Flame Retardancy of Cellulose Fabrics Treated with 3-(Hydroxyphenyl Phosphinyl) Propanoic Acid

  • 3-(Hydroxyphenyl phosphinyl) propanoic acid (HPPA) has been one of the most commonly used durable flame retardant agents for polyethylene terephthalate (PET) for many years. We intended to explore the application of HPPA to cellulose fabrics as formaldehyde-free phosphorus based flame retardants (FRs) through green chemistry process. The flame retardancy of the flame-retardant treated cellulose fabrics were characterized by using inductively coupled plasma spectroscopy (ICP) and limiting oxygen index (LOI). Structural changes of the treated cellulose fabrics were carried out by thermogravimetric analysis (TGA) and Fourier transform infrared (FT-IR) spectroscopy. To enhance the flame retardancy of HPPA treated cellulose fibers, glycerol polyglycidyl ether (GPE), a crosslinking agent was employed. Both HPPA and GPE treated cotton fabric imparted an LOI value over 26.

  • Benzoxazolo carbocyanine compounds were synthesized by condensation of a suitable ortho-ester with an appropriately substituted 2-methylbenzoxazole in the presence of triethylamine. This compounds used as green sensitizing dyes in photographic emulsions. The compounds are characterized by fast atom bombardment mass spectrometry. The values(m/z) of structurally significant ions observed in FAB spectra. It was showed tentative fragmentation pattern in FAB spectroscopy of $HN(C_2H_5)_3$ cations in glycerol/trifluoroacetic acid matrix.

Purification and Characterization of a 15S Arpase from Chick Skeletal Muscle (계 골격근에서 15S ATPase의 순수분리 및 특성연구)

  • An Arpase complex has been purified to apparent homogeniety from the extract of chick skeletal muscle using conventional column chromatographies and glycerol density gradient centrifugation. This eWe has a sedimentation coefficient of 15S as determined by the gradient centrifugation and therefore is referred to as the 15S ATPase. It behaves as a 600-kOa molecule upon gel filtration analysis using a Superose-6 column. However, the ATPase runs as a 95-kDa polvpeptide when analyzed by polvacrvlamide gel electrophoresis in the presence of sodium dodecyl sulfate. Thus, the Arpase is likely to consist of six identical subunits of 95 KDa. It has a Km value of 0.6 mM for ATP and is maximally active at pH 9.

  • Structured lipids (SLs) are defined as triacylglycerols to change the fatty acid composition in the glycerol backbone and lipases are known as a powerful tool for the syntheses of SLs. Structured lipid from corn oil, capric acid, and conjugated linoleic acid (CLA) by transesterification reaction and using several amounts of immobilized lipase RM IM (from Rhizomucor miehei) was studied, and 4% of lipase amount was selected for further study as the optimal amount. Comparison the chemical properties (free fatty acid value, iodine value, saponification value, tocopherols, and color analysis), solidification behavior, and volatile fractions (from headspace SPME GC-MS) between com oil and SL com oil was obtained.

  • The present paper suggests new method to know the effects of molecular diffusion and the helicity of microchannel flows on mixing in passive micromixers, which are essential components of a microfluidic chip. In this study, 'Helix Index' is newly defined as the magnitude of chaotic advection. Relationship between Helix Index and Mixing Index is analyzed numerically such as the wide range of Peclet and Reynolds numbers in three dimensional serpentine microchannel when using soluble solutions (water/glycerol). As a result, a simple algebraic equation is derived by this relationship based on a regression analysis. The algebraic equation is found to be able to accurately predict the mixing performance without solving the coupled, complex momentum and mass transfer equations.

Genomic Insight into the Salt Tolerance of Enterococcus faecium, Enterococcus faecalis and Tetragenococcus halophilus

  • To shed light on the genetic basis of salt tolerance in Enterococcus faecium, Enterococcus faecalis, and Tetragenococcus halophilus, we performed comparative genome analysis of 10 E. faecalis, 11 E. faecium, and three T. halophilus strains. Factors involved in salt tolerance that could be used to distinguish the species were identified. Overall, T. halophilus contained a greater number of potassium transport and osmoprotectant synthesis genes compared with the other two species. In particular, our findings suggested that T. halophilus may be the only one among the three species capable of synthesizing glycine betaine from choline, cardiolipin from glycerol and proline from citrate. These molecules are well-known osmoprotectants; thus, we propose that these genes confer the salt tolerance of T. halophilus.

