Antioxidant compound from the Lycopersicon esculentum (토마토(Lycopersicon esculentum)로부터 GSH 회복능을 보이는 항산화 화합물의 분리 동정)

  • The liquids of Lycopersicon esculentum were extracted with 70% aqueous MeOH and the concentrates were partitioned into EtOAc, n-BuOH, and H2O fractions. The repeated silica gel and octadecyl silica gel column chromatographies for the EtOAc fraction, whose activity was confirmed, led to isolation of one flavonol compound. The chemical structures of the compound were determined as quercetin (1) based on spectroscopic analyses including nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy. Through this study, the antioxidant efficacy was confirmed by demonstrating that the L. esculentum fraction showing an increase in glutathione mean (GM) and a decrease in glutathione heterogeneity (GH) uniformly raises the intracellular glutathione (GSH) level.

  • The fruits of tomato (Lycopersicon esculentum) were extracted with 70% aqueous methanol (MeOH) and the concentrates were partitioned into ethyl acetate (EtOAc), n-butanol (n-BuOH), and water (H2O) fractions. The repeated silica gel (SiO2) and octadecyl silica gel column chromatographies for the EtOAc fraction, whose activity was confirmed, led to isolation of one flavone compound. Nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy (MS) revealed the chemical structure of the isolated compound, 5,7,3'-trihydroxy-6,4',5'-trimethoxyflavone (1). LC-MS/MS analysis determined the content level of compounds 1 in the MeOH extract to be 4.02±0.12 ㎍/mg and in the TME-10 fraction to be 0.96±0.03 ㎍/mg. Through this study, the antioxidantive capacity was confirmed by demonstrating that the L. esculentum extract and their fractions showing an increase in glutathione mean and a decrease in glutathione heterogeneity uniformly raises the intracellular glutathione level.

전통 발효주로부터 glutathione 고함유 효모 Saccharomyces cerevisiae FF-8의 분리 동정 및 최적생산

  • 생체조직에 있어서 환원형 Glutathione(${\gamma}$-L-glutamyl-cysteinyl-glycine, GSH)은 항산화 작용, 간기능 회복 및 해독작용등의 생체 내에서 중요한 생리활성기능을 가지고 있다. 본 연구에서는 glutathione을 다량으로 함유하는 효모균주를 전통 발효주로부터 분리하여 최적 생산을 검토하였다. 분리된 균주는 형태학적, 생리학적 및 생화학적 특성을 검토한 결과 Saccharomyces cerevisiae로 동정되어 FF-8로 명명하였다. Glutathione 생산을 위한 Saccharomyces cerevisiae FF-8의 최적 생산조건은 YM(glucose 1.0%, acid peptone 0.5%, yeast extract 0.3%, malt extract 0.3%) 배지를 기본으로 하여 72시간 동안 진탕배양하면서 검토하였다. Glutathione 생산을 위한 최적 온도, 교반속도 및 pH는 각각 3$0^{\circ}C$, 100 rpm, 그리고 pH 6.0으로 나타났다. Glutathione 최대 생산배지조건은 탄소원으로 3%의 glucose, 질소원으로 3%의 yeast extract, 무기질원으로 0.06%의 KH$_2$PO$_4$및 전구체로 0.06%의 L-Cysteine으로 하였을 때 최대 생산량을 나타내었고, 최적 배지상에서 204$\mu\textrm{g}$/$m\ell$을 생산하였다.

Flavonoids from the arial parts of Artemisia agryi and their antioxidant capacity through GSH recovery effect (황해쑥(Artemisia agryi)으로부터 flavonoid 화합물들의 분리 동정과 세포 내 GSH 회복능을 통한 항산화 활성 평가)

  • The arial parts of Artemisia argyi were extracted with methanol : water (70:30), and the concentrates was partitioned into EtOAc (ethyl acetate), n-BuOH (normal butanol), and H2O (water) fractions. The repeated silica gel and ODS (octadecyl silica gel) column chromatographies for EtOAc and n-BuOH fractions led to isolation of four flavonoids without any ambiguity based on intensive interpretation of several spectroscopic data including nuclear magnetic resonance, and mass spectrometry. The chemical structure of the isolated compounds revealed to (2S)-naringenin (1), 3-methylkaempferol (2), 3,3'-dimethylquercetin (3), and 3,3',4'-trimethylquercetin (4). These four compounds were first isolated from A. argyi through this study. In this study, four compounds isolated from A. argyi showed an increase in glutathione mean and a decrease in glutathione heterogeneity so that the compounds uniformly raised the intracellular glutathione (GSH) level. Based on these results, it is considered that it can be used as a functional pharmacological material.

