Seedling emergence and .stand establishment in dry-seeded rice is unstable at present due to deep seeding, low temperature at seeding time, excessive drought or raining etc. This experiment was conducted to elucidate the effect of gibberellic acid(GA$_3$) on the seedling emergence and growth in dry-seeded rice. GA$_3$, IBA and Metalaxyl were treated by seed-soaking or-spray. Hwaseongbyeo, a japonica rice, was used. The seed-soaking treatment with GA$_3$ significantly increased the seedling emergence and plant height as compared with untreated seed. However, no difference was observed in IBA and Metalaxyl treatment. The optimum concentration of GA$_3$ to promote the seedling emergence and growth in dry-seeded rice was about 100ppm for seed-soaking and about 200ppm for seed-spray treatment. The seed-spray treatment of GA$_3$ with 200ppm promoted earlier and faster seedling emergence and elongated seedling height as compared with untreated seed.

Lychnis wilfordii (Regel) Maxim is a rare and valued ornamental plant. Germination rate reached 46.6% when seeds were treated with $100mg{\cdot}l^{-1}$ GA (Gibberellic acid). The highest callus induction was observed in the leaf explants of the seedlings on MS medium containing specific concentrations of $0.5mg{\cdot}l^{-1}$ BA ($N^6$-benzyladenine) and $3.0mg{\cdot}l^{-1}$ NAA (a-naphthalene acetic acid). The adventitious shoot was formed in 97.3% of calli on 1/2 WPM (Woody Plant Medium) medium. Shoot elongation of in vitro propagated plantlets was no difference among various medium. The plantlets grew well after transferring to the pot. This in vitro propagation protocol should be useful for conservation of this endangered plant.
In order to elucidate the effects displayed by the plant growth regulators on the senecence of tobacco leaves, the author applied gibberellic acid, Kinetin, Indole-acetic acid, uracil, and malefic hydrazide to the excised leaves in concentration of 25mg/l of each regulators. And author obtained the results as following, 1. Parallel to the decreases of chlorophyll, the amounts of protein and RNA were decreased. 2. The supression of decrease of the amounts of RNA and protein was displayed by the plant growth regulators, G.A., I.A.A.. The decrease of them in M.A. treatments was more than in non-treatments. 3. The ratio changes of chlorophyll a/b and the changes of protein and RNA content seemed to be no relations.

Plants are sessile and rely on a wide variety of growth hormones to adjust growth and development in response to internal and external stimuli. We have identified a gene, designated NAN, encoding a basic helix-loop-helix (bHLH) transcription factor that regulates cell elongation and seed germination in plants. NAN has an HLH motif in its C-terminal region but does not have any other discernible homologies to bHLH proteins. A bipartite nuclear localization signal is located close to the HLH motif. An Arabidopsis mutant, nan-1D, in which NAN is activated by the insertion of the 35S enhancer, exhibits growth retardation with short hypocotyls and curled leaves. It is also characterized by reduced seed germination and apical hook formation, symptomatic of GA deficiency or disrupted GA signaling. The phenotypic effects of nan-1D were increased by treatment with paclobutrazol (PAC), an inhibitor of gibberellic acid (GA) biosynthesis. NAN is constitutively expressed throughout the life cycle. Our observations indicate that NAN has a housekeeping role in plant growth and development, particularly in seed germination and cell elongation, and that it may modulate GA signaling.
Plant growth retardants reduce the plant height by inhibiting stem elongation in Lilium davuricum. To investigate the plant hormones related to stem elongation, we sprayed 50 $mg{\cdot}L^{-1}$ diniconazole to young plants of L. davuricum and quantified the contents of endogenous gibberellic acids (GA) and abscisic acid (ABA). In GA biosynthesis, L. davuricum had not only the early C-13 hydroxylation ($GA_{19}{\rightarrow}GA_{20}{\rightarrow}GA_1$) pathway resulting in $GA_1$ as the active form but also the non C-13 hydroxylation (NCH, $GA_{12}{\rightarrow}GA_{24}{\rightarrow}GA_9{\rightarrow}GA_4$) with $GA_4$ as the active form. However, the main pathway was NCH because $GA_4$ concentration of 55 $ng{\cdot}g^{-1}$ dry wt was much higher than $GA_1$ content of 0.23 $ng{\cdot}g^{-1}$ dry wt in control plant. Diniconazole inhibited GA biosynthesis through NCH pathway from its early stage. $GA_{12}$ content decreased by diniconazole up to 6% level of that of control and this effect continued to $GA_4$. Diniconazole reduced $GA_{12}$ content by 12.7 $ng{\cdot}g^{-1}$ dry wt, whereas that of control plant was 213.8 $ng{\cdot}g^{-1}$ dry wt. ABA content decreased up to one third of control by diniconazole application. From the contents of endogenous $GA_4$, $GA_1$, and ABA in this study, we could conclude that diniconazole reduces the plant height by inhibiting $GA_4$ biosynthesis in L. davuricum.

