Ceramide metabolism is known to be an essential etiology for various diseases, such as atopic dermatitis and Gaucher disease. Glucosylceramide synthase (GCS) is a key enzyme for the synthesis of glucosylceramide (GlcCer), which is a main ceramide metabolism pathway in mammalian cells. In this article, we developed a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to determine GCS activity using synthetic non-natural sphingolipid C8-ceramide as a substrate. The reaction products, C8-GlcCer for GCS, could be separated on a C18 column by reverse-phase high-performance liquid chromatography (HPLC). Quantification was conducted using the multiple reaction monitoring (MRM) mode to monitor the precursor-to-product ion transitions of m/z $588.6{\rightarrow}264.4$ for C8-GlcCer at positive ionization mode. The calibration curve was established over the range of 0.625-160 ng/mL, and the correlation coefficient was larger than 0.999. This method was successfully applied to detect GCS in the human hepatocellular carcinoma cell line (HepG2 cells) and mouse peripheral blood mononuclear cells. We also evaluated the inhibition degree of a known GCS inhibitor 1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP) on GCS enzymatic activity and proved that this method could be successfully applied to GCS inhibitor screening of preventive and therapeutic drugs for ceramide metabolism diseases, such as atopic dermatitis and Gaucher disease.
Gaucher disease (GD)는 glucocerebrosidase 유전자(GBA)의 돌연변이에 의하여 발병하는 전세계적으로 가장 유병율이 높은 리소좀 축적질환이다. GD는 신경학적인 증상의 유무에 따라 3가지 임상형으로 구분된다. 신경병증 GD인 2형과 3형의 경우는 대뇌에서 glucosylceramide (GlcCer)와 glucosylsphingosine (GlcSph)의 농도가 증가하면서 신경세포의 심각한 손실이 야기되는 특징을 보인다. 신경교종에서 유래한 H4 세포를 GD에서 증가하는 기질인 GluCer와 GlcSph를 첨가하여 배양하였을 때, 심각한 DNA손상과 더불어 세포의 사멸이 야기되는 것과 이러한 신경세포의 사멸은 GluCer 보다는 GlcSph을 처리하였을 때 더 현저하게 증가하는 것을 관찰하였다. H4 세포에 히스톤 탈아세틸화 효소(HDAC) 6의 저해제인 tubacin과 GlcSph을 함께 처리하였을 경우에는 DNA손상은 물론 GlcSph에 의하여 유도된 세포사멸과 관련된 단백질 인자들의 발현이 모두 감소되었다. 본 연구를 통해 GlcSph이 세포사멸을 통하여 신경병증 GD의 발병에 주요한 역할을 한다는 것을 알 수 있었고, HDAC6 저해제가 신경병증 GD 환자를 위한 치료제 후보물질로 제시될 수 있는 가능성을 확인하였다.
