다환 방향족 탄화수소(PAH) 분해 미생물 탐색을 위한 승화법의 개발 (Application of a Modified Sublimation Method to Screen for PAH-Degrading Microorganisms)

  • 다환 방향족 탄화수소(PAH; polycyclic aromatic hydrocarbon)는 발암성, 돌연변이 유발성, 유전독성을 지니기 때문에 인체위해성이 큰 물질로 알려져 있다. 기존 PAH 분해 미생물 탐색 방법중 독성이 강한 유기용매에 PAH를 용해시켜 미생물에 직접 분무하는 분무법, 미생물과 직접 혼합하는 한천중층법은 미생물 생장에 영향을 줄 뿐만 아니라, 특히 많이 쓰이는 분무법의 경우 분무되는 PAH의 양을 조절하기가 어렵고 멸균상태를 유지하기가 힘들다는 단점이 있다. 그래서 본 논문에서는 이와 같은 단점을 보완한 방법으로 승화법(Alley, Jeremy F. and Lewis R. Brown. 2000. Use of sublimation to prepare solid microbial media with water-insoluble substrates. Appl. Environ. Microbiol. 66, 439-442)을 도입하여 적용하였다. 그 결과 상용휘발유 및 태안유류유출지 시료로부터 분리한 350분리균주 중에서 7균주가 단일 PAH 또는 복수의 PAH 분해에 관여했다. 특히 Corynebacterium sp. SK20, Rhodococcus sp. TA24, Streptomyces sp. TA27은 시험한 pyene, phenanthrene, naphthalene에, Gordonia sp. H37는 pyrene, naphthalene에, Arthrobacter sp. S49는, naphthalene, phenanthrene에 활성이 있었다.

근권세균과 옥수수를 이용한 유류 및 중금속 복합 오염토양의 Rhizoremediation (Rhizoremediation of Petroleum and Heavy Metal-Contaminated Soil using Rhizobacteria and Zea mays)

  • 본 연구에서는 유류 분해능이 있고 식물 성장 촉진능력을 가진 Gordonia sp. S2RP-17, 중금속에 내성을 가지며 식물 성장촉진 능력이 있는 Serratia sp. SY5 및 옥수수를 이용하여 유류 및 중금속 오염 토양의 정화 특성을 조사하였다. 유류 및 중금속 오염 토양에서 51일간 재배한 옥수수의 평균 뿌리 건중량은 $1.9{\pm}0.2\;g$이었으나, 근권세균을 접종한 오염 토양에서 재배한 옥수수의 뿌리 건중량은 $5.6{\pm}0.7\;g$로, 근권세균 접종에 의해 옥수수의 뿌리의 중량이 유의적으로 증가함을 알 수 있었다(p<0.01). 초기에는 토양의 TPH 농도는 $21,576{\pm}3,426\;mg-TPH{\cdot}kg-dry\;soil^{-1}$이었는데, 51일 후 옥수수만을 식재한 토양의 잔류 TPH 농도는 $220{\pm}98\;mg-TPH{\cdot}kg-dry\;soil^{-1}$이었고, 옥수수와 함께 근권세균을 접종한 토양의 잔류 TPH 농도는 $20{\pm}41\;mg-TPH{\cdot}kg-dry\;soil^{-1}$이었다. 이러한 결과로부터 옥수수 식재에 의해 대부분의 TPH를 제거할 수 있으며, 옥수수와 함께 근권세균을 접종하면 TPH 효율이 조금 더 향상됨을 알 수 있었다. 그러나, 중금속 제거효율에 미치는 근권세균 접종 효과는 거의 없었다.

Twenty-five unrecorded bacterial species of the Republic of Korea belonging to the phylum Actinomycetota discovered during surveys in 2021

  • We isolated and identified 25 unrecorded bacterial species belonging to the phylum Actinomycetota found in the Republic of Korea. Sequence comparison of 16S rRNA was performed using the NCBI BLAST and EzBioCloud database to identify 25 species, which had a 16S rRNA gene sequence similarity of >98.8% and were allocated as unrecorded species in the Republic of Korea. Among the 25 unrecorded bacterial strains, Streptomyces was the most common with nine species, followed by Leifsonia with two species. Isoptericola, Nocardioides, Dermacoccus, Sinomonas, Patulibacter, Marmoricola, Allobranchiibius, Aldersonia, Actinokineospora, Agromyces, Aeromicrobium, Cellulomonas, and Gordonia with one species each were also found. Twenty-five unrecorded species were excavated in various environments, such as tidal flats, ferns, soil, pine cones, moss, mud, wetlands, and plants. These isolates were characterized on the basis of their phylogenetic, biochemical properties, and morphological data, and species descriptions were provided.

