Block of hERG $K^+$ Channel by Classic Histamine $H_1$ Receptor Antagonist Chlorpheniramine

  • Chlorpheniramine is a potent first-generation histamine $H_1$ receptor antagonist that can increase action potential duration and induce QT prolongation in several animal models. Since block of cardiac human ether-a-go-go-related gene (hERG) channels is one of leading causes of acquired long QT syndrome, we investigated the acute effects of chlorpheniramine on hERG channels to determine the electrophysiological basis for its proarrhythmic potential. We examined the effects of chlorpheniramine on the hERG channels expressed in Xenopus oocytes using two-microelectrode voltage-clamp techniques. Chlorpheniramine induced a concentration-dependent decrease of the current amplitude at the end of the voltage steps and hERG tail currents. The $IC_{50}$ of chlorpheniramine-dependent hERG block in Xenopus oocytes decreased progressively relative to the degree of depolarization. Chlorpheniramine affected the channels in the activated and inactivated states but not in the closed states. The S6 domain mutations Y652A and F656A partially attenuated (Y652A) or abolished (F656A) the hERG current block. These results suggest that the $H_1$ antihistamine, chlorpheniramine is a blocker of the hERG channels, providing a molecular mechanism for the drug-induced arrhythmogenic side effects.

An Effective Pattern Selection Process for Developing of Pattern Based Software (패턴 기반 소프트웨어 개발을 위한 효과적인 패턴 선정 프로세스)

  • Over the past decade, several types of design pattern have been defined in the software engineering area. But these patterns have not been used so often compared with data structure and algorithm. Likewise, methods to select and apply design patterns during analysis, design, and development are not mentioned in CBD methodologies such as CBD96, RUP, and MaRMI III. This paper suggests the process of effectively selecting design patterns which can be applied to analysis, design, and development for development of application software with based on those offered by GoF and J2EE pattern catalogs. It also demonstrates how to effectively pattern-based software and shows differences and relative advantages from RUP by applying the suggested process to the development of aviation job application.

  • 본 논문은 유해한 동영상을 실시간으로 분석하고 차단하기 위하여, 동영상의 비주얼 특징으로서 그룹 프레임(Group of Frame) 특징을 추출하여 SVM 학습모델을 활용하는 유해 동영상 분류에 관한 것이다. 지금까지 동영상 분류에 관한 연구는 주로 입력 동영상을 뉴스, 스포츠, 영화, 뮤직 비디오, 상업 비디오 등 사전에 정의한 몇 개의 장르에 자동으로 할당하는 기술이었다. 그러나 이러한 분류 기술은 미리 정의한 장르에 따른 일반적인 분류 모델을 사용하기 때문에 분류의 정확도가 높지 않다. 따라서, 유해 동영상을 실시간으로 자동 분류하기 위해서는, 신속하고 효과적인 동영상 내용분석에 적합한 유해 동영상 특화의 특징 추출과 분류 모델 연구가 필요하다. 본 논문에서는 유해 동영상에 대하여 신속하고, 정확한 분류를 위하여 유해 동영상의 대표 특징으로서 그룹프레임 특징을 정의하고, 이를 추출하여 SVM 학습 모델을 생성하고 분류에 활용하는 매우 높은 성능의 분석 방법을 제시하였다. 이는 최근 인터넷 뿐만 아니라 다양한 매체를 통하여 급속도로 번지고 있는 유해 동영상 차단 분야에 적극 활용될 수 있을 것으로 기대된다.

Identification of Antihypertensive Peptides Derived from Low Molecular Weight Casein Hydrolysates Generated during Fermentation by Bifidobacterium longum KACC 91563

  • Angiotensin-converting enzyme (ACE) inhibitory activity was evaluated for the low-molecular-weight fraction (<3 kDa) obtained from milk fermentation by Bifidobacterium longum KACC91563. The ACE inhibitory activity in this fraction was 62.3%. The peptides generated from the <3 kDa fraction were identified by liquid chromatography-electrospray ionization quantitative time-of-flight mass spectrometry analysis. Of the 28 peptides identified, 11 and 16 were identified as β-casein (CN) and αs1-CN, respectively. One peptide was identified as κ-CN. Three peptides, YQEPVLGPVRGPFPIIV, QEPVLGPVRGPFPIIV, and GPVRGPFPIIV, from β-CN corresponded to known antihypertensive peptides. We also found 15 peptides that were identified as potential antihypertensive peptides because they included a known antihypertensive peptide fragment. These peptides were as follows: RELEELNVPGEIVE (f1-14), YQEPVLGPVRGPFP (f193-206), EPVLGPVRGPFPIIV (f195-206), PVLGPVRGPFPIIV (f196-206), VLGPVRGPFPIIV (f197-206), and LGPVRGPFPIIV (f198-206) for β-CN; and APSFSDIPNPIGSENSEKTTMPLW (f176-199), SFSDIPNPIGSENSEKT- TMPLW (f178-199), FSDIPNPIGSENSEKTTMPLW (f179-199), SDIPNPIGSENSEKTTMPLW (f180-199), DIPNPIGSENSEKTTMPLW (f181-199), IPNPIGSENSEKTTMPLW (f182-199), PIGSENSEKTTMPLW (f185-199), IGSENSEKTTMPLW (f186-199), and SENSEKTTMPLW (f188-199) for αs1-CN. From these results, B. longum could be used as a starter culture in combination with other lactic acid bacteria in the dairy industry, and/or these peptides could be used in functional food manufacturing as additives for the development of a product with beneficial effects for human health.

