Germination Percentages of Different Types of Sweet Corn in Relation to Harvesting Dates

  • Germination of sweet and super sweet corn is lower than normal corn due to the higher sugar and lower starch contents of kernels. Sweet corn seeds are easily deteriorated in the field under the unfavorable condition, therefore it is important to identify the optimal harvesting time for seed production. This trial was conducted to investigate the responses of germination percentage of shrunken-2(sh2), brittle(bt), sugary(su), and sugary enhancer(se) hybrids in relation to harvesting dates. Eight hybrids of four different gene sweet corns were harvested at 15, 20, 25, 30, 35, 40, 45, and 50 days after silking(DAS). Germination test was performed using paper towel method. Mean germination percentages across eight hybrids showed the highest value at 45 DAS. There were significant differences among genes and within gene for germination. Shrunken-2 hybrid Mecca was higher than su hybrids for germination, indicating that sh2 would not be poorer than su Late harvesting beyond the optimal harvesting date might not be desirable because of more lodging and ear rots. Theoretical optimal harvesting date estimated from the regression equation was 40.9 DAS, however, practical date for harvesting would be a few days later than the estimated date if seedling vigor might be considered. Kernel dry weight per ear showed similar response to germination. Regression equation showed the highest kernel dry weight at 40.7 DAS. Significant correlations between kernel dry weight and germination were observed, impling that kernel dry matter accumulation would be an important factor for germination.

Effect of Soaking Time and Steeping Temperature on Biochemical Properties and γ-Aminobutyric Acid (GABA) Content of Germinated Wheat and Barley

  • The objective of this research was to investigate the effects of soaking time (6, 12 and 24 hr) and steeping temperature (25, 30 and $35^{\circ}C$) on germination time, germination percentage and weight loss of wheat and barley. Changes in chemical composition (such as protein, fat, and ash), reducing sugar content, enzyme activity and pasting profile and GABA ($\gamma$-animobutyric acid) content of germinated wheat and barley were also evaluated. The results clearly suggest that the short soaking time and lower steeping temperature significantly decreased germination time and weight loss, while germination percentage increased. Regarding the chemical composition, the protein content of wheat and barley was slightly increased after germination but there was no significant difference in content of crude fat and ash of both germinated cereals. The reducing sugar content of both germinated cereals decreased as the steeping temperature increased from $25^{\circ}C$ to $35^{\circ}C$. Increasing soaking time and steeping temperature led to increased amylase activity, and also corresponded to reduced paste viscosity. The highest GABA content that occurred with soaking times of 6 and 12 hr and a steeping temperature of $35^{\circ}C$ was 1,467.74 and 1,474.70 ${\mu}g/g$ for germinated wheat and 2,108.13 and 1,691.85 ${\mu}g/g$ for germinated barley. This study indicated that the optimum germination process for wheat and barley is a low steeping temperature and a short soaking time.

Variation of Seed Viability among Cone Harvest Times at Two Clonal Seed Orchards of Chamaecyparis obtusa

  • The timing of seed harvesting is an important decision in the management of seed orchards because it affects seed quality and yield. To investigate the effect of cone harvest time on seed quality and determine the optimal harvesting time, cones were regularly collected in seven times and germination tests were performed at each harvest time in two clonal seed orchards of Chamaecyparis obtusa. As cones developed, the percentage of seed germination increased before cone moisture content began to decrease significantly. The moisture contents of cones were highest at the first collection as 68.3% and 67.3% in Jeju and Gochang seed orchards respectively. At this time, germination speed was slowest, indicating poor seed vigour. The highest germination was found at the second stage in Jeju (36.5%) and at the seventh stage in Gochang (28.6%) seed orchard. The germination speed increased as cone moisture content decreased. Additionally, changes of seed vigour differed among the developmental stages in both seed orchards. Consequently, the optimal cone harvest time of C. obtusa seed orchards in Jeju was early September when high germination percentage was obtained. In Gochang seed orchards, late October was optimal cone harvest time when the germination speed was fast and the cone moisture content decreased.

