Nitric Oxide Production Ability and its Formation Mechanisms in Macrophage TIB 71 Cell Line by Polysaccharide Extracted from Ganoderma lucidum (영지버섯 다당체의 Nitric Oxide 생성능 및 생성기전 연구)

  • This study was carried out to get infomation on the nitric oxide production ability and its formation mechanisms of polysaccharides extracted from Ganoderma lucidum(PSG) by using murine macrophage cell line. The cultured mycelial cells of Ganoderma lucidum were extracted by alkali, and than neutralized by acid. The extract were passed through the column of DEAE cellulose for more purification. The neutral fraction was concentrated and precipitated with 95% ethanol. The precipitate was lyophilized and PSG was obtained. The immunomodulating effects of PSG on macrophage were performed by using murine macrophage cell line ATCC TIB 71 cells with PSG 0.5mg. PSG alone could not induce the production of nitrite, but it had a significant potential effect on nitrite secretion when the cells were primed and triggered with BCG and Interferon(IFN)-${\gamma}$. Also it was prominent by using calcium channel blocker(verapamil) and adenylate cyclase activator(forskolin).

Effect of Ganoderma lucidum Mycelial Fractions on the Functional Recovery of Primary Cultured Hepatocytes. (영지 균사체 분획이 일차배양 간세포 기능회복에 미치는 영향)

  • The cultured mycelia of Ganoderma lucidum were extracted and the extract was separated into six fractions by organic solvent fractionation. The antihepatotoxic activity of all the fractions was evaluated by measuring activities of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT). Among the fractions tested, the high-polarity fractions such as aqueous and n-butanol fractions significantly reduced activities of GPT and GOT in CCl4- and galactosamine-intoxicated rat primary hepatocytes. When intracellular synthetic activities were measured by pulsing the rate primary cultured hepatocytes with [3H]-uridine and [3H]-leucine, activities of DNA, RNA and protein. When direct toxicities of the fractions were measured against human hepatoma(SK-Hep-1), the non-polarity fractions such as n-butanol and ethyl acetate fractions showed potent direct cytotoxicities even at the concentration of 1 $\mu\textrm{g}$/ml. These data showed that Ganoderma lucidum has hepatoprotective and hepatotoxic recovery principles in its mycelia.

Neuroprotective Effects of Ethanol Extract of Ganoderma lucidum L. on murine hippocampal cells (영지 에탄올 추출물의 마우스 유래 뇌 해마세포 보호효과)

  • Ganoderma lucidum L. (GL) is a traditional oriental medicine that has been widely used as anti-inflammatory, antitumor and anti-oxidant in Korea and other Asian countries. In this study, we investigated the ethanol extract of GL has neuroprotective effects in murine hippocampal HT22 cells. GL ethanol extract has the potent neuroprotective effects on glutamate-intoxicated cells by inducing the expression of heme oxygenase (HO)-1 in HT22 cells. GL ethanol extract increased JNK phosphorylation. Obviously, When we treated the GL extract with c-Jun N-terminal kinase (JNK) inhibitor (SP600125), HO-1 expression was reduced. Moreover, we found that GL treatment caused the nuclear accumulation of Nrf2. In conclusion, the ethanol extract of GL significantly protects glutamate-induced oxidative damage by induction of HO-1 via Nrf2, JNK pathway in mouse hippocampal HT22. These results suggest that GL ethanol extract would be a good source for taking active compounds and may be a potential pharmaceutical products for brain disorder induced by neuronal damage and oxidative stress.

Analysis of Antioxidant Activity and Cytotoxicity against Human Cancer Cell Lines of Extract from Atractylodes rhizoma fermented with Ganoderma lucidum Mycelium (창출과 영지버섯 균사체 발효 추출물의 항산화 활성 및 인체 암세포주에 대한 세포독성 분석)

