These studies were carried out to survey the genetic contamination of six inbred mice (A, BALB/c, C3H, C57BL/6, CBA and KK) produced and supplied from the conventional breeding unit for improving the quality of mice as experimental animal. We examined alleles of five loci (Akp-1, Car-2, Hbb, Es-1 and Trf) by the use of biochemical markers with celluose acetate electrophoresis. As the results of test, BALB/c, A, C3H, C57BL/6, CBA and KK showed standard alleles in Akp-1, Car-2 and Hbb. But Es-1 of A and C57BL/6 and Trf of A, C3H, C57BL/6 and CBA did allelic divergence in loci. These results suggest that the colonies of A, C3H, C57BL/6 and CBA were genetically contaminated. Therefore, we recommend to eliminate the genetically contaminated mice thoroughly, to check on genetic monitoring regularly and to consider a counterpaln for improving the quality control as soon as possible.
A newly developed optimization method which uses the genetic algorithm combined with the sensitivity analysis is presented in this paper. The genetic algorithm is a probabilistic method, searching the optimum at several points simultaneously, requiring only the values of the object and constraint functions. It has therefore more chances to find global solution and can be applied various problems. Nevertheless, it has such shortcomings that even it approaches the optimum rapidly in the early stage, it slows down afterward and it can't consider the constraints explicitly. It is only because it can't search the local area near the current points. The traditional method, on the other hand, using sensitivity analysis is of great advantage in searching the near optimum. Thus the combination of the two techniques makes use of the individual advantages, that is, the superiority both in global searching by the genetic algorithm and in local searching by the sensitivity analysis. Application of the method to the several test functions verifies that the method suggested is very efficient and powerful to find the global solutions, and that the constraints can be considered properly.
Rapid development in computer and network technology these days has created in environment in which decisions for manufacturing companies can be made in a much broader perspective. Especially, better decisions on production and distribution planning(PDP) problems can be made laking advantage of real time information from all the parties concerned. However, since the PDP problem-a core part of the supply chain management- is known to be the so-called NP-hard problem, so heuristic methods are dominantly used to find out solutions in a reasonable time. As one of those heuristic techniques, many previous studios considered genetic a1gorithms. A standard genetic a1gorithm applies rules of reproduction, gene crossover, and mutation to the pseudo-organisms so the organisms can pass along beneficial and survival-enhancing trails to a new generation. When it comes to representing a chromosome on the problem, it is hard to guarantee an evolution of solutions through classic a1gorithm operations alone, for there exists a strong epitasis among genes. To resolve this problem, we propose a hybrid genetic a1gorithm based on Silver-Meal heuristic. Using IMS-TB(Intelligent Manufacturing System Test-bed) problem sets. the good performance of the proposed a1gorithm is demonstrated.
Different genetic parameters for weaning weight and wool traits were estimated using restricted maximum likelihood (REML) in Angora rabbits. Total wool yield of first (I), second (II) and third (III) clips were taken as a separate trait under study. The records from more than 2,700 animals were analysed through fitting six animal models with various combinations of direct and maternal effects. A log likelihood ratio test was used to select the most appropriate model for each trait. Direct heritability estimates for the wool traits were found to be moderate to high across different models. Heritability estimates obtained from the best model were 0.24, 0.22, 0.20 and 0.21 for weaning weight, clip I, II and III; respectively. Maternal effects especially due to permanent environment had higher importance at clip I and found to be declining in subsequent clips. The estimates of repeatability of doe effect on wool traits were 0.44, 0.26 and 0.18 for clip I, II and III; respectively. Weaning weight had moderately high genetic correlations with clip I (0.57) and II (0.45), but very low (0.11) with clip III. Results indicated that genetic improvement for wool yield in Angora rabbit is possible through direct selection. Further, weaning weight could be considered as desirable trait for earliest indirect selection for wool yield in view of its high genetic correlation with wool traits.
