Effects of Cheongjogupye-tang(淸燥救肺湯) and Yieum-jeon(理陰煎) on Secretion of Mucin from Respiratory Epithelial Cells (청조구폐탕(淸燥救肺湯)과 이음전(理陰煎)이 호흡기 접액분비에 미치는 영향)

  • Objectives : In this study, the author tried to examine whether Cheogjogupye-tang (淸燥救肺湯, CGPT) and Yieum-jeon (理陰煎, YEJ) significantly affect in vitro and in vivo mucin secretion, MUC5AC gene expression in airway epithelial cells and contractility of isolated tracheal smooth muscle of rabbit. Materials and Methods : For in vitro experiment, confluent hamster tracheal surface epithelial (HTSE) cells were chased for 30 minutes in the presence of CGPT and YEJ to assess the effects of the agents on mucin secretion by enzyme-linked immunosorbent assay (ELISA), with removal of oriental herbal medicine extract from each agent-treated sample by centrifuge microfilter. Also, the effects of the agents on TNF-alpha or EGF-induced MUC5AC gene expression in human airway epithelial cells (NCI-H292) were investigated. Possible cytotoxicities of the agent were assessed by examining both LDH release from HTSE cells and the rate of survival and proliferation of NCI-H292 cells. For in vivo experiment, hypersecretion of airway mucin and goblet cell hyperplasia was induced by exposure of rats to $SO_2$ over 3 weeks. Effects of CGPT and YEJ orally administered for 1 week on in vivo mucin secretion from tracheal goblet cells of rats and hyperplasia of goblet cells were assessed using ELISA and histological analysis after staining the epithelial tissue with alcian blue, respectively. Also, the effects of CGPT and YEJ on contractility of isolated tracheal smooth muscle were investigated. Results : (1) CGPT significantly inhibited in vitro mucin secretion from cultured HTSE cells. However, YEJ did not affect in vitro mucin secretion; (2) CGPT and YEJ did not affect hypersecretion of in vivo mucin and hyperplasia of tracheal goblet cells; (3) CGPT and YEJ slightly increased the expression levels of TNF-alpha or EGF-induced MUC5AC gene in NCI-H292 cells; (4) CGPT and YEJ inhibited acetylcholine-induced contraction of isolated tracheal smooth muscle of rabbit; (5) CGPT and YEJ did not affect LDH release from HTSE cells and the survival and proliferation of NCI-H292 cells. Conclusion : The results from the present study suggest that CGPT and YEJ mainly affect the expression of mucin gene rather than secretion of mucin and do not show remarkable cytotoxicity to respiratory epithelial cells.

A Study on the Defence Effect of Banhasasim-tang for White Rat's Acute Duodenal Injury (흰쥐의 급성 십이지장 손상에 대한 반하사심탕의 방어효과에 관한 연구)

  • Objectives : Banhasasim-tang has been clinically used to treat upper gastric intestinal discomfort. The object of this study is to examine the defense effect of Banhasasim-tang for acute duodenal injury of the mouse. Methods and Materials : Twenty-one rats were divided into 3 groups and treated as follows: the control group was untreated mice. The ADE group was acute duodenal-damage-elicited mice. The BST group was Banhasasim-tang treated mice before acute duodenal damage elicitation. The groups were examined with common morphology, paneth cells in intestinal crypt, absorptive cells and goblet cells in epithelium, cell division in mucose, COX-l as mucosal protector, COX-2 (which appears to play an important role in inflammation), IL-2R-inducing cellular immuno-chainreaction, and the distribution of apoptotic cells. Results : 1. Common morphology: the ADE group was observed with duodenal injury - loss of villi, infiltration of cells concerned to inflammation (lymphocytes, granular leukocytes) to submucosal layer - by hemorrhagic erosions, while the BST group was seen the same as normal in proportion to increasing treatment time before injury. 2. Histochemical change: the ADE group was observed with noticeable decreased distribution of absorptive cells with microvilli, acid mucin secreted goblet cell, neutral mucin secreted goblet cell, paneth cells compared to the normal group. The BST group was seen to have distribution of epithelium cells resembling normal in proportion to increasing treatment time before injury. 3. Imnunohistochemical change: the ADE group showed a change of factors leading to duodenal injury as reduce of cytokinesis, COX-1, increase of COX-2, IL-2R-. In contrast, the BST group tended to reduction of cytokinesis, COX-1, increase of COX-2, IL-2R- in proportion to increasing taking time before injury. 4. Apoptosis change: the ADE group showed increasing apoptosis cells, in contrast to the BST group which was the same as normal in proportion to increasing treatment time before injury. Conclusions : According to the above results, by increasing the defense system of mucosal epithelium, Banhasasim-tang is thought to effectively protect tissue against ulcers resulting from acute duodenal injury.

