Case-Control Study of Diet in Patients with Cervical Cancer or Precancerosis in Wufeng, a High Incidence Region in China

  • Purpose: To investigate the diet of patients with cervical cancer and precancerosis in the Wufeng area, a high-incidence region in China. Methods: In the case group, 104 patients diagnosed with cervical cancer or cervical intraepithelial neoplasias (CINII/III) were recruited from the Wufeng area. Nine hundred thirty-six healthy women were selected from the same area as the matched controls. A questionnaire, which included questions about general lifestyle conditions, smoking and alcohol status, source of drinking water, green tea intake, and diet in the past year, was presented to all participants. Results: Green tea intake (P=0.022, OR=0.551, 95% CI=0.330-0.919) and vegetable intake (P=0.035, OR=0.896, 95% CI=0.809-0.993) were identified as protective factors against cervical cancer or CINII/III. There was no indication of any associations of other lifestyle factors (smoking status, alcohol status, source of drinking water) or diet (intake of fruit, meat/egg/milk, soybean food, onion/garlic, staple food and pickled food) with cervical cancer. Conclusions: The results suggest that eating more fresh vegetables and drinking more green tea may help to reduce the risk of cervical cancer or CINII/III in people of the Wufeng area.

  • Functional food for prevention of chronic diseases is one of this century's key global challenges. Cancer is not only the first or second leading cause of death in China and other countries across the world, but also has diet as one of the most important modifiable risk factors. Major dietary factors now known to promote cancer development are polished grain foods and low intake of fresh vegetables, with general importance for an unhealthy lifestyle and obesity. The strategies of cancer prevention in human being are increased consumption of functional foods like whole grains (brown rice, barley, and buckwheat) and by-products, as well some vegetables (bitter melon, garlic, onions, broccoli, and cabbage) and mushrooms (boletes and Tricholoma matsutake). In addition some beverages (green tea and coffee) may be protective. Southwest China (especially Yunnan Province) is a geographical area where functional crop production is closely related to the origins of human evolution with implications for anticancer influence.

The protective effect of Halal food extract in pancreatic beta cell lines.

  • In Islamic dietary guidelines, Halal foods are allowed as edible blessed food. Most foods were categorized within halal for Muslims. The main point of Halal food is that foods are clean in every process and based on Halal standard which might be different in each country. Most pancreatic ${\beta}$ cells synthetize, store, and release insulin. Specific molecular, functional as well as ultrastructural traits of pancreatic ${\beta}$ cells could control their insulin secretion properties and survival phentoype. Insulin-secreting pancreatic ${\beta}$-cells are essential regulators of mammalian metabolism. In addition, the pancreatic ${\beta}$ cell plays an important role in the pathogenesis of type 1 and type 2 diabetes as improving glucose homeostasis by preserving, expanding and improving the function of this key cell type. However, the pharmacological effect of halal food has not been unclear yet, especially food habit-dependent diabetes. The aim of the this study was to determine the preventive effect of Iran plants extract (Almond, Garlic, Cumin, Ginkgo biloba, Holy basil, Psyllium, Satureja khuzistanica, Fenugreek, Green tea, Ipomoea betatas, Blueberry) on RINm5F cells and MIN6 cells as pancreatic ${\beta}$ cell line. The cytotoxicity of the extracts of Iran plants on RINm5F cells and MIN6 cells were measured by using MTT assays. The preventive effects of Iran plant extracts were measured by WST-8 cell proliferation assay on streptozotocin (STZ)-induced cell death in MIN6 cells. In presented result showed that all extract of Iran plants (0.01-10mg/ml) did not show cytotoxicity in RINm5F cells and MIN6 cells. Among non-cytotoxic extract, the protective effects could be detect in high dose concentration. These results suggest that the extract of Iran plants may serve as a potential therapy for diabetes.

Production of Mono-Hydroxylated Derivatives of Terpinen-4-ol by Bacterial CYP102A1 Enzymes

