Effect of light on fruit body primordium formation of Ganoderma lucidum on nutrient agar medium (광(光)이 Ganoderma lucidum의 자실체 원기 형성에 미치는 영향)

  • The objective of this research was to determine the effect of light quality on formation of fruit body primordia (FBPs) of Ganoderma lucidum. To achieve this 5 isolates of the fungus that develops fruit body primordia on nutrient agar media were incubated with or without continuous irradiation. The fluorescent lamps used different colors such as black light blue (BLB), pure blue (P-B), pure green (P-G), pure yellow (P-Y) and pure red (P-R). Effect of periodic light and dark exposures on FBP formation of isolate Gl-009 was investigated. The FBP formation in G. lucidum isolates was also tested under monochromatic light produced by the combination of interference filters and colored glass filters. Three isolates produced FBPs under all kinds of fluorescent lamps, whereas two induced FBPs only under visible light except for BLB fluorescent lamp. However, these isolate did not form FBPs in the dark. The FBP was formed at light intensity from 0.05 to $10.0\;{\mu}mol\;m^{-2}s^{-1}$, and begun to reduce its number as light intensity increase over $0.5\;{\mu}mol\;m^{-2}s^{-1}$. When the isolate was incubated under periodic light and dark exposures, the number and weight of FBP increased as compared with those under continuous light. Initiation of FBP requires at least 4 days of light illumination. Although isolate Gl-003 produced FBPs in a wide range of 400 to 800 nm, other four isolates had two effective regions 400 to 500 nm and 700 to 750 nm in FBP formation.

Antitumor Activity and Effect on Cell Proliferation and Differenciation of Exopolysaccharide Produced by Submerged Cultivation of Ganoderma lucidum (영지(Ganoderma lucidum)의 액체배양에 의한 세포외 다당의 항암활성과 세포증식 및 분화에 미치는 영향)

  • Exopolysaccharide (CBP) from submerged culture broth of Ganoderma lucidum mycelium and the water soluble (BWS) and water insoluble (BWI) fractions of CBP were prepared by gel filtration. Antitumor activity and effects on proliferation and differenciation of human cancer cells and mouse NIH 3T3 cells were studied. Cytotoxicity test of CBP, BWS and BWI fractions on human cancer cell lines was performed by using sulforhodamine B (SRB) assay. A549 (lung carcinoma), Colo320 DM and HSR (colon carcinoma), and NIH 3T3 cells were used. BWI fraction showed the strongest cytotoxicity (maximum 20% survival) to all human cells tested. However it did not induced apoptosis. Interestingly BWI fraction did not exert cytotoxic effect on NIH 3T3 cells at low concentration of cells ($5{\times}10^4$) but strong toxic effect at high concentration of cells($5{\times}10^5$) which showed transformed morphology. These results suggest that BWI may have cancer cell specific anticancer activity. However, BWI fraction did not effect the amount of pRb and c-myc protein, which implied that BWI fraction did not act at the early stage of signal transduction pathway. CBP fraction induced differenciation of human leukemic cell line, HL-60 cells suggesting the carcinogenesis prevention of normal cell and possible induction of normalization for cancer cell.

Screening of Inhibitory Activity of Edible Mushrooms on Dopamine ${\beta}-Hydroxylase$ (식용 버섯류의 도파민 베타 수산화효소에 대한 저해활성 검색)

  • Dopamine ${\beta}-hydroxylase\;(DBH)$ catalyses the enzymatic reaction of dopamine to norepinephrine. For the purpose of screening DBH inhibitory activity from edible mushrooms, Ganoderma lucidum, Agaricus bisporus and Lentinus edodes were examined by tracing inhibitory activities against bovine adrenal DBH, utilizing tyramine as a substrate. Among the three edible mushrooms tested, Ganoderma lucidum showed potent enzyme inhibitory activilies above 100% against DBH in chloroform fraction. Lentinus edodes and Agaricus bisporus showed inhibitory activities in ethylacetate fraction on 79.7% and 64.7%, respectively. Each solvent fraction of these mushrooms were assessed in the aspects of their inhibitory activities against DBH, and their $IC_{50}$ values were calculated. $IC_{50}$ value of chloroform fraction of Ganoderma lucidum was $1.60{\times}10^{-4}\;g$, and those of ethylacetate fractions of Agaricus bisporus and Lentinus edodes were $5.50{\times}10^{-4}\;g\;and\;2.35{\times}10^{-4}\;g$, respectively.

