인태아 상경신경절내 소형의 과립함유세포에 관한 전자현미경적 연구 (Ultrastructural Study on the Development of the Small Granule-Containing Cells in Superior Cervical Ganglion of Human Fetus)

  • The development of small granule-containing cell in the superior cervical ganglion was studied by electron microscopic method in human fetuses ranging from 40 mm to 260 mm crown rump length (10 to 30 weeks of gestational age). At 40 mm fetus, the superior cervical ganglion was composed of clusters of undifferentiated cells, primitive neuroblasts, and unmyelinated nerve fibers together with blood vessels. At 90 mm fetus, the superior cervical ganglion consisted of neuroblasts, satellite cell, small granule-containing cells, and unmyelinated nerve fibers. Two morphological types of the small granule-containing cells in the superior cervical ganglion were first indentified at 90 mm fetus, but were rare. Type I granule-containing cell occurred in solitary and had long processes, whereas type II cells tend to appeared in clusters near the blood capillaries. The granule-containing cells were characterized by the presence of dense-cored vesicles ranging from $150{\sim}300nm$ in diameter in both the cell bodies and processes. Other organelles included abundant mitochondria, rough endoplasmic reticulum, neurotubules, and widely distributed ribosomes. The granule-containing cells had long processes similar to those found in principal ganglionic cells. They could be identified by their content in dense-cored vesicles. The small granule-containing cells increased somewhat in size and number with increase of fetal age. Synaptic contacts were first found on the solitary granule-containing cell at 150 mm fetus. Synaptic contacts between the soma and processes of type I granule-containing cells and preganglionic axon terminals were observed. In addition, synaptic junctions between the processes of granule-containing cells and presumed dendrite of postganglionic neuron were also observed from 150 mm onward. On the basis of these features type I granule-containing cells could be considered as interneurons. The clusters of type II granule-containing cells were located in the interstitial or subcapsular portions of the ganglion, and had short processes which ended in close relation to fenestrated capillaries. Therefore it may be infer that clusters of type II granule-containing cells have an endocrine function.

The cytoplasm of the neuroblast contained rough endoplasmic reticulum, mitochondria, Golgi complex, Nissl bodies and dense-cored vesicles. As the neuroblasts grew and differentiated dense-cored vesicles moved away from perikaryal cytoplasm into developing processes. Synaptic contacts between the cholinergic axon and dendrites of postganglionic neuron and a few axosomatic synapses were first observed at 70 mm fetus. At 90 mm fetus the superior cervical ganglion consisted of neuroblasts, satellite cells, granule-containing cells, and unmyelinated nerve fibers. The ganglion cells increased somewhat in numbers and size by 150 mm fetus. Further differentiation resulted in the formation of young ganglion cells, whose cytoplasm was densely filled with cell organelles. During next prenatal stage up to 260 mm fetus, the cytoplasm of the ganglion cells contained except for large pigment granules, all intracytoplasmic structures which were also found in mature superior cervical ganglion. A great number of synaptic contact zones between the cholinergic preganglionic axon and the dendrites of the postganglionic neuron were observed and a few axosomatic synapses were also observed. Two morphological types of the granule-containing cells in the superior cervical ganglion were first identified at 90 mm fetus. Type I granule-containing cell occurred in solitary, whereas type II tended to appeared in clusters near the blood capillaries. Synaptic contacts were first found on the solitary granule-containing cell at 150 mm fetus. Synaptic contacts between the soma of type I granule-containing cells and preganglionic axon termials were observed. In addition, synaptic junctions between the processes of the granule-containing cells and dendrites of postganglionic neuron were also observed from 150 mm fetus onward. In conclusion, superior cervical ganglion cells and granule-containing cells arise from a common undifferentiated cell precursor of neural crest. The granule-containg cells exhibit a local modulatory feedback system in the superior cervical ganglion and may serve as interneurons between the preganglionic and postganglionic cells.