  • CYP102A1 from Bacillus megaterium is an important enzyme in biotechnology, because engineered CYP102A1 enzymes can react with diverse substrates and produce human cytochrome P450-like metabolites. Therefore, CYP102A1 can be applied to drug metabolite production. Terpinen-4-ol is a cyclic monoterpene and the primary component of essential tea tree oil. Terpinen-4-ol was known for therapeutic effects, including antibacterial, antifungal, antiviral, and anti-inflammatory. Because terpenes are natural compounds, examining novel terpenes and investigating the therapeutic effects of terpenes represent responses to social demands for eco-friendly compounds. In this study, we investigated the catalytic activity of engineered CYP102A1 on terpinen-4-ol. Among CYP102A1 mutants tested here, the R47L/F81I/F87V/E143G/L188Q/N213S/E267V mutant showed the highest activity to terpinen-4-ol. Two major metabolites of terpinen-4-ol were generated by engineered CYP102A1. Characterization of major metabolites was confirmed by liquid chromatography-mass spectrometry (LC-MS), gas chromatography-MS, and nuclear magnetic resonance spectroscopy (NMR). Based on the LC-MS results, the difference in mass-to-charge ratio of an ion (m/z) between terpinen-4-ol and its major metabolites was 16. One major metabolite was defined as 1,4-dihydroxyp-menth-2-ene by NMR. Given these results, we speculate that another major metabolite is also a mono-hydroxylated product. Taken together, we suggest that CYP102A1 can be applied to make novel terpene derivatives.

시판되는 어 .패류에 대한 Listeria 속균의 조사연구 (A Study on Listeria Strain Species for Fishes and Shellfishes on Sale)

  • 본 연구에서는 이 지역에서 유통되고 있는 해수 및 어패류를 직접 채취하여 Listeria spp.를 분리 하였으며, 각각의 검체에 대한 균종별 분리율, 생화학적시험과 표준균주를 사용한 pH에 대한 시간별 균의 생육관계를 조사하였으며, L. monocytogenes에 대한 마늘, 향신료, 녹차 등의 농도에 따른 시판용 TSB(0.6%ye, pH 7.3)에서의 시간별 균의 생육관계를 조사하였고, 또한 항균제 10종에 대하여 감수성 시험을 하여 다음과 같은 결과를 얻었다. 1. 총 검 체 200건에서 Listeria spp.가 검출된 것은 32건으로 분리율 16%이었고, 검체별로는 전어(japanese gizzard shad)에서 37.5%(6/16)로 가장 높았으며, 병어(pomfret)와 새우(metapenaeus shell) 에서 각각 31.3%(5/16). 맛(sinonoracula constricta)과 조기 (yellow tailrunner)에서 각각 18.8% (3/16), 해수(sea water)에서 14.3%(8/56), 바지락(little neck clams)에서 12.5%(2/16)이었으며, 홍합(hard- shelled mussels) 소라(top shell), 꼬막(cockle clam)에서는 검출되지 않았고, 균종별로는 L. innocua가 14.0%(28/200), L. monocytogenes와 L. seeligeri는 각 각 1.0%(2/200)가 분리 되었다. 2. L. monocytogenes를 비롯한 4종의 표준균주 모두는 pH 3.0에서 균의 생육이 정지되었고, pH 10.0에서는 미약하지만 균의 생육이 진행되었으며, 산성보다는 알칼리성에서 균의 생육이 활발함을 알 수 있었으며, pH 7.0∼8.0에서는 균의 생육이 가장 왕성하였고,4종의 표준균주 중 L. seeligeri가 최대의 0.D값인 0.627로 가장 높았다. 3. 마늘, 겨자, 와사비, 녹차 등의 농도에 따른 시판용 TSB(0.6%ye 포함, pH 7.3)에서 1. monocytogenes의 생육은 마늘추출물에서 0.D값이 0.078에서 0.210으로 가장 저해를 받았으며, 겨자와 와사비는 균의 생육에 큰 저해를 주지 못하였으나, 녹차 추출물은 미약하지만 18시간 이후에 균의 생육저해에 관여하고 있음을 알 수 있었다 4. 10종의 항균제 감수성시험에서 L . innocua 16주는 ampicillin, kanamycin, penicillin, trimethopriml/sulfamethoxale, vancomycin에 대하여 모두 100%의 감수성을 보였고, ciprofloxacin과 gentamicin은 각각 43.7%, 97.7%로 감수성 이 나타난 반면, tetracycline (5주, 31.3%), cefotaxime(12주, 75%)은 내성을 나타내었고, 특히 nalidixic acid는 100%모두 내성을 나타내었다.

