Analysis of Evolutionary Content in High School Biology Textbook (고등학교 생물 교과서에서의 진화내용분석)

  • This study analyzed the evolutionary content in 13 textbooks developed from the first to the 6th high school biology curriculum, The content analysis of textbooks, which were delineated nine component, was performed on the 80 evolutionary categories, According to the result, the proportion of the total evolutionary content in textbook increased from the textbooks developed by the Ist curriculum to the textbooks developed by the 6th curriculum, but the proportion of 'main narrative' in total evolutionary content was gradually decreased. It also showed that biology curriculum and points of view of textbook writers influenced on the proportion of evolutionary contents. On the whole, the topics of analysed textbooks exhibit insufficient diversity, Any categories- 'group selection', 'gene selection', 'gaps in fossil record', 'co-evolution', 'punctuated equilibrium', 'mosaic evolution', 'place of labor in human evolution', 'human race differentiation', 'criticism of "ontogeny recapitulates phylogeny", and 'human activities affecting evolution' - were not treated and others - 'theory of neutralism', 'theories of major episodes(excepting extinctions) found in the geologic time scale', 'sympatric speciation', 'clinal and area-effect speciation', 'polyploidy and evolution', 'gradualism' and 'evolution and origin of mammals' - were treated very lightly, the most emphasized topic was 'phylogeny in general' and 'formation of precells', 'miscellaneous' in the order of emphasis. 'Theory of natural selection' was lightly treated as just one of evolutionary theory though it should be emphasized as major theme of evolution. Also, the law of recapitulation, of which biologists doubt the validity, was discussed as an evidence of evolution in some textbooks. And the agents of genetic equilibrium disruption like genetic drift and migration were treated as of little importance. On the basis of above result, it was suggested that the textbook writers introduced the more meaningful evolutionary topics focused the theory of natural selection in explanation of evolution and evolution theory.

A comparative study of carcass characteristics and meat quality in genetic resources Pekin ducks and commercial crossbreds

  • Objective: The study was aimed to compare carcass traits, physicochemical and textural properties of meat in two different genotypes of Pekin ducks with regard to sex effect. Methods: The study involved 120 Pekin ducks: 30 males and 30 females of strain P33 (Polish native Pekin ducks) and 30 males and 30 females of Star 53 HY (commercial hybrid Pekin ducks). At 49 d of age, 48 birds (12 males and 12 females of each genotype) were selected for dissection. After the dissection, meat samples were collected to determine meat quality traits. Results: The studied Pekin ducks of different genotype showed significant differences in body weight, carcass weight, dressing percentage, as well as percentages of breast muscles, skin with subcutaneous fat, abdominal fat, neck, and remainders of eviscerated carcass with neck. Duck genotype influenced the content of crude protein, crude fat, Na, K, P, Zn, $pH_{24}$, electric conductivity ($EC_{24}$), cooking loss, $L^{\star}$, $a^{\star}$, most textural traits of breast muscle, and also Na, Mg and Fe content, $EC_{24}$, drip loss, cooking loss and $L^{\star}$, $a^{\star}$, and $b^{\star}$ colour coordinates of leg muscles. Regardless of genetic origin, males exhibited higher BW, carcass weight and carcass neck percentage, as well as lower redness, hardness, chewiness and gumminess of breast muscle compared to females. The genotype${\times}$sex interaction was significant for the crude fat content and cooking loss of breast muscle, and for the yellowness of leg muscle. Conclusion: Star 53 HY ducks are more suited for broiler production due to their higher body weight and dressing percentage. Their breast and leg meat are characterized by more beneficial chemical composition but has poorer sensory and textural properties compared to the meat of P33 ducks.

  • This paper deals with the genetic properties of R plasmids in Salmonella originated from pigs and cattle. The plasmid DNA was examined for incompatibility, stability and fertility inhibition(Fi), and gel electrophoresis was performed for isolation of plasmid DNA. The results obtained were summerized as follows: 1. Among the 66 conjugative R plasmids from 44 pigs and 22 cattle, 61 R plasmids (92.4%) were $Fi^-$, whereas the remainder were $Fi^+$. 2. The Inc groups of 66 R plasmids were determined with 7 standard plasmids. Twenty-six R plasmids were classified into Inc group $I{\alpha}$, H1, H2 or F1, 40 R plasmids being not classified with standard plasmids used, and the Inc group $I{\alpha}$ (57.7%) was most frequent. 3. Inc groups $I{\alpha}$, H1, and F1 were identified in strains from swine, Inc groups H2 and F1 from cattle. 4. The plasmid DNA profiles in 16 Salmonella isolated from pigs and cattle were confirmed as being 1 to 10 fragments by the gel eletrophoresis. Their molecular weight ranged 1.0 to 90 megadalton. 5. The molecular weight of conjugative plasmids ranged 1.0 to 80 megadalton in 4 Salmonella (P-4, P-5, P-7 and P-8) isolated from pigs.

