Isolation and Characterization of the Smallest Bacteriophage P4 Derivatives Packaged into P4-Size Head in Bacteriophage P2-P4 System

  • Bacteriophage P4, a satellite phage of coliphage P2, is a very useful experimental tool for the study of viral capsid assembly and cos-cleavage. For an in vitro cos-cleavage reaction study of the P2-P4 system, new shortened and selectable markers containing P4 derivative plasm ids were designed as a substrate molecules. They were constructed by swapping the non-essential segment of P4 DNA for either the kanamycin resistance (kmr) gene or the ampicillin resistance (apr) gene. The size of the genomes of the resulting markers were 82% (P4 ash8 delRI:: kmr) and 79% (P4 ash8 delRI:: apr) of the wild type P4 genome. To determine the lower limit of genome size that could be packaged into the small P4-size bead, these shortened P4 plasmids were converted to phage particles with infection of the helper phage P2. The conversion of plasmid P4 derivatives to bacteriophage particles was verified by the heat stability test and the burst size determination experiment. CsCl buoyant equilibrium density gradient experiments confirmed not only the genome size of the viable phage form of shortened P4 derivatives, but also their packaging into the small P4-size head. P4 ash8 delRI:: apr turned out to be the smallest P4 genome that can be packaged into P4-sized head.

고속철도 차량의 제동성능 해석에 대한 실험적 검증 (Experimental verification of the brake performance analysis for the high speed train)

  • In general, the braking system of a high speed train has an important role for the safety of the train. To stop safely the train at its pre-decided position, it is necessary to properly combine the various brakes. The train has adopted a combined electrical and mechanical (friction) braking systems. In order to design a good brake system, it is essential for designers to predict the brake performance. In this paper, the braking performance analysis program has been developed and verified by comparing the simulation results with the brake test results of HSR-350x; both results match very well. Also, the brake performances of high speed trains can be predicted by using this program under various conditions.

CFD를 이용한 Wake Equalizing Duct의 최적설계 (Design Optimization of Wake Equalizing Duct Using CFD)

  • In this paper, wake equalizing duct (WED) form optimization was carried out using computational fluid dynamics (CFD) techniques. A WED is a ring-shaped flow vane with a foil-type cross-section fitted to a hull in front of the upper propeller area. The main advantage of a WED is the power savings resulting from the uniformity of the velocity distribution on the propeller plane, a reduction in the flow separation at the aft-body, and lift generation with a forward force component on the foil section. This paper intends to evaluate these functions and find an optimized WED form for minimizing the viscous resistance and equalizing the wake distribution. In the optimization process, the study uses four WED parameters: the angle of the section, longitudinal location, and angles of the axes for the half rings against the longitudinal and transverse planes of the ship. KRISO 300K VLCC2 (KVLCC2) is chosen as an example ship to demonstrate the WED optimization. The optimization procedure uses genetic algorithms (GAs), a gradient-based optimizer for the refinement of the solution, and Non-dominated Sorting GA-II(NSGA-II) for Multiobjective Optimization. The results show that the optimized WED can reduce the viscous resistance at the expense of the uniformity of the wake distribution.

경상남도 상북면 양산단층 서부지역에 대한 습윤지수 특성 연구 (Characterization of Wetness Index in Western Area of Yangsan Fault, Sangbuk-myeon, Kyeongnam-do)

  • The study area adjoins with Yangsan fault in Sangbuk-myeon, Samsam-ri, Kyongsang-namdo and consist of the natural steep slope. After drawing data layer which have altitude by using digital topography data, it is converted to lattice DEM of $10m{\times}10m$ size. From this, gradient map of unit lattice, slant direction map and shadow relif map are made. Using flow apportioning algorithm, upper slope contributing area and wetness index by established lattice can be calculated. Area that have high wetness index shows lineament structure of northwest-southeast direction, and this agrees with shear fracture system. The result of electricity specific resistance survey in the study area shows that area of high wetness index has low electricity specific resistance anomaly. That is, wetness index conforms with distribution of fractured zone that accompanied chemical weathering of rock. Therefore, wetness index can be used as the method of detecting fractured zones and judging the stability of the area.

Molecular Dynamic Simulation for Penetration of Carbon Nanotubes into an Array of Carbon Nnantotubes

  • When two layers of carbon nanotube (CNT) arrays are loaded to mate, the free ends of individual CNTs come into contact at the interface of the two layers. This leads to a higher contact resistance due to a smaller contact region. However, when the free CNT ends of one array penetrate into the mating array, the contact region increases, effectively lowering the contact resistance. To explore the penetration of mating CNTs, we perform molecular dynamic simulations of a simple unit cell model, incorporating four CNTs in the lower array layer coupled with a single moving CNT on the upper layer. The interaction with neighboring CNTs is modelled by long-range carbon bond order potential (LCBOP I). The model structure is optimized by energy minimization through the conjugate gradient method. A NVT ensemble is used for maintain a room temperature during simulation. The time integration is performed through the velocity-Verlet algorithm. A significant vibrational motion of CNTs is captured when penetration is not available, resulting in a specific vibration mode with a high frequency. Due to this vibrational behavior, the random behaviors of CNT motion for predicting the penetration are confirmed under the specific gap distances between CNTs. Thus, the probability of penetration is examined according to the gap distance between CNTs in the lower array and the aspect ratio of CNTs. The penetration is significantly affected by the vibration mode due to the van der Waals forces between CNTs.

