Iron sulfate and molasses treated anthocyanin-rich black cane silage improves growth performance, rumen fermentation, antioxidant status, and meat tenderness in goats

  • Objective: This study investigated the effects of feeding anthocyanin-rich black cane treated with ferrous sulfate and molasses on animal performance, rumen fermentation, microbial composition, blood biochemical indices, and carcass characteristics in meat goats. Methods: Thirty-two Thai-native×Anglo-Nubian crossbred male goats (14.47±2.3 kg) were divided equally into two groups (n = 16) to investigate the effect of feeding diet containing 50% untreated anthocyanin-rich black cane silage (BS) vs diet containing anthocyaninrich black cane silage treated with 0.03% ferrous sulfate and 4% molasses (TBS) on average daily gain (ADG) and dry matter intake (DMI). At the end of 90 d feeding trial, the goats were slaughtered to determine blood biochemical indices, rumen fermentation, microbial composition, and carcass characteristics differences between the two dietary groups. Results: Goats fed the TBS diet had greater ADG and ADG to DMI ratio (p<0.05). TBS diet did not affect rumen fluid pH; however, goats in the TBS group had lower rumen ammonia N levels (p<0.05) and higher total volatile fatty acid concentrations (p<0.05). Goats in the TBS group had a higher (p<0.05) concentration of Ruminococcus albus but a lower (p<0.05) concentration of methanogenic bacteria. The TBS diet also resulted in lower (p<0.05) thiobarbituric acid-reactive substances concentration but higher (p<0.05) total antioxidant capacity, superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase concentrations in blood plasma, while having no effect on plasma protein, glucose, lipid, immunoglobin G, alanine transaminase, and aspartate aminotransferase. Meat from goats fed the TBS diet contained more intramuscular fat (p<0.05) and was more tender (p<0.05). Conclusion: In comparison to goats fed a diet containing 50% untreated anthocyanin-rich black cane silage, feeding a diet containing 50% anthocyanin-rich black cane silage treated with 0.03% ferrous sulfate and 4% molasses improved rumen fermentation and reduced oxidative stress, resulting in higher growth and more tender meat.

C57BL/6 Mice에서 이소플라본의 지질강하, 항산화, 항염증효과를 통한 항동맥경화 효과 (Anti-atherogenic Effect of Isoflavone through Hypolipidemic, Anti-oxidative and Anti-inflammatory Actions in C57BL/6 Mice)

  • 5주령의 C57BL/6 mice종 암컷 40수를 2주간의 적응기를 거쳐, 평균 체중 $17.9{\pm}0.9\;g$인 실험동물을 완전임의배치로 한 군당 10수씩 4군으로 나누어 실험식이는 동맥경화식이(control)에 이소플라본을 첨가한 실험식이를 각각 8주간 급여하면서 사육하였다. 이소플라본의 첨가량은 식이 100 g 당 10 mg(IF-10)과 40 mg(IF-40) 및 100 mg(IF-100)이 되도록 조정하여 제조하였다. 이소플라본의 섭취는 체중증가량에 영향을 미치지 않는 것으로 나타났으며 식이효율은 각 군 간의 유의적인 차이를 보이지 않았다. 이소플라본 섭취에 의한 혈장 TG 농도는 대조군에 비해 이소플라본을 첨가한 식이를 섭취한 군에서 감소하는 추세를 보였다. 총 콜레스테롤 수준은 모든 실험군에서 증가하는 경향을 보였으나 8주 후의 농도 변화는 대조군에 비해 이소플라본을 첨가한 식이를 섭취한 군에서 감소하는 추세를 보였다. 세포부착물질인 MCP-1과 지질과산화물인 TBARS의 생성량은 이소플라본의 첨가량이 높을수록 현저하게 감소되는 결과를 보여 농도의존적인 효과를 보였다. 간의 TG와 총 콜레스테롤 농도는 이소플라본의 첨가량이 높을수록 감소하는 것으로 나타났으며(p<0.05), 간의 항산화 영양소인 glutathione 함량은 이소플라본을 100 mg 제공한 IF-100에서 유의적으로(p<0.05) 높은 수준을 보였다. 간의 항산화 효소계 활성은 대조군에 비해 이소플라본을 첨가한 식이를 섭취한 군에서 대체로 높게 나타났으며 특히 이소플라본 100 mg을 첨가한 식이를 섭취한 IF-100에서 현저하게 높은 수준을 보였다. 이상의 결과들로 미루어 볼 때 이소플라본의 섭취가 혈장의 중성지방을 저하하고 항염증, 항산화효과를 보여 동맥경화예방에 효과가 있는 물질로 사료된다.

