Postmortem skeletal muscle metabolism of farm animals approached with metabolomics

  • Skeletal muscle metabolism regulates homeostatic balance in animals. The metabolic impact persists even after farm animal skeletal muscle is converted to edible meat through postmortem rigor mortis and aging. Muscle metabolites resulting from animal growth and postmortem storage have a significant impact on meat quality, including flavor and color. Metabolomics studies of postmortem muscle aging have identified metabolisms that contain signatures inherent to muscle properties and the altered metabolites by physiological adaptation, with glycolysis as the pivotal metabolism in postmortem aging. Metabolomics has also played a role in mining relevant postmortem metabolisms and pathways, such as the citrate cycle and mitochondrial metabolism. This leads to a deeper understanding of the mechanisms underlying the generation of key compounds that are associated with meat quality. Genetic background, feeding strategy, and muscle type primarily determine skeletal muscle properties in live animals and affect post-mortem muscle metabolism. With comprehensive metabolite detection, metabolomics is also beneficial for exploring biomarker candidates that could be useful to monitor meat production and predict the quality traits. The present review focuses on advances in farm animal muscle metabolomics, especially postmortem muscle metabolism associated with genetic factors and muscle type.

  • In this paper, we deal with a hierarchical location-allocation problem in designing the broadband convergence networks (BcN). The objective is to minimize the total cost of switch and cable while satisfying the quality of service (QoS). We formulate the problem as an integer programming model and develop the Tabu Search (TS) heuristic algorithm to find a good feasible solution within a reasonable time limit. Initial solution is obtained by using the tree structure. Three neighborhood generation mechanisms are used by local search heuristic: insertion, switch up, and switch down. In order to demonstrate the effectiveness of the proposed algorithm, we generate lower bounds from nonlinear QoS relaxation problem. We present promising computational results of the proposed solution procedures.

In this research, emission factors and additionality proving process are studied, which are important procedures of doing CDM project.

Hydriding Failure Analysis Based on PIE Data

  • Recently failures of nuclear fuel rods in Korean nuclear power plants were reported and their failure causes have been investigated by using PIE techniques. Destructive and physico-chemical examinations reveal that the clad hydriding phenomena had caused the rod failures primarily and secondarily in each case. In this study, the basic mechanisms of the primary and the secondary hydriding failures are reviewed, PIE data such as cladding inner and outer surface oxide thickness and the restructuring of the fuel pellets are analyzed, and they are compared with the predicted behaviors by a fuel performance code. In addition, post-defected fuel behaviors are reviewed and qualitatively analyzed. The results strongly support that the hydriding processes, primary and secondary, played critical roles in the respective fuel rods failures and the secondary hydriding failure can take place even in the fuel rod with low linear heat generation rate.

  • To investigate failure mechanisms observed in carbon-phenolic thermal insulators, differential equations which govern the decomposition process in a deformable anisotropic porous solid are derived for three-dimensional axisymmetric constructions. The governing equations not only couple the material deformation with pore pressure, but also couple pressure and temperature, which means that heat convected by the pyrolysis gases is properly accounted for. Then the Bubnov-Galerkin finite element method is applied to these equations to transform them into a semidescrete finite element system. A thermal insulation liner in the cowl region under typical operating conditions is analyzed to find a mechanism for plylift. The results from the structural analysis show across-ply failure in the cowl zone. The mechanism for plylift is hypothesized as a sequential procedure : 1) the across-ply failure which is the precursor to plylift and 2) the local fiber buckling caused by generation of excessive in-plane compressive stress. To prevent plylift, the across-ply stress can be reduced by using appropriate material ply angles in cowl zone design.

  • 환경세의 이중배당가설은 1980년대 중반 처음 제기된 이래, 최근까지 그 실현가능성에 대한 논의가 매우 활발한 연구주제이다. 이중배당가설의 실현가능성에 대해 부정적 입장을 견지하였던 2세대 연구들과는 달리, 3세대 연구들은 가설이 실현될 수 있는 조건에 초점을 맞추어 왔다. 3세대 연구들은 함수적 가정이나 환경세 부과대상 재화의 성질, 그리고 초기 조세체계에 대한 가정 등 다양한 경로를 통해 이중배당이 가능함을 주장하고 있다. 하지만 한 가지 흥미로운 점은, 3세대 연구들은 외형적으로 전혀 다른 접근방법을 취하고 있는 것으로 보이지만 그 작동기제를 살펴보면 노동시장에 유사한 영향을 미친다는 점에서 공통점이 발견된다는 것이다. 본 논문은 이러한 점에 착안하여 수행되었으며, 환경세 정책이 노동시장에 미치는 영향에 논의의 초점을 맞추고 이론적, 수리적 분석을 시도하였다. 분석결과는 이와 같은 이론적 직관과 일치하는 것으로 나타났다. 즉, 이중배당가설의 성립 여부는 정책변화가 노동시장에 미치는 영향이 매우 중요하며, 특히 가설이 성립하기 위해서는 환경세 정책이 노동공급의 증가를 유발해야 한다는 것이다.

