A Research on the origin and Daily Dose of Gardeniae Fructus in Shanghanlun (상한론(傷寒論)에서 치자(梔子)의 기원(基原)과 1일 복용량)

  • Objective : The purpose of this study is to determine the origin and daily dose of Gardeniae Fructus in Shanghanlun. Methods : Define the origin and daily dose of Gardeniae Fructus by searching Shanhanlun and Herbology Books(本草書). And Compared these with those of current Gardeniae Fructus. Results : According to Shanhanlun, the origin of Gardeniae Fructus is Gardenia jasminoides, but G. jasminoides f. longicarpa must excluded since originally it was used for dyeing. Among the 8 prescriptions containing Gardeniae Fructus, 15 Gardeniae Fructuses for the daliy dose in Chijabekpi-tang(Zhizibopi-tang) is assumed to be 14 Gardeniae Fructuses, as it is found 14 in many other prescriptions same in other prescriptions. The quantity of 14 Gardeniae Fructuses in Shananlun is 3 Ryang(兩), which is equal to 19.5 g in the current measurement. 1 Gardeniae Fructus is equal to 1.4 g. The length of the Gardeniae Fructus in the market which is produced in korea is 22-27 mm, and the diameter is 15-19 mm, whereas in China it is 20-25 mm, and the diameter is 13-18 mm. This meets the standard requirements of the Pharmacopoeia. Conclusions : As a result from above, the origin of Gardeniae Fructus is Gardenia jasminoides, but G. jasminoides f. longicarpa must be excluded. The recommended dose in a day is 14 Gardeniae Fructuses, which is equivalent to 19.5 g, the amount generally used in Shanhanlun.

The Effects of Gardeniae Fructus Aqua-Acupuncture on Liver Injury of Rats Induced by CCI4 (치자약침(梔子藥鍼)이 실험적(實驗的) 백서(白鼠)의 간손상(肝損傷)에 미치는 영향(影響))

  • This is the study of the effects of Aqua-acupuncture with Gardeniae Fructus on thc recovery of rat's liver which was damaged by 0.3ml/ea of $CCI_4$. Rats were divided into 4 groups; Normal-group(None treated group), Control-group(Not treated after $CCI_4$-intoxicated), Exp. I(Treated with Saline Aqua-acupuncture after $CCI_4$-intoxicated) and Exp. ll(Treated with Gardeniae Fructus Aqua-acupuncture after $CCI_4$-intoxicated). Biochemical assays for each serum enzyme activities of AST, ALT, Albumin, LDH, ${\gamma}$-GT, TG and Total cholesterol were performed. The results were summarized as follows: 1. AST activities in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. Albumin in serum increased in the Gardeniae Fructus Aqua- acupurkture treated group after $CCI_4$-intoxicated. 4. LDH in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after CClcintox icated, the Gardeniae Fructus Aqua acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. 5. ${\gamma}$-GT In serum significantly decreased In the Gardeniae Fructus Aqua-acupuncture trea ted group after $CCI_4$-intoxicated. In compan son with Saline-treated group after $CCI_4$-intoxicated, the Crardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated was not recognized significantly. 6. TG in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after $CCI_4$-intoxicated, the Gardeniae Fructus Aqua -acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. 7. Total cholesterol in serum decreased in the Gardenias Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after $CCI_4$-intoxicated, the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. The results from ahove show that the Gardeniae Fructus Aqua- acupuncture has highly effects on the damaged liver caused by $CCI_4$. Therefore it is expected that the Gardeniae Fructus Aqua- acupuncture could be used to cure the damaged liver.

