Lipid Metabolism and Fatty Liver in Poultry (닭의 지방대사와 지방간)

  • A great progress in genetic selection, nutrition and management practices has contributed to the improved growth rate of broilers and egg production in laying hens. For the increased productivity of modern poultry, a healthy chicken liver needs to cope with the increased metabolic demands. The liver is the major site of de novo fatty acid synthesis; therefore, hepatic lipogenesis is crucial for producing better quality meat and eggs. When de novo lipogenesis exceeds the capacity of lipid metabolism and secretion, large amounts of lipids accumulate in the liver of broilers, leading to a fatty liver. Upon onset of egg-laying in hens, lipids including free fatty acids, triglycerides, and phospholipids are dramatically increased in blood plasma for the synthesis of yolk precursors in oocytes. Productive hens with fatty liver often have hemorrhagic syndrome and sudden death due to the heavy demands of yolk synthesis, which burdens the liver. Understanding the lipid metabolism and hepatic lipid disorders is a key point in the improvement of the growth and production of chickens. This review focuses on the recent studies on lipid metabolism, the hepatic lipid disorders, and the prevention or reduction of fatty liver in poultry.

Renal transplantation in a patient with Bartter syndrome and glomerulosclerosis

  • Bartter syndrome (BS) is a clinically and genetically heterogeneous inherited renal tubular disorder characterized by renal salt wasting, hypokalemic metabolic alkalosis and normotensive hyperreninemic hyperaldosteronism. There have been several case reports of BS complicated by focal segmental glomerulosclerosis (FSGS). Here, we have reported the case of a BS patient who developed FSGS and subsequent end-stage renal disease (ESRD) and provided a brief literature review. The patient presented with classic BS at 3 months of age and developed proteinuria at 7 years. Renal biopsy performed at 11 years of age revealed a FSGS perihilar variant. Hemodialysis was initiated at 11 years of age, and kidney transplantation was performed at 16 years of age. The post-transplantation course has been uneventful for more than 3 years with complete disappearance of BS without the recurrence of FSGS. Genetic study revealed a homozygous p.Trp(TGG)610Stop(TGA) mutation in the CLCNKB gene. In summary, BS may be complicated by secondary FSGS due to the adaptive response to chronic salt-losing nephropathy, and FSGS may progress to ESRD in some patients. Renal transplantation in patients with BS and ESRD results in complete remission of BS.

  • Recently, due to molecular biology and engineering technology, DNA microarray makes people watch thousands of genes and the state of variation from the tissue samples of living body. With DNA Microarray, it is possible to construct a genetic group that has similar expression patterns and grasp the progress and variation of gene. This paper practices Cluster Analysis which purposes the discovery of biological subgroup or class by using gene expression information. Hence, the purpose of this paper is to predict a new class which is unknown, open leukaemia data are used for the experiment, and MCL (Markov CLustering) algorithm is applied as an analysis method. The MCL algorithm is based on probability and graph flow theory. MCL simulates random walks on a graph using Markov matrices to determine the transition probabilities among nodes of the graph. If you look at closely to the method, first, MCL algorithm should be applied after getting the distance by using Euclidean distance, then inflation and diagonal factors which are tuning modulus should be tuned, and finally the threshold using the average of each column should be gotten to distinguish one class from another class. Our method has improved the accuracy through using the threshold, namely the average of each column. Our experimental result shows about 70% of accuracy in average compared to the class that is known before. Also, for the comparison evaluation to other algorithm, the proposed method compared to and analyzed SOM (Self-Organizing Map) clustering algorithm which is divided into neural network and hierarchical clustering. The method shows the better result when compared to hierarchical clustering. In further study, it should be studied whether there will be a similar result when the parameter of inflation gotten from our experiment is applied to other gene expression data. We are also trying to make a systematic method to improve the accuracy by regulating the factors mentioned above.

