Background: Ginsenoside F2 (GF2), a minor component of Panax ginseng, has been reported to possess a wide variety of pharmacological activities. However, its effects on glucose metabolism have not yet been reported. Here, we investigated the underlying signaling pathways involved in its effects on hepatic glucose. Methods: HepG2 cells were used to establish insulin-resistant (IR) model and treated with GF2. Cell viability and glucose uptake-related genes were also examined by real-time PCR and immunoblots. Results: Cell viability assays showed that GF2 up to 50 μM did not affect normal and IR-HepG2 cell viability. GF2 reduced oxidative stress by inhibiting phosphorylation of the mitogen-activated protein kinases (MAPK) signaling components such as c-Jun N-terminal kinase (JNK), extracellular signal-regulated kinase 1/2 (ERK1/2), and p38 MAPK, and reducing the nuclear translocation of NF-κB. Furthermore, GF2 activated PI3K/AKT signaling, upregulated the levels of glucose transporter 2 (GLUT-2) and GLUT-4 in IR-HepG2 cells, and promoted glucose absorption. At the same time, GF2 reduced phosphoenolpyruvate carboxykinase and glucose-6-phosphatase expression as well as inhibiting gluconeogenesis. Conclusion: Overall, GF2 improved glucose metabolism disorders by reducing cellular oxidative stress in IR-HepG2 cells via MAPK signaling, participating in the PI3K/AKT/GSK-3β signaling pathway, promoting glycogen synthesis, and inhibiting gluconeogenesis.
쥐 간세포를collagenase perfusion method에 의해 분리한 뒤 collagen coated dish와floating collagen membrane에서 일차배양하여 간세포의 분화기능을 조사하였다. 두 경우 모두 간세포의 생존율은 5일 이후부터 점차 감소하였거나 간세포에 의한 암모니아 처리기능과 albumin생성기능은 약 7일간 유지되었다. 또 이러한 분화기능에 유지는 ollagen coated dish보다floating collagen membrane이 유리한 것으로 나타났다.
The effect of ginseng saponin on the activities of isocitrate lyase, palate synthase, succinate dehydrogenase, malate dehydrogenase and lipase have been investigated at the early phase of germinating soy-bean sprout and found that all the above enzymes were stimulated when the bean was rinsed for 24 hours with 10-4% saponin solution. The length of the saponin treated group was not longer than that of control group but the weight of the former was heavier (15%) than the latter. Total sugar content of test group was always much higher than that of control. From the above results, it was concluded that ginseng saponin might stimulate several enzymes of Soybean sprout during germination resulting in rapid growth of the Soybean sprout.
Sleep-disordered breathing (SDB) is associated with increased cardiovascular and cerebrovascular morbidity. Epidemiological and clinic-based studies have shown that SDB is related to impaired glucose tolerance and increased insulin resistance, independent of obesity. Despite of a consistent association between SDB and impaired glucose-insulin metabolism, the mechanism underlying this relationship has not been fully elucidated. It is recognized that hypoxemia and hypercapnia that occur in SDB provoke sympathetic nervous activity and catecholamine, epinephrine and norepinephrine, and cortisol are released. Sympathetic hyperactivity and increased catecholamines can impair glucose homeostasis by increasing glycogenolysis and gluconeogenesis, which can result in increased circulating insulin levels and increased risk of insulin resistance. A prospective study is needed to investigate the causal relationship between SDB and impaired glucose-insulin metabolism in a healthy population without diabetes, hypertension and obesity as etiologic risk factors.
My professional journey to understand the glucose homeostasis began in the 1990s, starting from cloning of the promoter region of glucose transporter type 2 (GLUT2) gene that led us to establish research foundation of my group. When I was a graduate student, I simply thought that hyperglycemia, a typical clinical manifestation of type 2 diabetes mellitus (T2DM), could be caused by a defect in the glucose transport system in the body. Thus, if a molecular mechanism controlling glucose transport system could be understood, treatment of T2DM could be possible. In the early 70s, hyperglycemia was thought to develop primarily due to a defect in the muscle and adipose tissue; thus, muscle/adipose tissue type glucose transporter (GLUT4) became a major research interest in the diabetology. However, glucose utilization occurs not only in muscle/adipose tissue but also in liver and brain. Thus, I was interested in the hepatic glucose transport system, where glucose storage and release are the most actively occurring.