  • The present study was performed to clarify the histochemical compositions and fine structure of the mucus secreting cells in the gastrointestinal mucosa of normal mice. The mucus cells in the surface epithelium of stomach body had neutral mucin and some quantity of weak acid mucin. And the mucus cells in gastric pits and mucus neck cells had neutral mucin. The goblet cells in villial epithelium of small intestine contain strong acid sulfated mucin as their main content and a little of neutral mucopolysaccharide. However, the goblet cells in intestinal glands-Liberkuhn crypt were confirmed to contain non-sulfated weak acid mucin. The goblet cells in the surface epithelium of colon had the same component as the small intestine did. But the cells in the crypts of colon contained neutral and weak acid mucin as their main contents. The majority of secretory granules of the surface epithelial cells of the stomach body had high electron density, and some granules with low electron density appeared too. While the mucin granules in the mucus neck cells were low in its electron density, and some of those granules were frequently found to have dense core in them. Secretory granules in goblet cells of small and large intestines had low electron density. The mode of secretion in mucin-containing cells in gastro-intestinal tract was found to be merocrine.

Effect of Piryongbanggamgil-tang on Airway Mucin Secretion, Production, Gene Expression and Hypersecretion of Mucus (필용방감길탕이 기도 뮤신의 분비, 생성, 유전자 발현 및 점액 과다 분비에 미치는 영향)

  • Objectives In this study, the author tried to investigate whether piryongbang-gamgil-tang (PGGT) significantly affect in vitro airway mucin secretion, PMA- or EGF- or TNF-${\alpha}$-induced MUC5AC mucin production / gene expression from human airway epithelial cells and increase in airway epithelial mucosubstances and hyperplasia of tracheal goblet cells of rats. Materials and Methods For in vitro experiment, confluent RTSE cells were chased for 30 min in the presence of PGGT to assess the effect of PGGT on mucin secretion by enzyme-linked immunosorbent assay (ELISA). Also, effect of PGGT on PMA- or EGFor TNF-${\alpha}$-induced MUC5AC mucin production and gene expression from human airway epithelial cells (NCI-H292) were investigated. Confluent NCI-H292 cells were pretreated for 30 min in the presence of PGGT and treated with PMA (10 ng/ml) or EGF (25 ng/ml) or TNF-${\alpha}$ (0.2 nM) for 24 hrs, to assess both effect of PGGT on PMA- or EGF- or TNF-${\alpha}$-induced MUC5AC mucin production by ELISA and gene expression by reverse transcription-polymerase chain reaction (RT-PCR). For in vivo experiment, the author induced hypersecretion of airway mucus and goblet cell hyperplasia by exposure of rats to $SO_2$ during 3 weeks. Effect of orally-administered PGGT during 2 weeks on increase in airway epithelial mucosubstances from tracheal goblet cells of rats and hyperplasia of goblet cells were assesed by using histopathological analysis after staining the epithelial tissue with alcian blue. Possible cytotoxicities of PGGT in vitro were assessed by examining LDH release from RTSE cells and the rate of survival and proliferation of NCI-H292 cells. In vivo liver and kidney toxicities of PGGT were evaluated by measuring serum GOT/GPT activities and serum BUN/creatinine concentrations of rats after administering PGGT orally. Results (1) PGGT did not affect in vitro mucin secretion from cultured RTSE cells. (2) PGGT significantly inhibited PMA-, EGF-, and TNF-${\alpha}$-induced MUC5AC mucin productions and the expression levels of MUC5AC mRNA from NCI-H292 cells. (3) PGGT decreased the amount of intraepithelial mucosubstances and showed the tendency of expectorating airway mucus already produced. (4) PGGT increased LDH release from RTSE cells. However, PGGT did not show in vivo liver and kidney toxicities and cytotoxicity to NCI-H292 cells. Conclusion The result from this study suggests that PGGT can regulate the production and gene expression of airway mucin observed in diverse respiratory diseases accompanied by mucus hypersecretion and do not show in vivo toxicity to liver and kidney functions after oral administration. Effect of PGGT with their components should be further studied using animal experimental models that reflect the diverse pathophysiology of respiratory diseases through future investigations.