  • Fourteen bacterial strains, low10-4-1, J11015, 17J27-22, 17G22-9, 17G9-4, 17Bio_15, 17gy_33, 17SD1_21, Strain8, 17Sr1_17, J21014T, H31021, 17J49-9, and 17J80-6 assigned to the phylum Actinobacteria, Bacteroidetes, Deinococcus-Thermus, and Firmicutes were isolated from soil samples. Phylogenetic analysis based on 16S rRNA gene sequence revealed that strains low10-4-1, J11015, 17J27-22, 17G22-9, 17G9-4, 17Bio_15, 17gy_33, 17SD1_21, Strain8, 17Sr1_17, J21014T, H31021, 17J49-9, and 17J80-6 were most closely related to Marmoricola aurantiacus (98.9%), Calidifontibacter indicus (99.8%), Gordonia soli (98.8%), Rhodococcus globerulus (99.5%), Pseudarthrobacter siccitolerans (99.1%), Hymenobacter qilianensis (98.7%), Hymenobacter terrae (99.0%), Deinococcus yunweiensis (99.2%), Deinococcus proteolyticus (99.7%), Domibacillus indicus (99.2%), Exiguobacterium mexicanum (100.0%), Kurthia senegalensis (99.1%), Lysinibacillus composti (99.6%), and Bacillus loiseleuriae (99.3%). These fourteen species have never been reported in Korea, therefore we report them here for the first time.

Bioremediation Potential of a Tropical Soil Contaminated with a Mixture of Crude Oil and Production Water

  • A typical tropical soil from the northeast of Brazil, where an important terrestrial oil field is located, was accidentally contaminated with a mixture of oil and saline production water. To study the bioremediation potential in this area, molecular methods based on PCR-DGGE were used to determine the diversity of the bacterial communities in bulk and in contaminated soils. Bacterial fingerprints revealed that the bacterial communities were affected by the presence of the mixture of oil and production water, and different profiles were observed when the contaminated soils were compared with the control. Halotolerant strains capable of degrading crude oil were also isolated from enrichment cultures obtained from the contaminated soil samples. Twenty-two strains showing these features were characterized genetically by amplified ribosomal DNA restriction analysis (ARDRA) and phenotypically by their colonial morphology and tolerance to high NaCl concentrations. Fifteen ARDRA groups were formed. Selected strains were analyzed by 16S rDNA sequencing, and Actinobacteria was identified as the main group found. Strains were also tested for their growth capability in the presence of different oil derivatives (hexane, dodecane, hexadecane, diesel, gasoline, toluene, naphthalene, o-xylene, and p-xylene) and different degradation profiles were observed. PCR products were obtained from 12 of the 15 ARDRA representatives when they were screened for the presence of the alkane hydroxylase gene (alkB). Members of the genera Rhodococcus and Gordonia were identified as predominant in the soil studied. These genera are usually implicated in oil degradation processes and, as such, the potential for bioremediation in this area can be considered as feasible.

A report of 42 unrecorded bacterial species isolated from fish intestines and clams in freshwater environments

  • Nine fish and one clam species were collected from freshwater environments in Korea, including four lakes, two streams, and the Nakdong River, to investigate the host-associated bacteria. Hundreds of bacterial strains were isolated from the samples using a cell sorter and a dilution plating method. After identification of the bacterial strains using 16S rRNA gene sequences, 42 strains with greater than 98.7% sequence similarity with validly published species were determined to be unrecorded bacterial species in Korea. These strains were phylogenetically diverse and assigned to four phyla, six classes, 17 orders, 27 families, and 32 genera. At the genus level, the unrecorded species were classified as Corynebacterium, Mycobacterium, Mycolicibacterium, Gordonia, Williamsia, Modestobacter, Brachybacterium, Sanquibacter, Arthrobacter, and Mycolicibacterium of the class Actinobacteria; Empedobacter, and Flavobacterium of the class Flavobacteriia; Fictibacillus, Psychrobacillus, Cohnella, Paenibacillus, Rummeliibacillus, Enterococcus, and Vagococcus of the class Bacilli; Aquamicrobium, Paracoccus, and Sphingomonas of the class Alphaproteobacteria; Achromobacter, Delftia, and Deefgea of the class Betaproteobacteria; and Aeromonas, Providencia, Yersinia, Marinomonas, Acinetobacter, and Pseudomonas of the class Gammaproteobacteria. The 42 unrecorded species were subjected to further taxonomic characterization using gram staining, cellular and colony morphological determination, biochemical analyses, and phylogenetic analyses. This paper provides detailed descriptions of the 42 previously unrecorded bacterial species.

