  • The oocyst wall is severed by means of mechanical injury or chemical agents. This study reports the percentage of in vitro sporocyst release following mechanical shaking in the presence of varying sizes of glass beads. Glass beads measured 0.5, 1, and 3 mm in diameter and were shaken with the oocysts for different times ranging from 5 sec to 5 min. Approximately 80% of sporocysts were released with 5 min of shaking in the presence of 3 mm glass beads, as well as 30 sec with 0.5 mm beads and 1 mm glass beads. The release of sporocysts of E. tenella was most efficient using 1 mm glass beads and treatment times of 30 sec to 1 min. Therefore, the use of 1 mm glass beads with 30 sec to 1 min of agitation is recommended in order to maximize sporocyst release and recovery and to improve the yield of viable sporozoites for use in biochemical, tissue culture, and immunological applications of coccidia.

  • Histamine is a kind of primary biogenic amine arising from the decarboxylation of the amino acid L-histidine. The toxicology of histamine and its occurrence and formation in foods are especially emphasized in fermented foods. In this study, the biosensor for detection of histamine with functionalized multi-walled carbon nanotubes (MWCNT) was developed. We also searched for an appropriate insoluble substrate to immobilize the enzyme. The developed biosensor showed a detection limit of $0.1{\mu}M$ hydrogen peroxide. The enzyme reactor was prepared with diamine oxidase immobilized on insoluble carriers including CNBr-activated sepharose 4B, calcium alginate, and controlled pore size glass beads. The coupling efficiency of CNBr-activated sepharose 4B, calcium alginate, and controlled pore size glass beads were 48.5%, 40.3%, and 51.0%, respectively. In addition, the response currents on histamine with each immobilized enzyme reactor prepared with CNBr-activated sepharose 4B, calcium alginate, and controlled pore size glass beads were 120 nA, 110 nA, and 140 nA at $100{\mu}M$ of histamine concentration, respectively. Therefore, it is suggested that controlled pore size glass beads are the best carriers for immobilizing diamine oxidase to detect histamine in this biosensor.

  • We developed a microfluidic immunoassay platform for the detection of various analytes such as bacterial pathogen by packing antibody-immobilized glass beads in spatially-isolated microchambers on a microfluidic device. Primary amines of antibody were covalently conjugated to carboxyl-terminated glass beads previously treated with aminosilane followed by glutaraldehyde. Through this covalent binding, up to 905 $\mu$g immunoglobulin G (IgG) per gram of glass beads was immobilized. For application, glass beads attaching antibody specific to Escherichia coli O157:H7, a foodborne pathogen, were packed into a microfluidic device and used for the detection of the serotype. This prototype immunoassay device can be used for the simultaneous detection of multiple analytes by sequentially packing different-sized glass beads attaching different antibody in discrete microchambers on a single microfluidic device.

  • The application of 3D printing technology is expanding due to the production of the complex-shape parts and the one-step manufacturing process. Moreover, various technical solutions in 3D printing are emerging through continuous research and development. Representative technologies include SLS technology, in which a desired area is sintered and laminated by irradiating a powder-type material with a laser. In addition, high-performance engineering plastic parts are being manufactured in increasing numbers. In this study, tensile specimens were fabricated from polyamide 12, a widely available polymer, and the glass bead-reinforced polyamide 12. The specimen-build orientation was divided into 0°, 45°, and 90° on the fabrication platform, and the tensile test temperature was -25℃, 25℃, and 60℃. The test results showed that the tensile modulus of both materials decreases as the build orientation becomes closer to 90°. In addition, the tensile strength of glass bead-reinforced PA12 showed more dependence on the build orientation than PA12. In addition, the tensile modulus and tensile strength decreased with increasing test temperature.

