High frequency somatic embryogenesis through leaf explant-derived callus culture in Muscari armeniacum cv. 'Early Giant' (무스카리 'Early Giant' 잎 절편 유래 캘러스 배양을 통한 고빈도 체세포배 발생)

  • Using calli of $Muscari$ $armeniacum$ cv. 'Early Giant' that is monocotyledonous ornamental bulb crop with increasing demand in Korea, we carried out current studies to establish an in vitro multiple propagation protocol via somatic embryogenesis. We found that soft pale yellow green calli were induced from leaf explants cultured on all media containing 0.1~3.0 $mg{\cdot}L^{-1}$ auxins such as 1-naphthalene acetic acid (NAA) and 2,4-dichlorophenoxyacetic acid (2,4-D). However, induced calli showed vigorous growth only when they further transferred on same media containing 2,4-D, 4-amino-3,5,6-tri-chloropicolinic acid (picloram), or 3,6-dichloro-o-anisic acid (dicamba). Although frequency of somatic embryo induction depended on callus source and PGR composition in somatic embryo induction media, somatic embryogenesis was initiated on surface of proliferated calli after transferring on media with no PGR or 0.01 $mg{\cdot}L^{-1}$ NAA co-supplemented with various cytokinins such as $N^6$-benzylaminopurine (BAP). Highest number of embryo at 9.3 per callus clump was obtained when calli which were grown under 0.1 $mg{\cdot}L^{-1}$ picloram supplementation were sub-cultured on medium with 0.01 $mg{\cdot}L^{-1}$ NAA and 0.5 $mg{\cdot}L^{-1}$ BAP. In addition, morphological characteristics of somatic embryo were categorized into following nine phases: globular, biased heart, biased torpedo, early cotyledonary, middle cotyledonary, late cotyledonary, early sprouting, middle sprouting, and late sprouting embryos.

  • Several types of endosperm and embryo mutants were induced by the treatment of MNU (N -methly- N -nitrosourea) on fertilized egg cell of rice plant. These mutants were named as dull. waxy, white core, floury, sugary, shrunken, colored seed coat and giant embryo according to their appearence, micro-scopic feature on SEM and amylose content. White cored mutant was the most frequent one among them. All of the mutants were segregated as controlled by a single recessive gene except 47320 (dull). Futher studies on the genetics and physico-chemical properities of the mutants are ongoing.

Characteristics of Pop-rice and Rice Tea Using Black Sticky Rice with Giant Embryo (흑찰거대배아미를 이용한 팝라이스와 흑미차의 품질 특성)

  • Recently developed black waxy rice with a giant embryo ('Nunkeunheukchal', BGE) was selected and processed to produce high quality nutritional food. BGE contains high levels of several phytochemicals with antioxidant activities, as well as other reported health beneficial properties. In addition, the giant embryo has high protein, lipid, and amino acids contents. Within the free amino acids, ${\gamma}$-aminobutyric acid (GABA), a major inhibitory neurotransmitter, has long been used for treating the aftereffects of brain injuries and stroke. A method for manufacturing pop-rice and black rice tea by popping process in BGE is provided to increase a taste, nutrition and functionality. The produced 'pop-rice' showed increased protein (11.3%) and lipid (3.7%) contents compared with control variety, IB ('Ilmibyeo'). In addition, melanoidin related products, polyphenol and functional amino acid contents were increased by the popping process. Pop-rice tea made of BGE showed the highest extraction of total sugar, glucose, raffinose and sucrose (4 times higher than brown rice) by hot water. Scavenging activity ($SC_{50}$) of processed BGE rice powder showed strong antioxidative activity of 0.24 mg/ml using DPPH and 1.82 mg/ml using ABTs method. Thereafter, these results suggested that the popping processed rice of BGE could be one of the promising materials for healthy food development.