Polyphasic Analysis of the Bacterial Community in the Rhizosphere and Roots of Cyperus rotundus L. Grown in a Petroleum-Contaminated Soil

  • Cyperus rotundus L. is a perennial herb that was found to be dominating an area in northeast Brazil previously contaminated with petroleum. In order to increase our knowledge of microorganism-plant interactions in phytoremediation, the bacterial community present in the rhizosphere and roots of C. rotundus was evaluated by culture-dependent and molecular approaches. PCR-DGGE analysis based on the 16S rRNA gene showed that the bacterial community in bulk soil, rhizosphere, and root samples had a high degree of similarity. A complex population of alkane-utilizing bacteria and a variable nitrogen-fixing population were observed via PCR-DGGE analysis of alkB and nifH genes, respectively. In addition, two clone libraries were generated from alkB fragments obtained by PCR of bulk and rhizosphere soil DNA samples. Statistical analyses of these libraries showed that the compositions of their respective populations were different in terms of alkB gene sequences. Using culturedependent techniques, 209 bacterial strains were isolated from the rhizosphere and rhizoplane/roots of C. rotundus. Dot-blot analysis showed that 17 strains contained both alkB and nifH gene sequences. Partial 16S rRNA gene sequencing revealed that these strains are affiliated with the genera Bosea, Cupriavidus, Enterobacter, Gordonia, Mycoplana, Pandoraea, Pseudomonas, Rhizobium, and Rhodococcus. These isolates can be considered to have great potential for the phytoremediation of soil with C. rotundus in this tropical soil area.

제주 연안의 괭생이모자반(Sargassum horneri)에서 분리된 세균의 계통학적 다양성 및 군집 구조 분석 (Phylogenetic Diversity and Community Structure of Microbiome Isolated from Sargassum Horneri off the Jeju Island Coast)

  • 최근 괭생이모자반(Sargassum horneri)은 제주의 해안가와 해변, 연근해 양식장 등 매년 대량으로 속출되어 인근 양식업자 및 주민에게 막대한 피해를 주고 있는 추세이다. 본 연구에서는 제주 인근 연안으로 흘러 들어온 괭생이 모자반에 서식하고 있는 미생물의 다양성을 탐색하고 동정을 통한 미생물의 기능을 파악함으로써 생태학적 문제에 관한 기초자료를 제공하고자 하였다. Proteobacteria는 분리 된 균주 중 88%를 차지한 우점문으로 ${\alpha}-proteobacteria$강은 6속 10종으로 Pseudorhodobacter속이 40%를 차지하고 Paracoccus속 20%, Rhizobium, Albirhodobacter, Skermanella 및 Novosphingobium 속은 각각 10%를 차지했다. ${\beta}-proteobactera$강은 5속 10종으로 Hydrogenophaga속이 50%, Azoarcus 20%, 나머지 Oxalicibacterium, Duganella, Xenophilus속은 각각 10%를 차지했다. ${\gamma}-proteobacteria$강은 13속 57종으로 Proteobacteria문에서 74%로 우점강을 차지했고, Shewanella는 23%, Pseudomonas속 12%, Cobetia 19%, Rahnella, Vibrio 및 Serratia속은 4%, 나머지 Rheinheimera, Raoultella, Pantoea, Acinetobacter, Moraxella 및 Psychrobacter속은 2%를 차지했다. Actinobacteria는 1속 2종으로 Gordonia와 Nocardioides속이 각각 50%를 나타냈다. Bacteroidetes문은 3속 5종으로 Lacihabitans, Mariniflexile속은 33%, 나머지 Dyadobacter, Cellulophaga와 Ferruginibacter속은 11%를 차지했다.