Concurrent Validity between Figure-of-8 Walking Test and Functional Tests Included Tasks for Dynamic Balance and Walking in Patient with Stroke (뇌졸중 환자에서 8자 모양 경로 보행 검사의 동시 타당도 연구)

  • Purpose: The purpose of this study was to determine the concurrent validity between Figure-of-8 Walking Test (F8W), Berg Balance Scale (BBS), Four Squared Step Test (FSST), and Timed UP and GO Test (TUG) in patients with stroke. Methods: Forty two participants (26 men, 16 women, $55.0{\pm}11.72$) with at least three months post stroke who were able to walk at least 10 m without walking aid participated in this study. Assessment of concurrent validity between the F8W (time and steps) and BBS was performed using Spearman rank order correlation and between the F8W (time and steps), FSST and TUG assessed using Pearson correlation. Results: The time of the F8W showed correlation with BBS (r=-0.46, p<0.01), FSST (r=0.64, p<0.01), and TUG (r=0.81, p<0.01), and steps of the F8W showed correlation with BBS (r=-0.43, p<0.01), FSST (r=0.47, p<0.01), and TUG (r=0.51, p<0.01). Conclusion: The F8W is a valid measure of balance and walking skill among patients with stroke and may provide complementary information with regard to dynamic balance and functional walking for the real life of stroke patients.

A Comparative Study of [F-18] Florbetaben (FBB) PET Imaging, Pathology, and Cognition between Normal and Alzheimer Transgenic Mice

  • Alzheimer's disease (AD) is highly prevalent in dementia, with no specifically effective treatment having yet been discovered. Amyloid plaques are one of the key hallmarks of AD. Transgenic mouse models exhibiting Alzheimer's disease-like pathology have been widely used to study the pathophysiology of Alzheimer's disease. In this study, we showed an age-dependent correlation between cognitive function, pathological findings, and [F-18] Florbetaben (FBB) PET images. Nineteen transgenic mice (12 with AD, 7 with controls) were used for this study. We observed an increase in ${\beta}$-Amyloid deposition ($A{\beta}$) in brain tissue and [F-18] FBB amyloid PET imaging in the AD group. The [F-18] FBB data showed a mildly negative trend with cognitive function. Pathological findings were negatively correlated with cognitive functions. These finding suggests that amyloid beta deposition can be well-monitored with [F-18] FBB PET and a decline in cognitive function is related to the increase in amyloid plaque burden.

Enhanced electrocapacitive performance and high power density of polypyrrole/graphene oxide nanocomposites prepared at reduced temperature

  • An attempt was made to investigate the effect of the preparation temperature on the electrocapacitive performance of polypyrrole (PPY)/graphene oxide (GO) nanocomposites (PNCs). For this purpose, a series of PNCs were prepared at various temperatures by the cetyltrimethylammonium bromide-assisted dilute-solution polymerization of pyrrole in presence of GO (wt%) ranging from 1.0 to 4.0 with ferric chloride as an oxidant. The formation of the PNCs was ascertained through Fourier-transform infrared spectrometry, X-ray diffraction spectra, scanning electron microscopy and simultaneous thermogravimetric-differential scanning calorimetry. The electrocapacitive performance of the electrodes derived from sulphonated polysulphone-bound PNCs was evaluated through cyclic voltammetry with reference to Ag/AgCl at a scan rate (V/s) ranging from 0.2 and 0.001 in potassium hydroxide (1.0 M). The incorporation of GO into the PPY matrix at a reduced temperature has a pronounced effect on the electrocapacitive performance of PNCs. Under identical scan rates (0.001 V/s), PNCs prepared at $10{\pm}1^{\circ}C$ render improved specific conductivity (526.33 F/g) and power density (731.19 W/Kg) values compared to those prepared at $30{\pm}1^{\circ}C$ (217.69 F/g, 279.43 W/Kg). PNCs prepared at $10{\pm}1^{\circ}C$ rendered a capacitive retention rate of ~96% during the first 500 cycles. This indicates the excellent cyclic stability of the PNCs prepared at reduced temperatures for supercapacitor applications.