  • 도룡뇽 피부점액선의 미세구조를 전자현미경으로 관찰하여 다음과 같은 결과를 얻었다. 1. 피부점액선은 선체부와 분비관으로 구성되며, 선체부는 선상피세포와 근상피세포로 구성되었다. 2. 피부점액선의 선체부의 선상피세포는 명세포, 암세포, mitochondria-rich cell, endoplasmic reticulum-developed cell, secretory granule-containing cell 및 vesiculated cells들로 구성되었다. 3. 명세포, 암세포 및 mitochondria-rich cell은 미분화세포, endoplasmic reticulum-developed cell은 분비전기세포, secretory granule-containing cell은 성숙세포 그리고 vesiculated cell은 분비후기세포라고 생각된다.

흰쥐의 소뇌에서 selenium 방법에 의한 아연이 함유된 세포의 확인 (Identification of the Zinc-containing Cells in the Cerebellum of Rat by Selenium Method)

  • A zinc-specific method (selenium method) has been employed to identify the zinc-containing cells in the cerebellum of the rats. When rats were allowed to survive 24 hours after the sodium selenite administration, zinc selenide reaction products formed in zinc-containing cellular boutons are retrogradely transported to the somata of those boutons. And the zinc selenide products accumulated in somata of the cells can be rendered visible by silver amplification of developer. Zinc-containing cells identified by the method were Bergmann glial and granule cells. Labeled zinc-containing cells were absent in molecular layer and white matter of the cerebellum. In ultrastructural level, the zinc selenide products were located in lysosomes of somata of the zinc-containing cells.

개구리(Rana catesbeiana) 피부점액선(皮膚粘液腺)에 관한 전자현미경적(電子顯微鏡的) 연구(硏究) (Electron Microscopic. Study on Mucous Glands in Frog Skin)

  • This study was accomplished to investigate the ultrastructure of mucous glands in dorsal skin of frog (Rana catesbeiana) by means of scanning and transmission electron microscopes. The dorsal skin of Rana catesbeiana is composed of epidermis and dermis. The cutaneous mucous glands consist of inner glandular epithelial cells and outer myoepithelial cells. Glandular epithelial cells are divided into four types by the microscopic ultrastructure; ER-rich cell, round secretory granule-containing cell, foam-like granule mass-containing cell, mitochondria-rich cell. Myoepithelial cell has a long elliptical nucleus and filled with fibrous materials in the cytoplasm. As a result of scanning microscopic observation, the surface of dorsal skin is covered with cutaneous protrusions. The opening sites of the mucous glands are irregularly distributed in dorsal skin.

납 내성균의 균체내 납 축적 특성 (Characteristics of Lead Accumulation in Lead-Tolerant Pseudomonas aeruginosa)

  • 납 내성균인 P. aeruginosa의 균체내 납 축적 특성, 축적형태 및 균체내에 축적된 납의 회수 방법을 조사한 결과는 다음과 같다. 납 농도가 100mg/l인 용액중에서 처리 48시간 후 납 내성균주의 납 처리율은 약 97% 이상이었다. 납이 축적된 균체를 전자현미경으로 관찰한 결과, 균체의 cell wall과 cell membrane에 많은 electron-dense granule들이 형성되어 있었으며, electron-dense granule들을 EDS로 분석한 결과, 이 granule들은 lead complex인 것으로 확인되었다. 공시균주의 균체내 축적된 납은 증류수에 의해서는 거의 탈착이 되지 않았으나 EDTA 처리에 의해서는 약 87% 이상 탈착되었다. 납이 축적된 균체를 $550^{\circ}C$에서 회화시켰을 때 건조균체량의 약 30%에 해당하는 작열잔류화합물들이 생성되었으며, 이 작열잔류화합물들을 EDS로 분석한 결과, 납이 약 86% 이상 함유되어 있는 비교적 순수한 납 화합물인 것으로 확인되었다.