Ranunculus asiaticus characterizes colorful and attractive flower shapes that are related with the ornamental value of bulbous plants. Improving ornamental value of bulbous flowers has been the general goal of floricultural market. Gibberellic acid ($GA_3$) and benzyladenine (BA) play an important role in growth and developmental processes in floriculture. Combinational treatments of these two hormones have been used in floriculture to improve flower quality. We assessed the effects of combined $GA_3$ and BA, as well as the individual effects of each hormone, on growth characteristics using soil drench application to eight R. asiaticus cultivars, 'Giallo Millepetali', 'Bianco Millepetali', 'Arancio Millepetali', 'Rosa SC', 'Arancio Pratolino', 'Giallo Pratolino', 'Bianco Pratolino', and 'Rosa Ch Pratolino'. $GA_3$ treatments increased plant height and first flower size of R. asiaticus cultivars. Moreover, about 5 to 9 days to flowering were averagely shortened by $GA_3$ treatments compared to controls. On the other hand, the opposites, including first flower size and days to flowering, were observed for cultivars treated with BA, compared with controls. Treatments of $GA_3$ + BA generally affected growth traits, such as plant height, flower size, and the timing of flowering on some R. asiaticus cultivars. In particular, about 5 to 6 days to flowering were reduced on average by Treatments of $GA_3$ + BA. Our results showed positive growth effects, including plant height, days to flowering, first flower height, number of flowers from the application of individual and combined hormones to R. asiaticus cultivars and demonstrate a role for these hormones in future bulbous floriculture.

Perilla frutescens var. japonica Hara was collected from farmland in Seongju-gun. Fifteen endophytic fungal strains with different colony morphologies were isolated from roots of P. frutescens. Waito-c rice seedlings were treated with the concentrated culture filtrates (CF) of endophytic fungi for observation of their plant growth-promoting activities. In the results, the CF of Y2H001 fungal strain promoted the growth of the waito-c rice seedlings. The phylogenetic tree of Y2H001 strain was analyzed by the combined sequences of the partial internal transcribed spacer region (ITS) and partial betatubulin gene. Molecular and morphological studies identified the Y2H001 strain as belonging to Aspergillus flavus. In gas chromatography mass spectrometry (GC/MS) analysis of the CF of Y2H001 strain, gibberellic acid (GA) was detected and quantified. Therefore, we describe Y2H001 strain as a new $GA_3$-producing A. flavus based on morphological, molecular characteristics and analysis of secondary metabolite.
This experiment was conducted to evaluate the effect of gibberellin biosynthesis retardants as used by rice lodging inhibitors on the gibberellin antagonism, auxin interation, ethylene evolution and growth of second crops. Results obtained can be summarized as follows. Inabenfide, paclobutrazol and uniconazole markedly inhibited the epicotyl elongation of mung bean. Inhibiting effect of epicotyl by these chemicals was markedly stimulated by gibberellic acid, thus showing clear antagonism between these chemicals and gibberellic acid. Significantly large number of roots were formed in the mung bean cuttings which were rooted in the paclobutrazol and uniconazol of 1 ppm. The higher the concentration, the more the number of roots forms. It was guessed that these effect was closely related with auxin. Ethylene evolution was a little stimulated in the leaf of rice under the treatment of inabenfide, paclobutrazol and uniconazole at earlier stage(5 DAT), however it was suppressed at later stage(10, 30 DAT) at higher concentration. The effect of gibberllin biosynthesis inhibitors to second crops retarded tomato plants without influencing the height of barley. The treatment of paclobutrazol and uniconazol which is triazole-type more severely inhibited than that of inabenfide which is isonicotinanilide-type. The more the concentration, the less the height of tomato plants.