Prominin-1 (PROM1), also called CD133, is a penta-span transmembrane protein that is localized in membrane protrusions, such as microvilli and filopodia. It is known to be expressed in cancer stem cells and various progenitor cells of bone marrow, liver, kidney, and intestine. Accumulating evidence has revealed that PROM1 has multiple functions in various organs, such as eye, tooth, peripheral nerve, and liver, associating with various molecular protein partners. PROM1 regulates PKA-induced gluconeogenesis, TGFβ-induced fibrosis, and IL-6-induced regeneration in the liver, associating with Radixin, SMAD7, and GP130, respectively. In addition, PROM1 is necessary to maintain cancer stem cell properties by activating PI3K and β-Catenin. PROM1-deficienct mice also show distinct phenotypes in eyes, brain, peripheral nerves, and tooth. Here, we discuss recent findings of PROM1-mediated signal transduction.
Lactiplantibacillus plantarum K9은 굼벵이에서 분리된 다양한 생리활성물질에 기인하여 프로바이오틱스 균주로 활용 가능한 유산균이다. L. plantarum K9 유전체 분석결과로써 박테리아 염색체와 3 plasmid가 존재하는 것으로 나타났다. L. plantarum K9의 핵심 대사경로 분석 결과 해당과정, 오탄당대사(pentose phosphate pathway)는 정상적으로 수행되는 것으로 나타났다. 그러나 포도당신생합성과 ED pathway의 핵심 효소인 fructose-1,6-bisphosphatase (EC: 3.1.3.11)와 6-phosphogluconate dehydratase (EC: 4.2.1.12) / 2-keto-de- oxy-6-phosphogluconate (KDPG) aldolase (EC: 4.2.1.55)가 각각 결여되어 있기 때문에 포도당신생합성과 ED pathway는 수행하지 못하는 것으로 제의된다. 또한, TCA 회로에서 fumarate 및 malate를 형성하는 일부 효소만 존재하는 반면에 나머지 TCA 회로에 연관되는 효소들이 모두 결여되어 있었기 때문에 TCA 회로는 진행되지 못하는 것으로 추정되었다. 산화적 전자전달계는 NADH dehydrogenase complex I과 cytochrome reductase complex IV에 해당하는 요소들을 보유하고 있기 때문에 class IIB 타입(bd-type)의 전자전달시스템을 수행할 것으로 예측되었다. 종합적으로, L. plantarum K9은 lactic acid 동형발효를 수행하며, 포도당신생합성 및 오탄당대사가 가능하며, class IIB 타입(bd-type) 산화적 전자전달시스템에 의해 에너지 대사를 수행하는 것으로 제의된다. 따라서, L. plantarum K9은 다른 유산균주에 비교하여 lactic acid 생성량이 비교적 높아 생리활성도가 우수할 것으로 제의된다. 다른 한편으로, L. plantarum K9은 산화적 전자전달이 가능한 것으로 추정되어 산소에 대한 내성이 높아서 배양 특성이 양호하여 프로바이틱스로써 활용가능성이 높은 것으로 제의된다.
본 총설에서는 아미노산의 공업적 생산균인 Corynebacterium glutamicum의 탄소 대사 및 이와 관련된 총체적 조절 메커니즘에 대한 최근의 연구를 정리하였다. C. glutamicum의 산업적 발효을 위한 기질로서 사용되는 당밀은 주로 sucrose, glucose, fructose로 이루어져 있으며, 이들 당은 phosphotransferase system을 통해서 수송된다. C. glutamicum의 탄소 대사 특징은 glucose가 다른 당이나 유기산 등과 함께 존재할 때, glucose와 이러한 탄소원 들을 동시에 대사한다. 그러나 glucose/glutamate 혹은 glucose/ethanol 등의 혼합물에서 는 탄소원의 순차적 이용으로 인해 나타나는 diauxic growth 현상을 나타내며, 이러한 carbon catabolite repression(CCR) 현상은 E. coli나 B. subtilis 등에서 알려진 것과는 다른 독특한 분자적 메커니즘과 조절 circuits을 가지고 있음이 밝혀지고 있다. C. glutamicum의 CRP homologue인 GlxR은 acetate 대사를 포함하여 glycolysis, gluconeogenesis 및 TCA cycle 등을 포함하는 중심탄소대사 조절 뿐만 아니라, 다양한 세포 기능의 조절에 관여하는 총체적 조절 단백질로서의 역할이 제시되고 있다. C. glutamicum의 adenylate cyclase(AC)는 막과 결합된 class IIIAC 로서, 막 단백질의 특성상 아직 규명되어 있지 않은 세포 외부의 환경 변화에 대응하여 세포 내의 cAMP합성 수준을 조절할 수 있는 sensor로 추정할 수 있다. 특히 C. glutamicum의 경우 배지내 glucose 를 비롯한 탄소원과 cAMP 농도와의 관련성이 E. coli에서 알려진 교과서적 지식과는 상반되게 변화하는 경향을 보이고 있어, cAMP signaling에 의한 세포 내 regulatory network 등은 향후 풀어야 할 의문으로 남아있다. 탄소대사 조절의 최상위에 존재하며 global 조절자인 GlxRcAMP 복합체 이외에도 차상위 전사조절 단백질로서 RamB, RamA, SugR 등이 존재하여 다양한 탄소대사를 조절한다. 최근 들어서는 새로운 탄소원으로서 대두되고 있는 biomass 관련 기질들을 이용할 수 있는 C. glutamucum 균주 구축을 통하여 이용 기질의 범위를 확대시키고자 하는 연구 및 탄소 대사와 관련하여 L-lysine의 발효 수율 혹은 생산성을 향상시키고자 하는 다양한 분자적 균주 육종 연구 등이 수행되고 있다.