  • Isozyme patterns of leucine aminopeptidase, esterase and protein from 16 isolates of Ganoderma lucidum were observed by electrophoresis for characterization of the isolates. Even in the same isolate, the patterns of the isozymes from mycelium and cap were different. The esterase patterns from the mycelium could differentiate the isolates. Of 16 isolates, four isolates showed identical patterns. It was assumed that these had the same genetic background.

  • The anticoagulant polysaccharide was screened from the ediable mushrooms. Among them, alkali extract of Ganoderma lucidum showed the highest activity in aPTT. A crude polysaccharide fraction (GL-I) was prepared from the 1N NaOH solution extract of Ganoderma lucidum followed by methanol-reflux, precipitation with ethanol, dialysis and lyophilization. GL-I inhibited the intrinsic pathway in blood coagulation pathway and exhibited concentration dependent anticoagulation effects. The anticoagulant activity of GL-I was decreased greatly by periodate oxidation, but was not changed by pronase digestion. These suggest that carbohydrate moiety may be related to the anticoagulant activity. GL-I consisted of glucose, galactose, fucose, xylose, mannose, arabinose in a molar ratio of 19.3:3.0:2.3:1.3:1.0:0.3. GL-I was partially purified on the DEAE-Toyopearl 650C(GL-IalongrightarrowGL-If) and on the Sephadex G-100(GL-Ic-ilongrightarrowGL-Ic-II).

Studies on the Trace Elements, Heavy Metals and Organic acids Content of Edible Mushrooms (버섯류의 미양금속, 중금속 및 유기산 함양에 관한 연구)

  • To investigate on the element content of twelve edible mushrooms and Aloe arborescent, i.e., Lentinus edodes, Ganoderma lucidum (culturing in wood and soil), Tricholoma matsutake, Agaricus auricula-Jude (produced in Korea and China), sarcodon asparatus, Pleurotus ostreatus, Coriolus bisporus, Cyrophora esculenta, Auricularia auricula-Jude (produced in Korea and China), Sarcodon asparatus, Pleurotus ostreatus, Coriolus rersicolor, Smilax rotundifolia and Aloe arborescenstyl were analyzed by Atomic absorption spectrometer. The obtained results were summarized as follows : 1. Potassium, sodium, magnesium and iron content for the most part samples were in large quantities, especi phosphorus content of those was highest ammount for the all samples. 2. Sodium content f·as much ammount in the Lentinus edodes(39 mg, and Ganoderma lucidum(20 mg) , Culturing in wood and soil, while potassium was very high ammount in the Aloe arborescens and other samples. Mush ammount of magnesium as compared with others was Lentinus edodes(144 mg) , Ganoderma lucidum(128 mg), Aloe arborescens(50 mg) and Pleurotus ostreatus (60 mg) . 3, Phosphorus content of Lentinus edodes, Gyrophora esculenta, Auricularia polytricha and Agaricus bisporus was much ammount while iron content of all samples equality higher ammount. Sodium content of Aloe arborescens was not analyzed out for almost all, its potassium(82 mg), magnesium(50 mg) and iron(18 mg) content comparatively higher quentity than others nllnerals and phosphorus volume (4.9 mg) as compared with others, was conspicuously lower detect. 4. Cadimium and lead content of harmful metal element were detected on trace quantity for the most part samples. 5. Organic acids of samples i.e., Lentinus edodes, Agaricus bisporus, Pleuritus ostreatus and Ganoderma lucidum were Citrate, Malate, Fumalate, Succinate, Oxalate, Acetate, Lactate, and Tartarate and Citrate, Malate and Fumarate contents were higher amount remarkbly than other organic acids. Tartarate content was trace amount.

