Effect on Gene Expression Profile of Rat Hippocampus Caused by Administration of Memory Enhancing Herbal Extract (육미지황탕가미방이 흰쥐의 기억능력과 중추신경계 유전자 발현에 미치는 영향)

  • The herbal extract (YMT_02) is a modified herbal extracts from Yukmijihwang-tang (YMJ) to promote memory-enhancing. The YMJ extracts has been widely used as an anti-aging herbal medicine for hundred years in Asian countries. The purpose of this study is to; 1) quantitatively evaluate the memory-enhancing effect of YMT_02 by behavior task, 2) identify candidate genes responsible for enhancing memory by cDNA microarray and 3) assess the anti-oxidant effect of YMT_02 on PC12 cell. Memory retention abilities are addressed by passive avoidance task with Sprague-Dawley (SD) male rat. Before the training session, the rats are subdivided into four groups and administrated with YMT_02, Ginkgo biloba, Soya lecithin and normal saline for 10 days. The retention test was performed. 24 hours after the training session. The retention time of the YMT_02 group was significantly (p<0.05) delayed (~100%), whereas Ginkgo biloba and Soya lecithin treatment delayed 20% and 10% respectively. The hippocampi of YMT_02 and control group were dissected and mANA was further purified. After synthesizing cDNA using oligo-dT primer, the cDNA were applied to Incyte rat GEMTM 2 cDNA microarray. The microarray results show that prealbumin(transthyretin), phosphotidylethanolamine N-methyltransferase, and PEP-19 are expressed abundantly in the YMT_02 treated group. Especially, PEP-19 is a neuron-specific protein, which inhibits apoptotic processes in neuronal cell. On the other hand, transcripts of RAB15, glutamate receptor subunit 2 and CDK108 are abundant in control group. Besides, neuronal genes involved in neuronal death or neurodegeneration such as neuronal-pentraxin and spectrin are abundantly expressed in control group. Additionally, the YMT_02 shows an anti oxidative effect in the PC12 cell. The list of differentially expressed genes may implicate further insight on the action and mechanism behind the memory-enhancing effect of herbal extracts YMT_02, for example, anti-apoptotic, anti-oxidative, and neuroprotective effects.

Effect of Phytogenic Feed Additives in Soybean Meal on In vitro Swine Fermentation for Odor Reduction and Bacterial Community Comparison

  • The effect of different phytogenic feed additives on reducing odorous compounds in swine was investigated using in vitro fermentation and analyzed their microbial communities. Soybean meal (1%) added with 0.1% different phytogenic feed additives (FA) were in vitro fermented using swine fecal slurries and anaerobically incubated for 12 and 24 h. The phytogenic FAs used were red ginseng barn powder (Panax ginseng C. A. Meyer, FA1), persimmon leaf powder (Diospyros virginiana L., FA2), ginkgo leaf powder (Ginkgo biloba L., FA3), and oregano lippia seed oil extract (Lippia graveolens Kunth, OL, FA4). Total gas production, pH, ammonianitrogen ($NH_3$-N), hydrogen sulfide ($H_2S$), nitrite-nitrogen ($NO_2{^-}$-N), nitrate-nitrogen ($NO_3{^-}$-N), sulfate (${SO_4}^{--}$), volatile fatty acids (VFA) and other metabolites concentration were determined. Microbial communities were also analyzed using 16S rRNA DGGE. Results showed that the pH values on all treatments increased as incubation time became longer except for FA4 where it decreased. Moreover, FA4 incubated for 12 and 24 h was not detected in $NH_3$-N and $H_2S$. Addition of FAs decreased (p<0.05) propionate production but increased (p<0.05) the total VFA production. Ten 16S rRNA DGGE bands were identified which ranged from 96 to 100% identity which were mostly isolated from the intestine. Similarity index showed three clearly different clusters: I (FA2 and FA3), II (Con and FA1), and III (FA4). Dominant bands which were identified closest to Eubacterium limosum (ATCC 8486T), Uncultured bacterium clone PF6641 and Streptococcus lutetiensis (CIP 106849T) were present only in the FA4 treatment group and were not found in other groups. FA4 had a different bacterial diversity compared to control and other treatments and thus explains having lowest odorous compounds. Addition of FA4 to an enriched protein feed source for growing swine may effectively reduce odorous compounds which are typically associated with swine production.