Inhibitory Effects of Gardeniae Fructus on Apoptosis induced by 4-HNE in PC-12 Cell (치자(梔子)의 Apoptosis유발 억제효과)

  • Objectives : The purpose of this study was to observe the effects of Gardeniae Fructus on 4-HNE-induced apoptosis in PC-12 cell. Methods : A MTT assay was conducted to observe the cytotoxicity of Gardeniae Fructus on the PC-12 cell viability and the cytoprotective effects of Gardeniae Fructus on PC-12 cell against oxidative stress caused by 4-HNE. And western blot was conducted to observe the expression of Bax, Bcl-2, Caspase-3, $TNF-{\alpha}$ proteins which are involved in intrinsic and extrinsic apoptosis pathway. Results : 25, 50, 100, 200 and $400{\mu}g/m{\ell}$ of Gardeniae Fructus water extract had no cytotoxicity on PC-12 cell. $200{\mu}g/m{\ell}$ of Gardeniae Fructus water extract had significant cytoprotective effect on PC-12 cell against oxidative stress caused by 4-HNE. The expression of Bax protein in 50, 100 and $200{\mu}g/m{\ell}$ of Gardeniae Fructus was significantly decreased in PC-12 cell. The expression of Bcl-2 protein in $200{\mu}g/m{\ell}$ of Gardeniae Fructus was significantly increased in PC-12 cell. The expression of Caspase-3 protein in 100 and $200{\mu}g/m{\ell}$ of Gardeniae Fructus was significantly decreased in PC-12 cell. The expression of $TNF-{\alpha}$ protein in $50{\mu}g/m{\ell}$ of Gardeniae Fructus was significantly decreased in PC-12 cell. Conclusions : These results suggest that Gardeniae Fructus water extract is effective to protect PC-12 cell from 4-HNE induced apoptosis.

Inhibitory effect of Gardeniae Fructus extracts on oxidation and motility of keratinocytes (치자추출물이 산화 반응 및 피부각질형성 세포 운동성에 미치는 영향)

  • Objectives : Gardeniae Fructus extract is used as a component of various cosmetics. However, the effect of the extract on the motility of keratinocytes has not been studied. The aim of this study is to investigate the inhibitory effect of ethanol extract of Gardeniae Fructus (GFET) or ethyl acetate extract of Gardeniae Fructus (GFEA) on oxidation and motility of human keratinocyte HaCaT cells. Methods : Antioxidant activity of Gardeniae Fructus extracts were determined by the 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay. To investigate the cytotoxicity of Gardeniae Fructus extracts, 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed. The mRNA expression levels of tight junction related genes were analyzed using quantitative RT-PCR analysis. Cell migration assay was employed to determine the activity of Gardeniae Fructus extracts on motility of human keratinocyte HaCaT cells. Results : GFET and GFEA showed strong antioxidant activity. GFEA showed stronger cytotoxicity in HaCaT cells than GFET until $2.0mg/m{\ell}$ concentration. Cell migration assay demonstrated that GFET and GFEA decreased the motility of HaCaT cells. In addition, the mRNA expression level of claudin 8 among tight junction genes was significantly reduced by GFET or GFEA treatment. Conclusions : We investigated the physiological activities of the extracts of Gardeniae Fructus extracts on human keratinocytes by two different extraction methods. In addition, the mRNA expression level of claudin 8 among tight junction genes was significantly reduced by either GFET or GFEA treatment. This study provides basic information on the application of Gardeniae Fructus extract to cosmetics component.