스트렙토조토신 유발 당뇨 쥐의 산화스트레스에 대한 매생이 추출물의 신장 보호 효과 (In vivo Study of the Renal Protective Effects of Capsosiphon fulvescens against Streptozotocin-induced Oxidative Stress)

  • 당뇨 모델 쥐에서 매생이 추출물과 그 지표물질인 pheophorbide A의 신장 보호 효과를 확인하였다. 5주령의 SD 랫드를 일주일간 순화시킨 뒤 40 mg/kg BW의 농도로 streptozotocin을 복강 투여하였으며, 한달 동안의 유도기간을 거쳐 모든 당뇨 유발군에서 혈당이 400-500 mg/dL가 된 것을 확인한 이후 9주 동안의 샘플 투여를 진행하였다. 신장 손상과 관련한 혈액 생화학적 지표인 BUN, creatinine에서는 당뇨 유발군 사이에 개선되는 경향을 보이진 않았다. 신장 조직에서 산화스트레스에 의해 유발되는 지질 과산화물을 측정 하였을 때 당뇨 유발군인 STZ군($0.19{\pm}0.04$)은 정상 대조군인 Con군($0.05{\pm}0.00$)에 비하여 3.5배 많은 MDA를 생성하였지만 매생이 추출물 4, 20 mg/kg BW투여한 CL, CH군은 MDA의 생성량이 $0.05{\pm}0.02$, $0.08{\pm}0.02$, 지표물질인 pheophorbide A를 0.2 mg/kg BW을 투여한 PhA군은 0.12{\pm}0.01 nmol/mg protein로 샘플 투여군에서 과산화물이 유의적으로 감소하는 것으로 보아(p<0.05) CFE와 PhA의 항산화 작용을 확인할 수 있었다. 항산화 바이오 마커인 GSH와 GSSH 함량 및 GPx, GR, GST 활성을 측정한 결과 CFE와 PhA는 산화스트레스에 의한 신장 조직의 GSH 함량이 감소하는 것을 막아 당뇨로 인한 신장 손상에 보호 효과가 있는 것을 확인하였다(p<0.05). 또한 STZ에 유도된 당뇨 쥐의 신장 조직엔 많은 양의 과산화물이 축적되어 있으며 이를 소거하기 위해 GPx의 활성이 높아졌는데, AC군과 PhA군에서 GPx 의 증가 정도를 유의적으로 완화시켰다(p<0.05). GR 활성은 CL, CH, PhA군에서 Con군 수준까지 올라오는 것으로 보아 신장의 산화적 손상을 정상수준까지 회복 시키는 것으로 보였으며, GST 결과로 미루어 보아 CFE와 PhA는 신장 조직에서 일차적으로 과산화물을 포집하고 GSH를 이용하여 체내의 독성 물질을 분해시켜 STZ로 유도된 당뇨로부터 신장 보호 효과를 나타내었다. 또한, 조직 내에서 SOD와 CAT의 활성 측정 결과, CFE와 PhA는 지질과산화물의 증가로 산화적 손상이 가속화 될 때, 항산화 효소계의 활성을 증가시키고 산화 스트레스에 의한 조직 손상을 완화 시켜 주는 것으로 확인되었다. 신장 조직의 병리학적 관찰 결과 CL, CH 및 PhA군에선 STZ군에 비해 비이상적인 글리코겐의 축적이 유의적으로 저해 되는 것을 관찰할 수 있었다(p<0.05). 위와 같은 실험 결과를 통하여 매생이 추출물과 PhA가 당뇨병성 신장 손상에 보호 효과가 있다는 것을 확인하였다.

