To improve the growth of mouse hybridoma 2c3.1 secreting anti-Hepatitis B surface antigen monoclonal antibody (anti-HBsAg MAb), we had constructed an enriched medium and observed the effects of fetal bovine serum and serum-free supplements including human serum albumin, 'insulin and transferrin', and monoethanolamine. For further enhancement of growth, the concentrations of two major energy sources, glucose and glutamine, were strengthened with various ratios in the enriched medium. Maximum cell growth and monoclonal antibody production obtained in various ratios of glucose/glutamine with an inoculation concentration of 2$\times$105 cells/ml were 0.73$\times$106-4.62$\times$106 cells/ml and 65.1-422.6 $\mu\textrm{g}$/ml, respectively. Glutamine was round to be a major energy source and a limiting nutrient in comparison to glucose for 2c3.1 cell cultivation in enriched media with low serum.
The excretion of ammonia and glutamine synthetase activities were measured in aposymbiotic Paramecium and symbiotic Paramecium. The uptake of nitrate and ammonia, and specific enzyme activities of nitrate reductase, glutamate dehydrogenase and glutamine synthetase were investigated in symbiotic Chlorella from Paramecium bursaria and Chlorella ellipsoidea. The ammonia concentration in the culture media of aposymbiotic Paramecium was increased according to the growth of the Paramecium but it was not changed in symbiotic Paramecium. Nitrate, the major nitrogen source, was taken up at a rate of 0.635 nmol/ 106 Chlorella/hr in Chlorella ellipsoidea. Most of ammonia was assimilated to glutamine by glutamine synthetase, of which acitivty was 1,467 $\mu$mol/mg protein/min in Chlorella elliposidea. Contrary to Chlorella ellipsoidea, ammonia and glutamine transported from the Paramecium were the nitrogen source of symbiotic Chlorella and ammonia was taken up at a rate of 3.854 nmo./106 Chlorella/hr into synmbiotic Chlorella. Most of ammonia were assimilated to glutamate by glutamate dehydrogenase in symbiotic Chlorella. The glutamate dehydrogenase (GDH/NADH) activity was 0.851 $\mu$mol/mg protein/min.

Herein we report multiple ion mobility (IM) peaks in electrospray ionization IM mass spectrometry (ESI-IM-MS) produced by glutamine residue in peptide. The mobility features of 147 peptides were investigated using ESI-IM-MS combined with liquid chromatography. Of these peptides, 66 presented multiple IM peaks, and analysis of their sequence using collision induced dissociation (CID) revealed that glutamine (Gln), as well as proline (Pro), plays a critical role in generating multiple IM peaks. Mutant-based investigations using Gln-containing peptides indicate that the side chain of Gln promotes intermolecular interactions, inducing multiple structures of the peptide ions in the gas phase. Consequently, the present study demonstrates that the distinct ion mobility signatures identified herein can potentially be used to characterize glutamine-containing peptide ions.
Batch suspension cultures of hybridoma cell were performed with various initial glutamine concentrations to investigate the effects of glutamine on cell growth and death, monoclonal antibody production, glucose and glutamine consumption, and the production of lactate and ammonium ion. An mathematical kinetic model was formulated to describe the kinetics of cell growth, the consumption of nutrients (glucose and glutamine), and the production of monoclonal antibody and waste metabolites (lactate and ammonium ion) based on experimental data. An equation for the specific growth rate was developed such that superimposed Monod equation in glucose and glutamine, with non-competitive type inhibition relations in ammonium ion and lactate. The inhibition constant for lactate was inversely proportional to the lactate concentration. The specific death rate was considered to be a function of glucose, glutamine, ammonium ion and lactate concentration.