  • Ganoderma lucidum has been traditionally used as a medicine for treatment of bronchitis, arthritis, and high blood pressure, and it has been reported to display many biological activities including anticancer and immune activities. Since mushroom mycelium is known to have excellent biological activities together with mushroom fruiting body, studies on biological activities of mushroom mycelium have been actively conducted. Thus, the present study compared the biological activities before and after the cultivation of Ganoderma lucidum mycelium on Atractylodes rhizoma. When the radical scavenging activity was assessed by the DPPH assay, ARGL (ethanol extract of Atractylodes rhizoma mycelium fermented with Ganoderma lucidum) showed radical scavenging activity of 5.58~82.56% at concentrations of $10{\sim}500{\mu}g/assay$, while AR (ethanol extract of Atractylodes rhizoma) showed radical scavenging activity of 5.27~72.08% at the same concentrations. When measured by using the ABTS assay, ARGL showed higher radical scavenging activity than AR, which was consistent with the result obtained by the DPPH assay. In the MTT assay, the cytotoxicity of ARGL against all cell lines was higher than that of AR. In particular, the cytotoxicities of AR and ARGL against Hep3B at a concentration of $400{\mu}g/assay$ were 71.81% and 86.40%, respectively. In addition, the result obtained by the SRB assay was consistent with the result obtained by the MTT assay. According to the results mentioned above, there is a high probability that medicinal herb cultures using mycelium can be used as sources of functional foods since the cytotoxicities against cancer cells and antioxidant activities increased when the mycelium was fermented with Atractylodes rhizoma.

The Effects of Ganoderma lucidum Extract on Osteoblast in Rat Fetus Calvarial Cells (영지(靈芝) 추출물이 Rat fetus 두개골로부터 분리한 조골세포에 미치는 영향)

  • Objectives: In this study, the author aimed to evaluate the effect of EtOH extract of Ganoderma lucidum (GLE) on osteoblast proliferation in rat fetus calvarial cells. Methods: The osteoblast separated from rat fetus calvariae was cultivated for 6~21 days and evaluated the cell function. After the addition of GLE on the culture medium, we determined the effect of GLE on the cell viability, cell proliferation, bone matrix protein synthesis, alkaline phosphatase (ALP) activity, collagen synthesis and calcified nodule formation of the cultivated osteoblast. Results: GLE did not change the survival rate of rat calvarial osteoblast. GLE increased the proliferation of rat calvarial osteoblast. GLE increased ALP activity of rat calvarial osteoblast. GLE increased bone matrix protein synthesis of rat calvarial osteoblast. GLE increased collagen synthesis of rat calvarial osteoblast. GLE slightly affected calcified nodule formation of rat calvarial osteoblast. Conclusions: This study suggests that Ganoderma lucidum might improve the osteoporosis resulted from augmentation of osteoblast proliferation.

Studies on Inorganic Composition and Immunopotentiating Activity of Ganoderma lucidum in Korea (한국산(韓國産) 영지(靈芝)의 무기(無機) 성분(成分) 및 면역(免疫) 증강(增强) 작용(作用)에 관한 연구(硏究))

  • To determine contents of inorganic elements of Ganoderma lucidum, the horn-shaped carpophores and the pileus of Ganoderma lucidum were incinerated and analyzed by inductively coupled plasma atomic emission spectrophotometry. The ash contents of the pileus and the horn-shaped carpophore were 1.48% and 1.40%, respectively. The pileus contained calcium, magnesium, sodium, manganese, iron, zinc and germanium in that order. The horn-shaped carpophore contained magnesium, calcium, zinc, manganese, iron, copper and germanium in that order. To examine the protein-bound polysaccharide from Ganoderma lucidum for immunopotentiating activity, its fruit bodies were extracted with hot water. Purification of the extract was carried out by acetone precipitation and dialysis. The fraction obtained during the purification procedure consisted of a polysaccharide moiety (51%) and a protein moiety (5%). When the compound was administered intraperitoneally to the mice at a dose of 50mg/kg, it enhanced the accumulation of the peritoneal exudate cells, macrophage and polymorphonuclear leucocytes, thereby indicating immunopotentiation.

Effect of Ganoderma lucidum Extract on Experimentally Induced Hepatic Damage and Hyperlipemic Rats. (영지(靈芝)엑기스가 백서(白鼠)의 실험적(實驗的) 간장중독(肝臟中毒) 및 고지혈증(高脂血症)에 미치는 영향(影響))

  • This study was attempted to investigate the effect of Ganoderma lucidum (Young-Jii) extract on the activities of GPT GOT Al. P LDH and the level of total bilirubin and total cholesterol in serum of $CCl_4$-intoxicated rats, and on the level of total lipids triglyceride phospholipids and total cholesterol in the serum of experimentally induced hyperlipemic rats, and on the effect of body and liver weight in rats. The results were shown as follows; In $CCl_4$-intoxicated rats, the extract showed a significant decrease in the activities of GPT and Al.P, a slight decrease in the activity of GOT and LDH; The level of total bilirubin was slightly affected, but significantly decreased at a dose as high as 500 mg/kg; the level of total cholesterol was increased dose dependently. In hyperlipemic rats, the extract caused a significant decrease in the level of total lipids and triglyceride and the rate of decrease was more pronounced with repeated treatments for 10 days; the level of phospholipids and total cholesterol were slightly decreased with repeated treatment of the extract at a dose of 300 mg/kg for 10 days; A significant body weight gain was shown with the treatment of the extract.