Purpose: The purpose of this research, using descriptive correlation design was to identify the extent to which the mothers having children with rare genetic metabolic diseases(MPS, PWS) have parenting stress and guilt feeling. Method: This study used PSI /SF(Abidin, 1995) and Guilt Index as devised herein. From 156 mothers, data were collected from February to July 2006, using self-administered questionnaires. This study received the approval from IRB at S Hospital (IRB File No: 2006-02-014). Data were analyzed with descriptive statistics, t-test, ANOVA, and correlation. Results: Mothers felt very high level of parenting stress and sense of guilt. Parenting stress was related positively to guilt feeling. Conclusion: These findings could help understand the families of children with rare genetic metabolic diseases and those provide basic information in developing effective counseling and education programs for relief of parenting stress and guilt feeling. This study would be significant in the fact that it is the first research, targeting on the families of children with rare genetic metabolic diseases in Korea.
Inbred lines of Chinese cabbage KU-101 (resistant line against Plasmodiophora brassicae race race 6) and CS-113 (susceptible line) were crossed and their progeny lines F$_1$, BC$_1$F$_1$, F$_2$, and F$_3$ were produced for the construction of the genetic linkage map of R brassicae race 6-resistant Brassica campestris ssp. pekinensis genome. Restriction fragment length polymorphism (RFLP) was applied to compare between parents and their f$_2$ progenies with a total of 192 probes and 5 restriction enzymes. The constructed RFLP map covered 1,104 cM with a mean distance between genetic marker of 8.0 cM, and produced 10 linkage groups having 121 genetic loci. The loci of P. brassicae race 6 (CR6)-resistant Brassica genome were determined by interval mapping of quan-titative trait loci (QTL), which resulted from bioassay using the same race of the fungi in P3 population. Resistant loci were estimated in numbers 1 (Gl) and 3 (G3) linkage groups. In the regression test, Gl had a value of4.8 logarithm of odd (LOD) score, while C3 had values of 4.2-7.2. Given these results, the location of the CR6-resistant loci within the Brassica genome map can now be addressed.
Fusarium graminearum Schwabe causes Fusarium head blight (FHB), a devastating disease that leads to extensive yield and quality loss of wheat and other cereal crops. Twelve isolates of F. graminearum were collected from naturally infected spikes of wheat from Assiut Egypt. These isolates were compared using SRAP. The results indicated distinct genetic groups exist within F. graminearum, and demonstrated that these groups have different biological properties, especially with respect to their pathogenicity on wheat. There were biologically significant differences between the groups; with group (B) isolates being more aggressive towards wheat than groups (A) and (C). Furthermore, Trichoderma harzianum (Rifai) and Bacillus subtilis (Ehrenberg) which isolated from wheat kernels were screened for antagonistic activity against F. graminearum. They significantly reduced the growth of F. graminearum colonies in culture. In order to gain insight into biological control effect in situ, highly antagonistic isolates of T. harzianum and B. subtilis were selected, based on their in vitro effectiveness, for greenhouse test. It was revealed that T. harzianum and B. subtilis significantly reduced FHB severity. The obtained results indicated that T. harzianum and B. subtilis are very effective biocontrol agents that offer potential benefit in FHB and should be harnessed for further biocontrol applications. The accurate analysis of genetic variation and studies of population structures have significant implications for understanding the genetic traits and disease control programs in wheat. This is the first known report of the distribution and genetic variation of F. graminearum on wheat spikes in Assiut Egypt.
For the purpose of securing of strains which can be usefully utilized to study symbiosis between Rhizobium and legume plant, A. tumefaciens T7 was isolated and characterized and then subgroup biovar was determined. A. tumefaciens T7 induced smooth tumor like nopaline type one and did not grow at $37^{\circ}C$ and in the presence of 2% NaCl on yeast extract mannitol medium. The strain was able to grow on the New and Kerr selective media and utilize erythritol but not phenylalanine, tryptophan, and tartarate as a sole carbon source. Negative results were obtained from 3-keto-lactose production and oxidase test. The strain produced alkalifrom malonate and citrate and showed acid litmus milk reaction At least two large plasmids were detected in the cell lysate. According to all of these results, it could be concluded that subdivision of isolated strain was biovar 2.
There was no a difference of genetic analysis among methods(means, joint scaling test, 3 Parameter model) . The magnitude of additive effects generally paralleled the magnitude of difference between parental means and appeared to be more independent from non-allelic interaction than did dominance effects, whereas the magnitude of dominance effects were inflated by non-allelic interaction. Additive effects were significant for all characteristics observed and it was a major effects in inheritance of number of leaves. Dominance effects were higher than additive effects for plant height, days to flower, flesh leaf weight per plant, curing rate, total alkaloid and total nitrogen.