본 연구는 PFEG(글루코실세라마이드 4.0% 함유)가 저 미네랄 특수사료(HR-AD)를 섭취시킨 헤어리스 마우스에서 발생하는 피부장벽 붕괴에 대한 억제작용이 있는지를 확인하고자 하였다. 4주간의 투여를 통해 정상군(Rabo MR Stock)과 비교하여 대조군(HR-AD)에서는 경피 수분 손실량(TEWL)이 서서히 상승하여 피부장벽기능의 붕괴가 발생함과 동시에 표피 수분량 및 각층 수분량도 서서히 저하되는 것이 확인되었다. 육안적으로는 깊은 주름 형성이 확인되었으며 조직학적 소견에서는 표피의 비후와 각화 항진이 확인되었다. 그러나 PFEG를 섭취시킨 시험군에서는 대조군에서 유발되는 피부장벽기능 붕괴에 대한 억제 효과가 확인되었다. 즉, 글루코실세라마이드 0.01% 배합군 및 0.1% 배합군에서는 섭취 시작 2주일째 이후 야기되는 TEWL 상승을 완전히 억제하였다. 또한, 저하되는 표피 수분량, 각층 수분량에 대해서도 유의하게 억제하여 피부 보습 기능을 유지하고 있었다. 육안적으로도 깊은 주름 형성은 억제되었으며, 조직학적인 검토에서도 표피의 비후나 각화 항진은 확인되지 않았다. 이러한 결과는 HR-1 마우스의 피부 세라마이드 생산 개선에 기인한 결과라고 예상된다. 즉, HR-AD 섭취에 의해 HR-1 마우스는 세라마이드를 포함한 피부 지질의 함량 및 구성에 이상이 생기고, 피부 수분량이 급격히 줄어들어 피부 보습 능력이 저하된다. 그런데 PFEG의 혼합 투여는 손상된 피부의 세라마이드 함량 및 구성을 정상과 가깝게 만들어, 대조군에서 나타나는 증상의 발현을 억제하는 것으로 판단된다. 이상의 결과로부터 PFEG의 섭취는 피부장벽기능의 손상으로 발생하는 다양한 피부 수분 손실을 억제함으로써 피부 보습 개선에 우수한 효능이 있음을 알 수 있었다. 따라서 PFEG가 피부 보습에 도움을 주는 건강기능성 원료로서 이용 가능성이 높음을 확인할 수 있다.
Glycosphingolipids are structural components of mammalian cell membranes and are involved in essential cellular physiology such as cell-cell interaction, recognition, transmembrane signaling, proliferation and cell death. In this study, the simple quantitative method of monoglycoceramides-containing glucosylceramide and galactosylceramide was developed. The glycosylceramides extracted from culture cells and rat plasma were resolved by TLC, deacylated by SCDase and analyzed by HPLC-fluorescence detector at an excitation wavelength of 340 nm and an emission wavelength of 455 nm. Limit of detection was approximately 0.1 pmol and limit of quantification was about 1 pmol for both monoglycoceramide standards. The recoveries of standard glucosylceramides from intra- and inter-day assays were 113.8 and 88.8% and those of galactosylceramides were 110.7 and 123.9%, respectively. The monoglycoceramide contents of SW-620 cells and rat plasma were $141.5{\pm}5$ pmol/$1{\times}10^6$ cells and $3.9{\pm}0.3{\mu}M$, respectively. The present analytical method provides a reproducible quantification and total content of monoglycoceramide which may be as a potential biomarker for lipid imbalance-related human diseases.

Recently, several reports suggest that ceramide formation has been implicated in the apoptosis signaling in response to chemotherapeutic agents. In this study, to enhance paclitaxel-mediated cytotoxicity and endogenous ceramide levels, we blocked ceramide metabolism using an inhibitor of glucosylceramide synthase, l-phenyl-2- dacanoylamino-3-morpholino-l-propanol(PDMP) and SM synthase, D609. Exposure of human breast cancer cells to paclitaxel accumulated de novo ceramide synthesis by enhancement of SPT activity 1.2-fold, whereas ceramide synthase activity was not altered. (omitted)
신령버섯(Agaricus blazei Murill)은 항암, 면역력 개선, 항비만 등 다양한 효능이 알려져 있으며, 피부 효능으로는 항산화, 항염, 미백 등에 대하여 보고되었다. 이러한 효능을 뒷받침하기 위하여, 신령버섯의 성분 연구가 진행되었다. 따라서 본 연구에서는 신령버섯 추출물의 피부 효능을 재확인하고 유용한 효능성분을 밝혀내어 이를 화장품 소재로서 이용하고자 하였다. 신령버섯 추출물은 100 ㎍/mL에서 멜라닌 합성 저해, 콜라겐 합성 증진, 보습과 관련된 유전자(hyaluronan synthase-2, 3와 aquaporin-3) 발현 증가를 나타내었다. 이들 화합물은 분광학적 데이터와 문헌값을 비교하여 ergosterol (1), 5-dihydroergosterol (2), cerevisterol (3), cerebroside B (4), cerebroside D (5), adenosine (6), 및 benzoic acid (7)로 동정하였다. 이들 중 비교적 효능이 알려지지 않은 3종의 sterol (1-3)과 2종의 glucosylceramide (4, 5)에 대하여 피부효능을 평가하였으며, 그 결과 5-dihydroergosterol (2)이 마우스 흑색종 세포에서 멜라닌 생성을 억제하였으며, 또한 인간 진피 섬유아세포 세포에서 콜라겐 생합성 촉진하였다. 그리고 cerevisterol (3), cerebroside B (4), cerebroside D (5)는 마우스 대식세포에서 NO 생성을 억제하였으며, 특히 cerebroside D (5)는 인간각질형성세포에서 hyaluronan synthase-2와 aquaporin-3 유전자 발현을 증가시켰다. 따라서 신령버섯 추출물과 분리 화합물은 화장품 소재로 활용 가능 할 것으로 생각된다.