Possible cytotoxicities of MA in vitro were assessed by examining LDH release from RTSE cells and the rate of survival and proliferation of NCI-H292 cells. In vivo liver and kidney toxicities of MA were evaluated by measuring serum GOT/GPT activities and serum BUN/creatinine concentrations of rats after administering MA orally. Results : 1. MA decreased the amount of intraepithelial mucosubstances of rats exposed to sulfur dioxide inhalationally. 2. MA decreased in vitro mucin secretion from cultured RTSE cells. 3. MA significantly inhibited PMA-, EGF-, and TNF-alpha-induced MUC5AC mucin productions and the expression levels of MUC5AC mRNA from NCI-H292 cells. 4. MA did not show either in vitro or in vivo hepatic or renal toxicities. Conclusion : The results from this study suggests that MA can regulate the secretion, production and gene expression of airway mucin observed in diverse respiratory diseases accompanied by mucus hypersecretion and does not show in vivo toxicity to liver and kidney functions after oral administration. Effects of MA should be further studied using animal experimental models that simulate the diverse pathophysiology of respiratory diseases via future research.

Histopathological changes of small intestine of cats infected with helminth (연충감염 고양이 소장의 병리조직학적 변화)

  • Helminth and their ova or protozoan oocysts from fecal samples of cats in Chonju and Daejon were examined from December 1995 to January 1996. Out of the 43 samples, 41(95.35%) were positive and 66(153.49%) were cumulative positive for parasite infection. The identified parasites were summerized as follows; Worms of Toxocara cati(62.79%), Taenia taeniformis(48.88%), Spirometra sp(23.25%) and Diphyllobothrium latum (6.97%), eggs of Clonorchis sinensis(2.32%), Ancylostoma tubaeformae(2.32%) and Capillaria sp(4.65%), oocyst of Isospora (2.32%) sp. In haevy infected intestine, mast cells(38 cells/10 villi) were detected in villi and goblet cells(131 cells/$0.1mm^2$) were severely increased in mucosa. Epithelium of intestine was partially deteched B. The end of villi were necrosed and hyperaemia was widely recognized in lamina propria. T taeniaeformis have not the neck, but have 2 layed hooks(anterior : 19, posterior : 19, total : 38).

The Morphological Study of the Midgut Epithelium during the Metamorphosis of Pine Caterpillar (變態에 따른 솔나방(Dendrolimus spectabilis Butler) 中腸上皮의 形態的 考察)

  • The morphological changes of the midgut epithelium during the metamorphosis of pine caterpillar are observed with light and electron microscope, being divided into 5 stages from the 8th instar larva to just after pupation. The midgut epithelium of 8th instar larva is composed of columnar cell, goblet cell, regenerative cell, and endocrine cell. The secretorials are arranged on the nuclear membrane in the columnar cell of the midgut epithelium in the 8th instar larva, and lysosomes are augmented in the apical portion. Cytoplasmic extrusions are observed in the apical surface of columnar cell but they have no cell organells. Nucleus, mitochondria, rER, Golgi complex, and free ribsomes are observed in the regenerative cell. Regenerative cells are differentiated into the form of goblet cell, and vacuoles are gradually increased in the cytoplasm. Just pupa stage, the materials, which appears to be mainly composed of Ca, are observed in the circular form and goblet cavity of regenerative cell are detached to lumen. As a result, it reflects the process of the degeneration of the midgut epithelium that lysosomes are gradually augmented in the columnar cell, that nuclear materials are removed to cytoplasm, and that cytoplasmic extrusions are observed in the apical surface. And though regenerative cells are differentiated into the form of the goblet cell, it is believed that goblet cavity is detached from regenerative cell to the lumen and midgut epithelium of pupa stage is formed.

Secretory Differentiation of Hamster Tracheal Epithelial Cells Increases Activation of Matrix Metalloproteinase-2

  • In chronic airway inflammatory diseases such as asthma and chronic bronchitis, it has been suggested that matrix metalloproteinases secreted from infiltrating neutrophil contribute the pathogenesis of the disease and have been a focus of intense investigation. We report here that hamster tracheal surface epithelial goblet cells (HTSE cells) produce matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2). Matrix metalloproteinase activities were investigated using [$^3H$]collagen-digestion assay and gelatin zymography. The subtype of matrix metalloproteinases expressed from HTSE cells was MMP-2 (gelatinase A), which was determined by Western blot with various subtype selective anti-matrix metalloproteinase antibodies. The MMP-2 and TIMP-2 cDNAs from HTSE cells were partially cloned by RT-PCR and they reveal more than 90% of sequence homology with those from human, rat and mouse. The collagenolytic activity was increased with the secretory differentiation of the HTSE cell and it was found that zymogen activation was responsible for the increased MMP-2 activity in HTSE cells. The results from the present study suggest that the metaplastic secretory differentiation of airway goblet cells may affect chronic airway inflammatory process by augmenting the zymogen activation of MMP-2.

