Higher Protein Digestibility of Chicken Thigh than Breast Muscle in an In Vitro Elderly Digestion Model

  • This study investigated the protein digestibility of chicken breast and thigh in an in vitro digestion model to determine the better protein sources for the elderly in terms of bioavailability. For this purpose, the biochemical traits of raw muscles and the structural properties of myofibrillar proteins were monitored. The thigh had higher pH, 10% trichloroacetic acid-soluble α-amino groups, and protein carbonyl content than the breast (p<0.05). In the proximate composition, the thigh had higher crude fat and lower crude protein content than the breast (p<0.05). Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of myofibrillar proteins showed noticeable differences in the band intensities of tropomyosin α-chain and myosin light chain-3 between the thigh and breast. The intrinsic tryptophan fluorescence intensity of myosin was lower in the thigh than in the breast (p<0.05). Moreover, circular dichroism spectroscopy of myosin revealed that the thigh had higher α-helical and lower β-sheet structures than the breast (p<0.05). The cooked muscles were then chopped and digested in the elderly digestion model. The thigh had more α-amino groups than the breast after both gastric and gastrointestinal digestion (p<0.05). SDS-PAGE analysis of the gastric digesta showed that more bands remained in the digesta of the breast than that of the thigh. The content of proteins less than 3 kDa in the gastrointestinal digesta was also higher in the thigh than in the breast (p<0.05). These results reveal that chicken thigh with higher in vitro protein digestibility is a more appropriate protein source for the elderly than chicken breast.

  • The aim of this study is to investigate effects of Prunus mume extract on gastric secretion in rats and experimentally carbon tetrachloride-induced liver damage of rabbits. The groups of Prunus mume extract showed excellent increasing effect in total acidity and peptic activity and decreasing effect on gastric volume than the control group whereas clear differences were not found between sample adminstered group and control group in gastric pH. In liver function test Prunus mume extract exhibited rapid recuperation of liver function that is sGPT activity showed an apparant decreasing effect from the 6th day total cholesterol and alkaline phosphatase level from the 10th day respectively compared to the control group in carbon tetrachloride-intoxicated rabbits. It is, is suggested that Pruns mume extract can be used as the stomachics to promote the gastric digestion by increasing the peptic activity and health food to shorten the recovery time from liver diseases.

익모초(益母草) 녹즙(綠汁)이 흰쥐의 위액분비기능(胃液分泌機能)에 미치는 영향(影響) (Effect of Leonurus sibiricus green juice on gastric sceretion of Rats)

  • These studies were carried out to investigate effects of Leonurus sibiricus green juice on gastric secretion of rats and acute toxicity in mice ; $LD_50(mg/kg)$ of Leonurus sibiricus extract was 625mg/kg intraperitoneally in mice. The groups administered green juice and water extract of Leonurus sibiricus showed more excellent increasing effect in total acidity and peptic activity, and decreasing effect on gastric volume than control group. But any differences were not found between sample and control group in gastric pH. It is suggested that Leonurus sibiricus can he used as the bitter stomachics to promote the gastric digestion.

양파 열수추출물의 항산화능 및 인공소화후의 항돌연변이 효과 (Antioxidant and Antimutagenic Activities of Hot Water Extract from White and Yellow Onions after Simulated Gastric Digestion)

  • 양파 열수 추출물의 항산화능 및 인공소화 전과후의 항돌연변이 효과를 BHT와 ascorbic acid와 비교하여 조사하였다. 총 페놀 및 플라보노이드의 함량은 노란색 양파 추출물이 횐색 양파 추출물보다 많았으며, 양파 추출물의 과산화수소 소거능은 농도가 증가함에 따라 비례적으로 증가하였다. $\beta-carotene-linoleate\;system$에서 항산화능과 reducing power는 증가되었으나, BHT와 ascorbic acid보다는 낮았다. 양파열수 추출물의 인공소화 후에 IQ와 MNNG로 처리된 Salmonella typhimurium TA80과 TA100에 대하여 항돌연변이 효과가 나타났으며, 노란색 양파가 횐색 양파보다 높게 나타났다. 결론적으로 양파 추출물을 소화시키면 항산화 및 항돌연변이의 효과는 양파의 페놀과 플라보노이드의 성분 변화에 의한 것으로 판단된다.

