Transcriptome analysis of the livers of ducklings hatched normally and with assistance

  • Objective: "Hatchability" is an important economic trait in domestic poultry. Studies on poultry hatchability focus mainly on the genetic background, egg quality, and incubation conditions, whereas the molecular mechanisms behind the phenomenon that some ducklings failed to break their eggshells are poorly understood. Methods: In this study, the transcriptional differences between the livers of normally hatched and assisted ducklings were systematically analyzed. Results: The results showed that the clean reads were de novo assembled into 161,804 and 159,083 unigenes (${\geq}200-bp$ long) by using Trinity, with an average length of 1,206 bp and 882 bp, respectively. The defined criteria of the absolute value of log2 fold-change ${\geq}1$ and false discovery rate${\leq}0.05$ were differentially expressed and were significant. As a result, 1,629 unigenes were identified, the assisted ducklings showed 510 significantly upregulated and 1,119 significantly down-regulated unigenes. In general, the metabolic rate in the livers of the assisted ducklings was lower than that in the normal ducklings; however, compared to normal ducklings, glucose-6-phosphatase and ATP synthase subunit alpha 1 associated with energy metabolism were significantly upregulated in the assisted group. The genes involved in immune defense such as major histocompatibility complex (MHC) class I antigen alpha chain and MHC class II beta chain 1 were downregulated in the assisted ducklings. Conclusion: These data provide abundant sequence resources for studying the functional genome of the livers in ducks and other poultry. In addition, our study provided insight into the molecular mechanism by which the phenomenon of weak embryos is regulated.

Effects of Salviae miltiorrhizae Radix (SMR) on Serum Lipid Level in Hyperlipidemic Rats (단삼(丹蔘)이 고지혈증 흰쥐의 혈중 지질 변화에 미치는 영향)

  • Objective : Salviae miltiorrhizae Radix (SMR) has widely used to treat patients with cardiovascular diseases such as coronary arteriosclerosis, angina pectoris and hyperlipidemia. This study was designed to investigate the effects of SMR on changes in serum cholesterol and protective effects on liver tissue damage in Hyperlipidemic rats. Methods : The present author investigated changes in serum glucose, cholesterols, AST/ALT and histopathological changes of liver tissue by oral administration of SMR in Rats. Results : In this study, body weights of hyperlipidemic rats induced high fat diet did not changed, and treatment with SMR did not affect body weights in hyperlipidemic rats. For experimental period, Food and Water uptake in SMR administered group were the same as those in hyperlipidemic control group. In this experiment, treatment with SMR decreased total cholesterol and AST in serum which elevated by high fat diet respectively. In addition, SMR administration protected liver tissue from damage induced by induction of hyperlipidemia. Conclusions: These results suggest that SMR is useful to treat patients with disease related to cardiovascular diseases including hyperlipidemia, because SMR can decrease cholesterol and AST in serum and also have non-specific protective effect on tissues including liver.

Health Status by Anthropometrie Values and Serum Biochemical Indices of Preschool Children in Day-Care Centers in Gumi (구미지역 보육시설 유아들의 신체계측과 혈액생화학적 지표에 따른 건강상태)

  • This study was conducted to investigate health status of preschool children utilizing anthropometric values and serum biochemical indices. The subjects were 1,159 boys and 1,019 girls in Gumi day-care centers aged 4 to 6 years. The mean height and weight of 4, 5 and 6 year old children were 103.0 cm and 17.1 kg, 107.2 cm and 18.3 kg and 113.4 cm and 20.6 kg, respectively. By the obesity index criteria, $5\%$ of boys and $4\%$ of girls were obese whereas $10\%$ of boys and $9\%$ of girls were underweight. By the Kaup index criteria, $7\%$ of boys and $4\%$ of girls were obese. By the percentage of body fat criteria, $5\%$ of boys and girls were obese. The mean concentrations of serum total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, Atherogenic Index and serum glucose were 154.1-156.5 mg/dL, 80.1-81.9 mg/dL, 46.8-47.4 mg/dL, 90.9-92.8 mg/dL, 2.34-2.39 and 76.4-77.8 mg/dL, respectively. Serum triglyceride level, AI and body fat percentage of obese children were significantly higher than other children and were significantly lower in HDL-cholesterol level (p < 0.05). In addition there were significant correlation between anthropometric values and serum biochemical indices : Kaup index and obesity index were positively correlated to the systolic blood pressure and diastolic blood pressure in boys and girls (p < 0.01) and to the triglyceride concentration in boys (p < 0.05). These results indicate the need of developing and implementation of nutrition education program for preschool children to prevent further development of childhood obesity and to maintain normal serum lipid profiles.

