Effects of Setaria italica on Gap Junction-Mediated Intercellular Communication for the Development of Cancer Chemopreventive Agents

  • Inhibition of gap junction-mediated intercellular communication (GJIC) has been considered as an important factor in the tumor promotion phase of carcinogenesis. Recovery effects of natural products on gap junctional intercellular communication are measured by scrape-loading and dye transfer method using Lucifer yellow after administration of phorbol-12-myristate-13-acetate (PMA) on WBF344 cells. Among tested natural products, the hexane fraction and subfractions (F-01 and F-04) of Setaria italica were relatively effective for recovery of GJIC. The hexane fraction of Setaria italica $(EC_{25},\;12.14\;{\mu}g/ml)$ and subfractions $(F-01:EC_{50},\;10.74\;{\mu}g/ml;EC_{25},\;1.58\;{\mu}g/ml,\;F-04:EC_{50},\;11.03\;{\mu}g/ml;\;EC_{25},\;3.12\;{\mu}g/ml)$ revealed dose-dependent recovery effects on GJIC. Our data show GJIC activity measurement by Lucifer yellow spread on cells can be an effective tool for the screening of natural products with possible cancer chemopreventive effects.

Evaluation of Toxicity of Anthocyanin from Black Soybean by Feeding Test in Mice (검정콩 안토시아닌의 마우스를 이용한 안전성 연구)

  • Over the past few years, anthocyanin has been demonstrated to exert potentially anti-oxidative and bioavailability effects including anti-cardiovascular disease and cancer-preventive effects in humans. This study was conducted to assess the toxicity of anthocyanin extracts from black soybean seed coats via oral administration in a mice feeding test. The final anthocyanin-containing products induced signs of toxicity on mortality in the 4-week feeding test. The incidence of clinical signs and changes in body and organ weight were also not observed in all anthocyanin-treated groups as compared with the control groups. In hematology analysis, RBC indices concluding MCV, MCH, and MCHC and WBC differential counting such a NEU, LYM, MONO, EOS, and BASO evidenced no significant differences between the anthocyanin treatment and control groups. The biochemical parameters in serum analysis - ALT, AST, BUN, Crea etc.- were also not significantly altered in those groups. Absolute and relative organ weights were not increased after 4 weeks of treatment with anthocyanin extracts in mice. In conclusion, anthocyanin extracts from the black soybean has biological activity without any toxic effects, which also suggests that the consumption of soy containing anthocyanin products may be a good source for health and medical supplies.

The Effect of Voice Therapy in Vocal Polyp Patients (성대용종 환자의 음성치료 효과)

  • Vocal polyps are benign phonotraumatic lesions which are traditionally treated using phonomicrosurgical techniques. In the case of hyperfunctional voice use, voice therapy is effective and results in voice improvement. However, the utility of voice therapy about vocal polyp is in great demand. The purpose of this study was to evaluate the effects of voice therapy in patients with vocal polyps. The authors reviewed the medical records of 193 patients with vocal nodules or vocal polyps, and 64 patients (31 nodules and 33 polyps) were enrolled. All of the subjects had received explanation of problems, vocal hygiene education, and been treated by the $SKMVTT^{(R)}$ (Seong-Tae Kim's multiple voice therapy technique) ranging from 4 to 16 sessions (mean: 8.6 sessions). All subjects were examined by perceptual assessment, acoustic and aerodynamic measures, and VRP (voice range profile). In perceptual assessment, patients with vocal nodules had more breathy and strained voices than the vocal polyp group. Both groups significantly reduced rough, breathy voice after voice therapy. Patients with vocal polyps had worse voice quality than patients with nodules in acoustic measures. Both groups showed reduced jitter and shimmer after voice therapy. In aerodynamic measures, MPT and Psub were increased, and MFR was reduced (p<.05). Participants' frequency range and intensity range were increased after voice therapy, but only frequency range resulted in a significant difference (p<.05). In conclusion, the therapeutic effect of voice therapy in patients with vocal nodules and polyps was demonstrated perceptually and acoustically. We can suggest that voice therapy, including advice, vocal hygiene, and $SKMVTT^{(R)}$ is a useful as an initial choice of treatment for patients with vocal polyps before considering a surgical approach.

  • This study was performed to evaluate repeated-dose oral toxicities of Flos lonicerae extract in Fischer 344/n rats. Flos lonicerae was administered orally to rats at dose levels of 0, 37, 111, 333, 1,000 and 2,000 mg/kg/day. Each group consisted of 10 rats of each gender. The Flos lonicerae extract was given once a day, 5 times a week, for 90 day repeatedly. This study was conducted in accordance with the Protocol of Korea National Toxicology Program and The Standards of Toxicity Study for Medicinal Products. In the present study, there were no toxicologically significant changes in mortality, clinical signs, body weight gains, ophthalmoscopy, urine analysis, hematology, serum biochemistry, necropsy findings, organ weights, histopathology, estrus cycle and sperm examination of all animals treated with Flos lonicerae extract. These results suggest that the oral no observed adverse-effect level of the test item, Flos lonicerae extract, in rats is higher than 2,000 mg/kg/day in both genders. The target organs were not established.