  • In oriental medicine, Rhei Radix et Rhizoma & Gardeniae Fructus are frequently used materials for anti-inflammatory effect especially urinary tract disease and for neuropsychosis. Bang-Yak-Hap-Pyun(方藥合編) is widely used for clinical herbal prescription book. In Bang-Yak-Hap-Pyun(방약합편), Rhei Radix et Rhizoma is used 0.8g~16g. and Gardeniae Fructus is used 1.2g~6g. The most important fact is that the prescriptions used Rhei Radix et Rhizoma & Gardeniae Fructus at the same time are 4(Yang-Gyuk-San, Pal-Jung-San, Sa-Chung-Hwan, Suk-Gyul-Myung-San. The ratio of Rhei Radix et Rhizoma & Gardeniae Fructus are 2:1, 1:1, 1:1, 1:2 ). The number of prescription Rhei Radix et Rhizoma & Gardeniae Fructus in Bang-Yak-Hap-Pyun is total 45. 1 in sang-tong(上統), 5 in jung-tong(中統), 39 in ha-tong(下統). The group of mostly used disease is about urinary tract. The ratio of Rhei Radix et Rhizoma in Bang-Yak-Hap-Pyun is 3.33%~57.14%. And the ratio of Gardeniae Fructus is 3.47%~98%. The mostly quoted medical literature what prescription included Rhei Radix et Rhizoma & Gardeniae Fructus in Bang-Yak-Hap-Pyun are Dong-Yi-Bo-Gam and Man-byung-hoe-chun.

Quality Monitoring of Specification Standard of Gardeniae Fructus in the Korean Pharmacopoeia and Studies HPLC Standard Chromatogram (치자(梔子)의 규격 기준 모니터링 및 HPLC 표준크로마토그램 연구)

  • Objectives : Gardeniae Fructus is a ripe fruit of Gardenia jasminoides Ellis, which has been used as traditional medicines for anti-inflammatory, diuretic, antipyretic, and antibacterial activity. The aim of this study was to compare of Gardeniae Fructus in South Korea collected during three years according to the standards in monographs of the Korean Pharmacopoeia Eleventh edition (KP11). Methods : 30 items of Gardeniae Fructus from two cultivation regions were classified into dried(n=15) & steamed (n=15) and tested according to the standards in monographs of the KP11. Gardeniae Fructus was carried out identification(comparison of colors, thin layer chromatography), heavy metals, residual pesticides, total ash, and assay registered at KP11. Add to we tested loss on dry, contents of ethanol-soluble extracts, and HPLC profiling. Results : In TLC chromatogram of identification test, the spot of gardenoside and geniposide were observed at $R_f$ value of about 0.3 and 0.5. Heavy metals and residual pesticides met the requirements of the standards for all samples. The results of total ash of each samples are measured maximum 4.87 %. According to HPLC for assay, the samples contain 4.80~6.10 % of geniposide and 0.45~1.83 % of gardenoside. Conclusion : We have verified the current specification standard of Gardeniae Fructus and standard that is not set. By the results, it is proposed a new draft of loss on drying and confirmed the content of gardenoside revised. HPLC standard chromatogram of Gardeniae Fructus is proposed. We hope that it will help the standardization of Gardeniae Fructus.

Standardization of Geniposide Content of Gardeniae Fructus (치자의 Geniposide 함량 기준 설정)

  • Gardeniae Fructus (Gardenia jasmmoides Ellis) is widely used as a natural food colorant and as a traditional medicine for the treatment of hepatic and inflammatory diseases. The quality evaluation of Gardeniae Fructus was carried out by HPLC using geniposide as a marker. The geniposide was isolated from commercial Gardeniae Fructus on ODS column using a mixed solvent of acetonitrile-water (v/v 9:91) as mobile phase and a detection wavelength 240 mm. The content of geniposides in twenty different samples was in a range of 1.20% to 7.17% of total tissues and the average was 4.97%.

A Study on the prescriptions including Gardenia fructus in Dongeuibogam (동의보감(東醫寶鑑) 중 치자(梔子)가 배오(配伍)된 방제(方劑)의 활용(活用)에 대한 고찰(考察))