An inhibitory of seed germination by an extracellular metabolite of Pseudomonas sp. F721 (Pseudomonas sp. F721의 세포외 대사산물에 의한 종자의 발아억제)

  • Pseudomonas sp. F721 isolated from soil produced a substance related in seeds germination inhibition. Addition of phytohormone, and GA (gibberellin acid) in the culture broth elevated production of the germination inhibition substance. The production of the substance was optimized in the culture conditions of $35^{\circ}C$, pH 9.0, 150 rpm, 48 hr, glucose 0.5% (w/v), and innoculation ratio 1.0% (v/v). The physical and chemical stability of the substance in the variety of pH ranging from 2.0 to 12.0 and from freezing to $100^{\circ}C$ were shown. The germination inhibition substance suppressed 90% of germination compared with that of the control experiment in a few days.

Effects of Selected Stabilizers on the Color Deterioration of Crude Pigment Extract from Schizandra fruit (Schizandra Fructus) (여러 가지 안정화 물질이 오미자 색소 추출물의 가열 변색에 미치는 영향)

  • The effects of selected stabilizers and sugars on color deterioration of anthocyanin in Schizandra fructus were examined at $100^{\circ}C$ for $120{\sim}180$ min. Among four test sugars, it was found that fructose accelerated the thermal color deterioration while maltose retarded the color deterioration by 40%. Maltodextrin and ${\gamma}$-cyclodextrin showed the highest stabilizing effect on the thermal color deterioration of crude pigment extract from Schizandra fructus(CPES) containing 2 mg% of anthocyanin. Addition of maltodextrin or ${\gamma}$-cyclodextin at 5% retarded thermal color deterioration of CPES. In gel system, 5% of maltodextrin also retarded the color deterioration by $15{\sim}20%$ during storage at $25^{\circ}C\;and\;35^{\circ}C$.

  • The purpose of this study was to improve the conductivity of $Li_3V_2(PO_4){_3}$ by adding carbon source so that the discharge rate and cyclic properties were improved. Glucose and CNT were added to $Li_3V_2(PO_4){_3}$ and the structure and electrochemical properties were studied. $Li_3V_2(PO_4){_3}$, $Li_3V_2(PO_4){_3}$/C and $Li_3V_2(PO_4){_3}$/CNT were synthesised by solid state reaction using hydrogen reduction method at 600, 700, 800, $900^{\circ}C$. The cathode materials were assembled to coin cell type 2032 with Lithium metal as a counter electrode. The coin cell was galvanostatically evaluated in the voltage range of 3.0~4.8 V.

Changes of Plasma Metabolites, Hormones, and mRNA Expression of Liver PEPCK-C in Spontaneously Ketotic Dairy Cows

  • The objective of this study was to understand changes of plasma metabolites, hormones, and mRNA level of cytoplasmic phosphoenolpyruvate carboxykinase (PEPCK-C) in liver in spontaneous clinical ketosis; 10 clinically ketotic cows and 10 healthy cows were chosen from the same dairy farm. Eleven blood parameters and liver fat content were measured in all cows, and mRNA levels of PEPCK-C in liver were measured by semi-quantitative reverse transcription (RT) polymerase chain reaction (PCR). In ketotic cows, concentration of plasma glucose decreased (p<0.01), concentration of plasma nonesterified fatty acids (NEFA) and $\beta$-hydroxybutyric acid (BHBA) increased (p<0.01), liver fat content (18.8% wet weight) and activity of plasma aspartate aminotransferase (AST) increased (p<0.01), but concentration of plasma total bilirubin (TBIL), $\gamma$-glutamyl transpeptidase ($\gamma$-GT), and cholinesterase (CHE) increased (p>0.05). In addition, concentration of plasma insulin decreased (p<0.05), concentration of plasma glucagons decreased (p>0.05), and mRNA level of PEPCK-C in liver increased (p<0.05). It is concluded that the adaptative changes of metabolites, hormones, and mRNA level of PEPCK-C in ketotic cows were in favor of the enhancement of gluconeogenesis, the decrease of fat mobilization and the relief of ketosis, but these were still inadequate to relieve ketosis.

