Single-minded 1 Gene Mapping and Its Variants Association with Growth, Carcass Composition and Meat Quality Traits in the Pig

  • Single-minded 1 gene (SIM1) is a homolog of Drosophila SIM1 gene which plays a key role in the midline cell lineage of the central nervous system and is implicated in the regulation of feeding behavior and obesity in the human and mouse. In this study, porcine SIM1 gene was firstly mapped to chromosome 1p13 using radiation hybrid (RH) mapping and two polymorphisms were detected at position 607 (A/G) in SIM1 intron7 and position 780 (C/T) in SIM1 exon8. The last substitution was genotyped in a 364 F2 animal-population and an association analysis of these genotypes was performed with growth, carcass and meat quality traits by the statistical animal model. The results showed the significant influence of the SIM1 genotype on growth (p<0.05): live weight at birth, later period of growth and average daily gain; and effects on carcass composition (p<0.05): weight of head and buck kneed foreleg, backfat depth, loin eye area, carcass leaf fat and ham fat weights; and traits related to intramuscular fat content (p<0.05).

Grain Size Relate Gene in CNDH, and Identification Of Shape Based on QTL Mapping in Rice

  • Rice is 34% of the world's population used as a staple food. But the world population is increasing. Food security is not well protected. Improving cultivar development can address food security. Quantitative trait locus (QTL) mapping is a statistical analysis using both phenotypic and genotypic dates. The purpose of QTL mapping is to determine a gene. Increasing grain size is a way to increase yield in rice. Grain size-related genes were mapped using CNDH population obtained by cross-breeding Cheongcheong (Indica) and Nagdong (Japonica) through anther culture. Grain harvested from experimental field of Kyungpook National University in Gunwi in 2021. Genes related to grain length were detected between RM5964-RM12285, RM20924-RM20967 in chromosome 1, 7. LOD score is 5.88 and 5.6. Genes related to grain width was detected between RM289-RM18130 in chromosome 5. LOD score is 4.57. Genes related to grain length/width ratio were detected between RM5459-RM3482, RM5699-RM1211 and RM3838-RM3381 in chromosome 1, 2, 5. LOD score is 3.75, 3.14 and 3.41. 4 genes was detected in chromosome 1 and 2 genes was detected in chromosome 2 and 7 genes was detected in chromosome 5. 2 genes related to grain shape and quality were detected. 4 genes related to grain length were detected. 4 genes related to grain size were detected. 1 gene related to grain size and weight was detected. 2 genes related to grain length and weight were detected. By finding the gene related to grain size, it provides food to people threatened by food security and solves the food shortage.

  • Advances in molecular biology have renewed hope for the discovery of disease relevant gene. The basic strategy is gene mapping and likely to have on important role in psychiatric research and practice. Recent linkage studies of chromosomal loci to psychiatric diseases shed light on the potential for new genetics in psychiatric science. This article reviews molecular application to psychiatrymethodological issues in genetic linkage, study of gene expression by analysis of mRNA, and current linkage studies in psychiatric diseases.

E. coli B. MQ1844 균주의 오존감수성 유전자의 특성 (A gene responsible for ozone sensitivity (ozrB) in chromosome of escherichia coli B. MQ 1844)

  • An ozone-sensitive mutant of Escherichia coli strain B, MQ 1844 is described. Its properties, including high sensitivity to ozone and radiation, inducible filamentation, extensive DNA degradation and impaired DNA synthesis following ozonation, are attributable to a mutation in ozrB, a gene which is cotransducible with malB. Based on differences in phenotypic expression as well as on the particular location of this gene on the bacterial chromosome, ozrB appears as distinct from the other ozone-or radiation-sensitivity genes previously described.

Mapping, Tissue Distribution and Polymorphism Study of the Porcine SOCS2 and SOCS3 Genes

  • Using the somatic cell hybrid panel (SCHP) and radiation hybrid (IMpRH) panel, porcine SOCS2 gene was mapped at SSC5 (1/2) q21-q24 and closely linked with SW1383 marker (47 cR in distance), while SOCS3 gene was assigned to SSC12p11-(2/3p13) and closely linked with SW2490 (43 cR). The reverse transcriptase-polymerase chain reaction (RT-PCR) was performed to detect the expression of these two genes in the different tissues and the results showed that both SOCS2 and SOCS3 genes were widely expressed in tissues investigated (heart, liver, spleen, lung, kidney skeletal muscle, fat and brain), although some tissues showed lower gene expression. Moreover, SOCS2 and SOCS3 genes had different expression levels at different stages, in different tissues and in different breeds. A G/A substitution, which can be recognized by restriction enzyme of Cfr421, was observed in 5' untranslated region (5'-UTR) of SOCS2 gene. The allele frequencies was investigated by PCR-restriction fragment length polymorphism (PCR-RFLP) method and it showed that the allele frequency among Dahuabai, Erhualian, Yushan, Qingping, Large white and Landrace tested were different. Association analysis in a cross experimental populations revealed no significant association between the SOCS2 gene polymorphism and the economic traits investigated. The full-length coding regions (CDs) of porcine SOCS3 gene was obtained by RT-PCR.