  • Silkworm (Bombyx mori) pupae have been widely worked in wound dressing, hepatoprotective activity, antigenotoxicity, control of glucose level and anti-cancer activity. To investigate the anti-obesity activity of ethanol extract of silkworm pupae powder fermented with Cordyceps militaris (ESfC), the free glycerol release and cAMP concentration as well as fat accumulation were measured in the primary adipocytes of SD (Sprague Dawley) rats and high fat diet (HFD)-treated C57BL/6 mice treated with 12 weeks. Firstly, the presence of the cordycepin with lipid lowering effect was confirmed in ESfC using HPLC analysis. The level of free glycerol and cAMP concentration was significantly increased in the primary adipocytes treated with high dose of ESfC ($400{\mu}g/ml$) although these levels were consistently maintained in other dose ESfC treated groups. In HFD-induced obesity model, the increased fat weight and size of adipocytes in HFD+Vehicle treated group was recovered in HFD+ESfC treated group. Also, the liver weight and the number of lipid droplets were higher in HFD+Vehicle treated group than No treated group. But, this level was significantly decreased in HFD+ESfC treated group compared with HFD+Vehicle treated group. Furthermore, a similar recovery was detected on the phosphorylation of periliphin and HSL, and ATGL expression. Overall, the results of the present study provide some scientific evidences that ESfC can stimulate lipolysis in primary adipocytes and prevent fat accumulation in HFD-treated obesity model, and therefore have the potential for use as anti-obesity agents to treat obese patient.

Characterization of Microbial Communities in a Groundwater Contaminated with Landfill Leachate using a Carbon Substrate Utilization Assay (탄소원 이용도 평가를 활용한 매립지 침출수로 오염된 지하수의 미생물 군집 특성 해석)

  • The microbial community properties of groundwater samples contaminated with landfill leachates were examined using Ecoplate including 31 sole carbon sources. The samples were KSG1-12 (leachate), KSG1-16 (treated leachate), KSG1-07 (contaminated groundwater), KSG1-08 (contaminated groundwater), and KSG1-13 (uncontaminated groundwater). Among the carbon sources used as substrates, 2-hydroxy benzoic acid, D,L-$\alpha$-glycerol phosphate, and D-malic acid were not utilized in any sample, while D-xylose, D-galacturonic acid, L-aspargine, tween 80, and L-serine were utilized in all 5 samples. The rest of substrates showed very different patterns among the samples. Average well color development (AWCD) analysis demonstrated that the potential activity on 31 substrates was in the order of KSG1-16 > KSG1-12 > KSG1-07 > KSG-08 > KSG1-13, which generally agrees with the degree of pollution, except KSG1-16. Principal component analysis (PCA) on similarity between samples showed two groups (KSG1-12, -07 and -08 vs KSG1-16 and -13), coinciding with contaminated and uncontaminated groups. Shannon index showed that the microbial diversities were similar among the samples.

Neutralizing Chimeric Mouse-human Antibodies against Burkholderia pseudomallei Protease: Expression, Purification and Characterization

  • A recombinant Fab monoclonal antibody (Fab) C37, previously obtained by phage display and biopanning of a random antibody fragment library against Burkholderia pseudomallei protease, was expressed in different strains of Escherichia coli. E. coli strain HB2151 was deemed a more suitable host for Fab expression than other E. coli strains when grown in media supplemented with 0.2% glycerol. The expressed Fab fragment was purified by affinity chromatography on a Protein G-Sepharose column, and the specificity of the recombinant Fab C37 towards B. pseudomallei protease was proven by Western blotting, enzyme-linked immunosorbent assay (ELISA) and by proteolytic activity neutralization. In addition, polyclonal antibodies against B. pseudomallei protease were produced in rabbits immunized with the protease. These were isolated from high titer serum by affinity chromatography on recombinant-Protein A-Sepharose. Purified polyclonal antibody specificity towards B. pseudomallei protease was proven by Western blotting and ELISA.

Transcriptome Analysis of Phosphate Starvation Response in Escherichia coli

  • Escherichia coli has a PhoR-PhoB two-component regulatory system to detect and respond to the changes of environmental phosphate concentration. For the E. coli W3110 strain growing under phosphate-limiting condition, the changes of global gene expression levels were investigated by using DNA microarray analysis. The expression levels of some genes that are involved in phosphate metabolism were increased as phosphate became limited, whereas those of the genes involved in ribosomal protein or amino acid metabolism were decreased, owing to the stationary phase response. The upregulated genes could be divided into temporarily and permanently inducible genes by phosphate starvation. At the peak point showing the highest expression levels of the phoB and phoR genes under phosphate-limiting condition, the phoB- and/or phoR-dependent regulatory mechanisms were investigated in detail by comparing the gene expression levels among the wild-type and phoB and/or phoR mutant strains. Overall, the phoB mutation was epistatic over the phoR mutation. It was found that PhoBR and PhoB were responsible for the upregulation of the phosphonate or glycerol phosphate metabolism and high-affinity phosphate transport system, respectively. These results show the complex regulation by the PhoR-PhoB two-component regulatory system in E. coli.
