Metabolomic approach for discrimination of processed ginseng genus (Panax ginseng and Panax quinquefolius) using UPLC-QTOF MS

  • Discriminating between two herbal medicines (Panax ginseng and Panax quinquefolius), with similar chemical and physical properties but different therapeutic effects, is a very serious and difficult problem. Differentiation between two processed ginseng genera is even more difficult because the characteristics of their appearance are very similar. An ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF MS)-based metabolomic technique was applied for the metabolite profiling of 40 processed P. ginseng and processed P. quinquefolius. Currently known biomarkers such as ginsenoside Rf and F11 have been used for the analysis using the UPLC-photodiode array detector. However, this method was not able to fully discriminate between the two processed ginseng genera. Thus, an optimized UPLC-QTOF-based metabolic profiling method was adapted for the analysis and evaluation of two processed ginseng genera. As a result, all known biomarkers were identified by the proposed metabolomics, and additional potential biomarkers were extracted from the huge amounts of global analysis data. Therefore, it is expected that such metabolomics techniques would be widely applied to the ginseng research field.

  • Background : Ginseng is mainly grown as a break crop in paddy fields after rice has been cultured for approximately 4 - 5 years, because it reduces the negative effects of continuous rice cropping. However, physiological disorders, such as leaf discoloration, occur in ginseng grown in paddy fields with poor drainage and excessive levels of inorganic components. Methods and Results : This study investigated the effect of ridge height on the growth characteristics and yield of 6 year old Panax ginseng. Ridge height was varied by making 20, 30, and 40 cm high ridges in a pooly drained paddy field. Soil moisture content decreased, while electrical conductivity (EC) as the ridge height increased. The $NO_3$, K, Ca, Mg, and Na levels also rose as ridge height increased, but organic matter and $P_2O_4$ levels did not. The leaf discoloration ratio rose as the ridge height increased, and root yield reached a peak when the ridge height was 30 cm. Conclusion : A ridge height of 30 cm in poorly drained paddy field improved ginseng growth by reducing leaf discoloration and increasing root survival, owing to more suitable soil moisture and EC levels.

  • To investigate the effect of light intensity on the growth of shoots and roots, water and chlorophyll contents in the ginseng leaf were determined at 5%, l0%, 20%, 30% light transmittance rate (LTR) with 1,2 and 4 year-old ginseng plants in the field. Stem length, size of the leaf, water and chlorophyll contents were decreased as the increase of the light intensity in all ages of ginseng plants. The degree of decrease was severe in the one year-old ginseng compared to that of 2, or 4 year-old ginseng, while there was no difference between the 2 and 4 year-old plants. Root weight per plant was highest at 5% LTR in the one year-old plants, while it was at 20% LTR in the 2 and 4 year-old plants. Generally, demand of light for the growth of one year-old ginseng was lower than those of 2 or 4 year-old plants there was no difference of it among the ginseng plants older than 2 years.

  • American ginseng (Panax quinquefolium) is herbaceous perennial plants indigenous to North American forests. This is highly valued as medicinal herbs with a long history of collection from wild populations since 1716. Wild American ginseng distributed from Quebec in Canada to northern Florida in USA. A heavy concentration is found in the Appalachian mountains, although wild American ginseng is considered endangered. The price paid for field cultivated ginseng has dropped dramatically in the past 10 years, while the price for wild or woods cultivated ginseng has rised significantly. The price curve for ginseng resembles a roller coaster, reflecting not only supply and demand but many other factors. This information will be useful to understand American ginseng compared to Korean ginseng.

석회류제의 시용이 엽연형 황피발생 억제에 미치는 영향 (Effects of Some Liming Materials Application on the Alleviation of the Symptoms of Mn Toxicity Inculding Marginal Leaf Chlorosis)

  • Effect of liming material application on the prevention or cure of Mn toxicity symptoms including marginal leaf chlorosis in 3-year-old ginseng plants grown in acidic soils were investigated. It was found that the ratio of Fe to Mn was above about 0.5 and the ratio of Mn to Fe was below about 2.00 in 4-year old ginseng leaves when liming materials were applied in field experiments. It was apparent that the occurrence of marginal leaf chlorosis was decreasing affected by application of Ca, Mg and Fe. The appearance of marginal leaf chlorosis was decreased to about 78 percent in 4-year-old ginseng and to about 69 percent in 5-year-old ginseng, and then the fresh root weight was increased up to 66 percent in 4-year ginseng plants.

사료작물 윤작재배에 의한 연초포지의 제염효과 (Soil Desalinization by Pasture Crops in Tobacco Field)

  • The field experiment for field desalinization by precultivation of orchard crops were carried out to evaluate relationship between the varieation of chlorine contents of soil and crop uptake in the upland diverted from paddy field. After harvest of grass crops, soil samples were taken for analysis of chlorine contents of soil layers. Regardless of kinds of grass crops cutivated, contents of soil chlorine were decreased comparing to non-crop plot. Chlorine content in plant harvested at just before the flowering stage was much higher than that of after flowering. Chlorine uptake and dry matter were increased in order of Italian ryegrass, Perennial ryegrass, and Sudan grass. Positive correlations were showed between chlorine uptake and dry matter. The content of soil chlorine decreased by higher yield of dry matter.

Characteristics of Absorption and Accumulation of Inorganic Germanium in Panax ginseng C. A. Meyer

  • The characteristics of absorption and accumulation of inorganic germanium in Panax ginseng C. A. Meyer were examined. In 4-year-old P. ginseng, the germanium content of the field soil increased with increased amounts and frequencies of inorganic germanium application, while chemical components of the soil, such as available phosphate and exchangeable calcium, potassium, and magnesium, decreased with the increased inorganic germanium application. In the 4-year-old P. ginseng, the germanium content was highest in the rhizome and increased in the order of stem, leaf, lateral root, and main root, suggesting that inorganic germanium was absorbed from the root and translocated to the stem and leaf via the rhizome. As for changes in ginsenosides in 4-year-old P. ginseng rhizomes, the contents of ginsenosides $Rb_1$, $Rb_2$, Re, and Rf decreased as the germanium content in soil increased. Ginsenosides $Rb_1$, $Rb_2$, Rc, Re, and Rf in the main root also decreased with increasing germanium content in the main root. The results suggest that inorganic germanium treatment may increase organic germanium in harvested P. ginseng, thus enhancing the medicinal effi cacy of ginseng products.

A Study on the Current Status of Mountain-Grown Ginseng

  • For the current study, 2,000 questionnaire sheets were printed and distributed while at the same time postal questionnaires were also conducted. The questionnaire survey was conducted July 25 through September 25, 2008, whereby 206 copies of desirable responses were secured. Analysis of the survey made it possible to grasp the overall current status and prospects of the mountain-grown ginseng business, and the level of technology required for cultivating mountaingrown ginsengs. It was learned that, with no legal and institutional arrangements now in force, no precise facts and figures concerning the total area cultivated and the quantity produced are currently available, and that the products are being marketed under the table. Under such circumstances, it is high time for the mountain-grown ginseng cultivation business to contribute to the promotion of incomes of the farming households and the generation of national wealth by developing the business into a systematic industry. This study conducted a survey on the current status of mountain-grown ginseng producers and of their production, thereby contributing to the introduction of policies for mountain-grown ginsengs.