나선코일 튜브 내에서의 압력손실에 관한 연구 (A Study on the Pressure Loss in Helically Coiled Tubes)

  • The resistance coefficient and heat transfer performance are studied for the turbulent water flow in a smooth coiled tube having variable curvature ratios and a corrugated-coiled tube having a ratio of coil to tube diameter of 22. Experiments are carried out for the fully developed turbulent flow of water in tube coils on the uniform wall temperature condition. This work is limited to tube coils of R/a between 22 and 60 and Reynolds numbers from 13000 to 53000. The tube having a ratio of coil to tube diameter of 27 among the 3 smooth tube coils shows the best heat transfer performance. A corrugated-coiled tube(R/a=60) shows more excellent performance than a smooth coiled tub (R/a=60) at a similar curvature ratio. The friction factor f is sensitive to changes in the velocity profile caused by a temperature gradient. Allowance was made for the pressure loss in the short inlet and outlet lengths and due to the presence of the thermocouple inlet and outlet as a result of separate experimental on a straight tube. It is to be expected that the allowance at the exit will be somewhat too low because of secondary flow effects carried over from the coil.

11층 열장벽 피막의 고온물성에 관한 연구 (A study on the thermal properties of the 11 layer thermal barrier)

  • The purpose of this study is to evaluate the properties of the functional gradient thermal barrier coatings by plasma spray process. The evaluations of mechanical and thermal properties such as fatigue, oxidation and wear-resistance at high temperatures have been conducted. Furthermore, residual stress and bond strength have been evaluated. The range of thickness of coated layers was 550~600$\mu\textrm{m}$. The range of hardness of layers was 800~900 Hv and the porosity range of coatings was about 7 to 14%. The top coating layer of $ZrO_2$ in thermal barrier was composed of tetragonal structure after spraying. The coated layers of $ZrO_2$ on the Inconel substrate is the best resistance for thermal fatigue. Those coatings had the least compressive stress in comparison with other coatings. In high temperature oxidation test, the coatings on Inconel substrate was better than the coatings on SUS substrate. The bond strength of the concave type was greater than that of linear types and convex types coatings.

플립칩 Sn-3.5Ag 솔더범프의 Electromigration과 Thermomigration 특성 (Electromigration and Thermomigration Characteristics in Flip Chip Sn-3.5Ag Solder Bump)

  • Electromigration test of flip chip solder bump is performed at $140^{\circ}C$ C and $4.6{\times}10^4A/cm^2$ conditions in order to compare electromigration with thermomigration behaviors by using electroplated Sn-3.5Ag solder bump with Cu under-bump-metallurgy. As a result of measuring resistance with stressing time, failure mechanism of solder bump was evaluated to have four steps by the fail time. Discrete steps of resistance change during electromigration test are directly compared with microstructural evolution of cross-sectioned solder bump at each step. Thermal gradient in solder bump is very high and the contribution of thermomigration to atomic flux is comparable with pure electromigration effect.

슬래브 하부 수평저항을 고려한 지반위의 콘크리트 슬래브 해석 모델 및 온도하중에 의한 거동 분석 (Analysis Models of Concrete Slabs-on-Grade Considering Horizontal Resistance at Slab Bottom and Behavior under Thermal Loads)

  • 지반위에 놓인 콘크리트 슬래브가 온도하중을 받을 때 지반의 전단저항과 슬래브 하부와 지반과의 마찰 등에 의해 생기는 슬래브 하부의 수평저항을 고려하여 지반위에 놓인 콘크리트 슬래브의 거동을 분석하였다. 지반위의 콘크리트 슬래브와 강성도로포장의 해석에 널리 사용되는 탄성지반위의 얇은 판을 이용하여 슬래브 하부의 수평저항을 고려하기 위한 해석 공식을 유도하였다. 이를 이용하여 판요소와 쉘요소를 이용한 유한요소법에 의한 모델을 개발하여 수치해석 결과를 도출하였다. 해석 공식과 수치해석 모델을 이용한 해석 결과를 비교 분석하였고 매우 비슷한 결과가 도출 되는 것을 알 수 있었다. 슬래브의 상부와 하부에 온도 차이가 있을 때와 슬래브의 온도가 전체적으로 감소할 때, 콘크리트 슬래브의 응력 분포에 슬래브 하부의 수평저항이 미치는 민감성을 여러 가지의 다른 슬래브의 두께, 탄성계수, 그리고 지반의 수직탄성계수 등을 고려하여 분석하였다. 해석 결과에서 온도하중을 받을 때 슬래브 하부의 수평저항은 슬래브의 응력에 매우 큰 영향을 미칠 수 있다는 것을 발견하였다.

박테리오파아지 P2-P4 시스템을 위한 tetracyclin resistance marker 함유 P4 유도체 벡터 플라스미드 조성 (Construction of New P4-Derived Vector Plasmid Containing Tetracyclin Resistance Marker for the Bacteriophage P2-P4 System)

  • 바이러스 조립 과정 기작 연구를 위한 좋은 재료인 박데리오파아지 P2-P4시스템에 이용될 벡터 플라스미드를 개발하기 위하여, P4 ash8 sid71을 출발 물질로 삼아 새로운 P4 유도체 벡터를 조성하였다. 유전자 재조합 기법을 써서 쉽게 선택 가능한 tetracyclin내성 유전자(tetR)를 도입하고 플라스미드P 4의 크기를 조절하였다. 이를 통해 얻어진 P4 ash8(sid7l) tetR은 12.09 kb의 크기를 가지며, 필요할 때 P2로 induction하면 생물학적 활성을 가지는 박데리오파아지로 전환 가능하였다. 전환된 파아지의 burst size를 결정하고, CsCi 부양균등밀도 편차실험을 수행하였다. 균등밀도 실험 분포도에서 P2크기 파아지 머리의 packaging 상한을 추정할 수 있었다.
