Purification and Characterization of Myristoyl-Acyl Carrier Protein Thioesterase from Iris tectorum

  • The myristoyl-acyl carrier protein (ACP) specific thioesterase from Iris tectorum was purified to a considerable homogeneity and characterized. The enzyme was eluted with a considerable stability by double-gradients using Triton X-100 and low ionic KCl or Na-phosphate through DEAE-52, Octyl-Sepharose, Q-Sepharose, and hydroxyapatite chromatoraphy. SDS-PAGE analysis showed a single band of 39 kDa. The native molecular weight was estimated to be 82 kDa by Sephacryl S-200 chromatography, indicating that the enzyme was a dimer. The thioesterase showed a chain-length specificity to myristoyl-ACP in preference to other-ACPs. The enzyme activity decreased by 1.0 mM myristate to about 27% of the original activity, whereas the remaining activity with decanoate was about 90%. The purified thioesterase was inhibited by myristoyl-CoA more than by myristate, suggesting that the myristoyl-AGP thiolesterase might be controlled by myristic acid and/or a subsequent product myristoyl-CoA. In addition, some biochemical characteristics of the enzyme were described.

  • Recently, a variable valve timing system has been widely adopted in internal combustion engine in order to improve the fuel economy and torque at low engine speed. In addition, it is known that varying valve timing according to the various engine operations could reduce exhaust gas, especially NOx, because of residual gas by valve overlap. In this study, to improve the low exhaust gas and fuel economy at part load condition, the residual gas and back flow of exhaust gas due to valve overlap were calculated computationally. Moreover, the characteristics of engine performances and NOx formations were investigated with the experiment of combination of intake and exhaust valve timing condition. Under these various valve operating conditions, the effects of both the positive valve overlap and negative valve overlap(valve underlap) were examined simultaneously. Finally, the characteristics of cyclic THC emission were analyzed by using Fast Response FID(FR-FID) in the cylinder, intake port and exhaust port positions. Besides, the effect of the different gradients of the valve timing change on engine performance was investigated and an optimum control strategy was suggested.

  • In this paper, the natural convection in a square enclosure, partly heated from below, with two adiabatic vertical wall and one upper horigental wall is studied nomerically. In numerical study, SIMPLE(Semi-Implicit for Pressure Linked Equation) algorithems are applied for the integration of momentum and energy equation. The grid size used in this study is the coordinates of size (22$\times$22). As a result of numerical analysis, the initial fluid flow depends on the thermal diffusion, but, as time passes, the fluid flow depends on convection and buoyancy of the enclosure. In Case 1, the heating region was been in the central position of the bottom wall. In case 2, the heating region was in the left position of the bottom. In case of Case 1, the lapse time of sensing the temperature of 72$^{\circ}C$ is approximately 15 sec almost at the same time in the coordinates (6, 22), (11, 22). In case of Case 2, the lapse time in the coordinates (6, 22), (11, 22) was 27 sec, 25 sec repectively. Also in case of Case 1 or Case 2, the gradients of y-position of the two sensors are transposed each other.

Observation of Water Property Variations in the Western Channel of the Korea Strait During 2006-2010 (2006~2010년 기간 동안 대한해협 서수도에서 관측된 해수 물성의 변동)

  • Seasonal to inter-annual variations of water properties in the western channel of the Korea Strait are investigated using quasi-monthly hydrographic observations collected during 2006-2010. Weak vertical temperature and salinity gradients are observed during the winter months and these remain until May. At the upper layer, temperature increases from March and reaches a maximum in August, while salinity decreases during the same period. Near-bottom water shows low temperatures during late winter and fall with a minimum peak in September. Korea Strait Bottom Cold Water produces thick layers (>20 m) in 2006 and 2010, while it is observed very near the bottom with relatively high temperature in 2008 and 2009.

Seepage Characteristics of Embankment as with/without Gravel Layer under the Earth Fill (성토층 하부의 자갈층 유·무에 따른 침투특성 변화)

  • A series of laboratory tests was carried out fur analyzing of seepage characteristics of two-layers embankment model which consists of gravel and earth fill layers. Gravel layers were built under the earth fill for a half and one-third width of earth fill of the model. Permeability of earth fill was ranged between $5.00\times10^{-5}\~3.00\times10^{-4}\;m/s$.. The tests were performed with hydraulic gradients(i), $0.10\~0.55$. From the test results, hydraulic head of earth fill with gravel layer was 1.6 times higher than that of earth fill without gravel layer. Seepage rate was increased up to $4\~22$ times and safety factor for piping was decreased to $13\~43\;\%$ comparing the earth fill with gravel layer to that without gravel layer. The gravel layer under the earth fill could, in general, give some serious seepage problems to seadike embankment.

