The relationship between Gly71Arg and TATA box polymorphism of GT1A1 gene and prolonged hyperbilirubinemia of breast milk feeding infant in Korean (지속성 고빌리루빈혈증과 연관된 모유 황달에서 UGT1A1(Gly71Arg, TATA box) 다형성에 대한 연구)

  • Purpose : It has been known that breast milk cause prolonged unconjugated hyperbilirubinemia. UGT1A1 is a important gene of uridine diphosphate glucuronosyltransferase (UGT) which has a major role of bilirubin metabolism. These findings suggest that there is a relationship between UGT1A1 gene mutation and prolonged jaundice of breast feeding infant. The aim of study was to investigate whether a polymorphism of the UGT1A1 gene exist in prolonged hyperbilirubinemia of breast milk feeding Korean infant. Methods : The genomic DNA was isolated from 50 full term Korean neonates, who had greater than a 10 mg/dL of serem bilirubin after 2 weeks of birth with no significant cause, and the other genomic DNA was isolated from 162 full term Korean neonates of the control population. Both group fed breast milk. We performed direct sequencing of TATA box and Gly71Arg polymorphism of the UGT1A1 gene. Results : Two of the 50 neonates with hyperbilirubinemia had AA polymorphism, and 40 had GA polymorphism. Five of the 129 neonates of the control group had AA polymorphism, and 4 had GA polymorphism. The allele frequency of G>A polymorphism in the hyperbilirubinemia group was 44.0%; it was significantly higher than 5.4% of the control group. TATA box polymorpism was not different both group significantly. Conclusion : Our result indicated that Gly71Arg polymorphism is associated with the prolonged hyperbilirubinemia of breast milk-feeding infant in Korean, while TATA box polymorphism is not associated with the prolonged hyperbilirubinemia of breast milk-feeding infant in Korean.

The Association of Neonatal Hyperbilirubinemia with UGT1A1 and CYP1A2 Gene Polymorphism in Korean Neonates (한국인의 신생아 황달과 UGT1A1 및 CYP1A2 유전자 다형성과의 연관성)

  • Purpose : The incidence of nonphysiologic neonatal hyperbilirubinemia is twice as high in East Asians as in whites. Recently, UGT1A1 mutation was found to be a risk factor for neonatal hyperbilirubinemia. In congenitally-jaundiced Gunn rats, which lack expression of UDP-glucuronosyltransferase, alternative pathways can be stimulated by inducers of CYP1A1 and CYP1A2 enzymes. CYP1A2 plays a major role in bilirubin degradation of the alternate pathway. We studied the relationship between UGT1A1 and CYP1A2 gene polymorphism of neonatal hyperbilirubinemia in Koreans. Methods : Seventy-nine Korean full term neonates who had hyperbilirubinemia(serum bilirubin >12 mg/dL) without obvious causes of jaundice, were analyzed for UGT1A1 and CYP1A2 gene polymorphism; the control group was sixty-eight. We detected the polymorphism of Gly71Arg of UGT1A1 gene by direct sequencing and T2698G of CYP1A2 by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) using MboII and direct sequencing. Results : Allele frequency of Gly71Arg mutation in the hyperbilirubinemia group was 32 percent, which was significantly higher than 11 percent in the control group(P<0.0001). Mutant gene frequency of T2698G was 41.8 percent in patients and 32.3 percent in the control group(P=0.015), but allele frequency was 21 percent in patients and 19 percent in the control group, which was not significantly higher(P=0.706). There was no relationship between mutations of two genes(P=0.635). Conclusion : The polymorphism of UGT1A1 gene(Gly71Arg) and CYP1A2 gene(T2698G) was detected in Korean neonatal hyperbilirubinemia. Only polymorphisms of Gly71Arg in UGT1A1 were significantly higher than control group.

Association between Arg16Gly Mutation in the ${\beta}_2$-Adrenergic Receptor Gene and Hypertension in the Korean Population

  • $\beta$$_2$-Adrenergic receptors($\beta$$_2$-AR) contribute to cardiovascular regulation by influencing several functions and a several studies suggest that a decreased function of the $\beta$$_2$-AR may be involved in essential hypertension. We investigated the Arg16Gly mutation of $\beta$$_2$-AR gene, which show enhanced agonist-promoted downregulation of the receptor and yielded different results in terms of association with essential hypertension. We studied the relationship between genetic variation in the $\beta$$_2$-adrenergic receptor gene and hypertension in a Korean population using Nde I restriction fragment length polymorphism (RFLP) analysis. There were significant differences in allele and genotype frequencies between essential hypertensive and normotensive group (Odds ratio(CI) = 1.71 (1.09-2.70)). Therefore, our result suggests that the Nde I RELP of the $\beta$$_2$-adrenergic receptor gene may be useful as a genetic marker in hypertension diagnostics in Korean population.