  • This study was conducted to investigate the plants status of the landscape plant species of 10 elementary schools in the Chungju city, and to obtain the fundamental informations for proper use of planting system in the elementary school landscape. The results are summarized as follows : 1. 199 species were planted in the 10 elementary schools, It was constituted 89 of woody plants, 108 of herbaceous plants and 2 of others. The average number of plants per school was about 60 species. The rate of plants species of Evergreen Trees to the Deciduous was 23 : 60 and the rate the trees to the shrubs was 59 : 24. 2. Almost the same species were planted in school landscaping zone. Juniperus chinensis var. kaizuka, Ginkgo biloba, Zelkova serrata were mainly planted in the species of trees. To improve the present conditions of school landscape should be considered for the purpose of function and the aspect of ecology. 3. To see the selected school trees and flowers, Zelcova serrata was selected to the school tree in the 4 of schools. Ginkgo biloba was selected 3 of schools, the others were Juniperus chinensis Pinus densiflora, etc. Rhododendron lateritium was appointed to the school flower in the 6 of schools, the others were Rosa spp., Forsythia koreana etc. To set the school tree and flower consider the preserved trees of the community.

  • Ten to thirty percent of ginko nut shell(GNS) were added to poplar sawdust to investigate mycelial growth. Mycelial growth on the medium with GNS was 93~97(mm/28days) and was slower than that of the control without GNS as that of 101(mm/28 days). Mycelial growth time on medium of sawdust with 10% GNS was same with control without GNS as 28 days but that on medium with 20 to 30% GNS was delayed for 2 days. The time of pinhead formation of sawdust with 10 to 30% GNS was 7 days, the growth time to harvest was 5 days. Yields per volume of GNS added treatments were 80.5g to 91.8g, which was increased 14 to 30% compared to 70.8g of the control without GNS.

Effect of $SO_2$ on Leaf Tip Blight of Ginkgo biloba caused by Coniothyrium sp. (은행나무의 잎끝마름병에 미치는 $SO_2$ 의 영향)

  • To examine the relationship between air pollution and occurrence of disease in plants, we investigated the alteration of physiology and biochemistry of Ginkgo biloba by inoculating with Coniothyrium sp. and fumigating with 0.1 ${\mu}l/l\;SO_{2-}$ Visual damage did not appear but photosynthesis, $CO_2$ use efficiency(CUE) and water use efficiency(WUE) were reduced when G. biloba was exposed to $SO_{2-}$for 7days (8 hours a day). When inoculated with Coniothyrium sp., the seventies of damage in G. biloba showed a threefold difference between $SO_{2-}$ treatment (SI) and $SO_{2-}$ free treatment (SFI) at day 42 from initial inoculation. Little difference was observed in sugar contents that may be used pathogens feed, among control, SFI and SI. In spite of the reduction in photosynthetic rate, sugar contents and CUE were maintained. WUE was enhanced 13% more at SI than SFI. The $CO_2$fixation boosted because of enhanced WUE, and thus sugar synthesis was not affected. In addition, sugar transport seems to be retarded for some internal alteration. Consequently, the severity of SI was more serious than that of SFI because Coniothyrium sp. easily invades into the physical texture of G. biloba weakened by $SO_2$ fumigation and because sugar was accumulated in leaves of G. biloba.

