겉보리에서 배양한 영지버섯 추출물의 항산화 및 항염증 효능 평가 (Evaluation of Anti-Oxidant and Anti-Inflammatory Activities of Ganoderma lucidum Cultured on Hulled Barley)

  • Background: Ganoderma lucidum cultured on hulled barley was investigated as a potential natural source of antioxidants and anti-inflammatory agents. Methods and Results: The yields from Ganoderma lucidum cultured on hulled barley water and ethanol extract were 17.69% and 25.77%, respectively. The antioxidant activity of Ganoderma lucidum cultured on hulled barley extracts was confirmed by various methods including assayss of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS), nitrite radical scavenging, and $Fe^{3+}$ to $Fe^{2+}$ reducing power activity. The ethanol extract of Ganoderma lucidum cultured on hulled barley showed improved DPPH, ABTS and nitrite radical scavenging activity compared with the water extract. After treatment of RAW264.7 cells with Ganoderma lucidum cultured on hulled barley ethanol extracts, the cell viability compared with the control was 92.82%, even at a concentration of $3,000{\mu}g/m{\ell}$. The ethanol extract inhibited reactive oxygen species (ROS) generation in RAW264.7 cells stimulated with $H_2O_2$, even at low concentrations. In addition, the ethanol extract showed an inhibitory effects on the production of lipopolysaccharide-induced nitric oxide (NO) in RAW264.7 cells. Conclusions: This study suggests that the extract of Ganoderma lucidum cultured on hulled barley is a potential source of natural antioxidants and anti-inflammatory agents.

영지(靈芝) (Ganoderma lucidum) 추출물(抽出物)이 유당(乳糖) 발효성(醱酵性) 효모(酵母)의 발효(醱酵)에 미치는 영향(影響) (The Effect of Korean Ganoderma Lucidum Extract on the Fermentation of Lactose-Fermentation Yeast)

  • 영지 extract가 유당발효성 효모인 Kluyveromyces fragilis KCCM 35458의 발효에 미치는 영향을 조사하기 위하여 탈지유 배양액에 영지 1호의 extract를 0%, 0.1%, 0.5% 및 1.0%의 농도별로 첨가하고 $30^{\circ}C$에서 72시간을 정치배양하면서 조사한 결과는 다음과 같다. 1. 영지 extract를 1.0% 첨가한 시험구(試驗區)는 대조구(對照區)에 비하여 시간이 경과함에 따라 pH에서 빠른 감소를 보였으며 적정산도에서는 빠른 증가를 보였다. 2. $CO_2$ 발생량(發生量)은 영지 extract를 첨가한 전(全) 시험구(試驗區)에서 높았으며 1.0% 첨가구의 효과가 월등하였다. 3. Alcohol 생성량(生成量)은 72시간 배양함에 따라 영지 extract 첨가구(添加區)에서 높게 나타났으며 1.0% 첨가구(添加區)에서 가장 높은 alcohol 생산성을 보였고 발효시간을 단축시켰다. 4. 효모수는 영지 extract 0.5%, 1.0%첨가구에서 대조구에 비하여 현저한 증식효과를 나타내었다. 5. 탈지유 배양액에 있어서의 Kluyveromyces fragilis의 유당발효 상태는 배양시간 경과에 따라 영지 extract 첨가구가 대조구에 비하여 발효가 촉진되었으며 1.0% 영지 extract 첨가구에서 효과적이었다.

의약품의 병용투여에 관한 연구 (VII) (Studies on the Concurrent Administration of Medicines(VII))

  • These studies were attempted to investigate the preventive effect of Ganoderma lucidum extract administered concurrently with glutathione on the liver damage induced by carbon tetrachloride ($CCl_4$) in rats. S-GOT and S-GPT activities of all the pre-treatment groups were significantly decreased, as compared with those of the control intoxicated by $CCl_4$. The concurrent administrations of Ganoderma lucidum extract with glutathione (100+100mg/kg, 200+100mg/kg, and 400+100mg/kg, respectively) were more effective than the individual administrations. i.e., Ganaderma lucidtcm extract (100, 200 and 400mg/kg, respectively) and glutathione (100, 200 and 400mg/kg, respectively). On the determination of lipid-peroxidation in liver, the concurrent administrations of Ganoderma lucidum extract with glutathione (100+100mg/kg, and 200+100mg/kg, respectively) significantly reduced the liver TBA values. Although hepatic cellular necrosis and fatty acid deposit were remarkably increased by $CCl_4$ intoxication, the concurrent administration of Ganoderma lucidum extract with glutathione (200+100mg/kg) reduced the pathological changes of parenchymal cell necrosis and fatty changes around centrilobalar area of the control. These findings indicate that the concurrent administrations of Ganoderma lucidum extract with glutathione showed better improvements than the individual administrations of them in all pathological aspects, in particular, against hepatitis and hepatic necrosis due to the cellular necrosis and fatty infiltration.