Global ginseng research

  • 지금까지 Web of Science의 core collection에 나타난 인삼 관련 연구논문의 수를 분석하였다. 인삼논문이 처음 나타난 1905년부터 2019년까지 전세계에서 총 8,090편의 SCI(E) 논문이 출판된 것으로 나타났다. 그중 최근 24년, 즉, 1996년부터 2019년까지의 논문이 7,385편이었다. 1980년 18편에 불과했던 인삼 논문이 1990년에는 53편, 2000년에는 97편, 2010년에는 369편, 2019년에는 678편으로 비약적으로 증가하였다. Web of Science의 core collection에 수재된 전체 학술논문에서 인삼 논문이 차지하는 비중도 1970년 0.0008%, 1980년 0.0044%, 1990년 0.101%, 2000년 0.0141%에서 2019년에는 0.0422%로 비약적으로 증가하였다. 지난 24년간 출판된 인삼 연구논문 중 원보(노트 포함)는 7,099편, 리뷰는 286편이었다. 총 78개국 3,286개 기관에서 연구가 이루어졌으며, 1,274개 학술잡지에 논문이 수재되었다. 전체 논문 중 중국에서 연구된 논문이 40.3%로 가장 높았고, 대한민국에서 연구된 논문이 34.7%로 한국과 중국의 연구가 전체의 75%를 차지하였다. 그 다음은 미국(6.0%), 일본(4.1%), 캐나다(2.9%) 순이었다. 2013년까지는 한국에서 연구된 논문이 가장 많았으나 2014년부터 중국에서 연구된 논문의 수가 더 많아졌다. 지난 24년간 인삼은 전세계에서 가장 많이 연구된 약용식물로 인삼 다음으로는 차(6,499편), 마늘(3,641편), 은행(2,590편), 생강(1,945편) 순이었다.

Maillard 반응에 의한 미더덕 체액의 풍미개선 (Flavor Modification of Mideoduck (Styela clava) Drips by Maillard Reaction)

  • Maillard 반응의 기본이 되는 당과 아미노산을 결정하기 위하여 미더덕 체액, 6탄당인 glucose와 5탄당인 ribose를 기본으로 하여, thiamine, 아미노산으로는 methionine, taurine, glycine, alanine, threonine, cystine 등을 사용하였고, 실용적인 방안을 강구하기 위하여 식용하는 양파, 마늘, 감귤, 생강, 녹차, 파 등을 첨가하여 반응액을 만들었다. Ribose와 thiamine 구가 패널로부터 가장 높은 평가를 받았고, 이어서 glucose와 thiamine 구가, ribose와 methionine 구는 아주 나쁜 풍미를 형성하는 것으로 나타났다. 함황화합물인 taurine을 첨가한 glucose 및 ribose 구는 보통의 평가로 나타났고, 체액에 당과 아미노산을 첨가한 경우도 thiamine 구만 보통으로 평가되었으나 나머지 아미노산의 경우 싫어 하는 것으로 나타났다. 기본의 액즙-당 반응구에서는 장쇄지방산의 함량이 31.32-62.71%로 가장 높은 함량을 보였고, A, B, C구에서는 1,2-bezenedicarboxylic acid dibutyl ester의 함량도 20%를 상회하는 것으로 나타나 이들 화합물이 전체 향기성분의 50% 이상으로 이들이 체액-당 반응계의 주된 화합물로 생성되어, 체액을 활용한 조미료의 생산을 저렴한 비용으로 할 수 있는 것으로 나타났다.

항균성 식품을 이용한 간편섭취 농산물 미생물오염의 감소 및 분자생물학적 분석 (Bacterial Contamination Reduction of Minimally Processed Agricultural Products using Antibacterial Foods and Molecular Biological Analysis)

  • 가열하지 않은 간편섭취 농산물의 섭취를 통한 세균 감염의 가능성을 줄이고자, 가정에서 쉽게 구할 수 있고 잔류에 대한 염려가 없는 천연 항균성 식품을 이용하여 세척할 경우, 물 세척에 비해 세균의 감소효과가 있는지 확인하고자 하였다. 여러 가지 항균성 식품성분 중 마늘의 allicin, 생강의 gingerol, 녹차의 catechin, 계피의 cinnamic aldehyde, 고추냉이의 allyl isothiocyanate의 단계별 희석액으로 세척한 후, 단순 물 세척의 세척효과와 비교한 결과, 단순 물 세척에 비하여 세균 감소의 효과가 큰 것으로 통계적으로 확인되었다. 간편섭취 농산물에서 검출된 Bacillus cereus 오염의 유래 및 연관성을 확인하고자 유전적 상관성을 분석해 본 결과 유사도가 매우 높은 균주들이 많이 검출되었다. 이 사실을 통해 간편섭취 농산물의 생산, 가공, 포장, 유통 등의 과정에서 공통적인 요인에 의한 지속적인 오염의 가능성 및 소비자의 감염 가능성을 파악할 수 있었으며, 감염을 예방하기 위한 사전적이고 체계적인 관리가 필요함을 확인하였다.