Consideration of Herb Cultivating in the Farms of Korea (국내에서 재배되고 있는 허브에 대한 고찰)

  • Herb found in the farm of Korea were classified into 3 divisions, 5 classes, 31 orders, 51 families, 134 genera and 226 kinds. In the taxon of family, labiatae, compositae and umbelliferae have larser number of herbs than another families. In the taxon of genus, Rosa (34 kinds), Mentha (8), Thymus (7), Pelargonium (6), Salvia (6), Lavandula (5), Citrus (4), Ocimum (4), Allium (4) in order contain many species. In the geographical floristic regions, Holarctic were 149 kinds, and next came Paleotropical of 26 kinds, Neotropical of 18 kinds, Capetown of 8 kinds, and Australian of 6 kinds in order. Plants growing naturally over 3 geographical region were 4 kinds including Passiflora caerulea. And 166 kinds were growing in the only 1 geographical region. Among the all farms, (1) farm (in Cheongwon) were highest number of 137 kinds. On the other hand, (3) farm (in Cheongsong) were lowest number of 59 kinds. Herb cultivating in all farms were 37 kinds including Pelargonium crispum. Among these, plants of 24 kinds belong to the labiatae family. Also, herb cultivating in the only 1 farm were 50 kinds including Humulus lupulus. About the numbers of usage of herb, the first in numbers was for food $(54.4\%,\;122\;kinds)$ including Ginkgo biloba, the second for medicinal purpose $(39.8\%,\;90\;kinds)$ including Equisetum arvense, the third for cosmetic $(15.9\%,\;36\;kinds)$, th fourth for flavor $(14.2\%,\;32\;kinds)$, the next for perfume $(12.4\%,\;28\;kinds)$, the next for dye $(10.8\%,\;23\;kinds)$, the next for insecticide $(6.6\%,\;15\;kinds)$, the next for ornament $(1.8\%,\;4\;kinds)$, the last for antiseptic $(0.9\%,\;2\;kinds)$.

A Study on the Evaluation and Improvement of Permeability in Radial and Tangential Section of Domestic Softwoods (국산 침엽수의 방사, 접선단면의 투습성 평가와 개선방안에 관한 연구)

  • The purpose of this research was to evaluate the vapor permeability of nine different species of domestic softwood and the vapor permeability evaluation through the production of small wall structures for the developing applications, utilizing the vapor permeability of wood. In addition, the permeability evaluation was attempted by means of the production of a small wall structure injected with a waste material, bottom ash, as a moisture absorbent for improving the permeability. Consequently, the results of the vapor permeability evaluation by means of Sd value are as follows: (1) It was observed that Abies holiphylla, Picea jezoensis, Ginkgo biloba, Pinus koraiensis and Pinus rigida are permeable to moisture among 9 species of domestic softwood in Korea. (2) By means of this, semi-permeability efficiency was evaluated when producing a small wall structure. (3) Besides, improved effects of permeability were evaluated when producing a small wall structure inserted with bottom ash. As a result, it was confirmed that the Sd value of Pinus koraiensis turned out to be 1.63, which is superior to other 8 tree species.

Studies on the Lipid Components of Ginkgo Nut (은행종실(銀杏種實)의 지질성분(脂質成分)에 관한 연구(硏究))

  • Lipids, extracted with chloroform-methanol (2:1 by vol.) and purified, from nut and leaf of Ginkgo biloba were identified and quantitatived by column, thin layer and gas liquid chromatography. The results were summarized as follow: 1) The total content of purified lipids in the nut and leaf on the fresh weight basis were 1.32% and 2.24%, respectively. 2) The lipid fractions in the nut obtained by silicic acid colum chromatography were found to be composed of about 89% neutral lipids and about 10% compound lipids, and in the leaf were found to be composed of about 28% neutral lipids and about 72% compound lipids. 3) Among the neutral lipid fractions, triglycerides (86.2%) were the major component in the nut, but esterified sterols (53.3%) were the major component in the leaf. 4) The main fatty acids of the total lipids were oleic(37.5%) and linoleic acid(44.5%) in the nut, but linolenic(45.2%) and palmitic acid (25.1%) were main fatty acids in the leaf. The patterns of fatty acid composition of the neutral lipid fractions in the nut and leaf were found to be similar, and oleic, linoleic and palmitic acid were the predominant. A large amount of oleic and linoleic acid in the glycolipid fractions was found in the nut compared with those in the leaf, but linolenic acid content in the leaf was significantly higher than in the nut. And patterns of fatty acid composition of the phospholipid fractions in the nut and leaf were found to be similar to that of glycolipid fractions.

