Replacing critical point drying with a low-cost chemical drying provides comparable surface image quality of glandular trichomes from leaves of Millingtonia hortensis L. f. in scanning electron micrograph

  • Sample preparation including dehydration and drying of samples is the most intricate part of scanning electron microscopy. Most current sample preparation protocols use critical-point drying with liquid carbon dioxide. Very few studies have reported samples that were dried using chemical reagents. In this study, we used hexamethyldisilazane, a chemical drying reagent, to prepare plant samples. As glandular trichomes are among the most fragile and sensitive surface structures found on plants, we used Millingtonia hortensis leaf samples as our study materials because they contain abundant glandular trichomes. The results obtained using this new method are identical to those produced via critical-point drying.

  • Pulmonary papillomas are rare benign epithelial neoplasms arising in bronchial surface epithelium. They are categorized by a variety of cell types including squamous, glandular, and mixed squamous and glandular type. Among them, glandular papilloma is extremely rare and has not been reported in Korea. The patient was a 52 year-old man presenting with a 4-months' history of recurrent hemoptysis. Bronchofiberoscopy revealed a whitish, glistening, and polypoid mass lesion at the proximal bronchus in the basal segment of the left lower lung. Bronchoscopic biopsy was performed; papillary fronds lined by ciliated or nonciliated pseudostratified columnar epithelium were noted on histologic findings. We present the first case of glandular papilloma in Korea. Two years later, the patient visited our hospital again due to hemoptysis. On follow-up bronchoscopy, a mass that had been found previously showed an increase in size.

여성형유방증 치료에 있어서 절제술 전 초음파 지방흡입술의 역할 (The Role of Ultrasound-Assisted Liposuction before a Surgical Excision in the Treatment of Gynecomastia)

  • Purpose: The method of using ultrasound-assisted liposuction and excision of the remaining glandular tissue is the preferred method for treating gynecomastia and is currently used worldwide. Herein, this article described the role of ultrasound-assisted liposuction before a surgical excision in the treatment of gynecomastia. The cosmetic results were objectively evaluated. Methods: 11 patients (22 breasts) underwent ultrasoundassisted liposuction and suction-assisted lipectomy between April 2007 and January 2009. At the end of the liposuction, the remaining glandular tissue was removed through the incision used for liposuction. We evaluated the cosmetic results using ordinary scale methods on the basis of four categories (recurrence, symmetry, contour irregularity, and scar). Results: The volume of aspirates ranged between 50 and 200 cc per breast and the average weight of tissue removed by excision was 65g per breast. No complications were recorded. Regarding the cosmetic evaluation, the recurrence, contour irregularity, and scar were excellent, the symmetry was good, and the overall results represented all those cases were mostly excellent. Conclusion: Ultrasound-assisted liposuction has many advantages in the treatment of gynecomastia. When excising the remaining glandular tissue, bleeding is decreased by the use of a tumescent technique. The glandular tissue is easily mobilized and excised after being "morselized" with ultrasound-assisted liposuction. The glandular tissue is simply dissected via the suction surface. Compared the residual mound of glandular tissue beneath the nipple and areola to the periphery, it facilitates precise control of the excision.

히스타민 유도 넙치(Paralichthys olivaceus) 위선부의 확장성 내강에 대한 조직학적 관찰 (Histamine-induced dilation of gastric glandular tubules of flounder, Paralichthys olivaceus)

  • 본 실험은 양식 넙치에서 임상적으로 빈번히 발생하는 위선세관의 확장성 변화가 위축성 변화인지를 알기 위하여 넙치의 정상 위선구조를 분명히 하는 한편 확장성 위선세관에 대하여 광학 및 전자현미경적 소견을 추구하고, 위액분비를 촉진하는 물질로 알려진 히스타민을 주사한 후 그 형태학적인 변화 내용을 서로 비교검토하였다. 외관상 건강한 넙치의 위선세관을 포함한 위선부의 조직학적 구조에 있어서는 이미 밝혀진 다른 어종과 큰 차이가 없었다. 광학현미경상에서 양식 넙치는 위선세관의 내강이 확장되고 소관 상피의 세포질의 양이 줄어든 위축성 변화를 보였고, 히스타민 주사 넙치에서도 유사한 소견을 보였다. 그러나 semi-thin 표본상에서 확장된 위선세관의 내강은 밝은 영역으로 나타났고, 이 밝은 영역은 주자전자현미경상에서 가늘고 긴 다수의 세포질 돌기로 이루어진 것임을 알 수 있었다. 따라서 광학현미경 하에서 관찰되는 위선세관의 확장성 변화는 위축성 병변이기 보다는 위액분비활동 증가 내지 과다에 따른 형태학적 변화의 하나인 것으로 추정되었다.