Heat Shock Causes Oxidative Stress and Induces a Variety of Cell Rescue Proteins in Saccharomyces cerevisiae KNU5377

  • In this study, we attempted to characterize the physiological response to oxidative stress by heat shock in Saccharomyces cerevisiae KNU5377 (KNU5377) that ferments at a temperature of $40^{\circ}C$. The KNU5377 strain evidenced a very similar growth rate at $40^{\circ}C$ as was recorded under normal conditions. Unlike the laboratory strains of S. cerevisiae, the cell viability of KNU5377 was affected slightly under 2 hours of heat stress conditions at $43^{\circ}C$. KNU5377 evidenced a time-dependent increase in hydroperoxide levels, carbonyl contents, and malondialdehyde (MDA), which increased in the expression of a variety of cell rescue proteins containing Hsp104p, Ssap, Hsp30p, Sod1p, catalase, glutathione reductase, G6PDH, thioredoxin, thioredoxin peroxidase (Tsa1p), Adhp, Aldp, trehalose and glycogen at high temperature. Pma1/2p, Hsp90p and $H^+$-ATPase expression levels were reduced as the result of exposure to heat shock. With regard to cellular fatty acid composition, levels of unsaturated fatty acids (USFAs) were increased significantly at high temperatures ($43^{\circ}C$), and this was particularly true of oleic acid (C18:1). The results of this study indicated that oxidative stress as the result of heat shock may induce a more profound stimulation of trehalose, antioxidant enzymes, and heat shock proteins, as well as an increase in the USFAs ratios. This might contribute to cellular protective functions for the maintenance of cellular homeostasis, and may also contribute to membrane fluidity.

Effects of Fertilization on Physiological Parameters in American Sycamore (Platanus occidentalis) during Ozone Stress and Recovery Phase

  • American sycamore seedlings were grown in chambers with two different ozone concentrations ($O_3$-free air and air with additional $O_3$) for 45 days. Both the control and the $O_3$ chambers included non-fertilized and fertilized plants. After 18 days of $O_3$ fumigation, seedlings were placed in a clean chamber for 27 days. Seedlings under ozone fumigation showed a significant decrease in pigment contents and photosynthetic activity, and a significant increase in lipid peroxidation. Fertilization enhanced physiological damage such as the inhibition of photosynthetic activity and the increase of lipid peroxidation under ozone fumigation. During the recovery phase, the physiological damage level of seedlings increased with ozone fumigation. In addition, physiological damage was observed in the fertilized seedlings. Superoxide dismutase (SOD) and glutathione reductase (GR) activities of $O_3$-treated seedlings increased up to 33.8% and 16.3% in the fertilized plants. The increase of SOD activity was higher in the fertilized plants than in the non-fertilized plants. Negative effects of ozone treatment were observed in the biomass of the leaves and the total dry weight of the fertilized sycamore seedlings. The $O_3$-treated seedlings decreased in stem, root and total dry weight, and the loss of biomass was statistically significant in the fertilized plants. In conclusion, physiological disturbance under normal nutrient conditions has an effect on growth response. In contrast, in conditions of energy shortage, although stress represents a physiological inhibition, it does not seem to affect the growth response.

생화학적 지표로 본 알코올성 간질환 환자의 비타민 및 무기질 영양상태 (Biochemical Evaluation of Nutritional Status of Vitamins and Minerals in Patients with Alcoholic Liver Disease)