Gaucher disease (GD, OMIM #230800 OMIM#230800) is a rare, autosomal recessive inherited metabolic disorder caused by mutation in GBA1 encoding the lysosomal enzyme, glucocerebrosidase. The deficiency of glucocerebrosidase leads to an accumulation of its substrate, glucosylceramide in macrophages of various tissues. Common clinical manifestations include cytopenia, splenomegaly, hepatomegaly, and bone lesions. The phenotype of GD is classified into three clinical categories: Type 1 (non-neuronopathic) is characterized by involvements on the viscera, whereas types 2 and 3 (neuronopathic) are associated with not only visceral symptoms but also neurological impairment, either severe in type 2 or variable in type 3. A diagnosis of GD can be confirmed by demonstrating the deficiency of acid glucocerebrosidase activity in leukocytes. Mutations in the GBA1 should be identified as they may be of prognostic value in some cases. Biomarkers including Chitotriosidase, CCL18, and glucosylsphingosine (lyso-GL1) are useful in diagnosis and treatment monitoring. Currently available disease-specific treatment in Korea consists of intravenous enzyme replacement therapy and substrate reduction therapy. For enhancing long-term prognosis, the onset of Parkinson's disease and Lewy body dementia, or the occurrence of a blood disease or cancer (hepatocellular carcinoma) should be monitored in older patients. The development of new strategies that can modify the neurological phenotype are expected, especially in Asia including Korea, where the prevalence of neuronopathic GD is relatively higher than that in western countries.
Here we have investigated how lactosylceramide (LacCer) modulates gene expression of adhesion molecules in TNF-${\alpha}$ and IFN ${\gamma}$ (CM)-stimulated astrocytes. We have observed that stimulation of astrocytes with CM increased the gene expression of ICAM-1 and VCAM-1. D-Threo-1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP) and N-butyldeoxynojirimycin (NBDNJ), inhibitors of glucosylceramide synthase (GLS) and LacCer synthase (galactosyltransferase, GalT-2), inhibited the gene expression of ICAM-1 and VCAM-1 and activation of their gene promoter induced by CM, which were reversed by exogenously supplied LacCer. Silencing of GalT-2 gene using its antisense oligonucleotides also attenuated CM-induced ICAM-1 and VCAM-1 expression, which were reversed by LacCer. PDMP treatment and silencing of GalT-2 gene significantly reduced CM-induced luciferase activities in NF-${\kappa}B$, AP-1, GAS, and STAT-3 luciferase vectors-transfected cells. In addition, LacCer reversed the inhibition of NF-${\kappa}B$ and STAT-1 luciferase activities by PDMP. Taken together, our results suggest that LacCer may play a crucial role in the expression of ICAM-1 and VCAM-1 via modulating transcription factors, such as NF-${\kappa}B$, AP-1, STAT-1, and STAT-3 in CM-stimulated astrocytes.
