabrics and color fastness of cotton dyed fabrics to washing and light were measured. The fastness of dyed experimental fabrics was increased by mordanting of protein fermentation and the color of dyed cotton was light red purple. The color of dyed fabric found with the optimum mordant treatment when treated with pre milk-mordant at $40^{\circ}C$ for 30min and 3% grape seed extract. On the whole, reddish tone very slightly increased with the milk pre-mordant. The color fastness of dyed cotton fabrics to light and washing was increased after fermentation.

  • The resveratrol contents in grapes showed a great variation according to the cultivars. In spite of it, a high resveratrol content in fruit was found generally in cultivars of Shigyoku, Pione, Muscat of Alexandria (black colored cultivars) and Muscatel Ruso, Gold Finger, Niunai (green colored cultivars). But in case included fruit stem and rachis, some cultivars, i.e. Shigyoku, Muscat of Alexandria and Baby Finger, showed relatively higher resveratrol content, and these cultivars could be advantageous for cultivation without chemical fungicide, because resveratrol was known as a phytoalexin. While resveratrol was not detected in fruit flesh, fruit stem and rachis contained considerably high content of resveratrol; that shows resveratrol may be synthesized in these parts.

  • Contents of resveratrol in different parts of Gerbong, Campbell, and Sheridan grapes were measured through HPLC analysis. Resveratrol contents in the flesh, skin, seed, and fruit stem of each grape showed significant differences, whereas little difference was observed among the cultivars. In fruit stem, and skin and seed, resveratrol contents were 170 to 440 and 4 to $8\;{\mu}g/g-dry$ weight, respectively, an indication that the fruit stem may be a good resource for resveratrol production.

Improvement Effect of Corn Silk, Perilla Leaf and Grape Stem Extract Mixture against UVB-Induced Skin Damage and Compound 48/80-Induced Pruritus (옥수수수염, 들깻잎 및 포도줄기 복합추출물의 UVB 조사 피부 손상 및 Compound 48/80 유도 가려움증 개선 효과)

  • This study was conducted to evaluate the synergistic protective effects of mixtures of corn silk, perilla leaf and grape stem extract (CPG mixture) against UVB-induced skin damage and compound 48/80-induced pruritus in mice. The results showed that treatment with CPG mixture exhibited much stronger suppressive effect on erythema and melanin index as well as melanin formation than treatment with ascorbic acid (AA) in UVB-irradiated mice. Moreover, the treatment with CPG mixture showed ameliorative effect on immune cell infiltration and collagen fiber destruction in UV-irradiated mice. The treatment with CPG mixture inhibited glutathione (GSH) depletion, lipid peroxidation and production of pro-inflammatory cytokines in UVB-irradiated mice. Furthermore, the treatment with CPG mixture inhibited compound 48/80-induced scratching behavior and histological changes in mice. Taken together, these results indicated that CPG mixture has potentials as functional and therapeutic materials against skin damage and itch-related skin diseases.

Comparison of the change in quality indices during distribution period by import season in three grape cultivars

  • As a result of analyzing the quality of imported grapes during the 2018 season, the hardness of the grape berry was found to be 10 N or less in total. In the case of Chilean grapes, the soluble solids tended to be lower as imports and distribution periods were delayed. The berry weight was the largest at 14.4 - 14.8 g for the 'Red Globe', 7.1 - 7.4 g for the 'Thompson Seedless' and 6.0 - 7.0 g for the 'Crimson Seedless'. The 'Crimson Seedless' grapes imported from Chile, which had a high berry shatter rate, had a shorter pedicel length, pad width and brush length than that of the other 2 varieties. Regardless of the grape varieties, the weight loss during the distribution at room temperature ($25^{\circ}C$) increased rapidly as the import season was delayed. Additionally, it was found that the later the distribution seasons, a higher stem browning index was observed regardless of the variety. The 'Thompson Seedless' was revealed to be more susceptible to browning than that of the other varieties. The shrinkage of the stem and the pedicel browning progressed within a short time during shelf-life as the distribution time was delayed. The incidence of berry decay of the imported grapes tended to increase with the progression of the distribution period. In the fruits distributed on April, except for the 'Crimson Seedless', the grapes were rapidly corrupted in the second half of the distribution. The 'Red Globe' grapes completely lost marketability due to a berry decay of 26.1% and 69.9% at 9 and 12 days after shelf-life, respectively.