  • Germination and vegetative growth of Enteromorpha multiramosa Bliding from Pyoson, Cheju Island were investigated in laboratory under various combinations of temperature (5-$25^{\circ}C$) and salinity (8-48$^{\circ}C$). Percent level of germination was relatively high at all combinations of the two factors. The highest value among the combinations was revealed at 15$^{\circ}C$ and 32$\textperthousand$. Dry weight also was fairly high at all levels of combination with maximum value at 2$0^{\circ}C$ and 32$\textperthousand$. Analysis of variance for germination and growth was completed respectively and polynomial prediction models were constructed. F ratio revealed that all factors had a significant effect (p<0.001) on percentage of germination and dry weight, and their interactions also were significant (p<0.001), although the F ratio of interactions was far less than that for either the separate effect of temperature or salinity. Response surface of polynomial equation represented that temperature influenced less than salinity on germination, while it effected remarkably on vegetative growth, so the Enteromorpha multiramosa was kept to visible macrothalli from winter to spring in Cheju Island.

  • Germination of Italian ryegrass and sorghum were measured at a range of temperatures and with PEG treatment at several different levels of concentration and treatment period to evaluate the effects of osmoconditioning with polyethylene glycol(PEG-8000). The results obtained are summarized as follows: 1. Cumulative germination percentage of intact Italian ryegrass seeds did not show much difference among temperatures, while that of sorghum seeds decreased markedly at low temperature. 2. Italian ryegrass seeds osmoconditioned with 20% PEG solution for two days at 10^{\circ}C.$ accelerated germination at 10^{\circ}C.$, with injurious effect of osmoconditioning at higher temperature for many days. 3. Osmoconditioning with 20% PEG solution for two or four days at 10^{\circ}C.$ increased germination of sorghum seeds, while the seeds treated at higher temperatures for long period suppressed germination.

An Ozone Micro-bubble Technique for Seed Sterilization in Alfalfa Sprouts

  • The efficacy of ozone micro-bubble water (OMBW) in reducing microbial populations on alfalfa seeds was investigated in this study. We observed the surface of alfalfa seeds using microscopy and found that many cracks and crevices existing on the surface could harbor pathogens. Alfalfa seeds were treated with tap water (TW), micro-bubble water (MBW), ozone water (OW), ozone micro-bubble water (OMBW), and chlorine water (CL) for 5 min, and total microbial population, E. coli and Salmonella spp. colonies were determined. Also, the sterilized seeds were germinated and cultivated for 5 d after sowing to investigate the percentage of germination and the growth of alfalfa sprouts. The treatments with OMBW and CL were most effective in reducing total microbial populations and E. coli was eliminated by OW, OMBW, and CL treatments. CL treatment reduced the percentage of germination and fresh weight of alfalfa sprouts, but OMBW did not cause any negative effects on the germination and growth of alfalfa sprouts. These results indicate that OMBW can be used as an effective sanitizer for eliminating seed-borne pathogens without detrimental effects on seed viability.

서울지역에서 사라져 가는 장억새(Miscanthus changii)의 서식환경조사 및 대량번식 방안 연구 (Habitat Environment and Massive Propagation Method of Rare Species Miscanthus changii Disappeared in Seoul Area)

  • This study was conducted to investigate the draft data for the habitat environment and massive propagation method of Miscanthus changii, which is classified as rare species. The in-site analysis of the native soil give a indication of pH 5.24~5.48, electric conductivity (EC)0.03~0.02 $dS{\cdot}m^{-1}$, organic matter (OM) 1.13~1.15%, Ca 3.4 $mg{\cdot}kg^{-1}$, K 0.25 $mg{\cdot}kg^{-1}$, Mg 0.53 $mg{\cdot}kg^{-1}$, N 0.23%, P 2.15 $mg{\cdot}kg^{-1}$ showed low and 0.33~9.12 $cmol{\cdot}kg^{-1}$ CEC. Growth and tiller number in rhizome propagation of Miscanthus changii showed that horticulture media (perlite 1+vermiculate 1+peat moss 1) is the most suitable soil to habitat. As for seed propagation, germination percentage is 0% (0/20) at first seeding, 2% (1/50) at second seeding and 2.1% (6/280) at third seeding. The average germination percentage was below 2%. This is the reason why Miscanthus changi disappear at habitat of poor soil environment and low germination. We gained 61 plants that 60 rhizome propagation and 1 by seed propagation, renatural to habitat.