포스파티딜세린(Phosphatidylserine; PS)은 생체막에서 구조적인 역할을 담당하는 인지질로서, 생체 내 다양한 세포작용에 필수적인 신호전달 효소의 보조인자로서 작용하는 것으로 알려져 있다. 하지만, PS의 생리활성에 대한 연구는 거의 이루어지지 않았고, 특히 피부에서의 생리활성에 대한 연구는 전무한 실정이다. 본 연구에서는 무모생쥐의 피부에 tape-stripping으로 경표피수분손실(TEWL)의 증가를 유도한 후, PS를 도포함으로써 그 손실을 현저히 감소시켰다. 또한, PS 도포군의 피부에서 세라마이드 함량이 증가된 사실을 확인한 바 있다. PS 도포군에서 non-hydroxyl 세라마이드와 glucosyl 세라마이드의 함량이 비처리군과 비교하여 각각 1.4배와 1.6배로 증가하였다. PS는 또한 피부각질세포의 분화를 촉진하였다. 피부각질세포에 PS를 처리함으로써 세포 형태가 분화상을 띄고 있음을 현미경 상에서 확인하였고, 표피분화의 특이적 표지 단백질인 Involucrin (INV)과 Transglutaminase 1 (TG'ase 1)의 발현이 각각 3.5배와 3배로 현저히 증가하였음을 웨스턴 블랏을 통하여 확인하였다. 또한 무모생쥐 피부에 PS를 도포한 결과 INV와 loricrin 단백질 발현이 증가하였다. 본 연구는 PS가 피부에서 생리활성을 나타낸다는 최초의 증거를 제시하며, 구체적으로는 각질세포 분화를 촉진함으로써 피부 세라마이드 함량을 증가시키고 경표피 수분손실을 감소시켜 궁극적으로 피부장벽을 강화하는 작용을 한다는 것을 보여준다.
본 연구에서는 기니피그에 8주간 필수지방산 결핍 식이 공급에 의해 표피 과증식을 유도한 후 계속된 2주간의 보라지유 또는 홍화유 식이 공급에 의한 과증식 억제 및 Cer, GlcCer, SM의 표피 함량 및 세라마이드 대사 관련 효소들의 단백질 발현 변화를 정상대조군인 BO군 및 필수지방산 결핍 군인 HCO군과 비교하였으며 그 결과는 다음과 같다. 1. 8주간의 필수지방산 결핍 식이 공급 후 계속된 2주간의 보라지유 식이 공급 (HCO + BO군)은 2주간의 홍화유 식이 공급에 (HCO + SO군) 비해 더욱 현저히 표피 과증식을 억제하였다. 2. 10주간 필수지방산 결핍식이를 공급한 HCO군은 정상대조군인 BO군에 비해 Cer의 총 함량 및 Cer1/2/5/6/7, GlcCer-A/B의 함량이 유의적으로 감소되었다. HCO + BO군의 Cer과 GlcCer의 총 함량 및 Cer1/2, GlcCer-A/B, SM1의 함량은 HCO군에 비해 유의적으로 증가한 반면 HCO + SO군의 이들 함량은 HCO군과 유사하였다. GlcCer-C/D, SM의 총 함량 및 SM 2/3의 함량은 군간 변화가 없었다. 3. HCO군에서는 GCase 발현이 감소한 반면 HCO + BO군에서는 현저히 증가하였다. HCO + SO군의 aSMase의 단백질 발현이 HCO군에 비해 유의적으로 증가하였으나 aCDase의 발현 또한 다른 군에 비해 현저히 증가하였다. HCO + BO군과 HCO + SO군 모두 SPT의 단백질 발현이 HCO군에 비해 유의적으로 증가하였다. 결론적으로 표피 과증식이 유도된 기니피그에 보라지유 식이 공급은 GlcCer-A/B을 포함하는 GlcCer 총 함량, SM1 및 GCase의 발현 증가와 더불어 Cer1/2를 포함하는 Cer 총 함량을 증가시켜 궁극적으로 표피 과증식을 현저히 억제하였다.