Seed size traits are controlled by multiple genes in crops and determine grain yield, quality and appearance. However, the molecular mechanisms controlling the size of plant seeds remain unclear. We performed functional analysis of BrPATL4 encoding Sec14-like protein to determine the genetic architecture of seed size, shape and their association analyses. We used 60 $T_3$ transgenic rice lines to evaluate seed length, seed width and seed height as seed size traits, and the ratios of these values as seed shape traits. Pleiotropic effects on general architecture included small seed size, erect panicles, decreased grain weight, reduced plant height and increased sterility, which are common to other mutants deficient in gibberellic acid (GA) biosynthesis. To test whether BrPATL4 overexpression is deleterious for GA signal transduction, we compared the relative expression of GA related gene and the growth rate of second leaf sheath supplied with exogenous $GA_3$. Overexpression of BrPATL4 did not affect GA biosynthesis or signaling pathway, with the same response shown under GA treatment compared to the wild type. However, the causal genes for the small seed phenotype (D1, SRS1, and SRS5) and the erection of panicles showed significantly decreased levels in mRNA accumulation compared to the wild type. These results suggest that the overexpression of BrPATL4 can control seed size through the suppression of those genes related to seed size regulation. Although the molecular function of BrPATL4 is not clear for small seed and erect panicles of BrPALT4 overexpression line, this study provides some clues about the genetic engineering of rice seed architecture.

This study was carried out to realize the effect of gibberllic acid$(GA_3)$, 1-naphthaleneacetic acid(NAA) and indole-3-acetic acid(IAA) on the biosynthesis of vitamin C. The relation between carbohydrate metabolism and vitamin C production in soybean sprouts was also investigated. Growth, vitamin C content, protein, galactonolactone dehydrogenase(GLD), ribulose diphosphate carboxylase(RuDpCO) and RNA level in the plastid and cytoplasm were determined. The effects of protein and respiratory inhibitors on the growth and vitamin C production were also examined. The most favourable growth of soybean sprouts was observed at the level of NAA $10^{-8}M,\;IAA10^{-6}M\;and\;GA_3\;10^{-5}M$ in the single treatment, respectively, and also favourable at levels of $GA_3\;10^{-5}M+NAA\;10^{-9}M\;and\;GA_3\;10^{-5}M+IAA\;10^{-9}M$ in the case of mixed treatment. The excellent growth was observed at the level IAA $10^{-6}M$ among all the single and mixed treatments. When the soybean sprouts were treated with NAA $10^{-8}M,\;IAA\;10^{-6}M\;GA_3\;10^{-5}M,\;GA_3\;10^{-8}M+IAA\;10^{-6}M,\;and\;GA_3\;10^{-5}M+IAA\;10^{-9}M$, the maximum growth rate was observed at the level of IAA $10^{-6}M$ and the conten of vitamin C was 24.26mg% which was 1.6 times higher than that of the control. RuDpCO was inhibited by the chloramphenicol at the concentration that did not inhibit the growth but the activities of NADP-GDH, GLD and vitamin C content were not affected. These results showed that the biosynthesis of viamin C had nothing to do with the activity of chloroplastic RNA but with cytoplasm. The highest vitamin C content was found at the the level of IAA $10^{-6}M$, where the GLD activity increased up 1.8 times of the control. The concentration of IAA $10^{-6}M$ promoted the biosynthesis of RNa and protein both in chloroplast and cytoplasm, especially in the cytoplasm. Thus it suggeted that IAA affected vitamin C biosynthesis by regulating RNA level in the cytoplasm. 2,4-Dinitrophenol as an uncoupler of oxidative phosphorylation did not inhibit the vitamin C biosynthesis, however, all of the respiratory inhibitors severely inhibited the growth and vitamin C biosynthesis.
