Carbon-Nanotube-Modified Glass Micropipette for Simultaneous Drug Injection and Neural Monitoring

  • Glass micropipettes are widely used for drug injection in neurological studies. To enable these devices to monitor neural activity simultaneously with drug injection, an electrode such as Ag/AgCl must be located near or inserted into the glass micropipette to detect electrical signals in vivo. Here, we report carbon-nanotube-modified glass micropipettes (CNGs), which have excellent electrochemical properties such as low impedance and large electrochemical surface area suited for neural recording. In addition, using a standard pressure pump, CNGs can deliver drugs to the target region without bending. Because they are based on standard glass micropipettes, CNGs can readily be applied to traditional equipment, creating opportunities to monitor precisely the drug-injected area.

소 체외수정란의 배반포기배의 OPS 대 GMP Vitrification의 비교 (Comparison of Open Pulled Straw (OPS) vs Glass Micropipette (GMP) Vitrification in IVP Bovine Blastocysts)

  • 본 연구는 체외생산된 배반포기배의 vitrification 을 위한 용기로서 glass micropipette(GMP)을 이용할 수 있는지, GMP 와 OPS 로 동결융해 후 생존율의 비교 및 GMP vitrification 후 hatching 율의 향상을 위하여 실시하였다. GMP vessel은 열전도율과 수정란을 포함하는 적은 질량 때문에 OPS 보다 동결 및 융해속도를 높일 수 있다. 3개의 체외수정란을 vitrification 용액에 노출시키고 OPS 또는 GMP vessel에 loading 시킨 후 액체질소에 침적하는데까지 20~25초 이내에 실시하였다. 동결ㆍ융해한 배반포기배는 0.25와 15 M sucrose solution 및 TCM 1999에 각각 5분씩 차례로 희석한 후 10% FCS가 첨가된 TCM 199에 24시간동안 배양하였다. OPS(75.9%)와 GMP(80.0%) 방법간의 re-expanding 율은 유의적 (P<0.05)인 차이가 없었다. OPS(34.1%)와 GMP(37.5%) 방법에서 hatching 율은 intact group(54.3%) 보다도 유의적 (P<0.05)으로 낮았다. 비록 GMP straw 당 3개 이하의 blastocysts 를 loading 하였더라도 narrow portion(83.3%) 보다도 wide portion(S6.7%)에서 vitrified 되었다면 re-expanding 율이 유의적 (P<0.05)으로 낮았다. 비록 30초 처리군과 무처리군 간에는 유의적인 차이가 없었지만 0.05% pronase 용액에 30, 60 및 90초간 처리군 (45.9, 54.7 및 57.5%)의 hatching 율은 무처리구 (35.0%) 보다 유의적 (P<0.05)으로 높았다. 이러한 결과들은 OPS와 GMP vitrification vessel은 체외생산된 배반포기배의 높은 생존율을 얻을 수 있다. 그러나 GMP vessel은 L$N_2$침적 후 vessel의 floating을 방지하기 위한 또 다른 cap 이 필요하지 않다는 유리한 점을 가지고 있다. 수정란의 loading 위치, 즉 narrow 또는 wide portion에 따라 소 체외 생산된 배반포기배의 생존력에 제한적인 요인으로 고려된다. 0.5% pronase 용액에 60 또는 90초간의 노출은 융해후 hatching 율을 향상시킬 수 있었다.

Effect of Warming Temperature on the Viability of Bovine IVP Blastocyst Cryopreserved by Glass Micropipette (GMP) Vitrification