Evaluation of the Apparent Ileal Digestibility (AID) of Protein and Amino Acids in Nursery Diets by In vitro and In vivo Methods

  • The objective was to evaluate in vitro prediction of ileal digestibility of protein and amino acids (AA) for current nursery pig diets (n = 10) by using pepsin and pancreatin incubations. To compare in vivo ileal digestibility, forty nursery pigs (4 pigs per diet) with an initial BW of $12.2{\pm}2.7$ kg were surgically equipped with T-cannula in the distal ileum. In all cases, the values of in vitro digestibility were higher than those of in vivo digestibility (p<0.05). With regard to the relationships of essential and non essential AA (CP), the $r^2$ value was 0.76. With regard to AA, high relationships were observed in Ile, Thr, and Gly (0.85, 0.83, and 0.89, respectively). Also, there was a lower relationship for Arg, Met, Ala, Asp, Glu, Pro, Ser, and Tyr with $R^2$ values of 0.56, 0.54, 0.40, 0.54, 0.45, 0.24, 0.49, and 0.35, respectively between in vitro and in vivo digestibility. The EAA relationship ($R^2$ = 0.71) was generally higher than that of NEAA ($R^2$ = 0.50) numerically. In conclusion, there were strong linear relationships between in vivo and in vitro ileal digestibility (CP, Ile, Thr, and Gly). In vitro prediction of ileal digestibility (CP, Ile, Thr, and Gly) seems to have significant potential for practical application.

  • Bacillus thuringiensis was mutagenized with UV light irradiation and nitrosoguanidine. Twenty-four tem perature-sensitive ts mutants were isolated at 42$^{\circ}C$ and classified into two groups by growth on nutrient agar at 42$^{\circ}C$. First is the lethal group, which did not grow at the nonpermissive temperature, the second is the reduced group whose growth was restricted from one-half to one-fourth, Thirteen ts mutants belong to the lethal group and eleven ts mutants belong to the reduced group. Auxotrophic mutant, A-N28 required five amino acids as growth factors, A-N65 also five amino acids, A-N92 seven, A-N115 four and A-N156 three. Bacillus thuringiensis wild type is resistant to penicillin, ampicillin, and cephalothin. The ts-Ul7l, A-N92 and A-Nl15 are sensitive to the three antibiotics. The ts -U601, -U603, -U604 and -Ul71 did not grow at the permissive temperature after temperature-shifting from 42$^{\circ}C$. Four auxotrophic mutants (A-N38, A-N65, A-N92 and A-Nl15) did not form spores in their cells.

Components of White and Dark Muscle of Skipjack for Canning (통조림용 가다랑어육의 식품성분)

  • The lipid components, nitrogenous extracts and amino acids of dark muscle(DM) of ski-pjack (Katsuwonus pelamis) were analyzed and compared with those of white muscle (WM). WM was higher in moisture and crude protein content, and lower in crude lipid and ash content than those of DM. Contents of volatile basic nitrogen in WM and DM were 22.7mg/100g and 46.9mg/100g. Total lipid(TL) of WM and DM consisted of $79.7\%,\;71.9\%$ neutral lipid(NL), $6.8\%,\;9.5\%$ glycolipid(GL), and $13.5\%,\;18.6\%$ phospholipid(PL), respectively NL was mainly com-posed of free fatty acid, triglyceride, and PL was mainly occupied by phosphatidyl ethanolamine, phosphatidyl choline. Also Iysophosphatidyl choline and Iysophosphatidyl ethanolamine were identified in PL. In fatty acid composition of TL, NL, GL and PL, WM revealed higher contents in saturates and monoenes such as 16 : 0, 18 : 1, while DM showed higher contents in polyenes such as 22 : 6 especially. The major fatty acids of these samples were generally 16: 0, 18:0, 18:1, 20:5 and 22 : 6. Contents of total free amino acids from WM and DM were 5,982.3mg/100g and 4,450.7 mg/100g (dry base). Of free amino acids, Tau concentration was much higher in DM than in WM, Ala, Gly, Met, Arg, Thr were also high in DM. But His was much higher in concentration in W. Content of inosinic acid(IMP) in WM(680.9mg/100g) was higher than that of DM(73.1mg/100g). The degradations of IMP proceeded very rapidly in DM. DM contained much higher trimethylamine oxide and trimethylamine than those of WM. The profile of combined amino acids in these samples, were very similar, and main amino acids were Glu, Asp, Lys, Ala, Ile and Arg.