Effect of Lactic Fermentation and Spray Drying Process on Bioactive Compounds from Ngoc Linh Ginseng Callus and Lactobacillus plantarum Viability

  • Ngoc Linh ginseng is one of the most valuable endemic medicinal herbs in Vietnam. In this study, Ngoc Linh ginseng callus was fermented by Lactobacillus plantarum ATCC 8014 (at 6, 7, and 8 log CFU/ml) to evaluate the extraction efficiency of bioactive compounds. The post-fermentation solution was spray-dried using maltodextrin with or without Stevia rebaudiana (3% and 6% v/v) as the wall material. Bioactive compounds such as polyphenols, polysaccharides, and total saponins, and L. plantarum viability during fermentation and after spray-drying, as well as under simulated gastric digestion, were evaluated in this study. The results showed that probiotic density had a significant effect on bioactive compounds, and L. plantarum at 8 log CFU/ml showed the best results with a short fermentation time compared to other tests. The total content of polyphenols, polysaccharides, and saponins reached 5.16 ± 0.18 mg GAE/g sample, 277.2 ± 6.12 mg Glu/g sample, and 4.17 ± 0.15 mg/g sample, respectively after 20 h of fermentation at the initial density of L. plantarum (8 log CFU/ml). Although there was no difference in the particle structure of the preparation, the microencapsulation efficiency of the bioactive compound in the samples containing S. rebaudiana was higher than that with only maltodextrin. The study also indicated that adding S. rebaudiana improved the viability of L. plantarum in gastric digestion. These results showed that S. rebaudiana, a component stimulating probiotic growth, combined with maltodextrin as a co-prebiotic, improved the survival rate of L. plantarum in simulated gastric digestion.

멸치젓갈의 인공소화시 N - Nitrosamine 생성과 돌연변이의 상관성 (Correlation of N - Nitrosamine Formation and Mutagenicity in Fermented Anchovy under Simulated Gastric Digestion)

  • 우리나라 전통 발효식품인 젓갈류의 생체내 소화과정에서 니트로사민 생성과 돌연변이원성의 가능성을 예측하기 위하여 실온(18~20$^{\circ}C$)에서 60일간 숙성시킨 멸치 젓갈을 인공적인 소화타액과 위액에 nitrate, thiocyanate, formaldehyde 및 ascorbic acid를 8 mM 이하의 각 농도별로 첨가하여 Nnitrosodimethylamine(NDMA) 생성과 돌연변이원성의 상관성을 조사하였다. 인공소화시 NDMA의 생성은 nitrite,thiocyanate 및 formaldehyde의 첨가농도에 따라서 촉진되었으나, ascorbic acid는 억제제로서 그 첨가농도에 비례하여 생성저해 효과가 나타났으며, 대체로 4 mM 농도에서 무첨가구에 비하여 71.3% 정도가 저해되었다. 돌연변이원성은 숙성시킨 젓갈에 nitrite, thiocyanate 및 formaldehyde를 여러 농도로 첨가하여 인공소화시킬 때에는 증가되었으나, ascorbic acid를 첨가하였을 때에는 현저하게 감소하였다.

인공위액조건과 예열처리에 따른 GM 콩 도입단백질(CP4EPSPS)의 소화성 평가 (In vitro Digestibility Assessment of CP4EPSPS in GM Soybean under Different Conditions of Simulated Gastric Fluid and Preheating)

  • 본 실험은 제초제 내성을 가지는 CP4EPSPS를 도입시킨 유전자재조합 콩을 이용하여 인공위액의 비율에 따른 도입 단백질의 소화특성을 평가하고, 열처리가 유전자재조합 콩의 도입단백질의 소화성에 미치는 영향을 살펴보았다. 유전자재조합 콩과 펩신의 비율에 따른 도입단백질 CP4EPSPS의 소화성은 인공위액 즉 펩신의 양에 따라 차이를 나타내 유전자재조합 식품의 도입단백질에 대한 소화성 평가에서 펩신의 양이 중요한 요인이 됨을 알 수 있었다. 또한 유전자 재조합 콩에 대한 예열처리가 도입단백질 CP4EPSPS의 펩신에 대한 내성을 많이 감소시켜 소화성을 높이는 것으로 나타났다. 따라서 앞으로 유전자재조합식품의 안전성을 평가하는 in vitro 소화평가 시험에서 생리학적 측면에 근거한 인공위액의 비율을 반영함으로써 보다 정확한 도입단백질의 소화성에 대한 평가가 필요하리라 사료되며, 또한 콩의 주요 섭취방법인 열처리에 따른 도입단백질의 소화특성을 평가함으로써 유전자재조합 콩의 안전성에 대한 신뢰도 제고를 위한 기초자료가 될 것으로 기대된다.