  • Many studies for verifying angiogenesis have been in progress, especially in the field of abnormal vascular proliferation to explain the pathogenesis and to develop a treatment of several diseases. In our previous experiments, endothelial cell proliferations were induced by DMH stimulation in vitro, and the 177 factors(142 up-regulated and 35 down-regulated factors) were identified. Among the up-regulated factors, 9 substances (EFEMP1, CTGF, CYR61, $ITG{\beta}1$, FHL2, SERPINE1, MYC, PTTG1 and MSH6) were selected, which were related to cell proliferation and showed high signal intensities. The RNA was isolated from HUVECs at the time of 0, 6, 12, 24 hours after the DMH treatment, and RNA of control group HUVECs was also isolated. Genetic information of selected molecules was used to make primer for each, and RT-PCR was performed to analyze both groups. In control and treatment groups, each substance presented variety of manifestation degree according to time differences. EFEMP1, CTGF, CYR61, $ITG{\beta}1$, FHL2 and MYC were related to abnormal vascular proliferation steadily and SERPINE1, PTTG1 and MSH6 were related secondarily. CTGF was related to both normal and abnormal proliferation, but it played a more significant role in abnormal proliferation from earlier stage. EFEMP1, CYR61, $ITG{\beta}1$, FHL2 and MYC were similar to CTGF, although the relation appeared lately. Further study should be performed to analyze the expressions and the interactions of growth factors, which could be utilized in the new therapeutic development.

A Case Report of MELAS syndrome Improved by Oriental Medicine Treatment (한방치료 후 호전된 MELAS 증후군 증례 1례)

  • Objective : Mitochondrial myopathy, encephalopathy, lactic acidosis, and stroke like episodes (MELAS) syndrome is a progressive neurodegenerative disorder. The typical presentation of patients with MELAS syndrome includes features such as mitochondrial encephalomyopathy, lactic acidosis, and stroke like episodes. Other features, such as seizures, diabetes mellitus, hearing loss, cardiac disease, short stature, endocrinopathies, exercise intolerance, and neuropsychiatric dysfunction are clearly part of the disorder. Approximately 80% of patients with the clinical characteristics of MELAS syndrome have a genetic mutation. This progressive disorder is reported to have a high morbidity and mortality. This case report is intended to estimate clinical effects of oriental meedicinal treatment of MELAS syndrome. Methods : A 44 year old female patient diagnosed as MELSA syndrome was treated with general oriental medicinal therapy including acupuncture, moxibustion, cupping, pharmacupunture and herbal medication in 20XX in Wonkwang Medical Center Gwangju. And the changes in symptoms and signs were evaluated as time dependently. Results : Although there is currently no curable treatment and MEALS syndrome is tend to progress, our treatment showed improvement in general weakness, gait disturbance and pain in the patient. Conclusion : Our case report suggests that various oriental medicinal treatment could be effective for improvement of MELAS syndrome and may represent a new potential therapeutic approach to control the disease. It could be applied to improve general condition, prevent relapse, enhance the quality of life and reduce complaints in the patient.

  • Abnormal distribution of the enteric nerves such as adrenergic, cholinergic and peptidergic nerves may cause the functional obstruction in Hirschsprung's disease (HD). Although the sustained contraction of the aganglionic segment is the main pathophysiology of HD, the etiology and pathogenesis is not thoroughly understood, With the recent progress of molecular biology and genetics,a more detailed approach to the pathogenesis of the HD can be undertaken. In this review, the roles of the nitric oxide, nitric oxide synthase and interstitial cells of Cajal on smooth muscle relaxation, the effects of extracellular matrix, cell adhesion molecules, neurotrophic factors on the migration and maturation of the neural crest cells are described. In the section of genetic factors, familial occurrences, association of chromosomal abnormalities, RET gene, glial cell line-derived neurotrophic factor gene, endothelin-3 gene and endothelin-B receptor gene and their r elationships to HD is briefly reviewed.

  • Forages are the backbone of sustainable agriculture. They includes a wide variety of plant species ranging from grasses, such as tall fescue and bermudagrass, to herbaceous legumes, such as alfalfa and white clover. Abiotic stresses, especially salinity, drought, temperature extremes, high photon irradiance, and levels of inorganic solutes, are the limiting factors in the growth and productivity of major cultivated forage crops. Given the great complexity of forage species and the associated difficulties encountered in traditional breeding methods, the potential from molecular breeding in improving forage crops has been recognized. Plant engineering strategies for abiotic stress tolerance largely rely on the gene expression for enzymes involved in pathways leading to the synthesis of functional and structural metabolites, proteins that confer stress tolerance, or proteins in signaling and regulatory pathways. Genetic engineering allows researchers to control timing, tissue-specificity, and expression level for optimal function of the introduced genes. Thus, the use of either a constitutive or stress-inducible promoter may be useful in certain cases. In this review, we summarize the recent progress made towards the development of transgenic forage plants with improved tolerance to abiotic stresses.