오이 씨앗의 발아는 세포의 지방체내 저장지방의 분해 결과인 acyl-CoA의 글라이옥시좀으로 이동 후 베타 산화의 결과물인 acetyl-CoA의 글라이옥실산 회로로의 유입과 지방의 유동으로 촉발된다. Acetyl-CoA는 글라이옥실산 회로의 가동을 위한 탄소원을 제공하며 시트르산과 말산을 생성하며 글라이옥실산 회로의 작동을 유도한다. 지방 저장 종자의 발아에 있어서 글라이옥실산 회로는 필수적 요소이며, 그 결과물인 말산 및 숙신산의 미토콘드리아로의 이동은 TCA 회로의 가동과 옥살초산의 생성 및 세포질로의 유동으로 PEPCK에 의한 당신생을 가능하게 한다. 즉, 저장 지방을 원료로 여러 대사물질의 생산 및 이동과 다중의 대사경로를 통하여 발아 시 사용 가능한 에너지원인 포도당의 형태로 전환이 이루어진다. 이에 동반하여 많은 유전자의 발현 조절이 이루어지고, 세포내 소기관 특히 미소체로 대표되는 글라이옥시좀은 말산 합성효소(malate synthase)와 이소 시트르산 분해효소(isocitrate lyase)로 특화된다. 또 다른 acetyl-CoA의 유동은 carnitine을 매개로 하는 BOU (A BOUT DE SOUFFLE)의 작동이다. 이것은 카니틴의 대사와 관련하여 고등식물의 발달과 대사과정에서의 중요성이 확인된 것으로 사료된다.
Brain-type natriuretic peptide (BNP)은 뇌나트륨이뇨펩티드로, 좌심실의 심근세포에서 분비되는 호르몬으로, 심장과 신장에 작용하여 혈관 확장과 나트륨 이뇨 작용 등을 하는 것으로 알려져 있으나, 최근에는 다양한 조직에서 대사 작용을 조절하는 것으로 보고된다. 본 연구에서는 간 조직에서 BNP의 영향을 알기 위해 BNP가 고지방식이에 의해 유도되는 인슐린 저항성을 개선하는지를 조사하였다. BNP을 주입한 쥐와 control로서 saline을 주입한 쥐들 간에는 몸무게, 체지방양(fat mass), 제지방량(lean body mass)의 변화는 없었다. 고인슐린혈증 정상혈당 글루코스 클램프(Hyperinsulinemic Euglycemic Glucose Clam) 동안, BNP를 주입한 고지방 식이 쥐들은 saline을 주입한 고지방식이 쥐에 비해 혈당(blood glucose)은 감소하였으며, 포도당 주입 속도(glucose infusion rate)는 증가하였다. 또한 BNP은 포도당 신생 및 중성지방 합성 관련 유전자들의 발현을 감소시켜, 간에서 포도당 생성과 중성지방의 양을 감소시켰다. BNP는 saline을 주입한 쥐에 비해 간 조직에서 Akt와 AMP-activated protein kinase (AMPK)의 인산화를 증가시켰는데, 이는 BNP을 처리한 AML12 간세포에서도 BNP는Akt와 AMPK 인산화를 증가시켰다. 이상의 결과는 BNP가 간에서 인슐린 저항성을 개선하여 포도당 생성과 중성 지방 생성을 억제함을 알 수 있었다.