Foliar Fertilization Effect of Environmentally-Friendly Organic Agricultural Materials for Grape Cultivation (포도재배를 위한 친환경 유기농자재의 엽면시비 효과)

  • This study was conducted to investigate foliar treatment effects of organic agricultural materials for the environmentally-friendly cultivation of grape. The organic materials applied were chitosan, wood vinegar (pyroligneous acid), amino acid solution, and ginkgo leaf extract. All the organic materials were relatively strong acidic ranging lower than pH 4.6. when comparing with other organic materials, amino acid solution contained relatively high contents of selected plant nutrients, such as N, P, K, Ca, Mg, Cu, Fe, Mn, and Zn. As comparison of selected soil properties in the grape cultivating field, soil pH values were lower at the harvest stage than at the first stage of grape growing (before treating the organic materials), and electrical conductivity (EC) and soil organic matter content were higher at the harvest stage in the all plots. The concentrations of available phosphorus increased in most of the plot soils except in control plot (conventional treatment). The concentrations of exchangeable K decreased in the plot treated with ginkgo leaf extract and the control plot. The exchangeable Mg concentrations decreased in soils of all the plots. On the other hand, the concentrations of N and K in the grape leaves were higher with the treatments of chitosan and amino acid solution, P concentrations were higher with the applications of chitosan, wood vinegar and amino acid solution, and Ca and Mg concentrations were higher with chitosan and amino acid solution treatments, respectively, than with others. The yields of grape were higher, $1,581{\sim}1,583kg\;10a^{-1}$, in the control and wood vinegar treatment plots than others. Sugar contents of grape were not different among all the plots.

BIOLOGICAL EFFECT OF MAGNOLIA AND GINKGO BILOBA EXTRACT TO THE ANTIMICROBIAL, ANTIINFLAMMATORY AND CELLULAR ACTIVITY (후박 및 은행잎 추출물의 향균, 향염 및 세포활성도에 미치는 영향)

  • Periodontal therapy for treatment of periodontitis involves the elimination of bacterial plaque and elimination of the anatomic defects by regenerative procedure. The purpose of this study was to evaluate on the biological effect of magnolia and Ginkgo biloba extract to the antimicrobial, antiinflammatory and cellular activity. Antimicrobial assay was performed with the diffusion method of the extract by measuring of growth inhibitory zone of B. cereus from blood agar plate. Effect of the extract to cellular activity of gingival fibroblast were examined using MTT method and measured the result with optical density on 570nm by ELISA reader. Inhibitory effects of $PGE_2$ production from gingival fibroblast was performed with the addition of $IL-l{\beta}$ and the extract to the well and examined to the product of $PGE_2$ from cell by ELISA reader. In vivo anti-inflammatory effect was performed with injection examined with clinically and histologically for their extent of mecrosis and inflammation. Antimicrobial activity of Magnolia extract showed significantly higher activity than that of control. However, GBE did not showed significant activity to compare with control, and mixture of Magnolia and GBE extract showed significantly higher activity than that of control. The effect of cellular activity to gingival fibroblast showed no significant differences of between control and Magnolia extract. However, GBE showed significantly higher rate of cellular activity to compare with control and even to PDGF-BB, and also showed same degree of cellular activity even though mixed with Magnolia extract. The inhibitory effect of $PGE_2$ production showed significantly reduction of $PGE_2$ production to compare with control, but its inhibitory effect was not much strong to compare with Indomethacin. In vivo, antiinflammatory effect of Magnolia extract to P. gingivalis injection of Hamster buccal check showed significantly reduction of inflammatory cell infiltration and tissue necrosis, but GBE showed no effect on the inhibition of inflammatory process. These results suggested that Magnolia and GBE extract possessed different kind of biological activity and also can be compensated on their activity with each other for elimination of bacterial plaque and anatonical defect.
