Properties of Cellulase Produced from Cellulomonas sp. YE-5 (Cellulomonas sp. YE-5가 생산하는 Cellulase의 특성)

  • Enzymatic properties of avicelase, carboxymethyl cellulase (CMCase) and P-glucosidase produced by Cellulomonas sp. YE-5 were studied. Optimal temperature and pH of avicelase were 40t and 6.0, and those of CMCase and P-glucosidase were $45^{\circ}C$ and 6.5. Avicelase and CMCase were stable between pH 5.0 and 9.5, and &glucosidase was stable between pH 5.5 and 8.0. Avicelase and P-glucosidase were inactivated when incubated at $35^{\circ}C$ for 6 hrs, and CMCase was at $40^{\circ}C$ for 6 hrs. All cellulases were strongly inhibited by $Cu^{2+} \; and \; Zn^{2+}. K_m$ values of avicelase for avicel, CMCase I and CMCase II for CM-cellulose, and ($\beta$-glucosidase for p-nitrophenyl-$\beta$-D-glucoside (PNPG) were 4.76, 16.4, 16.4 $\mu g$/ml and 3.51 mM, respectively.

Quantitative Determination of Five Phenolic Peroxynitrite-scavengers in Nine Korean Native Compositae herbs

  • Peroxynitrite (ONOO)-scavenging activities of nine Compositae herbs consisting of three Ixeris, two Youngia, two Cirsium and one of each Lactuca and Taraxacum species were evaluated. The contents of their ONOO scavengers in the extracts were also determined on a HPLC using seven standard compounds, chlorogenic acid (CGA), chicoric acid (CA), luteolin 7-glucoside (luteolin-7-glc), luteolin 7-glucuronide (luteolin-7-glcU), luteolin, linarin and pectolinarin. Five of those compounds exhibited potent ONOO-scavenging activities: IC50, CA (0.76 μM), CGA (1.34 μM), luteolin (0.81 μM), luteolin-7-glc (0.86 μM) and luteolin-7-glcU (3.13 μM). Both CA and luteolin-7-glc were highly contained in I. dentata (19.71 mg/g and 13.58 mg/g, respectively), I. dentata var. albiflora (17.58 mg/g and 23.83 mg/g, respectively) and I. sonchifolia (65.71 mg/g and 6.99 mg/g, respectively). Among the nine herbs, those three Ixeris species had very low IC50 values over the range of 0.48 - 1.74 μg/mL, suggesting that they could be potential therapeutic vegetables, particularly for preventing diabetic complications or obesity, which can be caused by an excess production of ONOO.

Rpi-blb2 Gene-Mediated Late Blight Resistance in Plants

  • Phytophthora infestans is the causal agent of potato and tomato late blight, one of the most devastating plant diseases. P. infestans secretes effector proteins that are both modulators and targets of host plant immunity. Among these are the so-called RXLR effectors that function inside plant cells and are characterized by a conserved motif following the N-terminal signal peptide. In contrast, the effector activity is encoded by the C terminal region that follows the RXLR domain. Recently, I performed in planta functional profiling of different RXLR effector alleles. These genes were amplified from a variety of P. infestans isolates and cloned into a Potato virus X (PVX) vector for transient in planta expression. I assayed for R-gene specific induction of hypersensitive cell death. The findings included the discovery of new effector with avirulence activity towards the Solanum bulbocastanum Rpi-blb2 resistance gene. The Rpi-blb2 encodes a protein with a putative CC-NBS-LRR (a coiled-coil-nucleotide binding site and leucine-rich repeat) motif that confers Phytophthora late blight disease resistance. We examined the components required for Rpi-blb2-mediated resistance to P. infestans in Nicotiana benthamiana. Virus-induced gene silencing was used to repress candidate genes in N. benthamiana and to assay against P. infestans infections. NbSGT1 was required for disease resistance to P. infestans and hypersensitive responses (HRs) triggered by co-expression of AVRblb2 and Rpi-blb2 in N. benthamiana. RAR1 and HSP90 did not affect disease resistance or HRs in Rpi-blb2-transgenic plants. To elucidate the role of salicylic acid (SA) in Rpi-blb2-mediated resistance, we analyzed the response of NahG-transgenic plants following P. infestans infection. The increased susceptibility of Rpi-blb2-transgenic plants in the NahG background correlated with reduced SA and SA glucoside levels. Furthermore, Rpi-blb2-mediated HR cell death was associated with $H_2O_2$, but not SA, accumulation. SA affects basal defense and Rpi-blb2-mediated resistance against P. infestans. These findings provide evidence about the roles of SGT1 and SA signaling in Rpi-blb2-mediated resistance against P. infestans.