  • Unlike the commonly used On/Off solenoid, constant attraction force which is independent of plunger displacement is a considerably important characteristic to proportional solenoid of the EPPR Valve. Attraction force uniformity is mainly affected by the internal shape design parameters. Due to a number of shape design parameters, the optimal parameter values are very complex and time consuming to find by trial and error method. Much research has been conducted or are still in progress to find the optimal parameter values by applying various optimization techniques like Genetic Algorithm, Evolution Strategy, Simulated Annealing, or the Taguchi method. In this paper, the design parameters which have primary effects on the attraction force uniformity and the average attraction force are decided by main effects analysis of Design of Experiments. Optimal parameter values are derived using finite-element analysis and a neural network model.

Development of a multiplex PCR method for identification of four genetically modified maize lines and its application in living modified organism identification

  • Advances in biotechnology have led to progress in crop genetic engineering to improve agricultural productivity. The use of genetically modified (GM) crops has increased, as have consumers' and regulators' concerns about the safety of GM crops to human health, and ecological biodiversity. As such, the identification of GM crops is a critical issue for developers and distributors, and their labeling is mandatory. Multiplex polymerase chain reaction (PCR) has been developed and its use validated for the detection and identification of GM crops in quarantine. Herein, we established a simultaneous detection method to identify four GM maize events. Event-specific primers were designed between the junction region of transgene and genome of four GM maize lines, namely 5307, DAS-40278-9, MON87460, and MON87427. To verify the efficiency and accuracy of the multiplex PCR we used specificity analysis, limit of detection evaluation, and mixed certified reference materials identification. The multiplex PCR method was applied to analyze 29 living, modified maize volunteers collected in South Korea in 2018 and 2019. We performed multiplex PCR analysis to identify events and confirmed the result by simplex PCR using each event-specific primer. As a result, rather than detecting each event individually, the simultaneous detection PCR method enabled the rapid analysis of 29 GM maize volunteers. Thus, the novel multiplex PCR method is applicable for living modified organism volunteer identification.

Comparison of Research Characteristics in Western, Chinese Traditional Medicine and Korean Medicine on Psoriasis (건선의 동서의학적 연구 특징의 비교)

  • Objectives: We compared research characteristics of western medicine, Chinese medicine and Korean medicine on causes, mechanisms, types, treatments and prevention of psoriasis. Methods: For western medicine, "Psoriasis" was used as keyword on Pubmed, for Chinese medicine, "銀屑病" and "中医" on CNKI (China National Knowledge Infrastructure" and for Korean medicine, "건선" on OASIS. Keyword searches were done for papers and books published after 2010. For Chinese medicine, there were more in-depth searches done for "從血論 (血熱, 血瘀, 血燥)" and "陽虛症". Results: Western medicine puts an emphasis on the foci, and approaches it from molecular and genetic levels based on molecular biology; while it views psoriasis as a disease with multiple possible causes, it ultimately sees it as an inflammation that is immunity-mediated. Western medicine seeks to suppress cytokine in order to prevent and eliminate inflammation at each stage of treatment While they are effective short-term, psoriasis recurs shortly after. Chinese and Korean medicines categorize psoriasis as an internal comprehensive systemic diseases that encompasses the patient's physical and mental characteristics, and defines it as a disease that has many causes and mechanisms such as "血熱, 血瘀, 血燥" and "陽虛". They use herbal medicine, acupuncture, and lifestyle interventions to improve the overall health of the patient in addition to treating psoriasis. Treatments are effective, but it takes relatively longer to see results, and can recur. Conclusion: In order for more progress to happen on psoriasis treatment, each branch of medicine must exchange knowledge and information more frequently.