  • Degranulation of the rat peritoneal mast cell induced by intraperitoneal injection of horseradish peroxidase(HRP) was studied using light and electron microscopes. 1. Rat peritoneal mast cells in the Tyrode's buffered salt solution injected control group did not show any particular morphological changes following the specified time course. 2. Under the light microscope, the majority of mast cells observed 10 minutes after HRP injection were nearly the same as those of the control group. However, after 30 minutes, granule densities or staining properties of certain cells began to decrease and these appearances increased gradually until 12 hours after injection, at which time small groups of granules being stained pale-red or pink with toluidine blue were easily identified in the cytoplasm of many cells, and numerous extruded granuleg were scattered around these cells. 3. In the mast cells representing the early stage of degranulation induced by HRP, the electron densities of certain granules decreased as the size enlarged, and perigranular cavities were formed by perigranular membrane expansion. As a result, a thin cytoplasmic septum was formed between the expanded perigranular membrane and the cytoplasmic membrane in the cell periphery, and fusion of the adjacent perigranular membranes was observed in the inner side of the cell. 4. In some mast cells, one or two changes in the peripheral cytoplasmic septum could be seen. One was a focal rupture of the peripheral septum and the other was the formation of a saccule containing one or more vesicles. This saccule was thought to be used for granule-extrusion site and/or material absorptive apparatus judging from the morphological characteristics. 5. As the degranulation proceeded, the granule was extruded from the cell after partial rupture of the peripheral cytoplasmic septum. This phenomenon proceeded to-ward the inner side of the cell through the fused perigranular cavities, and consequently several distinct cavities containing a few unextruded membrane-free granules were formed throughout the cytoplasm after 12 hours. As a rule, the granule-extrusion sites were relatively fewer while the cytoplasmic cavities resulting from degranulation were more numerously observed. Thus, it was thought that the granule-extrusion sites tended to be restricted in the HRP-induced degranulation.

초기계배 발생에 미치는 Serotonin의 영향에 관한 세포생물학적 연구 (Cell Biological Studies of the EfEect of Serotonin on Chick Embryogenesis)

  • Very early stage of chick embryo cultivated in the medium containing DLtryptophan by beaker method has been investigated in vitro morphologically using electron microscopy at cellular level and found that the development of tryptophan treated chick embryos corresponding to 18∼66 hrs incubation was impaired and york granule degradation was significantly delayed. It was also found that DNA, RNA and protein biosynthesis of tryptophan treated chick embryo was greatly lowered than those of control group. Conversion of L-tryptophan into serotonin was traced using 14C-L-tryptophan and found that 13.8cA of added radioactivity was recovered from serotonin formed during 18 hrs incubation and the amounts of serotonin formed were depend upon added amount of tryptophan in e99 yolk. It seemed that the serotonin formed from external tryptophan might inhibit the degradation of yolk granule by feedback mechanism, resulting in malformation of chick embryogenesis.

구리 내성균(Pseudomonas stutzeri)의 균체내 구리 축적특성 (Copper Accumulation in Cells of Copper-Tolerant Bacteria, Pseudomonas stutzeri)

  • 구리 내성균인 P. stutzeri의 균체내 구리 축적 특성, 축적형태 및 균체내에 축적된 구리의 회수 방법을 조사한 결과는 다음과 같다. 구리 농도가 100mg/l인 용액중에서 치리 48시간 후 구리 내성균주의 구리 처리율은 약 78% 이상이었다. 구리가 축적된 균체를 전자현미경으로 관찰한 결과, 균체의 cell wall과 cell membrane에 많은 electron-dense granule들이 형성되어 있었으며, electron-dense granule들은 EDS로 분석한 결과, 이 granule들은 copper complex인 것으로 확인되었다. 구리 내성균주의 균체내 축적된 구리는 증류수에 의해서는 거의 탈착이 되지 않았으나 EDTA 처리에 의해서는 약 80% 이상 탈착되었다. 구리가 축적된 균체를 $550^{\circ}C$에서 회화시켰을 때 건조균체량의 약 23.2%에 해당하는 작열잔류화합물들이 생성되었으며, 이 작열잔류화합물들은 EDS로 분석한 결과, 구리가 약 78.4% 이상 함유되어 있는 비교적 순수한 구리 화합물인 것으로 확인되었다.