  • The purpose of this study is to evaluate the nutritional status of vitamin and minerals in the patients with alcoholic liver disease and to obtain the materials for nutritional education for alcoholics. The subjects consist of 80 patients with alcoholic liver cirrhosis(ALC) and 12 patients with alcoholic fatty liver(AFL). The control group included 57 alcoholics without liver disease(A), 32 patients with viral liver cirrhosis(VLC) and 194 normal men(NL). Biochemical evaluation of nutritional status was investigated by the analysis of blood samples. The frequency of vitamin B1 deficiency in the ALC, AFL and A groups as indicated by the erythrocyte transketolase activity coefficient were 46.0%, 66.7% and 57.9% respectively. The frequency of vitamin B2 deficiency in the ALC, AFL and A groups as indicated by the erythrocyte glutathione reductase activity coefficient were 9.8%, 8.3% and 38.6% respectively. Vitamin A deficiency was not detected in the alcoholic subjects. The frequency of vi tamin E deficiency in ALC, AFL and A were 96.3%, 66.7% and 86.0% respectively. The levels of plasma lipid peroxidation products were significantly higher in the alcoholic subjects than in the normal subjects. The frequency of subjects below normal range of hemoglobin were 85.0% in ALC, 50.0% in AFL and 31.6% in A. The frequency of copper deficiency in the ALC, AFL and A groups were 48.4%, 16.7% and 17.5% respectively. The frequency of zinc deficiency in the ALC, AFL and A groups were 83.8%, 41.7% and 66.7% respectively. Overall, the vitamin and minerals status of the alcoholic subjects in this study was evaluated to be very poor on the basis of biochemical assessments. The results suggest that alcohol abuse and poor dietary intake could cause malnutrition and may be important risk factors in causing alcoholic liver disease in alcoholics. In addition, vitamin B1, vitamin B2, Cu, Fe and antioxidant supplementation may be effective in nutritional therapy for chronic alcoholics.

Acute phase protein mRNA expressions and enhancement of antioxidant defense system in Black-meated Silkie Fowls supplemented with clove (Eugenia caryophyllus) extracts under the influence of chronic heat stress

  • Background: The current study investigates the anti-stress effects of clove (Eugenia caryophyllus) extracts (0, 200, 400, and 600 mg/kg) on serum antioxidant biomarkers, immune response, immunological organ growth index, and expression levels of acute phase proteins (APPs); ovotransferrin (OVT), ceruloplasmin (CP), ceruloplasmin (AGP), C-reactive protein (CRP), and serum amyloid-A (SAA) mRNA in the immunological organs of 63-d-old male black-meated Silkie fowls subjected to 21 d chronic heat stress at $35{\pm}2^{\circ}C$. Results: The results demonstrated that clove extract supplementation in the diet of Silkie fowls subjected to elevated temperature (ET) improve growth performance, immune responses, and suppressed the activities of glutathion peroxidase (GSH-Px), superoxide dismutase (SOD), catalase (CAT), and thioredoxin reductase (TXNRD); reduced serum malonaldehyde (MDA) and glutathione (GSH) concentrations when compared with fowls raised under thermoneutral condition (TC). Upon chronic heat stress and supplementation of clove extracts, the Silkie fowls showed a linear increase in GSH-Px, SOD, CAT, and TXNRD activities (P = 0.01) compared with fowls fed diets without clove extract. ET decreased (P < 0.05) the growth index of the liver, spleen, bursa of Fabricius and thymus. However, the growth index of the liver, spleen, bursa of Fabricius and thymus increased significantly (P < 0.05) which corresponded to an increase in clove supplemented levels. The expression of OVT, CP, AGP, CRP, and SAA mRNA in the liver, spleen, bursa of Fabricius and thymus were elevated (P < 0.01) by ET compared with those maintained at TC. Nevertheless, clove mitigates heat stress-induced overexpression of OVT, CP, AGP, CRP and SAA mRNA in the immune organs of fowls fed 400 mg clove/kg compared to other groups. Conclusions: The results showed that clove extracts supplementation decreased oxidative stress in the heat-stressed black-meated fowls by alleviating negative effects of heat stress via improvement in growth performance, antioxidant defense mechanisms, immunity, and regulate the expression of acute phase genes in the liver and immunological organs.