HPLC를 이용한 지역 농산물의 비타민 B6 함량 분석 (Determination of Vitamin B6 Content using HPLC in Agricultural Products cultivated in Local Areas in Korea)

  • Contents of water soluble vitamin $B_6$ in a total of 62 agricultural products cultivated in local areas in Korea were analyzed by high performance liquid chromatography with fluorescence detector (HPLC/FLD). To verify the method of vitamin $B_6$ analysis, a quality control chart was formulated with in-house control using a mixture of broccoli and shiitake mushrooms. Among cereals, high content of vitamin $B_6$ measured $234.3{\sim}260.3{\mu}g/100g$ in dried mung bean and soybean. Vitamin $B_6$ content of non-glutinous and glutinous black rice measured $105.0{\mu}g/100g$ and $129.7{\mu}g/100g$, respectively. In vegetables, high content of vitamin $B_6$ were measured in passion fruit ($104.3{\mu}g/100g$), gat ($55.7{\sim}84.3{\mu}g/100g$), gomchwi ($31.3{\sim}88.0{\mu}g/100g$) and garlic ($72.7{\sim}98.3{\mu}g/100g$). Among fruits, gold kiwi 'Zespri' and green kiwi 'Hayward' revealed high vitamin $B_6$ content of $116.3{\mu}g/100g$ and $78.7{\mu}g/100g$, respectively. In persimmons, daebongsi had high vitamin $B_6$ content ($36.0{\sim}72.7{\mu}g/100g$) than bansi and sweet persimmon. Vitamin $B_6$ content in dried jujube and persimmon increased more than $86.7{\mu}g/100g$ compared to fresh materials. Among specialty crops, green tea powder ($64.7{\sim}251.0{\mu}g/100g$) and sansuyu ($172.3{\mu}g/100g$) revealed high content. Of mushrooms, vitamin $B_6$ content of Sparassis crispa ($139.3{\mu}g/100g$) was the highest. Vitamin $B_6$ content information of agricultural products in local areas in Korea collected from this experiment will be used as valuable preliminary data for grasp national nutritional status.

Does supplementing laying hen diets with a herb mixture mitigate the negative impacts of excessive inclusion of extruded flaxseed?

  • Objective: This study investigated the effects of extruded flaxseed with and without herbs mixture on egg performance, yolk fatty acids (FAs), lipid components, blood biochemistry, serological enzymes, antioxidants, and immune system of Hy-Line W-36 hens for nine weeks. Methods: Two hundred forty laying hens were randomly distributed to eight treatments, resulting in six replicates with five hens. Graded levels of dietary extruded flaxseed (0, 90, 180, and 270 g/kg) with and without herbs mixture (24 g/kg: garlic, ginger, green tea, and turmeric 6 g/kg each) were designed as treatments. Results: The two-way analysis of variance indicated that hens fed herbs mixture had a higher value of egg production, yolk high-density lipoprotein (HDL), superoxide dismutase, glutathione peroxidase, and white blood cell and lower contents of yolk cholesterol, glucose, and blood low-density lipoprotein than those fed diets without herb mixtures (p<0.05). The Flx27 (270 g/kg flaxseed) (153.5 g/kg n-3 FAs) and Flx27+H (270 g/kg flaxseed plus 24 g/kg herbs mixture) (150.5 g/kg n-3 FAs) groups were the most promising treatments in terms of yolk n-3 FAs content. In-teraction effect (herbs- flaxseed) for blood cholesterol, HDL, malondialdehyde, glutaredoxin, alanine transaminase, (ALT), aspartate transaminase (AST), haemoglobin and immune parameters was significant (p<0.05). The results showed layers fed herbs mixture (Flx9+H, Flx18+H, and Flx27+H) had a better value of total antibody, immunoglobulin M, immunoglobulin G, ALT, AST, and blood HDL as compared with representative flaxseed levels without herbs. Conclusion: High inclusion levels of extruded flaxseed (270 g/kg) without herbs to enrich eggs with n-3 appears to impair the antioxidant system, immunohematological parameters, and sero-logical enzymes. Interestingly, the herbs mixture supplementation corrected those effects. Therefore, feeding layers with flaxseed-rich diets (270 g/kg) and herbs mixture can be a promising strategy to enrich eggs with n-3 FAs.