Recent Progress in Methods of Generating Water Mist for Fire Suppression

  • To prevent the ozonosphere from being destroyed by Halon, it is an urgent task to find out Halon replacement. As one of the replacements water mist have showed broad applications by its advantages: little pollution to environment (not destroying the ozone layer or bring green house effect), extinguishing fire quickly, consuming a small quantity of water and having little damage to the protected objects. The methods of generating water mist strongly influence fire suppression effectiveness, which determine the cone angle, drop size distribution, flux uniformity, and momentum of the generating spray. The traditional water mist nozzle included pressure jet nozzles, impingement nozzles and twin-fluid nozzles. All of them have more or less disadvantages for fire suppression. Therefore, many research institutes and corporations are taking up with innovations in mist generation. This article provided some recent studies in State Key Laboratory of Fire Science (SKLFS) of University of Science and Technology of China. SKLFS have investigated new methods of generating water mist (i.e. effervescent atomization and ultrasonic atomization). and self developed a series of nozzles and developed advanced DPIVS (Digital Particle Image Velocimetry and Sizing) technique. Characteristics of water mist (the distribution of droplet sizes, flux density, spray dynamics and cone angle) produced by these nozzles were measured under different conditions (work pressure, nozzle geometry, etc.) using LDV/APV and DPTVS systems. A series of experiments were performed to study the fire suppression effectiveness in different fire scenario (different kindsof the fuel, fire size and ventilation conditions). The fire extinguishing mechanisms of water mist was also discussed.

Mechanical properties and failure mechanisms of sandstone with pyrite concretions under uniaxial compression

  • A uniaxial compression test was performed to analyse the mechanical properties and macroscale and mesoscale failure mechanisms of sandstone with pyrite concretions. The effect of the pyrite concretions on the evolution of macroscale cracks in the sandstone was further investigated through numerical simulations with Particle Flow Code in 2D (PFC2D). The results revealed that pyrite concretions substantially influence the mechanical properties and macroscale and mesoscale failure characteristics of sandstone. During the initial loading stage, significant stress concentrations occurred around the edges of the pyrite concretion accompanied by the preferential generation of cracks. Meanwhile, the events and cumulative energy counts of the acoustic emission (AE) signal increased rapidly because of friction sliding between the concretion and sandstone matrix. As the axial stress increased, the degree of the stress concentration remained relatively unchanged around the edges of the concretions. The cracks continued growing rapidly around the edges of the concretions and gradually expanded toward the centre of the sample. During this stage, the AE events and cumulative energy counts increased quite slowly. As the axial stress approached the peak strength of the sandstone, the cracks that developed around the edges of the concretion started to merge with cracks that propagated at the top-left and bottom-right corners of the sample. This crack evolution ultimately resulted in the shear failure of the sandstone sample around the edges of the pyrite concretions.

Activation of SAPK and Increase in Bak Levels during Ceramide and Indomethacin-Induced Apoptosis in HT29 Cells

  • It has been reported that activation of sphingomyelin pathway and nonsteroidal anti-inflammatory drugs (NSAIDS) inhibit the promotion of colon carcinoma. Ceramide, a metabolite of sphingomyelin, and indomethacin were shown to induce apoptosis in colon carcinoma cells. However, the mechanisms of ceramide- and indomethacin-induced apoptosis in the colon carcinoma cells are not clearly elucidated. Recent studys showed that indomethacin-induced apoptosis in colon cancer cells through the cyclooxygenase-independent pathways, and that may be mediated by generation of ceramide. In this study, we compared effects of ceramide and indomethacin on important modulators of apoptotic processes in HT29 cells, a human colon cancer cell line. Ceramide and indomethacin induced apoptosis dose- and time- dependently. Ceramide and indomethacin increased stress-activated protein kinase (SAPK) activity, and decreased mitogen-activated protein kinase (MAPK) activity. The expression of Bak was increased by the treatment of ceramide and indomethacin. The expression of other Bcl-2 related proteins (Mcl-1, $Bcl-X_L,$ Bax) which were known to be expressed in colon epithelial cells was not changed during the ceramide- and indomethacin-induced apoptosis. Our results suggest that ceramide and indomethacin share common mechanisms for induction of apoptosis in HT29 cells.