The effects of $Angelica$ $keiskei$ $Koidz$ on the expression of antioxidant enzymes related to lipid profiles in rats fed a high fat diet

  • This study was performed to examine the feeding effects of $Angelica$ $keiskei$ $Koidz$ (AK) and its processed products on serum, liver, and body fat content and the expression of antioxidant genes in rats fed a high fat diet. AK and its processed products were added at 3-5% to a high fat diet and fed to adult rats for 6 weeks. In experiment 1 (EXP 1), the rats were fed with one of six diets including a control diet (normal fat), high fat diet (HF), and HF + AK additives groups (four groups). In experiment 2 (EXP 2), the rats were separated into three groups of HF, HF + AK whole leaves, and HF + fermented juice (FS) + squeeze (SA). Body weight was not different among the groups in either experiment. The liver weight was lower in the FS and SA groups compared to that in the other groups (P<0.05). Serum luteolin was higher in the AK and processed products groups compared to that in the HF group (P<0.05). Gene expression of the antioxidative enzymes catalase and glutathione-s-reductase in the liver was higher in the AK processed products group than that in the other groups (P<0.05). The results suggest that the intake of AK and its processed products increased the expression of antioxidant enzymes in animals fed a high fat diet, reduced hepatic cholesterol content, and increased the effective absorption of luteolin.

Antioxidant Enzyme Responses against Abiotic and Biotic Stresses in Rehmannia glutinosa L. and Glycine max L.

  • Rehmannia glutinosa shows a high level of resistance to the non-selective herbicide paraquat. To characterize the antioxidant enzyme system of R. glutinosa, we comparatively examined the responses of antioxidant enzymes to UV, wounding and a general elicitor yeast extract in R. glutinosa and soybean. The levels of enzyme activities of the two plant species were drastically different between those per fresh weight (general activity) and per protein (specific activity) bases. The general activities of superoxide dismutase (SOD), peroxidase (POX), catalase (CAT), and glutathione reductase (GR) were lower, but that of ascorbate peroxidase (APX) was higher in R. glutinosa than in soybean. The specific activities of the enzymes, however, were about two- to seven-fold higher in R. glutinosa than in soybean, except that of CAT, which was about 12-fold higher in soybean. The general and specific enzyme activities of R. glutinosa relative to those of soybean showed a consistent increase in responses to the stresses only in SOD. The specific activities of SOD and APX were higher in R. glutinosa in all stress treatments. The results might suggest a relatively higher contribution of SOD and APX to the stress tolerance.

Germination and Biochemical Changes in Accelerated Aged and Osmoprimed Pinus thunbergii Seeds

  • The aim of this study was to investigate relationship among seed viability and enzymes activities involved in scavenging reactive oxygen species (ROS), especially, superoxide dismutase (SOD), glutathione reductase (GR), and catalase (CAT). In other respects, osmopriming has been demonstrated to reinvigorate aged seeds. Various viabilities of seeds that were ranged from 80 to 100% of germination rate could be produced using osmopriming and accelerated aging treatments. Priming treatment of Pinus thunbergii seeds for 3 days at $15^{\circ}C$ with a polyethylene glycol solution at -1.2 MPa improved their subsequent germination at $25^{\circ}C$. Accelerated aging (3, 6, 9, and 12 days at $41^{\circ}C$ and 100% relative humidity) decreased seed germination percentage depending on aging treatment duration. Electrolyte conductivities of seeds were measured as assay of membrane integrity. The conductivity from electrolyte leakage of P. thunbergii seed was also correlated with seed germinability. Conductivity for control seeds that had 95% of germination percentage was 3.48 ${\mu}S\;g^{-1}$, but jumped as doubled (7.98 ${\mu}S\;g^{-1}$) in 12-day-aged seed that had 80% of germination percentage. Our results demonstrate that aging of P. thunbergii seeds is associated with changes in the electrolyte leakage, lipid peroxidation, and antioxidant defense system. Priming of aged seeds progressively restored the initial germinative ability and resulted in a marked decrease in the levels of MDA and conductivity of seed leachate. These effects of priming were also well recovered of GR and CAT activities in aged seed. The improved seed quality by priming treatment appears at least partly attributable to reduced lipid peroxidation, resulting from enhanced antioxidative enzyme activities that are suggesting the antioxidant defense systems play a key role in seed vigor.