  • We defined a dam as a wetland for assessing wetland functions of man made dam. We compared Boryung dam with Upo wetland by HGM assessment. Hydrologic functions of Boryung dam are not good at short term water storage, but good at long term water storage. Biogeochemical functions of Boryung dam are about 50% of Upo wetland's functions. Boryung dam is a good wetland as a plant habitats and animal habitats. But functions as a habitat for birds were not good. With this study we look forward to making clear merits and demerits of Boryung dam’s functions as a wetland.

  • Recently, there have been a lot of studies for the value for wetlands with increased interest about wetlands. However, the value assessment for wetlands was usually conducted for 송 natural wetlands only, without consideration for Dam-wetlands. In this study, we assumed that a dam carried out a function of wetlands and defined such dams as Dam-wetlands. Choice Experiment(CE) was used in value assessment method. Study area of this study is Boryeong-dam. We performed questionnaire survey in six metropolitan cities and Boryeong city for the value assessment of Dam-wetland. The result of questionnaire survey was analyzed using a Conditional Logit(CL) and a Random Parameter Logic(RPL). Therefore, the total values of a Boryeong-dam is estimated as 72.8 billion-won when consider the wetlands of surrounding area.

  • The cyclohexanol dehydrogenase (ChnA), produced by Rhodococcus sp. TK6, which is capable of growth on cyclohexanol as the sole carbon source, has been previously purified and characterized. However, the current study cloned the complete gene (chnA) for ChnA and its flanking regions using a combination of a polymerase chain reaction (PCR) based on the N-terminal amino acid sequence of the purified ChnA and plaque hybridization from a phage library of Rhodococcus sp. TK6. A sequence analysis of the 5,965-bp DNA fragment revealed five potential open reading frames (ORFs) designated as partial pte (phosphotriesterase), acs (acyl-CoA synthetase), scd (short chain dehydrogenase), stp (sugar transporter), and chnA (cyclohexanol dehydrogenase), respectively. The deduced amino acid sequence of the chnA gene exhibited a similarity of up to $53\%$ with members of the short-chain dehydrogenase/reductase (SDR) family. The chnA gene was expressed using the pET21 a(+) system in Escherichia coli. The activity of the expressed ChnA was then confirmed (13.6 U/mg of protein) and its properties investigated.

  • Since ocean dumping of wastes was permitted by law in 1900, the amount of wastes dumped into the sea has increased ten times for 15 years. As a result, the dumping sites has been deteriorated so much that the pollution has become a social problem. The 96 Protocol to the 72 London Convention, which requires the contracting party's stricter control on the disposal of wastes at sea, took effect on March 24, 2006. Therefore, our country has become confronted to the situation on which it cannot delay putting a restraint on ocean dumping. Hereupon, Korea Coast Guard (KCG) initiates a dumping sites recovery program. The program is intended to curb the amount of wastes dumped at sea and to recover the polluted dumping sites. In this paper, the current status of our country's management of ocean dumping of wastes is examined and the future condition is anticipated KCG's ocean dumping management measures are also presented.

  • [ $Pb(Zr_{0.2}Ti_{0.8})O_3/SrRuO_3$ ] heteroepitaxial thin films were deposited at various temperatures on single crystal $SrTiO_3$ substrates by pulsed laser deposition and characterized for the microstructural and ferroelectric properties. The $SrTiO_3$ substartes etched by buffered oxide etch $(pH{\thickapprox}5.8)$ solution for 20s followed by the thermal annealing at $1000^{\circ}C$ for 1h showed the terrace ledges with a 0.4nm height. The $SrRuO_3$ bottom electrodes with a thickness of 52nm grown on $SrTiO_3$ single crystal also exhibit a terrace ledge similar to that of $SrTiO_3$. The PZT thin films were grown with an epitaxial relationship and showed typical P-E hysteresis loops shown at the epitaxial films. The 56nm thick-PZT films deposited at $650^{\circ}C$ exhibit a remanent polarization $(p_r)$ of $80{\mu}C/cm^2$ and a coercive field $(E_c)$ of 160kV/cm.

Rooting Promotion in Cutting Propagation of Tea (다(茶)나무 (Camellia sinensis L.) 삽목(揷木)시 발근솔(發根率) 향상(向上))

  • These experiments were conducted to promote the rooting in tea cutting bymeans of several root media, growth regulators, cutting material and cutting condition The results are summarized as follows. The rate of rooting was higher in the softwood than in the hardwood, and the best cutting sea­son was about August 10 in the hardwood, about April 10 in the softwood. The most suitable root medium was the Masato at which the rate of rooting was 71%, 87% each in the softwood and in the hardwood. When growth regulators, such as oxyberon, rootone were sprayed upon the hardwood, the rate of rooting was promoted to 4 - 9 % better than that of no treatment. But such a good effect was not recognized significantly in the softwood.
