Standardization of Gentianae Macrophyllae Radix (진교(秦?, Gentianae Macrophyllae Radix)의 품질표준화에 관한 연구)

  • The Radix of Gentiana macrophylla (Gentianaceae) has been used for dispelling wind-damp, relaxing the sinews, and clearing heat from deficiency. In order to evaluate its quality, the method for the quantitative determination of gentiopicroside as standard compound has been developed. Gentiopicroside was a major compound of Gentianae Macrophyllae Radix. It was analyzed by HPLC using 30% MeOH as a solvent system. The amount of gentiopicroside from Gentianae Macrophyllae Radix was in the range of $2.22{\pm}1.22%\;(n=18)$.

In vitro Variant Induction and Its Content of Gentiopicroside of Gentiana scabra BUNGE (용담(龍膽)의 기내변이주(器內變異株) 유도(誘導)와 변이주(變異株)의 Gentiopicroside 함량(含量))

  • This study was aimed to mutant induction in Gentiana scabra BUNGE through the tissue culture techniques and comparison of gentiopicroside content in each mutant plants derived from tissue culture. In order to induce the mutants of Gentiam scabra, seed, apical and lateral bud were soaked in $NaN_3$, EMS and MNU solutions and cultured on MS basal medium containing 0.5mg/l NAA, BA $NaN_3$, MNU-treated buds survived about 50% but not survived in EMS treatment. Seed germination rate was extremely low in EMS and MNU treatments but various types of mutant plantlets were obtained by those treatments. 63% of elongated type and 37% of dwarf type were obtained from tissue culture treated by various mutagen. Gentiopicroside contents of regenerated plantlets was analyzed. Root of dwarf type contained more gentiopicroside(1.383%) than that of elongation type (0.083%).

  • Gentiana uchiyamana Nakai is a perennial herb in Gentianaceous plants. From the root of Gentiana uchiyamana Nakai, the bitter secoiridoid glucosides were isolated and identified as gentiopicroside and swertiamarin. A high-performance liquid chromatographic method for quantitative determination of bitter component, i.e. gentiopicroside in the roots of Gentiana scabra and G. uchiyamana Nakai was developed, using a ${\mu}$-Bondapak $C_{18}$ column and methanol-water mobile phases. The content of gentiopicroside was calculated from calibration curve previously prepared using the standard, 3.55% in the roots of Gentiana scabra and 1.58% in Gentiana uchiyamana Nakai.

Gentiopicroside Ameliorates the Progression from Hepatic Steatosis to Fibrosis Induced by Chronic Alcohol Intake

  • In current study, we aimed to investigate whether the gentiopicroside (GPS) derived from Gentiana manshurica Kitagawa could block the progression of alcoholic hepatic steatosis to fibrosis induced by chronic ethanol intake. C57BL/6 mice were fed an ethanol-containing Lieber-DeCarli diet for 4 weeks. LX-2 human hepatic stellate cells were treated with GPS 1 h prior to transforming growth factor-β (TGF-β) stimulation, and murine hepatocyte AML12 cells were pretreated by GPS 1 h prior to ethanol treatment. GPS inhibited the expression of type I collagen (collagen I), α-smooth muscle actin (α-SMA) and tissue inhibitor of metal protease 1 in ethanol-fed mouse livers with mild fibrosis. In addition, the imbalanced lipid metabolism induced by chronic ethanol-feeding was ameliorated by GPS pretreatment, characterized by the modulation of lipid accumulation. Consistently, GPS inhibited the expression of collagen I and α-SMA in LX-2 cells stimulated by TGF-β. Inhibition of lipid synthesis and promotion of oxidation by GPS were also confirmed in ethanol-treated AML12 cells. GPS could prevent hepatic steatosis advancing to the inception of a mild fibrosis caused by chronic alcohol exposure, suggesting GPS might be a promising therapy for targeting the early stage of alcoholic liver disease.

Basic Studies on Culture Practice of Gentiana scabra BUNGE (용담(龍膽) 재배기술(栽培技術) 개발(開發)을 위한 기초(基礎) 연구(硏究))

  • This experiment was carried out to obtain the basic information on cultivation of Gentiana scabra BUNGE. Optimum temperature for germination, planting time effect of chemicals on rooting for cutting propagation, effect of cutting on root yield, and content of gentiopicroside were investigated. The germination ratio was highest at $25^{\circ}C$ but lower at $10,\;15,\;20\;,30^{\circ}C.$ The optimum planting time was March 20 the cutting in May and NAA treatment were effective on rooting for cutting propagation. Inhibition of flowering by cutting was effective for increasing root yield but not effective for gentiopicroside content of root.

  • For the clonal proliferation of Gentiana scabra Bunge. which is one of the medicinaland ornamental plant, establishment multiplication of shoot through tissue culture technique and transplantation into soil were carried out. The shoot proliferation increased on the MS medium containing 0.5mg/l NAA and 0.5mg/l BAP. Optimum pH for shoot growth was pH 5.9, consequently MS medium supplemented with 2g/l activated charcoal was most effective for plant growth. There are two types of somaclonal variants, tall type was 63% and dwarf type was 37%.
