A pathogenesis-related protein (PgPR5) gene that isolated from the leaf of Panax ginseng was characterized. The ORF is 756 bp with a deduced amino acid sequence of 251 residues. The calculated molecular mass of the matured protein is approximately 27.5 kDa with a predicated isoelectric point of 7.80. A GenBank BlastX search revealed that the deduced amino acid of PgPR5 shares highest sequence similarity to PR5 of Actinidia deliciosa (80% identity, 87% similarity). PgPR5 has a C-terminal and N-terminal signal peptide, suggesting that it is a vacuolar secreted protein. The expression of PgPR5 under various environmental stresses was analyzed at different time points using real-time PCR. Our results reveal that PgPR5 is induced by salt stress, chilling stress, heavy metal, UV, and pathogen infection. These results suggest that the PgPR5 could play a role in the molecular defence response of ginseng to abiotic and pathogen attack. This is the first report of the isolation of PR5 gene from the P. ginseng.

The cDNA of alcohol dehydrogenase (PgADH) was isolated and characterized from the leaf of Panax ginseng. The cDNA had an open reading frame of 801 bp and a deduced amino acid sequence of 266 residues. The calculated molecular mass of the mature protein is approximately 29 kDa with a predicated isoelectric point of 6.84. Homology analysis revealed that the deduced amino acid of PgADH shares a high degree of homology with the short-chain ADH proteins of other plants. Genomic DNA hybridization analysis indicated that PgADH represents a multi-gene family. The expression of PgADH under various environmental stresses was analyzed at different time points using real-time PCR. ABA, SA and especially JA (80-fold) significantly induced PgADH expression within 24 h of treatment. The positive responses of PgADH to abiotic stimuli suggest that ginseng ADH may protect against hormone-related environmental stresses.
Five ginsenosides $(A_1,\;A_2,\;B_1,\;B_2,\;C)$ and a yellow pigment were isolated from American ginseng stems and leaves. Ginsenoside $A_2,\;B_1,\;B_2$ and C were proven to be identical with Korean ginseng root ginsenoside $Rg_1,$ Rd, Re and $Rb_2,$ respectively. The yellow pigment proved identical with panasenoside isolated from Korean ginseng leaves. Ginsenoside $A_1$, which was also present in American ginseng roots, was not identical to any of the known root (ginsenoside $R_{0}-Rg_{2}$) and leaf (ginsenoside $F_{1}-F_{3}$) Korean ginseng saponins. A gas-liquid chromatographic method was developed to analyze ginsenosides and sapogenins in rabbit plasma and urine samples. Panasenoside and stigmasterol were found to be the best internal standards for ginsenosides and sapogenihs, respectively. Ginsenoside C had a significantly longer half-life, higher plasma protein binding, lower metabolic and renal clearance than ginsenoside $A_1,\;A_2\;and\;B_2$. Ginsenosides were not found in rabbit plasma and urine samples after oral administration. Ginsenoside C had a higher toxicity than ginsenoside $A_2$ after intraperitoneal administration to mice. Toxicity was not observed after oral administration of the ginsenosides.
Panax ginseng leaves are produced as the by-product when Panax ginseng roots were harvested. The Panax ginseng leaves was examed for the applicable possibility as the functional food. In this study, the changes in chemical composition of Panax ginseng leaves was examed by three methods as the hot-air dried(DRT), the aged tea(AGT) and the heat processed tea(HPT). The general composition of Panax ginseng leaves tea was shown as similar results in 3 different process methods. The level of the crude lipid and reducing sugar concentration were decreased slightly in HPT. The free sugar content of DRT was higher than the HPT and AGT. The existence of the higher content of free sugar composition in order are sucrose, fructose and glucose. The concentration of serine was the highest in the free amino acids, which were shown from 309.6 mg% to 336.6 mg%. The contents of free amino acid in Panax ginseng leaves made by DRT was higher than by AGT and HPT. The concentration of Ca was shown as the highest content among the minerals and was 2,115 mg%. The contents of minerals were existed in order of Ca, K, Mg, P, Na, Mn, Fe, Zn and Cu. But there were hardly any remarkable differences of mineral concentrations of Panax ginseng leaves tea made by different processing methods. The concentration of water soluble solid of Panax ginseng leaves tea processed by HPT was higher than by DRT and AGT. The concentration of ascorbic acid was shown the highest value of 424.4mg% in HPT. There was no differences in the fatty acid composition according to their processing methods. The concentration of palmitic acid was higher than that of other fatty acid. The order of fatty acid concentration were palmitic aicd, linoleic acid, linolenic acid, oleic acid and stearic acid, abundantly. As a conclusion, HPT was shown as the best process method for the production of Panax ginseng leaves tea.