  • This is a progress report of rice biotechnology including development of gene transformation system, gene cloning and molecular mapping in rice. The scope of the research was focused on the connection between conventional breeding and biotech-researches. Plant transformation via Agrobacterium or particle bombardment was developed to introduce one or several genes to recommended rice cultivars. Two chimeric genes containing a maize ribosome inactivating protein gene (RIP) and a gerbicide resistant gene (bar) were introduced to Nipponbare, a Japonica cultivar, and transmitted to Korean cultivars. The homozygous progenies of herbicide resistant transgenic plant showed good fertility and agronomic characters. To explore the genetic resourses in rice, over 8,000 cDNA clones from immature rice seed have been isolated and sequenced. About 13% of clones were identified as enzymes related to metabolic pathway. Among them, twenty clones have high homology with genes encoding enzymes in the photorespiratory carbon cycle reaction. Up to now about 100 clones were fully sequenced and registered at EMBL and GenBank. For the mapping of quantitative tarits loci (QTL) and eternal recombinant inbred population with 164 F13 lines (MGRI) was developed from a cross between Milyang 23 and Gihobyeo, Korean rice cultivars. After construction of fully saturated RFLP and AFLP map, quantitative traits using MGRI population were analyzed and integrated into the molecular map. Eighty seven loci were determined with 27 QTL characters including yield and yield components on rice chromosomes. Map based cloning was also tried to isolate semi-dwarf (sd-1) gene in rice. A DNA probe, RG 109, the most tightly linked to sd-1 gene was used to screen from bacterial artifical chromosome (BAC) libraries and five over lapping clones presumably containing sd-1 gene were isolated. Rice genetic database including results of biotech reasearch and classical genetics is provided at Korea Rice Genome Server which is accessible with world wide web (www) browser. The server provides rice cDNA sequences and map informations linked with phenotypic images.

Prospects of Application of Linkage Disequilibrium Mapping for Crop Improvement in Wild Silkworm (Antheraea mylitta Drury)

  • The wild silkworm, Antheraea mylitta Drury (Lepidoptera: Saturniidae) is a polyphagous silk producing insect that feeds on Terminalia arjuna, T. tomentosa and Shorea robusta and is distributed in the forest belts in different states of India. Phenotypically distinct populations of the A. mylitta are called "eco-race" or "ecotypes". Genetic improvement of this wild silkworm has not progressed much due to lack of adequate information on the factors that control the expression of most of the economically important traits. Considering the amazing technological advances taking place in molecular biology, it is envisaged that it is now possible to take greater control on these intractable traits if a combination of genetic, molecular and bioinformatics tools are used. Linkage disequilibrium (LD) mapping is one such approach that has extensively been used in both animal and plant system to identify quantitative trait loci (QTLs) for a number of economically important traits. LD mapping has a number of advantages over conventional biparental linkage mapping. Therefore, LD mapping is considered more efficient for gene discovery to meet the challenge of connecting sequence diversity with heritable phenotypic differences. However, care must be taken to avoid detection of spurious associations which may occur due to population structure and variety interrelationships. In this review, we discuss how LD mapping is suitable for the dissection of complex traits in wild silkworms (Antheraea mylitta).

Linkage Disequilibrium (LD) Mapping and Tagging SNP Selection of C-Fos Induced Growth Factor (Figf) Gene in Korean Population

  • We performed comprehensive SNP validation and linkage disequilibrium (LD) analysis of the c-fos induced growth factor (Figf) gene in Korean population. Out of 32 SNPs, only 9 SNPs were polymorphic in Korean population. Validated SNPs formed a single extended haplotype block with strong LD through the entire length of the gene. Tagging SNP analysis picked only 2 SNPs to represent most of the genetic variation information of the Figf gene. Our results demonstrate the utility of LD block and tagging SNP analysis for an efficient way of performing a candidate gene based association study.

Identification of Superior Single Nucleotide Polymorphisms (SNP) Combinations Related to Economic Traits by Genotype Matrix Mapping (GMM) in Hanwoo (Korean Cattle)

  • It is important to identify genetic interactions related to human diseases or animal traits. Many linear statistical models have been reported but they did not consider genetic interactions. Genotype matrix mapping (GMM) has been developed to identify genetic interactions. This study uses the GMM method to detect superior SNP combinations of the CCDC158 gene that influences average daily gain, marbling score, cold carcass weight and longissimus muscle dorsi area traits in Hanwoo. We evaluated the statistical significance of the major SNP combinations selected by implementing the permutation test of the F-measure. The effect of g.34425+102 A>T (AA), g.8778G>A (GG) and g.4102+36T>G (GT) SNP combinations produced higher performance of average daily gain, marbling score, cold carcass weight and the longissimus muscle dorsi area traits than the effect of a single SNP. GMM is a fast and reliable method for multiple SNP analysis with potential application in marker-assisted selection. GMM may prospectively be used for genetic assessment of quantitative traits after further development.

CAPS marker에 의한 Arabidopsis의 자외선 B 감수성 유전자 지도작성 (Mapping of UV-B sensitive gene in Arabidopsis by CAPS markers)

  • Arabidopsis thaliana columbia의 종자에 EMS를 처리하여 돌연변이체들을 만들었고 이중 UV-B에 감수성이 높은 돌연변이체를 골랐다. 이 UV-B 감수성 돌연변이체의 원인 유전자를 밝히기 위하여 교배 실험을 한 결과 이는 Mendel 유전법칙을 따르고 단일 유전자의 돌연변이에 의하여 나타나며 열성 유전을 하는 것으로 밝혀져 이 유전자를 uvs라 하였다. 염색체상의 uvs의 위치를 밝히기 위하여 CAPS maker를 이용한 연관분석을 하고자 하였고 이를 위하여 각각 maker의 primer 10종류를 제작하였다. 이를 이용, 각 PCR 산물에 대하여 uvs mutant와는 다른 제한효소 pattern를 갖는 Lansberg와 uvs mutant를 교배시켜서 얻은 것들로부터 DNA를 추출하여 PCR을 수행하였다. 이들과 자외선과의 감수성을 연관시켜 교차율을 계산한 결과 5번 염색체의 LFY3과 가장 가까웁게 연관되어 있었다.