  • Open beaker digestion method is routinely used as the sample preparation technique for trace element determination of rock samples by inductively coupled plasma mass spectrometry, With this method, however, dissolution of Zr and Hf is not always guaranteed especially when the samples contain refractory minerals. In this study, glass bead digestion technique was compared with conventional open beaker digestion technique for the sample preparation of three USGS rock standards such as AGV-2, BHVO-2, and G-3. Thirty trace elements including rare earth elements were analysed by ICP-MS and ICP-AES. There were no clear differences in analytical results for the AGV-2 and BHVO-2 standards between the two techniques, but Zr, Hf, Y, and middle- to heavy- rare earth element concentrations of the G-3 standard prepared by open beaker digestion technique were significantly lower than the recommended values. This can be attributed to the presence of refractory mineral zircon. On the contrary, all the analytical results of the G-3 standard prepared by glass bead digestion technique were in good agreement with the recommended values, indicating complete dissolution of zircon. The analytical results show that the volatile elements such as Pb and Zn were not lost during the preparation of glass bead. Low dilution glass bead digestion technique described here will be very helpful to enhance precision and accuracy of trace element analysis for geological samples containing refractory minerals.

  • Optimal bactericidal conditions of pilot scale UV-$TiO_2$ photoreractor were studied. As the inner diameter of reactor increased. the bactedcidal efficiency decreased. Similarly bactedcidal effect was elevated according to the higher concentration of $TiO_2$. however. the effect was not repressed by the highest concentration(6.000 $mg/{\ell}$)of $TiO_2$ Bactericidal effect of muscovite bead was higher than that of glass bead. When bacterial cells were applied to the photoreacter for 1. 5. and 15 min, bactericidal effects were 62, 94.3. and 99.8%, respectively. When 30 $mg/{\ell}$ of $H_2O_2$ was added to the reaction mixture and sterilized for 5 min, the bactericidal efficiency was 99.8%.

  • The extensive archaeological remains at the Baekje Kingdom site of Wanggung-ri, Iksan, in the South Jeolla Province, Korea, include a seventh century workshop area that was later covered by the construction of a Buddhist temple. Remains of glass, gold, and bronze artefacts, and the ceramic crucibles used in the working of these materials, provide evidence for a multi-craft, high-temperature technology. We will report the results of both chemical compositional and lead isotope analyses for Wanggung-ri glass, which is among the earliest lead-silica glass type in East Asia, as well as review the evidence for primary glass production at Wanggung-ri.

  • Here, we demonstrated simple nucleic acid, RNA, concentration method using polymer micro chip containing glass bead ($100\;{\mu}m$). Polymer micro chip was fabricated by PDMS ($1.5\;cm\;{\times}\;1.5\;cm$, $100\;{\mu}m$ in the height) including pillar structure ($160\;{\mu}m\;(I)\;{\times}\;80\;{\mu}m\;(w)\;{\times}\;100\;{\mu}m\;(h)$, gap size $50\;{\mu}m$) for blocking micro bead. RNA could be adsorbed on micro glass bead at low pH by hydrogen bonding whereas RNA was released at high pH by electrostatic force between silica surface and RNA. Amount of glass beads and flow rate were optimized in aspects of adsorption and desorption of RNA. Adsorption and desorption rate was measured with real time PCR. This concentrated RNA was applied to amplification micro chip in which NASBA (Nucleic Acid Sequence Based Amplification) was performed. As a result, E.coli O157 : H7 in the concentration of 10 c.f.u./10 mL was successfully detected by these serial processes (concentration and amplification) with polymer micro chips. It implies this simple concentration method using polymer micro chip can be directly applied to ultra sensitive method to measure viable bacteria and virus in clinical samples as well as environmental samples.

  • Despite significant effects on macroscopic migration and distribution of CO2 injected during geological sequestration, only limited information is available on wettability in microscopic scCO2-brine-mineral systems due to difficulties in pore-scale observation. In this study, a micromodel had been developed to improve our understanding of how scCO2 flooding and residual characteristics of porewater are affected by the wettability in scCO2-water-glass bead systems. The micromodel (a transparent pore structure made of glass beads and glass plates) in a pressurized chamber provided the opportunity to visualize scCO2 spreading and porewater displacement. CO2 flooding followed by fingering migration and dewatering followed by formation of residual water were observed through an imaging system. Measurement of contact angles of residual porewater in micromodels were conducted to estimate wettability in a scCO2-water-glass bead system. The measurement revealed that the brine-3M NaCl solution-is a wetting fluid and the surface of glass beads is water-wet. It is also found that the contact angle at equilibrium decreases as the pressure decreases, whereas it increases as the salinity increases. Such changes in wettability may significantly affect the patterns of scCO2 migration and porewater residence during the process of CO2 injection into a saline aquifer at high pressures.