Au-Ag Minerals and Genetic Environments from the Yeongdeog Gold-Silver Deposits, Korea (영덕(盈德) 금(金)-은광상(銀鑛床)에서 산출(産出)되는 금(金)-은광물(銀鑛物)과 광상(鑛床)의 생성환경(生成環境))

  • The Yeongdeog gold-silver deposits at Jipum, Gyeongsangbugdo, is of a middle Paleogene $(45.52{\pm}1.02Ma)$ vein type, and is hosted in shale and sandstone of Cretaceous age. Based on mineral paragenesis, vein structure and mineral assemblages, the ore mineralization can be divided into two distinct depositional stages. The early stage is associated with base-metals such as pyrite, arsenopyrite (27.99~30.99 at%), hematite, rutile, pyrrhotite, sphalerite (10.53~18.42 FeS mole%), chalcopyrite and galena with wallrock alteration such as chlorite, sericite and pyrite. The late stage is characterized by the Au-Ag mineralization such as electrum, Ag-bearing tetrahedrite, freibergite, pyrargyrite, unidentified mineral, pyrite, sphalerite (1.08~5.57 FeS mole%), chalcopyrite and galena. Fluid inclusion data indicate that fluid temperatures and salinities range from 343 to $227^{\circ}C$ and from 8.3 to 5.7 wt% eq. NaCl in early stage, respectively. Temperatures and salinities of NaCl eq. wt% range from 299 to $225^{\circ}C$ and from 12.9 to 4.3 in late stage, respectively. They suggest that complex cooling histories were occured by the mixing of the fluids. Sulfur fugacity $(-logfs_2)$ deduced by mineral assemblages and composition ranges from 8.3 to 14.7 atm. in early stage, and from 8.8 to 14.5 atm. in late stage. It suggests that the mineralization was related to decrease of temperature in early stage and fluctuations of $fS_2$ with decrease of temperature in late stage. Sulfur and oxygen isotope compositions are 4.48~5.60‰ and 9.25~10.8% in early stage, and late stage is 4.84~7.00‰ and 5.7‰, respectively. It indicated that hydrothermal fluids may be magmatic origin with some degree of mixing of another water during paragenetic time.

  • Citrus is a common term and genus (Citrus) of flowering plants in the rue family, Rutaceae. Citrus is believed to have originated in the part of Southeast Asia bordered by Northeastern India, Myanmar (Burma) and the Yunnan province of China. The taxonomy and systematics of the genus are complex and the precise number of natural species is unclear, as many of the named species are clonally propagated hybrids, and there is genetic evidence that even some wild, true-breeding species are of a hybrid origin. One of the most popular sequences for phylogenetic inference at the generic and infrageneric levels in plants is the chloroplast trnL-trnF region. We evaluated the seven taxa with the trnL-trnF region to estimate phenotypic relationships within the genera Citrus, Poncirus, and Fortunella in Korea. Alignment of the DNA sequences required the addition of numerous gaps. Sequence variation within Citrus was mostly due to insertion/deletion. Within the genus Citrus, C. lomonia and C. sinensis were relatives and sistered with C. aurantium in the four phylogenetic analyses (MP, ML, ME, and NJ). However, some external nodes were poorly supported.

Screening of Sclerotinia Rot Resistant Korean Origin Perilla (Perilla frutescens) Germplasm Using a Detached Leaf Method

  • Sclerotinia rot, caused by Sclerotinia sclerotiorum, is a devastating disease that poses a serious threat to perilla production in Korea. Identifying effective sources of resistance offers long term prospects for improving management of this disease. Screening disease resistant genetic resources is important for development of disease-resistant, new cultivars and conduct related research. In the present study, perilla germplasm were screened in vitro against S. sclerotiorum using detached leaf method. Among 544 perilla accessions, two were highly resistant (IT226504, IT226533), five were resistant (IT226561, IT226532, IT226526, IT226441, and IT226589), five were moderately resistant (IT226525, IT226640, IT226568, IT220624, and IT178655), 16 were moderately susceptible, 31 were susceptible, and 485 were highly susceptible. The resistant accessions in this study could serve as resistance donor in the breeding of Sclerotinia rot resistance or subjected to selection procedure of varietal development for direct use by breeders, farmers, researchers, and end consumers.