  • Objectives : This report describes 241 prescriptions related to the use of Gardenia Fructus blended prescriptions from Dongeuibogam. Methods : The following conclusions were reached through investigations on the prescriptions that use Gardeniae Fructus. Results : The Gardeniae Fructus is used in 49 fields of treatment such as urinary diseases, jaundice and furuncle, etc. Prescriptions that utilize Gardeniae Fructus are used in the treatment of fever, eye disease, hemorrhage, skin disease, ear disease, internal disease and so on. In the view of the causative agent of a disease, the prescriptions which are compounded with Gardeniae Fructus are related to endogenous agents such as jaundice, seven emotion, alcohol, exogenous agents such as cold, wind, heat and non-endo-exopathogenic factors like disease due to external factor, injury. The dosage of Gardeniae Fructus is 1pun(about 0.375) to 5don 5pun(about 21g), however 5pun(about 1.87g)~1don(about 3.75g) has been taken the most for clinical application. Conclusions : Hwangryunhaedok-tang is the most base prescriptions which use the Gardenia Fructus. Gardeniae Fructus is mainly used as a raw material.

Skin Permeation and Crosslinking with Biological Tissue of the Hydrolyzed Products of Gardeniae Fructus Extract (치자엑스 가수분해물 제제의 피부투과 및 생체 피부조직과의 교차결합에 관한 연구)

  • Gardeniae Fructus is consisted of geniposide and it's derivatives. For the purpose of treatment of skin disease, geniposide and hydrolyzed products (HP) of Gardeniae Fructus were studied on skin permeation and cross1inking with biological tissue. The hydrolyzed products (HP) and active ingredients of Gardeniae Fructus were identified and investigated about skin permeability. Genipin has provided low cytotoxic cross1inking reagents and formed stable and biocompatible crosslinked products. The permeation enhancing effects of geniposide and genipin under the hydrolyzed products of cream and hydrogel preparations were tested using Franz type diffusion cell and the skin of hairless mouse. The remaining proportions of geniposide and genipin were measured in the hydrolyzed products of cream and hydrogel preparations. The crosslinking of epidermic and endodermic tissue with genipin under the hydrolyzed prodcuts of cream and hydrogel preparation was observed using light microscopy. Increased absorption ratio of the skin of hairless mouse about genipin was higher than that of geniposide. Loads at break, tensile strengths and skin permeation rate of the hydrolyzed products (HP) of cream and hydrogel preparations were higher than the nonhydrolyzed products (NHP). The hydrolyzed products (HP) of cream and hydrogel of Gardeniae Fructus Extracts were proper preparations and crosslinking agents to increase the transdermal absorption with epidermic and endodermic tissue.

Anti-inflammatory Effect of the Processed Gardeniae Fructus in LPS-induced Macrophages (LPS로 유도된 대식세포에서 수치 치자의 항염효과)

  • Objective : Herbal processing is one of the traditional techniques used in Korean medicine to increase the effectiveness of herbs or reduce their toxicity. In this study, Gardeniae Fructus processed with ginger juice and alcohol was prepared to evaluate the anti-inflammatory effect on lipopolysaccharide (LPS)-induced macrophages. Methods : The processing of Gardeniae Fructus was performed by adding 40 % ginger juice or 10% alcohol to the total weight of Gardeniae Fructus and then roasting at 150℃ for 5 minutes. Cell viability was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. To detect nitric oxide (NO) production, culture media were mixed with Griess reagent and measured the absorbance at 540 nm. Prostaglandin E2 (PGE2) and pro-inflammatory cytokines were detected by enzyme-linked immunosorbent assay (ELISA). Western blot was applied to monitor protein expression levels. Results : LPS-induced NO, PGE2 and inflammatory cytokines were decreased by the treatment of normal or processed Gardeniae Fructus ethanol extracts (GFE). Compared to normal GFE, the processed GFE showed a stronger inhibitory effect on the production of NO and PGE2. These inhibitory effect of GFE was due to the suppression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) mediated from the inhibition of nuclear factor kappa B (NF-κB). Furthermore, processed GFE showed more suppressive effects on the expression of iNOS, COX-2 and IκBα proteins than normal GFE. Conclusion : From these results, it was concluded that GFE had an improved anti-inflammatory effect compared to normal GFE. These results provide an objective evidences for the use of herbal processing in Korean medicine.