Comparison of Pretreatment Methods for Extraction of Selected Components from Ganoderma lucidum (전처리 방법에 따른 영지버섯 추출액의 품질특성 변화)

  • This study was conducted to investigate the effects of pretreatment and extraction methods on the water extraction yields of sugar, protein and phenolic compounds, and the antioxidative activity of extract from Ganoderma lucidum. The herb was ground, sifted, and treated for 10 min with microwave (2,450 MHz) and ultrasound (20 kHz), and then extracted by boiling underatmospheric pressure and pressured boiling. Particle size $(0.8\~2.4\;mm)$, extraction time (2 hr) and solvent/solid ratio (20:1) were selected as optimal conditions for extraction.. Microwave and ultrasound treatment increased the extraction yields of total sugar, protein and phenolic compounds and the radical scavenging activity of extract. In comparison with microwave treatment and ultrasound treatment, microwave treatment was more effective than that of ultrasound treatment. The yields of all three compounds and the radical scavenging activity of extract were higher in pressured boiling extraction than in boiling underatmospheric pressure. The results showed that microwave radiation and sonication prior to extraction, and pressured extraction could be utilized for improving the extraction efficiency of G. lucidum.

The Protective Effects of Ganoderma lucidum on the DNA Damage and Mutagenesis (DNA손상 및 돌연변이에 대한 명지버섯의 방어효능)

  • Ganoderma lucidum is commonly known as medically potent mushroom, which has been widely used in China and other oriental countries for the treatment of various diseases, including cancer. In this report, we investigated the anti-oxidant and protective effect of Ganodema lucidum extract (GLE) against the DNA damage induced by free radical and U.V. In the assay of cell growth inhibition, the inhibitory cell growth rate induced by hydroxyl radical was dose-dependently decreased by GLE. This results support that GLE has a detoxifying activity against cytotoxicity of hydroxyl radical in E. coli cell. GLE also protected ColE1 plasmid DNA damage in the concentration of 200$\mu\textrm{g}$ per reaction on the DNA fragmentation assay. The nuclear tailing by hydrogen peroxide in single cell gel electrophoresis(SCGE) was decreased by GLE in the concentration of 50$\mu\textrm{g}$/ml. These data indicate that Ganoderma lucidum has an anti-oxidative activity to hydrogen peroxide. The mutation rate after irradiation of U.V. was reduced by 50$\mu\textrm{g}$/ml GLE and total number of Rif (Rifampicin) resistant mutants was decreased in a concentration dependent manner when added the GLE exogenously in a culture media. According to the results, it is likely that GLE has not only an anti-oxidative activity to hydroxyl radical but also an anti-mutagenic activity to U.V. mutagenesis.

  • This study was investigated the effects of water soluble extract of Ganoderma lucidum kale juice and soidium dextrothyroxine on lipid components in serum of hypercholesterolmemic rats in vivo in order to prevent in cardiovascular disease. Total cholesterol concentrations in serum were significantly increased after feeding cholesterol diet group compared with control group and were significantly increased after feeding cholesterol diet group compared with control group and were lower in Ganoderma lucidum kale juice and sodium dextrothyrioxine diet groups than in control group. Ratio of high density lipoprotein cholesterol concentration to total cholesterol were significantly highest in sodium dextrothyroxine fed groups among diet groups and were higher in Ganodrma lucidum and kale juice fed group than in control group. Phospholipid concentrations in seurm were significantly lower in Ganoderma lucidum kale juice and sodium dextrothyroxine(1.25mg/kg diet) fed groups than in control group and triglyceride concentra-tions were lower in Ganderma lucidum sodium dextrothyroxine fed groups than in other groups. Triiodothyronine concentration in serum were lower in Ganoerma lucdum and kale juice fed groups than in the other groups while it was higher in sodium dextrothyroxine diet group than in other groups. Tetraiodothyronine concentrations is serum were remakably higher in sodium dextrothyroxine fed group than in other groups. Blood glucose concentration was lower in cholesterol diet group than in other groups but was higher in sodium dextrothyroxine diet group than in other groups.