Background: It is known to reduce the mortality when glutamine is supplied to patients during the surgery or in intensive care unit through intravenous nutrition supply. The purpose of this study is to establish the appropriate basis for use of glutamine and guidelines of nutrition supply for critically ill patients in the hospital by examining the clinical effects of administration of glutamine with subjects of elderly critically ill patients receiving intravenous nutrition in one hospital in Korea. Method: Among elderly patients with age of 60 or more hospitalized in Yeuido St. Mary's Hospital from August 2012 to July 2013, those who stayed in the intensive care unit for more than a week and received TPN (Total Parenteral Nutrition) for more than 3 days during staying in the intensive care unit were classified to a test group using glutamine and a control group without glutamine. Duration of use of mechanical ventilator, duration of hospitalization, occurrence of infectious disease and death were compared between two groups. We would like to identify the clinical test figures affected by the use of glutamine by examining changes in SCr, Total Protein, Albumin, AST, ALT, TB, DB and GFR at the time of admission and discharge. Results: At the time of admission to intensive care unit, gender, physical measurement information and clinical test figures did not show any significant difference between 72 subjects in a test group and 24 subjects in a control group. Thus, two groups began in the same condition. There were no significant difference in duration of hospitalization, duration of intensive care unit, use of mechanical ventilator, occurrence of infectious disease and death. As the results of statistical analysis of the average changes of clinical test figures at the time of admission and discharge of intensive care unit, SCr and GFR were significantly changed in the test group. GFR was significantly changed in a control group. As the result of analysis of the clinical test figures at the time of discharge with reflection of average changes after clinical test figures were corrected at the time of admission of intensive care unit, TB and GFR were significantly increased in a test group compared with those in a control group. Other clinical test figures were not significantly changed. Conclusion: If glutamine is administered to critically ill patients over age 60 receiving TPN and careful monitoring for total bilirubin is made in the future, it is expected to give the positive effect on renal function andminimize the side effect of arise in total bilirubin.
This work was conducted to establish an efficient plant regeneration system for genetic transformation and the in vitro conservation of Sedum sarmentosum genetic resources. Effects of glutamine and $AgNO_3$ on plant regeneration between two genotypes were investigated using MS media supplemented with 0.2 mg/L NM and 3.0 mg/L BA. Calluses were formed on leaf explants placed on MS solid media supplemented with 3 mg/L 2,4-D and 1 mg/L BA. Calluses of Keumsan local strain produced shoots at a frequency of up to 100% after 50 days of culture on medium supplemented with glutamine. The highest number of shoots per callus was 17.6 at 350 mg/L glutamine. However, calluses of Wanju local strain gave rise to no shoots under the same culture conditions. Likewise, calluses of Keumsan local strain produced shoots at a frequency of up to 100% after 50 days of culture on medium supplemented with $AgNO_3$ whereas Wanju local strain sporadically produced shoots. The highest number of shoots per callus of Keumsan local stain was 16.1 at $15{\mu}M$$AgNO_3$. Regenerated shoots were subcultured on hormone-free MS medium for rooting and shoot growth, and then 3-5 cm high plantlets were transplanted to the artificial soils comprising vermiculite and perlite, where they survived at a frequency of 88-100%. After being transplanted into upland soil:sand (1:1, v/v) in a greenhouse, regenerated plants showed a morphologically normal growth.
Attempts were made if diol and triol fractions of ginseng saponin affect on glutamine transport into rat renal cortical mitochondria, swelling, phosphate dependent glutaminase activity, and consumption of oxygen. The following results were obtained. When mitochondrial preparation from rat renal cortex was incubated in medium containing 14C-glutamine and either triol or diol fractions, radioactivity was shown to increase at both 10-6% and 10-5% triol fractions of ginseng saponin, but reduce in case of diol fraction. The remarkable acceleration of the rate of swelling of renal cortical mitochondria was observed in the presence of 10-1% trios and diol fractions but no accerelation at lower concentrations. The activity of phosphate dependent glutaminase from renal cortical mitochondria was slightly activated at 10-2% of triol fraction. However, there was no effect in case of diol fraction. Oxygen consumption by mitochondria from renal cortex was remarkably increased at concentrations of 10-5% and 10-6% triol fractions, but reduced in the case of diol fractions. On the basis of these observations it was concluded that triol fraction of ginseng saponin might increase the transport of glutamine into mitochondria by accelerating the respiratory chain and supplying additional energy to mitochondria, and physiological role of triol fraction was entirely different from that of diol fraction of ginseng saponin.
Seventy-four piglets were used to investigate the effects of dietary glutamine (Gln) and glutamate (Glu) on the mucosal structure and active absorption of small intestinal, DNA and RNA concentrations of skeletal muscle tissue in piglets during d 28 to 42 of age. Postweaning piglets were fed for 14 d corn- and soybean meal-based diets supplemented with 0.0 or 1.0% L-Gln or L-Glu. On d 7 and 14 postweaning, pigs' small intestinal sections and longissimus dorsi were collected, at the same time, the D-xylose absorption test was conducted. The results suggested that in comparison to control piglets, jejunal atrophy during the first week postweaning was prevented by the glutamine and glutamate supplementation (1%) and the capability of small intestine to absorb Dxylose was improved. Furthermore the RNA concentration in skeletal muscle tissue was increased. These results provide an experimental basis for use of glutamine and glutamate on alleviating the weaning stresses and improving piglets' growth performance.

Nitrogen (N) is an important regulator of the expression of genes involved in carbon and N assimilation pathways in plants by selectively altering the levels of proteins and/or mRNAs. These in C4 plants include genes for such as phosphoenolpyruvate carboxylase, carbonic anhydrase, and pyruvate-Pi dikinase. The C4 genes are regulated in mesophyll cells by N availability both transcriptionally and posttranscriptionally through cytokinins and glutamine as signals. The level of both the signals is up-regulated by N availability: cytokinins in roots and glutamine in leaves. The level of glutamine is controlled by the differential expression by N of glutamine synthetase and ferrdoxin-dependent glutamate synthase genes which locate in the mesophyll cells of C4 plants. The results is discussed as molecular mechanism for the greater N use efficiency of the plants as well as N partitioning is the photosynthetic cells.