Germination of West African Ebony (Diospyros mespiliformis Hochst) Seeds: Effects of Dehydration and Different Pre-sowing Treatments

  • Diospyros mespiliformis is a highly valued and threatened tree species within the Sahelo-Sudanian zone of Africa, but its seed germination requirements under cultivation are not well researched. In a first experiment which aimed at determining germination response of seeds to dehydration, fresh seeds were dried at room temperature for 26 days during which their moisture content, their germinability, and their viability were monitored at two-day intervals. In the second experiment, 14 pre-germination treatments were tested for their effect on the germination of dried seeds. Results showed that fresh seeds had 52.7% moisture and achieved 97.7% germination. As seeds were dried, percentage germination gradually decreased with decreasing moisture content and reached 0% when moisture content had dropped to 18%. Meanwhile, seed viability remained at 100% over drying duration. Seeds that were not germinated after air dry also recorded 100% viability. The most effective treatment for inducing germination of dried seeds was scarification using 98% sulfuric acid for 30 min which resulted in 96.6% germination. This study reports for the first time in D. mespiliformis seeds a desiccation-induced dormancy which can be efficiently alleviated by acid scarification. This study provides useful information that will contribute to efficient management of D. mespiliformis seed resources for propagation.

해당화의 종자 발아 연구 (Improvement of Seed Germination in Rosa rugosa)

  • 장미의 발아율은 약 20% 정도이고 과피 안에 발아억제물질이 존재하며 물 투과성이 어렵기 때문에 육종의 한계가 있다. 본 연구는 장미속 식물인 해당화의 종자를 이용하여 위하여 $25^{\circ}C$ 배양실에 16시간 광주기 하에서 기내배양을 수행하였다. 해당화 종자를 half-cut하여 치상 방향에 따른 발아율을 확인하기 위하여 치상방향을 달리하여 2주간 배양하였다. 종자의 절단면이 배지에 닿지 않도록 치상하였을 경우 모두 발아가 되었으나, 반대로 치상한 경우에는 전혀 발아되지 않았다. Intact 종자와 half-cut된 종자의 수분 후 경과일수와 저온 요구도에 따른 발아율을 비교하기 위해 2주 동안 배양한 결과 intact 종자는 발아되지 않았으며 수분 후 경과 90일된 half-cut 종자에서는 저온처리의 유무에 관계없이 100% 발아하였으며, 수분 후 경과일수가 지날수록 발아율이 높았다. 또한 half-cut 종자를 적색, 청색, 흰색, 황색, 녹색 LED 하에서 배양한 결과 1주 후에 청색 LED에서 90%로 가장 높은 발아율을 보였다. 본 연구에서는 다양한 실험을 수행하여 층적 저온처리 없이도 해당화의 발아기간을 성공적으로 단축시켰고, 종자의 half-cut 방법을 장미의 육종연구에 적용한다면 더 효율적인 발아연구 및 육종연구를 할 수 있을 것이라 생각된다.

이탈리안 라이그라스 품종별 NaCl 처리가 발아 및 초기생육에 미치는 영향 (Effects of Sodium Chloride Treatment on Seed Germination and Seedling Growth of Italian Ryegrass Cultivars)

  • 이탈리안 라이그라스의 발아 및 초기 생육단계에서 내염성 수준 정도를 조사하기 위하여 in vitro 조건에서 다양한 염농도 조건에서 그린팜, 플로리다80, 코원어리 및 화산101호 품종을 이용하여 염해에 대한 반응을 발아율과 산화 스트레스 관련 효소들의 활성을 통해 조사하였다. 저농도의 NaCl 처리구에서는 모든 품종에서 발아율의 차이가 1~3% 정도 감소하는 경향을 보였으며 250 mM 이상의 NaCl 처리구에는 화산101호 품종을 제외한 모든 품종에서 50% 이상의 발아율 감소를 보였다. 또한, NaCl 농도가 높아질수록 발아에 걸리는 시간이 지연되었으며, 350 mM NaCl이 첨가된 처리구에는 화산101호 품종만이 치상 10일후에 발아가 시작하여 12% 정도 발아하였다. NaCl 농도에 따른 shoot과 root의 길이와 생초와 건조 후 무게의 변화도 만생종으로 갈수록 더 생육이 우수한 것으로 타나났다. 품종별간의 잎 조직에서 항산화효소 활성을 분석 결과 고염 조건 (150 mM NaCl)에서 POD 활성은 대조군과 유사한 수준으로 확인 되었다. 반면 APX 활성은 플로리다80과 코윈어리 품종에서는 증가하는 경향을, 화산101 품종에서는 감소하는 경향이 나타났으나 유의적인 수준에서 차이는 확인되지 않았다. 반면 그린팜 품종에서는 유의적인 수준에서 고염 처리에 의한 APX 활성이 감소되는 경향이 나타났다. 지속적으로 in vitro screening을 통해 선발된 계통들은 향후 특성평가와 인공교배를 통해 내염성 이탈리안 라이그라스 개발에 필요한 유용한 육종모재로 활용될 수 있을 것으로 판단된다.