영지(靈芝) 약침(藥鍼)과 영지(靈芝) 추출액의 항알레르기 효과에 대한 연구 (Study on Anti-Allergic Effecst of Ganoderma lucidum Herbal Acupuncture and Ganoderma lucidum Extract)

  • Objectives We studied on anti-allergic effects of Ganoderma lucidum herbal acupuncture(GHA) and Ganoderma lucidum extract(GE). Methods In vivo, Animals were herbal-acupunctured GHA at both B13s three times for 5 days. Then, we investigated compound 48/80-induced active systemic anaphylatic shock using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis using Sprague Dawley rat. In vitro, we measured cell viability, b-hexosaminidase release, IL-4 and TNF-a from RBL-2H3 cells, and nitric oxide from Raw264.7 cell after treatment of GE of various concentrations. Results In vivo, GHA pretreatments at both B13s inhibited compound 48/80-induced active systemic anaphylatic shock. Passive cutaneous anaphylaxis were inhibited by GHA10 and OP. In vitro, $0.1\;{\sim}\;2%$ GE treatments were not affect on cell viability and inhibited b-hexosaminidase release, IL-4, TNF-a and nitric oxide. Conclusions These results suggest that GHA and GE may be beneficial in the inhibition of allergic inflammatory response.

Protective effect of methanolic extract of Ganoderma lucidum P. Karst. Reishi from South India against doxorubicin-induced cardiotoxicity in rats

  • Doxorubicin is a powerful anticancer antibiotic extensively used in the treatment of several types of cancers. Long-term administration of this drug results in cumulative dose related cardiotoxicity due to enhanced production of free radicals leading to oxidative stress. Our earlier investigations have demonstrated significant antioxidant, anti-inflammatory and antitumour properties of Ganoderma lucidum extracts. We extended our investigations to evaluate the protective effect of Ganoderma lucidum extract against doxorubicin-induced cardiotoxicity. Administration of 3 doses of doxorubicin, 6 mg/kg body weights, i.p. per each dose, alternative days, showed dear signs of cardiotoxicity in rats. The drug enhanced serum creatine kinase (CK) activity and lipid peroxidation in tissue drastically. The drug also induced significant decrease in GSH level and activities of CAT, SOD and GPx. Administration of methanolic extract of G.lucidum (500 and 1,000 mg/kg body weight) significantly increased the level of GSH and activities of CAT, SOD and GPx. Activity of CK was significantly lowered in a dose dependent manner. The treatment also caused significant decrease in lipid peroxidation (MDA). The results thus indicated that methanolic extract of G.lucidum prevented oxidative stress caused by doxorubicin administration and the increase in serum CK activity and lipid peroxidation in the tissue. The experimental findings suggest the therapeutic potential of G.lucidum as adjuvant in cancer chemotherapy.

인삼 및 영지추출물이 유산균 생육에 미치는 영향 (Effects of Panax ginseng and Ganoderma lucidum Extract on the Growth of Lactic Acid Bacteria)

  • This study was carried out to Investigate the effects of Panax ginseng, Ganoderma lucidum extract and crude polysaccharide of G. lucidum on the growth of lactic acid bacteria. p. ginseng extract contained 60.7% carbohydrate and 27.5% protein, whereas G. lucidum contained 35.9% carbohydrate and 46.3% protein. The total sugar and protein content of crude polysaccharide of G. lucidum were 47.2% and 15.2%, respectively. Two amino acids(hg, Trp) were detected in p. ginseng extract and 11 amino acids (hg, Trp, Ua, Lys, Ser, etc.) in C. lucidum extract. By the addition of p. ginseng, 5. lucidum extract and crude polysaccharide, the cia. p. ginseng was more effective on the growth of 1. casei an: G. lucidum was more effective on that of S. thermophilus. The effect of free amino acids on the growth of tactic acid bacteria was also examined. Arginine and lysine stimulated the growth of L. casei, whereas Lysine, serine, arginine, and glutamic acid stimulated the growth of 5. thermophilus.