Weed control in the Korean ginseng(Panax ginseng C.A. Meyer) garden is more difficult than in other crops because the ginseng is a perennial crop and has to be cultivated under the artificial shading. As hand weeding is the only practical means to control weed after crop establishment, a selective herbicide would greatly simplify the control of weed in ginseng garden. In an exploratory experiment, the hervicide Fluazifop-butyl was found to be selectively safe for the Korean ginseng plants. Various rates of Fluazifop-butyl were sprayed on 2-, 3-, and 4-years old ginseng plants as a foliar spray to detect crop injury and to difine their critical concentration. No apparent injury to the ginseng plant was noticable even the doubled application rate of Fluazifop-butyl the based on recommended dosage. Neither abnormal foliar change nor any inhibition in leaf and stem growth was resulted for 2-, 3-, and 4-years old ginseng plants treated with Fluazifop-butyl tripled the recommended usage. The foliar treatments of Fluazifop-butyl did not influence the photosynthesis ability but inhibited the respiration of the ginseng leaf.
This study aims to establish the directions for the optimal nutrient contents determined by soil and leaf analyses. The study is to be used as a source for basic information in investigating the effects of nutritional environment on ginseng growth at the fields of Ansung region in Gyeonggi province. The physical property of soil porosity is closely related to ginseng growth, with more than 50% of good growth in the field. The optimal range of chemical properties in soil was analyzed as 0.20-0.28% T-N, 500-900 mg/kg Av.P$_2$O$_5$, and 2.3-3.5 cmol$^+$/kg Exch.Ca. The optimal range of inorganic nutrient contents in leaves was also analyzed. P value was less than 0.25%, and Mg was more than 0.22%. Other elements were not found clearly. The ratios of N/P, N/Mg, K/Mg, and Ca/P of leaves with good growth in the field showed 10<, 10-13, <14, 1<, respectively. In addition, the Ca:Mg:K ratio of the Exch.cation (cmol$^+$/kg) may be useful as an indicator in the assessment of plant growth in ginseng.
This study was conducted to define the effects of light intensity on the amount of saponin and free sugar and the ratio of triol group saponin and diol group saponin (PT/PD) in the leaf of Panax ginseng C.A. Meyer. 4-Year-old ginseng plants were grown under the shadings of different light transmittance rate(LTR) of 5%, 10%, 20% and 30% for 5 months and the leafiets were sampled from 2nd low at late August to determine the amount of saponin and free sugar.-Rd was main ginseuoside in the diol group saponin but in triol group saponin, ginsenoside-Re showed highest value and next was ginsenoside-$Rg_1$ and $Rg_2$ respectively. Up to 20% of light transmittance rate (LTR), the ginseng leaves grown under high light intensity showed an increase in the amount of total saponin and the ratio of PT/PD but the amount of total saponin and the ratio of PT/PD but the amount of total saponin and the ratio of PT/PD was decreased at the ginseng leaves grown under the shading of 30% LTR. The ginseng leaves grown under the shading of 20% LTR showed a significant increase in the amount of glucose and fructose but a significant decrease of sucrose content. A significant positive correlation ($r=0.992$^{**}$) was recognized between the of amount of total saponin and glucose.
This experiment was carried out to study the influence of the various soil water regimes on photosynthesis and growth and development of ginseng plant (3 years). The results were as follows: optimum soil water content for root dry weight and diameter appeared to be 62% of field capacity (13.9% fresh weight basis). The 62% field capacity showed superiority in leaf area, leaf dry weight and also in number of flower, fruit, seed per plant. Net photosynthesis rates per unit area increased with increasing soil water content but net photosynthesis rates per plant were superior in 62% field capacity. It is considered to control of the proper condition of temperature and humidity during the yellowing.