Studies on Screening and Comparison of Biological Activities fvom the Fruiting Body and Mycelium of Elfvingia applanata. (잔나비 걸상 버섯 자실체 및 균사체의 생리활성 탐색)

  • The biological activities of both ethanol and water extracts from the fruiting body of E. applanata and E. applanata mycelium and the three fractions of ethanol extracts from E. applanata were compared. 91% of MCF7 cell growth was inhibited by adding 0.5 g/l of water extracts of E. applanata and 81% of MCF7 cell growth was inhibited by adding 0.5 g/l of diethyl ether and chloroform fractions. It was also showed that above 60% of Hep3B cell growth was inhibited by adding all samples including the fractions. The ethanol extracts of E. applanata mycelium showed 33.3% of cytotoxicity on normal liver cell, WRL68 in adding 0.5 g/l of the samples and 40% in adding 0.5 g/l of chloroform fractions. The result of anti-mutagenicity of all extracts and fractions including ethanol extracts of Phelinus linteus were showed that diethyl ether fractions were most effective than any other samples. Hypoglycemic activities of diethyl ether and chloroform fractions were the most effective which scores were above 75%. The enhancement of glutathione-S-transferase activity was increased above 2.3 times by adding 1.0 g/l ethanol extracts of E. applanata and diethyl ether, chloroform fractions. It can be concluded that both biological activities of the fruiting body and mycelium of E. applanata were almost equivalent.

Effect of Fermented (Oenanthe stolonifera DC) Extract on the Activity of Enzymes Related to Liver Function of Alcohol-administered Rats and Mice (밭미나리 발효액이 알콜투여 흰쥐의 간기능관련효소활성에 미치는 영향)

  • It has long been recognized that dropwort contains specific funtional subtances for protecting human liver and preventing and curing its diseases, and thus it has been widely utilized in traditional folk remedy. In the present study. fresh of fermented extract of dropwort shoots grown on dryland and fresh extract of those grown on flooded fie1d were fed to the rats suffering from acute, subacute or chronical toxication induced by alcohol administration, and their affects were investigated. Administration of alcohol to rats and mice for 2 days at 5ml of 30% EtOH/kg/day raised total cholesterol and total glyceride which were, however, great1y supressed when alcohol was administered to the laboratory animals previously fed on fresh or fermented extract of dryland dropwort, or fresh extract of flooded field-grown dropwort for 20 days, without significant differences among the extracts. The levels of alanine aminotransferase and aspartate aminotransferase which were raised by alcohol adminstration were also lowered by feeding dropwort extract, among which that of fermented dryland-grown one was more effective than the other two. Chronic alcohol toxication was induced to rats by administering 10% alcohol for 10 months and fermented dropwort extract or tap water was fed to the rats for 5 days. The rats fed on fermented dropwort extract were lower in total cholesterol by 40% and in tota1 glyceride by 60% than the control. Alanine aminotransferase and aspartate aminotransferase in the rats fed on fermented dropwort extract were decreased by 87.2% and 91.7%, respectively, compared to the control, and the rats recovered almost to normal. Activity of alkaline phosphatase, catalase, superoxide dismutase or glutathione peroxidase changed greatly by alcohol administration in the rats suffering from chronic as well as acute toxication. The extract of fermented dry land dropwort significantly lowed the activity of those enzymes, especially, alkaline phosphatase and superoxide dismutase. The present results suggesting the possible medicinal effect of fermented dropwort extract to liver diseases.