RETINOL STABILIZATION BY PSEUDO-LIPOSOME AND LAMELLAR LIQUID CRYSTAL

  • It is well known that all-trans-retinol is not only very unstable in heat, light, air, and water, but also skin-irritant despite a good anti-wrinkle effect. Therefore, it is very difficult to stabilize retinol and make the safe retinol containing cosmetics by using a certain concentration of retinol with real effect. In order to dissolve these problems and apply retinol for skin care cream, firstly retinol is to be encapsulated in the vesicle called Liposphere (pseudo-liposome) which is made by homogenizing under high pressure the mixtures of lecithin, retinol, caprylic/capric triglyceride, and hydroalcoholic solution ; and then this retinol containing Liposphere is to be intercalated in lamellar liquid crystal layer which is prepared by emulsifying in an optimal ratio the mixtures composed of non-ionic emulsifier (cetearyl glucoside, sorbitan stearate & sucrose cocoate etc), cetearyl alcohol, stearic acid, cholesterol, and ceramide. In addition, the stability of the retinol containing oil in water cream by adding the polymeric emulsifier such as acrylate /C10-30 alkyl alkylate crosspolymer is to be ensured even at 55 C. Retinol containing oil in water cream prepared through above procedure could be very stable at 45 C for at least 50 days. The structure identification of lamellar liquid crystal was determined using polarized light microscope and electron microscope Conclusively, we could make the very stable retinol containing oil in water cream by triple procedure, that is, encapsulation of retinol in Liposphere, intercalation of retinol in lamellar liquid crystal layer, and assurance of the high temperature stability of cream even at 55 C.

THE EFFECT OF IRIDOID COMPOUND ON THE REMAINING PULP TISSUE AFTER PULPOTOMY (Iridoid 화합물이 치수절단 후 잔존치수 조직에 미치는 영향)

  • Aucubin, an iridoid glucoside, which is isolated from Aucuba japonica, has some biological effects. This study was to investigate the effect of aucubin on the remainig pulp tissues after pulpotomy. Mongrel dog's coronal pulps were mechanically exposed with a sterile round bur and excised with sterile sharp excarvator. After bleeding was controlled, in control group, $Ca(OH)_2$ powder was applied on the remaining pulps and the cavities were sealed with Z.O.E. cement. In experimental group 1, mixed powder with $Ca(OH)_2$ and aucubin(l : 1 by weight) was applied on the pulpotomized pulp surfaces. After the cavities were covered with sterile aluminum foil, they were sealed with Z.O.E. cement. In experimental group 2, only aucubin powder was applied on the remaining pulps and then they were treated the same as experimental group 1. In the all groups, the pulps were histopathologically observed by light microscope at the time intervals of 1, 2 and 4 weeks after experiment. The results were as follows : 1. In control and experimental groups, mild vascular congestion and bleeding were found in most of the specimens. Less inflammatory infiltration was observed in experimental groups than in control group. 2. Dentin bridge formation was found after 1 week at both control and experimental group 1. Dentin birdge had discontinuous osteodentin like appearance or contained some dentin chips. In experimental group 2, dentin bridge was not seen. 3. The coagulation necrosis layer on the remaining pulp tissues was seen in all groups. In experimental group 2, the thickest layer was observed. And in control group, coagulation necrosis layer was similar as in experimental group 1.

Inhibitory Effects of Flavonoids from Lespedeza cuneata on Aldose Reductase

  • Inhibition of aldose reductase (AR) has been shown to prevent the onset and progression of many diabetic complications wherein several AR inhibitors were isolated from plants abundant in polyphenolic compounds. Lespedeza cuneata (Fabaceae), a perennial plant indigenous in East Asian countries, is shown to be abundant in these polyphenolic substances such as flavonoids and tannins. However, there are no studies to date regarding its effects on AR. In this study, the inhibitory activity of the methanol extract and stepwise polarity fractions of Lespedeza cuneata on AR was investigated. The bioactive compounds purified from L. cuneata by repeated column chromatography were also tested for AR inhibition. Results show that the ethyl acetate and n-butanol fractions of L. cuneata exhibited potent inhibition against AR with $IC_{50}$ values of 0.57 and $0.49{\mu}g/mL$, respectively. Further analysis led to the isolation of acacetin (1), afzelin (2), astragalin (3), kaempferol (4), and scutellarein 7-O-glucoside (5). The AR inhibitory effects these five compounds were also determined in which compounds 2, 3, and 5 showed potent AR inhibitory effects with $IC_{50}$ values of 2.20, 1.91, and $12.87{\mu}M$, respectively. Quantitative analysis of astragalin (3) by HPLC-UV was also performed in the leaves and roots of L. cuneata (0.626 and 0.671 mg/g, respectively). This study reports that the flavonoids isolated from L. cuneata show promising AR inhibitory activities which can be further developed as natural therapies for treating and managing diabetic complications.