Impact of Sodium Copper Chlorophyllin on Mercury Absorption Using an in Vitro Digestion with Human Intestinal Cell Model

  • The effects of sodium copper chlorophyllin (SCC) on bioaccessibility and uptake of mercury from fish were investigated using an in vitro digestion coupled with a Caco-2 cell. Fish along with SCC was subjected to a simulated in vitro digestion, which simulates both the gastric and small intestinal phase in vivo. Mercury bioaccessibility, the amount of mercury released from fish to aqueous phase following a digestion, was measured. Various amounts of SCC (0.1-25 mg) significantly reduced mercury bioaccessibility in a dose dependent manner by 49-89% compared to the negative control (fish without SCC) (p<0.05). Mercury bioaccessibility in varying molar ratios of mercury to positive control, 2,3-dimercapto-1-propane sulfonate (DMPS) was between 24 and 52%. Mercury uptake by Caco-2 cells from test media containing aqueous phase following in vitro digestion was measured after 6 hr incubation at $37^{\circ}C$. Cellular mercury uptake with increasing amount of SCC ranged from 0.352 to $0.052\;{\mu}g$ mercury/mg protein, while those in DMPS treatment were between 0.14 and $0.27\;{\mu}g$ mercury/mg protein. Our study suggests that SCC can reduce mercury absorption following fish consumption and may be efficient as a synthetic chelating agent for long term chronic mercury exposure in fish eating populations.

Evaluation of the Digestibility of Korean Hanwoo Beef Cuts Using the in vitro Physicochemical Upper Gastrointestinal System

  • The aim of this study was to investigate the digestibility of different Korean Hanwoo beef cuts using an in vitro digestion model, in vitro physicochemical upper gastrointestinal system (IPUGS). The four most commonly consumed cuts - tenderloin, sirloin, brisket and flank, and bottom round - were chosen for this study. Beef samples (75 g) were cooked and ingested into IPUGS, which was composed of mouth, esophagus, and stomach, thereby simulating the digestion conditions of humans. Digested samples were collected every 15 min for 4 h of simulation and their pH monitored. Samples were visualized under a scanning electron microscope (SEM) to examine changes in the smoothness of the surface after digestion. Analysis of the amino acid composition and molecular weight (MW) of peptides was performed using reverse-phase high performance liquid chromatography and sodium dodecyl sulfate polyacrylamide gel electrophoresis, respectively. Following proteolysis by the gastric pepsin, beef proteins were digested into peptides. The amount of peptides with higher MW decreased over the course of digestion. SEM results revealed that the surface of the digested samples became visibly smoother. Total indispensable and dispensable amino acids were the highest for the bottom round cut prior to digestion simulation. However, the total amount of indispensable amino acids were maximum for the tenderloin cut after digestion. These results may provide guidelines for the elderly population to choose easily digestible meat cuts and products to improve their nutritional and health status.

베타-카로텐 탑재 하이드로 젤 농도와 저작에 따른 지방소화율과 생체접근율의 변화 (Influence of Starch Concentration and Mastication on the Lipid Digestion and Bioaccessibility of β-carotene loaded in Filled Hydrogels)

  • Purpose: This study was conducted to examine the effects of the starch concentration of filled hydrogel and the addition of ${\alpha}-amylase$ and simulated mastication processing in an oral phase on lipid digestion and ${\beta}-carotene$ bioaccessibility of filled hydrogels. Methods: Lipid digestion and ${\beta}-carotene$ bioaccessibility of the filled hydrogels were measured after the samples were passed through an in vitro gastrointestinal tract model consisting of oral, gastric, and small intestinal phases. Results: The initial rate and final extent of lipid digestion were higher in the filled hydrogels than in the emulsion when the filled hydrogels were treated in an oral phase without simulated mastication processing and addition of ${\alpha}-amylase$, regardless of starch concentration. However, when the filled hydrogels were minced using mortar and pestle for 2 min and were exposed to ${\alpha}-amylase$, the filled hydrogel fabricated with 5% starch showed the lowest lipid digestion rate and extent compared to the emulsion and other filled hydrogels. Bioaccessibility of ${\beta}-carotene$ was higher in the filled hydrogels than in the emulsion, regardless of the digestion method performed in an oral phase and starch concentration. However, there were appreciable differences in bioaccessibility of the filled hydrogels depending on whether or not simulated mastication and addition of ${\alpha}-amylase$ were employed. Conclusion: These results suggested that the rheological properties of initial filled hydrogels and simulated mastication processing in an oral phase plays an important role in determining the lipid digestion and ${\beta}-carotene$ bioacccessibility entrapped within filled hydrogels.