Effect of Feeding with High γ-Aminobutyric Acid (GABA) Containing Giant Embryo Black Sticky Rice (Oryza sativa L.) on Alcohol Intake in C57BL/6 Mice (GABA 고함유 흑찰거대배아미의 투여가 C57BL/6형 생쥐의 알코올 섭취량에 미치는 영향)

  • It is known that black sticky rice with giant embryo (BSRGB, Milyang 263) and giant embryo rice (GER) contains higher levels of GABA than rice. Therefore, feeding BSRGB, GER, or rice freely to C57BL/6 mice with two-hour alcohol intake for 16 days was investigated. For two-hour alcohol intake, a repeated measure ANOVA (three treatment groups repeated across 16 days; 8 two-day blocks) yielded a significant group by block interaction (df=16, F=3.109, p=0.004). The independent t-test showed that significant suppression of two-hour alcohol intake was observed when subjects were administered with BSRGB, compared with the rice alone across all the two-day blocks (p<0.05). The paired t-test revealed that a significant suppression of two-hour alcohol intake was observed starting 4 to 16 days after freely feeding with BSRGB compared to before feeding. However, there is no significant difference in the two-hour alcohol intake observed between the before and after administration of rice. A repeated measure ANOVA revealed no significant group by block interaction for 22-hour water intake and body weight. However, a repeated measure ANOVA revealed a significant grouping by block interaction for food intake. These results indicate a change of two-hour alcohol intake is presumably caused by GABA, which is found in higher levels in BSRGB than in rice.

Supplementary Effect of the Giant Embryonic Rice on Serum and Heaptic Lipid Levels of Streptozotocin-induced Diabetic Rats (Streptozotocin 유발 당뇨 쥐의 거대배아미 식이에 의한 혈액 및 간조직의 지질조성)

  • This study was carried out to investigate the supplementary effects of giant embryonic rice, which has over two times of embryo size compared the normal rice, on lipid metabolism of insulin dependent diabetic rats. Streptozotocin induced diabetic rats were fed four kinds of experimental diets com starch diet as a control (C-D), a polished rice diet (R-D), a brown rice diet (BR-D) and a giant embryonic rice diet (GER-D) respectively, for 6 weeks. Diet intake body weight, organ weights and lipid levels of serum, liver and feces were measured. There was significant difference in diet intake and body weight among experimental groups. The concentrations of serum triglyceride and total serum cholesterol of BR-D and GER-D groups were lower than those of others. The total hepatic cholesterol level was the lowest in GER-D group. The contents of total lipid and total cholesterol excreted in feces of BR-D and GER-D groups were higher than those of C-D and R-D groups. These results suggested the giant embryonic rice diets can effectively reduce serum triglyceride level and total hepatic cholesterol level in insulin dependent diabetic rats, and hypolipidemic effects be due to increasing fecal lipid excretion.

Effect of Germinated Black Sticky Rice with Giant Embryo on Alcohol Intake in C57BL/6 Mice (흑찰거대배아미 발아현미배아의 섭취가 C57BL/6 생쥐의 알코올 섭취에 미치는 영향)

  • Alcohol impacts many central nervous systems, such as dopamine, serotonin, opioids, and gamma-aminobutyric acid (GABA), leading to addiction. Many studies have investigated the relationship between GABA and alcoholism. The purpose of this study was to investigate the effects of GABA high and low rice intake on the alcohol intake behavior of mice. Black sticky rice with giant embryo (BSRGE), black sticky rice (BSR), giant embryo rice (GER), and rice (Rice) were germinated for 48 hr in brown rice. The embryos were then collected and used in the study. The diets were fed to respective C57BL/6 mouse groups ad libitum for 16 days and investigated for 2 hr alcohol intake, 22 hr water intake, 24 hr feed intake, and body weight. As a result of the repeated measure of ANOVA for the daily change of alcohol intake for 2 hr daily between the BSRGE and BSR groups, there was a significant difference in the number of days of intake (DF = 7, F = 4.812, p = 0.026). A significant daily decrease in alcohol intake was observed in the BSRGE group compared to the BSR group. This reduction was consistent from Day 10 to Day 16. Alcohol consumption also significantly decreased in the GER group compared to the Rice group. This decrease was observed from Day 12 to Day 16. In conclusion, BSRGE and GER resulted in decreased alcohol intake in C57BL/6 mice compared to BSR and rice. This suggests that BSRGE may prevent relapse in patients with alcohol use disorder.