  • The purpose of this study was to investigate the warming temperature and exposed time on the post-thaw survival rate and viability of bovine blastocyst cryopreserved by GMP vitrification. Groups of three bovine IVP blastocysts were sequentially placed into vitrification solution before being loaded into the GMP straws and immersed into LN$_2$within 20 to 25 sec. The warming rate was increased 2 times of warming temperature for improvement of post-thaw survival rates. The frozen embryos were warmed either at 35 or 70$^{\circ}C$ for 1 or 2 sec and then diluted in sucrose solution. Post-thaw blastocysts were serially washed in 0.25 and 0.15 M sucrose in holding medium (HM: TCM199 supplemented with 10% FCS) and TCM-199 for each 5 min, respectively, and then cultured in TCM199 for 24 h. The rate of re-expanded blastocyst was significantly different fer 35 and 70$^{\circ}C$ warming temporature (76.4 vs. 89.3%; P<0.05). The rate of re-expanded blastocyst at 70$^{\circ}C$ for 1 sec was significantly higher than that for 2 sec (91.1 vs. 70.9%; P<0.05). The number of nuclei counted were significantly different among control, 35 and 70$^{\circ}C$ (121${\pm}$8.5 vs. 104${\pm}$11.7 vs. 114${\pm}$10.3; P<0.05). These results indicated that the increasing of warming rate can provide high survival rates of bovine IVP blastocysts. Especially, the best viability of post-thaw blastocyst could be thaw at 70$^{\circ}C$ for 1 sec. The warming temperature and exposed time far warming was considered to be limiting factors to the viability of bovine IVP embryos. he purpose of this study was to investigate the warming temperature and expose.

  • Hydrodynamic focusing technique to generate focused flow has been used for flow cytometry in microfluidic devices. However, devices with circular capillary tubes made of glass are not suitable for flow visualization or optical signal detection because the rays of light are distorted at the curved interface. We devised a new acrylic chamber assembled with a pulled micropipette and a rectangular microchannel made of glass. This new channel geometry enabled us to visualize the three-dimensional (3D) flow characteristics with confocal imaging technique. We analyzed the 3D hydrodynamic focusing in a circular capillary tube and a rectangular microchannel over a practical range of flow rates, viscosities and pressure drops.

  • This paper reports low-cost microreactor $(10{\mu}{\ell})$ biochip for the DNA PCR (polymerase chain reaction). The microbiochip $(20mm{\times}28mm)$ is a hybrid type which is composed of PDMS (polydimethylsiloxane) layer with serpentine micochannel $(360{\mu}m{\times}100{\mu}m)$ chamber and glass substrate integrated with microheater and thermal microsensor. Undesirable bubble is usually created during sample loading to PMDS-based microchip because of hydrophobic chip surface. Created bubbles interrupt stable biochemical reaction. We designed improved microreactor chamber using microfluidic simulation. The designed reactor has a coner-rounded serpentine channel architecture, which enables stable injection into hydrophobic surface using micropipette only. Reactor temperature needed to PCR reaction is controlled within ${\pm}0.5^{\circ}C$ by PID controller of LabVIEW software. It is experimentally confirmed that SRY gene PCR by the fabricated microreactor chip is performed for less than 54 min.

미소전극어레이형 DNA칩을 이용한 유전자다형의 전기화학적 검출 (Electrochemical Detection of Single Nucleotide Polymorphism (SNP) Using Microelectrode Array on a DNA Chip)

  • In this study, an integrated microelectrode array was fabricated on glass slide using microfabrication technology. Probe DNAs consisting of mercaptohexyl moiety at their 5-end were spotted on the gold electrode using micropipette or DNA arrayer utilizing the affinity between gold and sulfur. Cyclic voltammetry in 5mM ferricyanide/ferrocyanide solution at 100 ㎷/s confirmed the immobilization of probe DNA on the gold electrodes. When several DNAs were detected electrochemically, there was a difference between target DNA and control DNA in the anodic peak current values. It was derived from specific binding of Hoechst 33258 to the double stranded DNA due to hybridization of target DNA. It suggested that this DNA chip could recognize the sequence specific genes. It suggested that multichannel electrochemical DNA microarray is useful to develop a portable device for clinical gene diagnostic System.