  • The aim of this investigation was to resolve the contradiction between the results of Sternberg and Mandels (1980, 1982)and those of Nisizawa et al., (1971) in cellulase induction by sophorose, and furthermore to study the conditional effects in sophorose-induced cellulase induction in Trichoderma reesei QM 9414. Sophorose could induce the synthesis of CMCase and ${\beta}-glucosidase$ simultaneously. Optimal induction medium by sophorose had the potassium citrate buffer solution of pH 3.0-4.0 for CMCase, but one of pH 5.0-6.0 for ${\beta}-glucosidase$. At this time, two different types of ${\beta}-glucosidase$ could be induced by sophorose: one was extracellular and had maximum at pH 5.0, the other was intracellular and had maximum activity at pH6.5. Induction study showed that $methyl-{\beta}-glucoside$ was not a true inducer of ${\beta}-glucosidase$ and that large ${\beta}-glucosidase$ induction could be obtained only by the addition of sophorose into the induction medium. Glucose repressed the induction of cellulase by sophorose. The repression of glucose could not be overcome by the addition of cyclic AMP into the induction medium.

Identification of Anthocyanin from The Extract of Soybean Seedcoat

  • Anthocyanins are naturally occuring phytochemicals and the main components of the coloring of plants, flowers and fruits. They are known to elicit antioxidative, anti-inflammatory and cancer preventive activity. In this study, we investigated anthocyanins in black / yellow soybean seedcoats using different methods of detection - thin layer chromatography (TLC), capillary zone electrophoresis (CZE) and HPLC analysis. The anthocyanins in soybean seedcoats were extracted by five independent methods of extraction and the aglycons (anthocyanidins) of the corresponding anthocyanins were prepared by acid mediated hydrolysis. The anthocyanin / anthocyanidin in black soybean seedcoat showed characteristic TLC mobility, CZE electrophoretic retention and HPLC migration time while little of anthocyanins were detected from yellow soybean seedcoat. The extracted anthocyanins showed pH dependent retention time in CZE and spectral change in UV-Vis spectrum. HPLC analysis of the hydrolyzed extract of black soybean seedcoat identified the presence of four anthocyanidins. The major anthocyanin in black soybean seedcoat was cyanin (cyanidin-3-O-glucoside), with the relative order of anthocyanidin in cyanidin > delphinidin > petunidin > pelargonidin.

The Effects of CD-product Specificity upon the Enzyme [CGTase] Reaction Condition (효소 [CGTase : Cyclodextrin glucanotransferase]의 반응 조건이 산물 [CD : Cyclodextrin]의 특이성에 미치는 영향)

  • Cyclodextrin glucanotransferase (EC 2.4.1.19, abbreviated as CGTase) is one of the most applied industrial enzymes that produces cyclodextrins from starch and related ${\alpha}$-1,4-glucans by intramolecular transglycosylation reaction upon Ca$\^$2+/ dependent manner. The reaction of CLEC, ${\alpha}$-CGTases from Bacillus macerans with the soluble starch as a substrate reveals that the surfactants (SDS, N-octyl-${\beta}$-D-glucoside) significantly affect not only the overall products of CDs but also their selectivity. The surfactants (SDS, Lubrol PX) trigger the increase of ${\alpha}$-CD production, but Triton x-100 and Tween 80 suppress ${\alpha}$-CD specificity. Organic solvents (dimethyl sulfoxide, formamide, 2-methyl-2,4-pentandiol, and ethylene glycol) also cause changes of total product and product selectivity.

Simultaneous Analysis of Bioactive Metabolites from Lonicera japonica Flower Buds by HPLC-DAD-MS/MS (HPLC-DAD-MS/MS를 이용한 금은화 생리활성 물질의 동시분석)

  • A high-performance liquid chromatography (HPLC) with diode array detector (DAD) and electrospray ionization mass spectrometry (ESI-MS) was established for the simultaneous determination of chlorogenic acid (1), sweroside (2), luteolin-7-O-glucoside (3), (E)-aldosecologanin (4) and 3,5-dicaffeoylquinic acid (5) from Lonicera joponica flower buds. The optimal chromatographic conditions were obtained on an ODS column (5 ${\mu}m$, 4.6${\times}$150 mm) with the column temperature $25^{\circ}C$. The mobile phase was composed of (A) water with 0.1% formic acid and (B) acetonitrile with 0.1% formic acid using a gradient elution, the flow rate was 0.3 ml/min. Detection wavelength was set at 250 nm. All calibration curves showed good linear regression ($r^2$>0.994) within test ranges. The developed method provided satisfactory precision and accuracy with overall intra-day and inter-day variations of 0.05${\sim}$1.95% and 0.15${\sim}$2.26%, respectively, and the overall recoveries of 97.71${\sim}$103.65% for the five compounds analyzed. The verified method was successfully applied to quantitative determination of the three types (phenolic compounds, iridoids and flavonoids) of bioactive compounds in 21 commercial L. japonica flower buds samples from different markets in Korea and China. The analytical results demonstrated that the contents of the five analytes vary significantly with sources.