Effects of Inhalation Anesthetics on Mucus Secretion of Goblet Cells at Lower Respiratory Tract in Rabbits (흡입마취제가 토끼의 하부호흡기도 점액분비세포에 미치는 영향)

  • Excess secretion of goblet cell stimulated by inhalation anesthetics have side effects during operation or postoperative care. Mucosubstances, which are almost secreted by goblet cells in the epithelium of the respiratory tract, are secreted by a direct irritation of inhalation anesthetics. This study was carried out to compare the differences of mucus secretion on lower respiratory tract stimulated by ethyl ether, halothane and isoflurane. Total of 24 rabbits were used as experimental animals. The trachea and the 1st bronchi were fixed in 10% neutral buffered formalin. After embedding in paraffin, the specimens were sectioned to a thickness of 6 ${\mu}{\textrm}{m}$, and PAS-H, Alcian blue pH 2.5 and Alcian blue pH 1.0 stains were performed for the observation of the composition and the quantity of the mucus. The results were as follows; Ethyl ether and isoflurane irritated the mucous membrane of the respiratory tract. Ethyl ether irritated more than isoflurane. Halothane irritated the mucous membrane, but its effect was minimal and had little influences during operation. In the specimens stained with PAS-H, Alcian blue pH 2.5 and Alcian blue pH 1.0, the mucosubstance lining the cilia and in the goblet cells of the trachea and 1st bronchi were the strongly PAS-H reactive mucosubstances, moderately Alcian blue pH 2.5 and Alcian blue pH 1.0. The PAS-H reactive mucosubstance were polysaccharides, neutral mucopolysaccharides, mucoproteins, glycoproteins and glycolipids. Trachea was easily affected than bronchi by inhalation anesthetics. Consequently, it is suggested that because halothane does not irritates respiratory mucosal secretion, its application may be efficient to the depressed respiratory system.

Effects of Several Oriental Medicines on Protein Kinase C Activator-Induced Production and Gene Expression of Airway Mucin and Animal Model for Airway Mucus Hypersecretion (단백질인산화효소 C 활성화제로 유도된 기도 뮤신 생성 및 유전자 발현과 점액 과분비 모델동물에 대한 수종(數種) 방제의 영향)

  • The author investigated whether Chwiyeon-tang(PC), Haengso-tang(PH), Jawanchihyo-san(PJ) and Gamisocheongryong-tang(PS) significantly affect both PMA-induced mucin production and MUC5AC gene expression in airway epithelial cells and sulfur-dioxide-induced airway goblet cell hyperplasia and mucus hypersecretion animal model using rat. Possible cytotoxicity of each herbal medicine was assessed by measuring the survival and proliferation rate of NCI-H292 cells. Confluent NCI-H292 cells were pretreated for 30 min in the presence of PC, PH, PJ and PS, respectively, and treated with PMA(10 $ng/m{\ell}$), to assess the effect of each herbal medicine on PMA-induced mucin production by enzyme-linked immunosorbent assay(ELISA). Effects of each herbal medicine on PMA-induced MUC5AC gene expression from the same cells were investigated. Also, hypersecretion of airway mucus and goblet cell hyperplasia were induced by exposure of rats to $SO_2$ during 3 weeks. Effects of orally-administered PC, PH, PJ and PS during 1 week on intraepithelial mucosubstances and hyperplasia of goblet cells were examined using histological analysis after staining the epithelial tissue with PAS-alcian blue. (1) PC, PJ, PS and PH did not show significant effects on the survival and proliferation of NCI-H292 cells ; (2) PC, PJ and PS significantly decreased PMA-induced mucin production from NCI-H292 cells ; (3) PC, PJ and PS significantly inhibit the expression levels of PMA-induced MUC5AC gene in NCI-H292 cells ; (4) Among PC, PJ, PS and PH, only PS decreased $SO_2$-induced hyperplasia of airway goblet cells and intraepithelial mucosubstances. This result suggests that PC, PJ and PS can not only affect the production of mucin but also affect the expression of mucin gene and this can explain, at least in part, the traditional use of PC, PJ and PS for controlling airway diseases showing hypersecretion of mucus in oriental medicine.