COLOR GRADIENT IN THE KING TYPE GLOBULAR CLUSTER NGC 7089

  • We use BV CCD images to investigate the reality of the color gradient within a King type globular cluster NGC 7089. Surface photometry shows that there is a strong radial color gradient in the central region of the cluster in the sense of bluer center with the amplitude of ~0.39 $\pm$ 0.07 mag/$arcsec^2$ in (B - V). In the outer region of the cluster, however, the radial color gradient shows a reverse case, i.e., redder toward the center. (B - V) color profile which was derived from resolved stars in VGC 7089 field also shows a significant color gradient in the central region of the clusters, indicating that lights from the combination of red giant stars and blue horizontal branch stars cause the radial color gradient. Color gradient of the outer region of NGC 7089 may be due to the unresolved background of the cluster. Similar color gradients in the central area of clusters have been previously observed exserved exclusively in highly concentrated systems classified as post core collapse clusters. We caution, however, to confirm the reality of the color gradient from resolved stars, we need more accurate imaging data of the cluster with exceptional seeing condition because the effect of completeness correlates with local density of stars.

  • We present an innovative method of multi-physics application involving energetic materials. We use an Eulerian methodology to address these problems. We have devised a new level set based tracking framework that can elegantly handle large gradients typically found in energetic response of high explosive and metals. Proper constitutive relations are employed to model the transient phases of gas, lliquid, and solid in the high strain rate regime. We use the confined and unconfined rate stick results to validate against the experimental data.

Adaptive mesh refinement/recovery strategy for FEA

  • This paper deals with the development of h-version adaptive mesh refinement and recovery strategy using variable-node elements and its application to various engineering field problems with 2D quadrilateral and 3D hexahedral models. The variable-node elements which have variable mid-side nodes on edges or faces are effectively used in overcoming some problems in connecting the different layer patterns of the transition zone between the refined and coarse mesh. A modified recovery technique of gradients adequate for variable-node elements and proper selection of error norms for each engineering field problems are proposed. In the region in which the error is greater than the permissible refinement error, the mesh is locally refined by subdivision. Reversely, in some parts of the domain having the error smaller than the permissible recovery error, the mesh is locally recovered (coarsened) by combination. Hierarchical structures (e.g. quadtrees and octrees) and element-based storage structures are composed to perform this adaptive process of refinement and recovery. Some numerical examples of a 3D heat conduction analysis of the concrete with hydration heat and a 2D flow analysis of vortex shedding show effectiveness and validity of the proposed scheme.

Relationship between Temperature Distributions and Outbreaks of Harmful Algal Blooms in Korean Waters

  • Harmful algal blooms (HABs) of Cochlodinium polykrikoides frequently occur around the South Sea of Korea, causing. economic losses in coastal breeding grounds. HAB outbreak scale usually changes each year depending on physical, biological and environmental conditions. Relatively large-scale HABs occurred in 1995, 1997, 1999, 2001, 2002 and 2003 with respect to spatial scale, duration and maximum density. Considering HAB scale and temperature distributions around the South Sea, we found that low coastal temperatures in August correspond to enormous HAB outbreaks. Cold waters created by coastal upwellings around the southeastern coast of Korea also corresponded to these outbreaks. Serial oceanographic investigations in August in the South Sea revealed that sea surface temperature anomalies had distinctively negative values when large-scale HAB outbreaks appeared. With regard to temperature differences between the surface and the 30-m layer, there was a tendency for large-scale outbreaks when temperature gradients around the seasonal thermocline weakened.

  • A Line Density algorithm was developed to quantify the sea surface temperature distribution using NOAA Sea Surface Temperature(SST) data and Geographic Information Systems(GIS), In addition, a GIS based automation model was designed to extract the Line Density Indices were determined by applying K-means Cluster. SST data in terms of March to May obtained on the coastal area of the Uljin from 2001 to 2004 in spring were used to make two data sets of average sea water temperature map in terms of year as well as month. From the result it was formed that water temperature gradient in April was the strongest among the other months, In particular very strog formation of oceanic front as well as temperature gradients were observed in front of the coastal area around Wonduk and Jukbyeon countries. Because those coastal area is a confront zone of two cold and a warm. It is expected that the development of a Line Density Algorithm would contribute to quantify of the SST for the research of Sea Surface Front(SSF) related to marine life management and the sea environmental conservation.