Amylose, Tocopherol, Free Sugar and Fatty Acid Content in Selected Mutant Lines of Oryza sativa cv. Shindongjin

  • To assess the potential as biofortified rice varieties, new endosperm and grain mutant lines were selected from $M_4$ generation seeds of the rice cultivar Shindongjin, which were either $\gamma$-irradiated or treated with N-methyl-N-nitrosourea(MNU) and lipid, sugar, and tocopherol content were analyzed. Amylose content in non-waxy mutants ranged from 8.8% in SM-4, a dull-type mutant, to 29.5% in SM-51, compared to 18.9% in the parental variety, Shindongjin. SM-23, a floury-type mutant, contained 0.09 ${\mu}g/g$ $\alpha$-tocopherol(40.9% of total tocopherol), was three times higher than in the parental variety. SM-32, a giant embryo-type mutant, had a 2.2-fold higher total tocopherol content, 2.1-fold higher $\alpha$-tocopherol, and 5.5-fold higher $\delta$-tocopherol content(47.3% of total tocopherol) than the parental variety(0.13 ${\mu}g/g$). Total free sugar content was elevated in all selected mutants and 1.2-8.6 times higher than in the parental variety(11.38 ${\mu}g/g$). These increased sugar levels were due to increase in sucrose concentration. SM-23(floury-type mutant) and SM-51(high amylose-type mutant) had 4.6 and 7.0 times more sugar, respectively, than the parental variety(11.38 ${\mu}g/g$). With relatively high concentrations, most mutants showed elevated fatty acid content in the SM 32(giant embryo-type) and SM-51(high amylose-type) mutants, at 124.56 and 89.59 mg/g, respectively. All selected mutants displayed valuable characteristics for the development of new varieties in rice-breeding programs.

Tissue-Specific Expression of Nebulin Isoform Proteins in Chicken (닭 조직에 따른 Nebulin Isoform 단백질의 특이적 발현)

  • The lengths of thick and thin filaments in the sarcomeres of most vertebrate skeletal muscles are precisely regulated and are important structural parameters in understanding muscle contraction. Nebulin is a usually large protein that spans the whole length of thin filaments in the sarcomeres of skeletal muscles. In this paper we used SDS-PAGE and immunoblot to identify nebulin isoform proteins in muscle and non-muscle tissues. We prepared embryonic chicken tissues including skeletal muscle, cardiac muscle, smooth muscle, brain, liver to compare nebulin isoform proteins. The proteins were divided into soluble and insoluble fraction. As a result, we identified tissue specific expression of various nebulin isoform proteins in muscle and non-muscle tissues of chicken. Nebulin was detected in skeletal muscle of adult chicken about 500 kDa. Nebulett was expressed in cardiac muscle of embryonic and adult chicken about 107 kDa. A giant protein with molecular mass of about 380 kDa was identified in brain of non-muscle of chicken. This giant protein was detected in the soluble fraction of chicken embryo. The unequal distribution of the nebulin isoform proteins suggests tissue specific regulation of the isoform expression and indicates a functional specialization of the encoded isoform subtypes.

  • A model of induction of neoplasia by viruses has develpoed from experimental studies in animals and in cultured cells and oncogenic transformation of cells is the result of integration of viral genetic information into the cellular DNA. The evidence for these associations was derived primarily from seroepidemiologic investigation. However, data indicating that the relation between HSV-2 and cervical cancer fits the model derived from experimental animal studies are not yet sufficient to draw conclusion with regard to the etiologic role the virus in the development of the neoplasms. In other hand, the K562 tumor cell is highly susceptible target for natural killer cell lysis by the lymphocytes of human and murine periperal blood. The characteristics of this effector cell type has been investigated. A study on natural killer cell mediated cytotoxicity(NKMC) against $^{51}Cr$-K562 as target cell was studed in HSV-2 infected ICR mouse. We have studied for susceptibility of HSV-2 against mouse embryo fibroblast(MEF) cells and NKMC from HSV-2 infected mouse. The results obtained that the mouse embryo fibroblast cells culture, the number and size of the cells were markedly increased and formed a monolayers relatively rapid, and become complete monolayer sheet around 72 hrs. Duration of cytopathic effect on MEF cells was rapid by serial passing of HSV-2. The morphology of the HSV-2 infected cells appear to be mainly round, ovium, spindle form and some of them was forming large giant cells. The NKMC was decrease in mouse with HSV-2 and comparison between effector/target cells ratio as 25:1 and 50:1 respectively, the NKMC was found to be more significantly decreased than normal control we have concluded that the natural killer cell activity of the viral infected mouse was shown as a suppressed during the HSV-2 infection, day 7th and 14th.