마이크로 프로브 기반 열전 센서 제작 기술 (Fabrication of a Micro-thermoelectric Probe)

  • 마이크로 수준에서의 온도측정을 위하여 유리 피펫 기반의 프로브형 온도센서를 제작하였다. 이를 이용하여 정밀도 ${\pm}$ 0.1 K 를 가지는 온도측정 보정 실험을 수행하였다. 본 연구에서 제작한 방식은 저온의 용융점을 갖는 솔더합금(Sn)을 피펫에 내부에 주입하고 외부에 니켈(Ni) 코팅을 하여 열전대를제작함으로써 저비용의 프로브형 온도센서를 구현하였다. 제작된 센서 팁 끝단의 지름은 5 Am 에서 30 Am 로 피펫을 가열하여 당기는 방식으로 프로브 끝단의 크기를 조절하였다. 제작된 온도센서는 챔버내에서 정밀하게 제어된 온도 조절기를 사용하여 보정하였으며, 이를 통하여 얻어진 열전계수의 범위는 8.46 에서 $8.86{\mu}V$/K 의 값을 얻었다. 이를 이용하면 MEMS 소자, 세포, 티슈 등의 바이오 소재의 온도 및 열특성 측정용 프로브로 활용할 수 있을 것으로 생각된다.

Comparison of Two Vitrification Methods of In Vitro Development Oocytes Collected from Porcine Antral Follicles Using Open Pulled Straw (OPS) Techniques

  • The advantages of the OPS techniques(Vajta G et al, Mol Reprod Dev 51: 53-58,1998) give 1) high survival rates of various types of eggs, 2) quick and simple process, 3) inexpensive equipment and reduced chilling injury. The efficiency of IVM/IVF technique in the porcine species is relatively lower than that obtained in other species such as ruminants. Two experiments were designed to investigate the effects of in-vitro fertilization of porcine oocytes matures using different OPS protocol for chilling and warming of vitrification. Porcine oocytes from ovaries collected at abattoir were matured for 44 hours in TCM199 Earle's salt supplemental with pyruvate, pff, L-cysteine, hormones and gentamycin. Oocytes were denuded and fertilized with frozen boar semen by common method. Porcine embryos produced routinely by in-vitro culture system of NCSU23 medium. The vitrification and the warming were conducted by OPS method with the glass micropipette instead of straw vessels and modified the protocol of G.Vajta(1999). In Exp 1, Chilling/Warming:Holding Medium(HM)+EG+DMSO/HM +sucrose Medium(SM) at 39$^{\circ}C$ warm stage. In Exp 2, : PBS+CS+EG+Ficoll+ Trehalose/PBS+Trehalose at 25$^{\circ}C$ stage. Filling, freezing, packing, thawing out and further culturing were performed to follow the basic protocol of G Vajta. During IVM-lVC and post-warming, fertilization parameter and developmental potential were compared to and statistically analysed. It was not significantly different from Exp 1 and Exp 2 but 25$^{\circ}C$ of stage was slightly higher on the morula/blastocyst forming rate and better atmosphere for worker than that at 39$^{\circ}C$ stage.

Mongolian 수란우에 한우 동결수정란의 이식 후 산자 생산 (Production of Korean Native Cow from Mongolian Cow following Transfer of Vitrified Blastocyst)

  • 본 연구는 체내, 체외 소 배반포기 배의 GMP vitrification 후 활력도의 비교와 한우 수정란을 몽골 소에 수정란이식 후 산자생산 가능성을 조사하고자 실시하였다. 한우 수정란은 체외수정란 또는 과배란처리에 의한 체내수정란을 생산하여 GMP vitrification 방법으로 동결 후 몽고로 수송하였다. 수란우는 CIDR과 $PGF_2\alpha$ 처리에 의하여 동기화를 유도하였다 체내수정란생산을 위하여 7두를 과배란처리하였다. 총 64개의 배반포기를 회수하였다. ($9.1\pm2.94$per cow). 체외수정란생산은 80.1% 분할율(174/217)과 40.8% 배반포기 발달율(71/174)을 얻었다. 체내수정란(93.7%; 45/48)의 동결융해 후 생존율은 체외수정란(82.5%; 52/63)보다 유의적으로 높았다(P<0.05). 8두의 몽골 소에 2개의 수정란을 이식하여 5두가 이식 후 60일째 임신이 확인되었으나, 그 중 1두는 240째 유산을 확인하였다. 그 중 2두의 수란우에서 2두의 산자를 275일째 생산에 성공하였다. 이러한 결과는 GMP vitrification 방법은 체내, 체외수정란의 동결보존방법으로 이용될 수 있을 뿐만 아니라 동결융해란의 몽골 소에 이식 후 한우를 생산할 수 가능성을 확인하였다.