Wall Rock Alteration and Genetic Environment of the Milyang Pyrophyllite Deposit (밀양 납석광상의 모암변질작용과 생성환경)

  • Milyang pyrophyllite deposit which was formed by hydrothermal alteration occurs in Late Cretaceous andesitic tuff in the Milyang area, Gyeongsangnamdo. The wall rock alteration and genesis of the Milyang pyrophyllite deposit were studied. The ore minerals are composed dominantly of pyrophyllite accompanied by small amounts of quartz, kaolinite, pyrite, dumortierite and diaspore. The alteration halo of this deposit can be divided into three zones on the basis of mineral assemblage; pyrophyllite, sericite and chlorite zone. The common mineral assemblages of each alteration zone are as follows: (1) pyrophyllite zone; pyrophyllite-quartz-kaolinite-pyrite-dumortierite-diaspore, (2) sericite zone; sericite-quartz-pyrite-kaolinite, and (3) chlorite zone; chlorite-plagioclase-quartz. Major element chemistry shows that characteristic depletion in MgO, CaO, and $Na_2O$ and relative increase in FeO from less altered chlorite zone to extensively altered pyrophyllite zone corresponding to variation in mineral assemblages. The paragenesis of ore minerals, oxygen isotope data, chlorite and illite geothermometry suggest that ore deposit was formed at about $250{\sim}330^{\circ}C$. Both hydrogen and silica activities are high in pyrophyllite zone. Potassium activity increases in sericite zone while hydrogen activity becomes low in chlorite zone. The pyrophyllite zone was formed relatively higher temperature than those of sericite and chlorite zones. The ore fluid was considered to be magmatic water in origin derived from the residual granitic magma which interacted with meteoric water.

Analysis of Genetic Diversity of Phytophthora infestans in Korea by Using Molecular Markers

  • A total of 367 isolates of Phytophthora infestans was collected from the leaf samples of late blight disease from five provinces in Korea over the three growing seasons of 2002-2004. Of the 367 isolates, 337 isolates were of the A1 mating type, and 30 isolates were of A2 mating type, showing that the majority was A1 mating type. Profiles of Gpi and Pep defined four allozyme genotypes among the total of 367 isolates. All four allozyme genotypes could be distinguished on the basis of Gpi profiles alone, whereas all isolates were homozygous at the Pep locus (100/100). The mitochondrial DNA haplotype of all isolates were the IIa haplotype. Amplification of the genomic DNAs extracted from eight isolates of each mating type by polymerase chain reaction with the selected primer (OPC-5 primer) produced a total of 20 DNA bands, of which 11 bands were polymorphic. According to the RAPD analysis using the OPC-5 primer, 106 isolates including two standard isolates were separated into 8 groups at the similarity level of 92.5%. The RAPD groups were not correlated with the allozyme genotypes and the isolated locations. All of the eight RAPD groups were identified in Gangwon-do, suggesting that Gangwon-do is the center of origin of the P. infestans in Korea. A 600-bp DNA band generated with the OPC-5 primer was specific to A1 mating type isolates, but not detected with A2 mating type, showing that the specific PCR primer can distinguish different mating types in P. infestans.

The New LM-PCR/Shifter Method for the Genotyping of Microorganisms Based on the Use of a Class IIS Restriction Enzyme and Ligation-Mediated PCR

  • This study details and examines a novel ligation-mediated polymerase chain reaction (LM-PCR) method. Named the LM-PCR/Shifter, it relies on the use of a Class IIS restriction enzyme giving restriction fragments with different 4-base, 5' overhangs, this being the Shifter, and the ligation of appropriate oligonucleotide adapters. A sequence of 4-base, 5' overhangs of the adapter and a 4-base sequence of the 3' end of the primer(s) determine a subset of the genomic restriction fragments, which are amplified by PCR. The method permits the differentiation of bacterial species strains on the basis of the different DNA band patterns obtained after electrophoresis in polyacrylamide gels stained with ethidium bromide and visualized in UV light. The usefulness of the LM-PCR/Shifter method for genotyping is analyzed by a comparison with the restriction endonuclease analysis of chromosomal DNA by the pulsed-field gel electrophoresis (REA-PFGE) and PCR melting profile (PCR MP) methods for isolates of clinical origin. The clustering of the LM-PCR/Shifter fingerprinting data matched those of the REA-PFGE and PCR MP methods. We found that the LM-PCR/Shifter is rapid, and offers good discriminatory power and excellent reproducibility, making it a method that may be effectively applied in epidemiological studies.

  • Gene therapy holds great promise for treating various forms of diseases with a genetic origin including cystic fibrosis, different forms of cancer, and cardiovascular disorders. The clinical use of gene therapy treatments is however restricted, mainly because of the absence of safe and efficient gene delivery technologies. In our group, with an aim of developing efficient and nontoxic polymeric gene delivery systems, several novel types of polymeric gene carriers have been designed, synthesized, and evaluated. Herein, I will mainly present our recent work on low molecular weight linear PEI-PEG-PEI triblock copolymers, degradable hyperbranched poly(ester amine)s, and reduction-sensitive poly(amido amine)s.
