Effects of Glutamine, Glycine and Nucleosides/Nucleotide Mixture on Intestinal Mucosal Growth in Rats (흰쥐의 소장 점막 세포의 성장에 미치는 Glutamine, Glycine과 Nucleosides/Nucleotide 혼합물의 효과)

  • Total parenteral nutritional effect was induced by surgical creation of Thiry-Vella fistula(TVFs) in rats. Glutamine, glycine or nucleosides/nucleotide mixture in solution was injected into the loops for 2, 4, 6, 8 days. Control animals received a 0.9% saline solution. Results include weight gain, total protein, DNA, [$^3$H] thymidine incorporation into DNA, morphometry of the intestine in both TVFs and intestine in continuity. Perfusion of nucleosides/nucleotide mixture into the bypassed loops caused an increase in total protein, DNA content, villous height, villous surface area in loops. The injection of glycine into loops caused an increase in [$^3$H] thymidine incorporation but the mean values of the protein and DNA contents were not significantly different from those in group Cont and group Nuc. Overall values for group Gln were slightly higher than those of the control but the differences were not statistically significant. This study suggests that this animal model may be useful for studying the effect of dietary factors on intestinal growth and maturation, separating the direct effect of diet from systemic effect on the intestine.

Effect of Supplementation of Zinc, Glutamine, Fiber, and Prebiotics in Presumed Healthy Indonesian Children Aged 1-3 Years

  • Purpose: Impaired intestinal mucosal integrity may affect the gastrointestinal function, especially in relation to nutrition, absorption, and barrier function. The purpose of this study was to measure the prevalence of impaired intestinal mucosal integrity in presumed healthy children aged 1-3 years and assess the effects of zinc, glutamine, fiber, and prebiotic supplementation in them. Methods: A cross-sectional study was conducted in 200 children aged 1-3 years in Pasar Minggu, South Jakarta, Indonesia. A randomized double-blind parallel group method clinical trial was then performed to assess the effects of zinc, glutamine, fiber, and prebiotic supplementation. Results: Elevated calprotectin was found in 91/200 subjects (45.5%) at the onset of the study. After 10 months, 144 subjects completed the study: 72 subjects received the trial formula, whereas the other 72 received the standard formula. A transitory decrease in fecal calprotectin (FC) was observed after 6 months in the subgroup with normal FC levels, who were fed the test formula (p=0.012). Conclusion: The prevalence of impaired intestinal mucosal integrity in this group of Indonesian children aged 1-3 years was high. Supplementation with zinc, glutamine, fiber, and prebiotics during 6 months reduced FC only in those who had low levels at baseline but not in those with impaired integrity.

  • The regulation of 3 ammonia assimilatory enzymes GDH(glutamate dehydrogenase), GS(glutamine synthetase) and GOGAT (glutamate synthase), have been examined in C. glutamicum for the biosynthesis of glutamate and glutmine. The cell free extracts of 3 kinds of arg, his and trp auxotrophs were investigated the activities of -ketoglutarate dehydrogenase, GDH, GS, and GOGAT on the media cultured with nitrogen excess and limiting conditions. Trp and his howed higher level of glutamate and glutamine than that of parental strain. The inhibition of GS activities by ADP suggested that GS is regulated by energy charge in C. glutamicum. The results with his, trp, glyc, ala, ser, and GMP implied that a system of feedback inhibition were effective. Three enzyme biosynthesis is repressed by nitrogen sources such as trp, pro, glyc, ala, ser and tyrosine.

  • ABA type block copolymers composed of poly($\gamma-benzyl$ L-glutamate) (PBLG) as the A component and poly (ethylene glycol) as the B component were obtained by polymerization of $\gamma-benzyl$ L-gletamate N -carboxyanhydride, initiated by amino groups at both ends of poly(ethylene glycol) . From circular dichroism measurements in ethylene dichloride solution as well as from infrared spectTa measurements in solid state, it was found that the polypep- tide block exists in the a-helical conformation, as in PBLG homopolymer. $Poly-N^5$ (3-hydroxypropyl glutamine) (PHPG)/poly(ethylene glycol)block copolymer hydrogel was obtained by the treatment of PBLG/PBG block copolymer with the mixture of 3-ammine-1-propanol and diamlnooctane. The water content of PHPG/PEG block copolymer hydrogel was about 80wt% when the concentration of crosslinking agent was below 5 mole % per polymer.

Glutamine synthetase (GS), a glial-specific enzyme. catalyzes the synthesis of glutamine from glutamate and ammonia and is associated with ischemic injury and neurological diseases. Preparation and Stability Measurement of Liposome-amino Acid Conjugates (리포솜-아미노산 결합체의 제조와 안정성 측정)

  • Liposome-amino acid conjugates were prepared using phopholipid (dipalmitoylphosphatidylcholine (DPPC) or distearoylph-osphatidylcholine(DSPC)) and hydrophobically modified amino acids (glutamic acid(glu), glutamine(gln) or asparagine(asn)). The size of liposomes was about 100 nm. According to the glucose-induced turbidity changes, liposomes composed of DPPC and glutamic acid have higher glucose binding affinity than liposomes of DPPC-glutamine or DPPC-asparagine. Also, the liposomes were more stable in terms of aggregation or fusion than the others (DPPC-glutamine, DPPC-asparagine and DSPC-amino acids). As a rdsult, stable liposomes with an affinity for glucose could be prepared with DPPC and glutamic acid.