효모를 이용한 발효영지 추출물이 흰쥐의 유영 운동 피로에 미치는 영향 (Effects of Yeast Fermented Ganoderma Lucidum Extract on the Recovery of Exercise Fatigue in Forced Swimming Rats)

  • Objectives : Ganoderma Lucidum has been used in traditional oriental medicine to treat various conditions including chronic fatigue and cough. This experimental study was designed to investigate the effects of yeast-fermented Ganoderma Lucidum extract on the fatigue of rats elicited by forced swimming. Materials and methods : The rat subjects were divided into 4 groups: normal group, control group administered with 0.9% normal saline, experimental I(Exp I) group administered with yeast-fermented Ganoderma lucidum extract and experimental II(Exp II) group administered with Ganoderma lucidum extract for 4 weeks. All the serum samples collected by heart puncture were used to measure biochemical factors such as glucose, lactate, lactate dehydrogenase(LDH), creatine kinase(CK), inorganic phosphorus, ammonia, total cholesterol(TC), triglycerides(TG), high-density lipoprotein(HDL), low-density lipoprotein(LDL), aspartate transaminase(AST), alanine transaminase(ALT). Results : 1. Weight change and swimming time were not significantly different in the experimental groups as compared with control group. 2. Lactate was significantly decreased in the experimental groups as compared with control group. Inorganic phosphorus was also significantly decreased in the Exp I group as compared with control group(p<0.05). 3. Serum lipid concentration was not significantly different in the experimental groups as compared with control group. 4. AST and ALT were significantly decrease in the experimental groups as compared with control group. Conclusions : Yeast-fermented Ganoderma lucidum in forced swimming rats would appear to reduce fatigue-related factors, AST, ALT of serum and promote anti-fatigue effect.

영지(靈芝)버섯(Ganoderma lucidum)의 소화기계(消化器系)에 대한 약효연구(藥硏究) (Studies on the Efficacy of Ganoderma lucidum in Digestive System)

  • This study was attempted to investigate the effect of Ganoderma lucidum extract on digestive system in experimental animals. Ganoderma lucidum water extract (GWE) was found to be promoted the charcol transport rate in the small intestine of mice. GWE exhibited the augmentation of spontaneus movement(motility) and contractile response(tension) in the ileum and colon strips of rabbit, and these action were inhibited by atropine. GWE given intraduodenaly(i.d.) exhibited the significant increase of gastric acid secretion in pylorus-ligated rats. GWE inhibited the formation of some experimental gastric ulcers(pylorus ligation ulcer i.d., indomethacin-induced ulcer p.o., i.d. and aspirin-induced ulcer p.o.) in rats, which are considered to relate to a protective action. GWE and EtOH extract(water soluble phase) were remarkably increase of bile excretion, when administration of i.d., intravenation(i.v.) and per os (p.o.) compared with normal-control group. GWE was observed antibacterial activity aginst several intestinal microoganisms and others bacteria in vitro test.

Ganoderma lucidum 균주에 의한 Laccase의 정제 및 효소적 특성 (Purification and enzyme characteristics of laccase from Ganoderma lucidum)

  • The production media and enzymatic characteristics of laccase from Ganoderma lucidum was investigated. Potato dextrose yeast extract media was proved to be the best for laccase production. The enzyme has optimum pH of 6.45km value of 6.71 mM and appeared to be stable at wide pH range. The enzyme was inactivated partially by methanol and ethanol and totally by sodium azide but not at all by acetone. Also the enzyme purification was performed and the data is given.

  • The effect of Ganoderma lucidum (Polyporaceae) extract on initiation and promotion of liver preneoplastic lesions in chemical carcinogenesis were examined in male rats. Male Fisher 344 rats, 8 weeks old, were injected i.p. with diethylnitro-samine(DENA` 200 mg/kg) as an initiator and were fed on diet containing 0.02%-acetaminofluorene (2-AAF) as a promoter for 2 weeks (group 1, 2` 2-4 week) and were fed on basal diet (group 3 and 4). The rats in test groups(group 1 and 3) were fed on diet containing 0.1% of Ganoderma lucidum extract for 6 weeks (2-8 week).