Ambient Variable Pressure Field Emission Scanning Electron Microscopy for Trichome Profiling of Plectranthus tomentosa by Secondary Electron Imaging

  • Glandular and nonglandular trichomes on the leaf surface of Plectranthus tomentosa were investigated by variable pressure field emission scanning electron microscopy (VP-FESEM). The segments of the plant's leaves were directly mounted without any specimen preparation, and examined at ambient temperature using a variable pressure secondary electron (SE) detector under ca. 15 Pa. Foliar trichomes maintained their shapes and structures without severe surface collapse or charging. The adaxial leaf surface was abundantly covered with different types of trichome. Nonglandular trichomes consisted of a basal cell and a long (up to ca. $300{\mu}m$) stalk. Meanwhile, capitate glandular trichomes had a secretory head and a short or long stalk. Peltate glandular trichomes with globose secretory heads were observed in close contact with the leaf epidermis. Spherical projections on the secretory head showed the secretion process of glandular trichomes. In addition to the trichomes, oval stomata were distributed on the abaxial leaf surface. These results suggest that ambient VP-FESEM can be used to classify the dehydration-sensitive foliar trichomes of succulent plants by SE imaging. At the FESEM resolution, this approach facilitates the rapid and detailed morphological analysis of a variety of trichomes in diverse plant taxa with reduced labor and preparation.

디지털 유방확대촬영술에서 Air gap technique의 유용성 평가 (Evaluation of the Usefulness for Air Gap Technique in Digital Magnification Mammography)

  • 이 연구는 디지털 유방확대촬영술에서 방사선 피폭을 감소시키며 영상의 질을 유지하기 위한 최적의 촬영조건을 알아보았다. 자동노출방식의 평균유선선량이 수동노출방식에 비해 높은 것으로 나타났다. 자동노출방식은 그리드(grid) 사용 여부에 따라 평균유선선량과 화질평가에 차이가 많았으나, 수동노출방식은 팬텀영상평가 점수에서만 차이가 있었다. 그러나 air gap technique을 사용할 경우 모든 노출방식에서 신호대잡음비의 증가가 뚜렷하게 나타났다. 따라서 유방확대촬영 시 air gap technique을 사용한다면 환자의 평균유선선량을 감소시키고 신호대잡음비를 높여 양질의 화질을 유지할 수 있을 것이다.

Fine Structure of the Glandular Epithelium during Secretory Silk Production in the Block Widow Spider Latrodectus mactans

  • Among the silk glands in the black widow spider Latrodectus mactans, the ampullate one is the most predominant gland in both sexes, and is com-posed of three functional parts - excretory duct, storage ampulla and convoluted tail regions. This experiment was performed using mechanical pulling stimulation with electric motor equipment to reveal a correlation between silk usage and silk producing system in this poisonous spider. The mature secretory products in glandular epithelium are closely packed and appear as electron-opaque spherical vesicles. A part of the vesicles with fine fibrillar paracrystalline texture seems to store some proteins which will function at the time of final assembly into fibrils. Most of the secretory silk products which originated from the rough endoplasmic reticula of the glandular epithelial cells are grown by fusion with surrounding small vesi-cles. However, the Golgi complex does not seem to play an important role in this process of secretion. According to progressive maturation of secre-tory silk product, these granules are progressively filled with a fine fibrillar material, and thus appear much more electron-dense than those of earlier states. When the secretory product is extruded from the glandular cavity, the epithelium is rapidly changed to a thinner layer of tall columnar cells with less definitive cell membranes. After extruding there ave a few secre-tory droplets within these cells, thus causing this region to stain much lighter.

선양치성낭의 임상 및 병리조직학적 분석 (CLINICAL AND HISTOPATHOLOGIC ANALYSIS OF GLANDULAR ODONTOGENIC CYSTS OF THE JAWS)

  • The glandular odontogenic cyst is an uncommon odontogenic cyst as a distinct entity. We reviewed a series of 7 glandular odontogenic cysts of the jaws experienced between 2003 and 2006 at the department of Oral and Maxillofacial surgery, Chosun university. The study group consisted of 3 females (42.9%) and 4 males (57.1%), with an age range of 31 to 75 years and mean age was 58.6 years. The maxilla was involved in 5 cases (71.4%) and the mandible in 2 cases (28.6%). Three cases involved impacted tooth. Clinically 6 cases showed swelling and tenderness. All the lesion presented well-defined unilocular radiolucent lesion radiographically. Provisional clinical diagnosis was varied, incisional biopsy was done in 1 case. Histopathologically, those were lined by non-keratinized stratified epithelium and thickened epithelial segments (plaques) are seen within the lining epithelium. And epithelial lining contains eosinophilic cuboidal type cells, mucous cells and mucin pools in microcystic areas are identified. All cysts were treated by enucleation. All cases are not recurred during follow up period.

Pelargonium peltatum 분비모의 미세구조 연구 (Ultrastructural study of Glandular Trichomes in Pelargonium peltatum)

  • 제라늄(Polargonium peltatum) 분비모의 미세구조를 광학현미경과 주사전자현미경을 사용하여 살펴보았다. Pelargonium peltatum의 분비모는 형태와 크기에 따라 short-stalked capitate glands (Type I)와 long-stalked capitate glands (Type II)의 두 가지 종류로 구분할 수 있었다. 두 종류의 분비모는 모두 배축면의 엽백과 엽병에 많이 분포하였다. 분비모는 1개의 분비세포, 3개의 병세포, 1개의 기부 세포로 이루어져 있었으며, 분비세포는 다수의 활면소포체와 색소체, 액포, 골지장치, 미토콘드리아 등을 가지고 있었다. 분비세포의 액포 내에서는 전자밀도가 높은 물질이 자주 관찰되었는데, 이들은 분비모의 주된 분비전구물질인 phenolic compound인 것으로 사료된다.