The Reliability and Validity of Figure-of-8 Walk Test in Patients with Stroke (뇌졸중 환자에서 Figure-of-8 walk test의 신뢰도와 타당도)

  • Purpose : The purpose of this study was to establish intra-rater, inter-rater, test-retest reliability, and concurrent validity of figure-of-8 walk test in people with stroke. Methods : The subjects of this study were 17 patients who were diagnosed with a stroke. Subjects were tested twice by the same raters, with 1 day between tests. Subjects were assessed by two physical therapists. Test-retest reliability was calculated using intraclass correlation coefficients (ICC). The concurrent validity was demonstrated by spearman correlation of F8WT with 10m walking test (10MWT), timed up and go test (TUG), Berg balance scale (BBS), dynamic gait index (DGI) and four square step test (FSST). Results : Intra-rater, inter-rater, test- retest of F8WT time, showed high reliability. Intra-rater, inter-rater, test-retest of F8WT steps demonstrated high reliability. Intra-rater, inter-rater, test-retest of F8WT total smoothness score showed below moderate reliability. There was a significant positive correlation of F8WT time with 10MWT, TUG, FSST. There was a significant negative correlation of F8WT time with DGI, BBS. There was a significant positive correlation of F8WT steps with 10MWT, TUG, FSST. There was a significant negative correlation of F8WT steps with DGI. There was a significant positive correlation of F8WT test total smoothness score with BBS. Conclusion : The time, and number of steps in F8WT show high inter, intra-rater, test-retest reliability. The F8WT smoothness shows below moderate reliability. The F8WT shows high concurrent validity with other comparable balance, and walking tests. The F8WT is a valid and reliable measure for assessing walking function in patients with a stroke.

The Prevalence Study of TMD and the Associated Factors in Korean Malocclusion Patients (한국인 부정교합자의 측두하악장애(TMD) 유병율과 그 기여요인에 관한 연구)

  • In order to investigate TMD prevalence in malocclusion patients and to study its relationship with occlusal factors, 205 malocclusion patients (M67, F138, 6Y1M-46Y8M) were examined. The following examinations were carried out, Questionnaire personal history, TMD symptoms, and the associated factors Clinical examination : TMJ sound and maximum mouth opening Orthopantomogram : condyle abnormalities, length of Co'-Inc' and Co'-Go', ratio Co'- Inc'/ Co'-Go', and depth of antegonial notch Transcranial view limitation of anterior movement of condyle Model Angle classification, overjet, overbite, midline discrepancy, missing of posterior teeth, posterior crossbite, attrition of palatal cusp of maxillary molars, crowding/spacing The results could be summarized as follows, 1. The prevalence of TMD showed that Helkimo Anamestic Inder(Ai) 0 was $46.8\%$, Ai I was $22.0\%$, Ai II was $31.2\%$ and subjective symptoms increased with aging (p<0.001) and were frequent in females (p<0.05). 2. Flattening ($4.4\%$) was the most frequent condyle abnormality on Orthopantomogram, and $8.3\%$ of subjects showed some abnormalities on Orthopantomogram. 3. The cases with neck and shoulder pain (p<0.001), clenching, lip biting (p<0.01), and headache (p<0.05) showed higher scores of Ai. 4. Angle class II showed high frequency of condylar abnormalities on Orthopantomogram, and subjects whose palatal cusp of maxillary molars had been attrided had the tendency to show high hi scores (p<0.05). The other occlusal factors had nothing to do with the symptoms of TMD. 5. In the cases that 1)the value of Co'-Inc', Co'-Go' or Co'-Inc'/Co'-Go' were low or 2)the differences of Co'-Go' or Co'-Inc'/Co'-Go' between the right and the left were large, condylar abnormalities were frequently obserbed on Orthopantomogram.

Effect of the Elderly Consumers' Financial Independency on Eating-out Decision Making Process (노인 소비자의 경제적 독립성이 외식 구매 의사 결정 과정에 미치는 영향에 관한 연구)

  • As Korea has approached the aging society, older Koreans have become an important force in restaurant sales today. To succeed with this silver market, it is important for restaurant managers to know who they are and which factor influence the older Koreans' eating-out decision making process. The purpose of this study was to investigate the effect of the elderly consumers financial independency on restaurant selection process. Data were collected from 178 older consumers above 55 years old and analyzed using the descriptive statistic analysis, MANOVA, and one-way ANOVA. The results showed that the elderly consumers financial independency significantly influenced the decision making process in determining where they eat out Significant differences were found between high income group and low income group in the Problem Recognition Step(Wilks' Lambda=0.776, F=3.796), Information Search Step(Wilks' Lambda=0.779, F=2.959), Alternative Evaluation Step (I :Wilks' Lambda=0.835, F=1.748/ II :Wilks' Lambda=0.764, F=3.212), and Purchase Decision Step(Wilks' Lambda=0.849, F=2.412), except the Post-Purchase Behavior(Wilks' Lambda=0.933, F=1.179). The more financially independent older consumers were, the more directly they were involved in the eating out decision making process. Older consumers with higher income and more personal property were likely to 'propose to eat out by themselves'(F=10.986), to obtain restaurant information from the 'printed materials'(F=9.707), to consider 'convenient location' as most important factor when they eat out(F=5.594), and to go to 'family restaurant'(F=7.067), 'Japanese restaurant'(F=7.391) and 'fine dining restaurants'(F-=6.382). In conclusion, we found that the elderly consumers financial independency did influence the eating-out decision making process. Considering that older Korean will become a financially independent consumer and will be eating away from home more often, food service operations should actively position themselves for this market and develop the market-driven menus and services to meet their needs and expectations.