  • The effect of ${\delta}-aminolevulinic\;acid$ (ALA) biosynthetic precursors and related compounds on the ALA productivity from a strain of Rhodobacter sphaeroides has been examined in vivo and in vitro systems. The relative ratios of ALA productivities by $C_{4}$- pathway to that by $C_{5}$-pathway in vivo and in vitro systems were 0.78 and 1.37, respectively. Although the expression rates of $C_{4}-$ and $C_{5}-pathways$ in cell-free systems prepared after precursors supplemented cultivations were increased 1.35 and 1.52 folds, respectively, the rate increase of $C_{4}-pathway$ was accompanied by the rate decrease of the $C_{5}-pathways$, and vice versa, as that the rates of both $C_{4}-$ and $C_{5}-pathways$ were lowered to be 0.91, 0.83, respectively. The order of cellular uptake rates of ${\gamma}-glutamyl$ derivatives relative to that found with L-glutamic acid were shown to be D-glutamic acid, 0.55: D-glutamine, 0.5: L-glutamine, 0.4: ${\gamma}-L-glutamyl$ ethylester, 0.3: GSH and Glu-pNA, 0. L and D configurations of glutamine were indicated as better substrates in vivo for ALA yields than those of glutamic acid, respectively.

Effects of Dietary Supplementation of Taurine, Carnitine or Glutamine on Endurance Exercise Performance and Fatigue Parameters in Athletes (타우린, 카르니틴 또는 글루타민 섭취가 운동선수의 지구력운동 수행능력 및 혈중 피로요소에 미치는 영향)

  • The effects of taurine, carnitine or glutamine supplementation on endurance exercise performance along with related fatigue factors were evaluated in male college students in the Department of Physical Education, who's maximal oxygen consumption rates (VO$_2$max) were equivalent to those of endurance athletes. Twenty four subjects were randomly divided into 4 groups (n=6), and given placebo, taurine (4 g/day), carnitine (4 g/day), or glutamine (4 g/day) tablets for 2 weeks. Subjects could run 6.9 min or 9.0 min longer until exhausted on a treadmill at the intensity of 75% VO$_2$max following taurine or camitine supplementation for 2 weeks, respectively, compared to the value measured prior to each supplementation. Glutamine or placebo supplementation did not improve the endurance exercise performance based on the running time until exhausted on a treadmill. Serum lactate concentrations measured 1 hr after the initiation of the endurance exercise, as well as at all-out state tended to be decreased by taurine, carnitine, or glutamine supplementation, and were significantly lowered (43% decrease) by carnitine supplementation (p < 0.05). Taurine supplementation significantly reduced the serum inorganic phosphorus concentration measured at all-out state (14% decrease, p < 0.05), while carnitine supplementation significantly lowered the resting state serum inorganic phosphorus level (20% decrease, p < 0.05). Taurine (32% reduction) or carnitine (23% reduction) supplementation significantly decreased serum ammonia concentration measured at all-out state (p < 0.05). From these results, 4 g/day of taurine or carnitine supplementation appears to improve the endurance exercise performance and related human fatigue factors.

Effect of Amino Acid, Polyamine, and Flavonoid on the Pollen Germination of Peach(Prunus persica SIEB under Low Temperature Conditions (아미노산, Polyamine 및 flavonoid 첨가가 복숭아 화분의 저온 발아에 미치는 영향)

  • The objective of this study was focused on the enhancement of pollen germination frequency in peach (Prunus $persica\;_{SIEB}$) under low temperature conditions. The effect of factors such as amino acid, polyamine, and flavonoid on the pollen germination was investigated, and the results are summarized as follows. When amino acid, polyamine or flavonoid was added to the germination medium at $10^{\circ}C$, pollen germination frequency was strongly promoted. Optimum concentration of each supplement for pollen germination enhancement was $100\;mg{\cdot}L^{-1}\;H_3BO_3$, 10 mM asparagine, 10 mM glutamine, 100 mM spermine, $1000\;{\mu}M$ putrescine, and $1.0\;{\mu}M$ kaemferol. The best combination of factors in pollen germination was $100\;mg{\cdot}L^{-1}\;H_3BO_3+10\;mM$ asparagine, followed by $100\;mg{\cdot}L^{-1}\;H_3BO_310mM$ glutamine, $100\;mg{\cdot}L^{-1}\;H_3BO_3+200mM$ spermine, and 10 mM asparagine. These combinations promoted pollen germination by 18% in 'Nagasawa-Hakuho', and 19% in 'Shuho' compared to their germination percentage on the basal medium.

  • The extracts of toha, a kind of freshwater shrimp (Caridina denticulata denticulata) being cultivated in Naju district, Chonnam of Korea and jeotsaeu, a kind of seawater shrimp (Acetes japonicus) being caught in Shinan district, Chonnam of Korea, were analyzed for the taste constituents-extractive nitrogen, free amino acids, oligopeptides, nucleotides and related compounds, quaternary ammonium bases, and guanidino compounds-using specimens collected from December in 1994 to November in 1995. The content of extractive nitrogen. free amino acids, oligopeptides, nucleotides and related compounds in toha extract was higher than that in jeotsaeu extract. On the contrary, the amount of homarine, trimethylamine oxide and trimethlamine in toha extract was lower than that in jeotsaeu extract. But in case of creatine and creatine both species showed almost the same level. Arginine, lysine, glutamine, glutamic acid, phenylalanine, leucine, alanine, asparagine and glycine were the major amino acids in toha extract, while arginine, glutamic acid, proline, glutamine, lysine, leucine, glycine, alanine and taurine were the major amino acids in jeotsaeu extract. Methionine, glutamine, valine were almost the same level between the two species. As for nucleotides and related compound, AMP was the principal constituent in toha extract, and inosine was the principal constituent in jeotsaeu extract. In conclusion, the content of major nitrogenous constituents in toha extract was more abundant than that in jeotsaeu extract. So toha was more excellent raw material than jeotsaeu.