Effects of Monensin on Metabolism and Production in Dairy Saanen Goats in Periparturient Period

  • This trial evaluated the effects of dietary supplementation with monensin sodium on dry matter intake, metabolic parameters and milk yield and milk composition of dairy Saanen goats in the periparturient period. Twelve Saanen pregnant dairy goats were assigned to a treatment and a control group. Saanen goats were fed monensin as its 10% sodium salt in the amount of 33 mg/kg of total dry matter intake during 30 d before parturition till d 42 in milk. Blood samples were collected from each goat at d 30, 15 and 7 before expected kidding time and also in d 1, 7, 13, 19, 21, 28, 35 and 42 postpartum. The serum concentrations of ${\beta}$-Hydroxybutyrate (BHBA), non-esterifed fatty acid (NEFA), calcium, magnesium, inorganic phosphorus, cholesterol, triglyceride, urea, total protein, albumin and glucose and aspartate aminotransferase (AST) activity were determined. The BHBA concentration significantly decreased in goats, which received monensin in comparison to controls in the postpartum period (p = 0.049). The concentration of sodium (Na) was significantly influenced by monensin treatment in the postpartum period (p = 0.048). In addition, the difference in dry matter intake was highly significant between the two groups during the pre-partum period. Controls had more dry matter intake (DMI) than treatment goats (p = 0.0001). Mean 3.5% fat corrected milk production was not influenced by monensin treatment. However, milk fat percentage was significantly decreased in monensin treated goats (p = 0.0017).

Biodegradation of Crude oil by Marine Bacterium Pseudomonas sp. CHCS-2 and Composition of the Biosurfactant (해양세균 Pseudomonas sp. CHCS-2에 의한 원유분해 및 생물유화제의 성분 분석)

  • marine bacterium Pseudomonas sp. CHCS-2 produced the biosurfactant in the culture broth which contained 2%(w/v) arabian light crude oil and the productivity of biosurfactant was increased with the addition of glucose. The crude oil in the culture broth was degraded by this strain and carbon chain of $_nC_{12}~_nC_{22}$ was completely degradaded during the incubation for 196 h. The crude biosurfactant was purified by Amberlite XAD-7, Sepharose CL-4B and DEAE-Sepharose CL-6B column chromatography. Therefore, 0.21g/L of the purified biosurfactnat was obtained. The purified biosurfactant was a type of lipoprotein and the molecular weight was estimated as 67kDa by SDS-PAGE. The lipid composition was identified as octadecanoic acid by gas chromatography/mass spectrometry. And then, the N-terminal amino acid sequence of the protein was determined as Ser-Val-lle-Asn-Thr-lle-X-Met-lle-Gly-Gln-Gln- and the sequence did not show homology to any other known lipoprotein. Therefore, the purified lopoprotein was predicted novel biosurfactant.

Purification and Characterization of Glycerate Kinase From the Thermoacidophilic Archaeon Thermoplasma acidophilum: An Enzyme Belonging to the Second Glycerate Kinase Family

  • Thermoplasma acidophilum is a thermoacidophilic archaeon that grows optimally at $59^{\circ}C$ and pH 2. Along with another thermoacidophilic archaeon, Sulfolobus solfataricus, it is known to metabolize glucose by the non-phosphorylated Entner-Doudoroff (nED) pathway. In the course of these studies, the specific activities of glyceraldehyde dehydrogenase and glycerate kinase, two enzymes that are involved in the downstream part of the nED pathway, were found to be much higher in T. acidophilum than in S. solfataricus. To characterize glycerate kinase, the enzyme was purified to homogeneity from T. acidophilum cell extracts. The N-terminal sequence of the purified enzyme was in exact agreement with that of Ta0453m in the genome database, with the removal of the initiator methionine. Furthermore, the enzyme was a monomer with a molecular weight of 49kDa and followed Michaelis-Menten kinetics with $K_m$ values of 0.56 and 0.32mM for DL-glycerate and ATP, respectively. The enzyme also exhibited excellent thermal stability at $70^{\circ}C$. Of the seven sugars and four phosphate donors tested, only DL-glycerate and ATP were utilized by glycerate kinase as substrates. In addition, a coupled enzyme assay indicated that 2-phosphoglycerate was produced as a product. When divalent metal ions, such as $Mn^{2+},\;CO^{2+},\;Ni^{2+},\;Zn^{2+},\;Ca^{2+},\;and\;Sr^{2+}$, were substituted for $Mg^{2+}$ the enzyme activities were less than 10% of that obtained in the presence of $Mg^{2+}$. The amino acid sequence of T. acidophilum glycerate kinase showed no similarity with E. coli glycerate kinases, which belong to the first glycerate kinase family. This is the first report on the biochemical characterization of an enzyme which belongs to a member of the second glycerate kinase family.