  • The medicinal properties of Ganoderma lucidum have been recognized in China for many centuries. Active pharmaceutical components include triterpenes. To elucidate the molecular regulation of triterpene biosynthesis in this mushroom, a 57-base pair DNA fragment encoding the fourth conserved domain SQ-4 (SMGLFLQKTNIIRDYNEDL) of squalene synthase was synthesized and cloned into the expression vector pET-32a(+). The recombinant fusion protein induced by IPTG (isopropyl-$\beta$-D-thiogalactopyranoside) was overexpressed in the Escherichia coli. Using the purified recombinant fusion protein of 20.9 kDa, a specific polyclonal antibody was obtained from immunized rabbit. Expression of squalene synthase at different development stages of Ganoderma lucidum was analyzed.

Screening of Hepatoprotective Substances from Higher Fungi by Primary Cultured Rat Hepatocytes intoxicated with Carbon Tetrachloride ($CCl_4$로 독성을 유발시킨 초대배양 간세포를 이용한 고등균류로부터 간세포 보호물질의 검색)

  • Hepatoprotective effects of polysaccharides extracted from liquid cultured mycelia were screened by measuring the glutamic pyruvate transaminase activity of the primary cultured rat hepatocytes intoxicated with carbon tetrachloride. Sixty among 75 isolates of higher fungi showed to be hepatoprotective effect, and these were 13 of Ganoderma lucidum, 5 of Lentinus edodes, 1 of Pleurotus ostereatus, 4 of Coriolus versicolor, 2 of Lyophyllum spp., 7 of Grifora frondosa, 3 of Agaricus spp., 14 of Schizophyllum commune and 11 of Cordyceps spp.. Especially, 10 isolates, Ganoderma lucidum IY003 and IY009, Lentinus edodes IY103, Lyophyllum sp. IY402, Agaricus sp. IY701 and IY703, Schizophyllum commune IY804, IY810 and IY818, Cordyceps sp. IY902, were indicated below 80% of glutamic pyruvate transaminase activity.

Hepatoprotective Evaluation of Ganoderma lucidum Pharmacopuncture: In vivo Studies of Ethanol-induced Acute Liver Injury

  • Objectives: Alcohol abuse is a public issue and one of the major causes of liver disease worldwide. This study was aimed at investigating the protective effect of Ganoderma lucidum pharmacopuncture (GLP) against hepatotoxicity induced by acute ethanol (EtOH) intoxication in rats. Methods: Sprague-Dawley (SD) rats were divided into 4 groups of 8 animals each: normal, control, normal saline pharmacopuncture (NP) and GLP groups. The control, NP and GLP groups received ethanol orally. The NP and the GLP groups were treated daily with injections of normal saline and Ganoderma lucidum extract, respectively. The control group received no treatment. The rats in all groups, except the normal group, were intoxicated for 6 hours by oral administration of EtOH (6 g/kg BW). The same volume of distilled water was administered to the rats in the normal group. Two local acupoints were used: Qimen (LR14) and Taechung (LR3). A histopathological analysis was performed, and the liver function and the activities of antioxidant enzymes were assessed. Results: GLP treatment reduced the histological changes due to acute liver injury induced by EtOH and significantly reduced the increase in the alanine aminotransferase (ALT) enzyme; however, it had an insignificant effect in reducing the increase in aspartate aminotransferase (AST) enzyme. It also significantly ameliorated the superoxide dismutase (SOD) and the catalase (CAT) activities. Conclusion: The present study suggests that GLP treatment is effective in protecting against ethanol-induced acute hepatic injury in SD rats by modulating the activities of ethanol-metabolizing enzymes and by attenuating oxidative stress.

Enzymatic Properties of a Cellulase from Ganoderma lucidum (불로초(不老草)가 생산(生産)하는 Cellulase의 효소학적(酵素學的) 성질(性質))

  • A cellulose-degrading enzyme from Ganoderma lucidum was partially purified by ammonium sulfate precipitation and its enzymatic properties were studied. The enzyme had an optimum pH for activity at 4.0, and its stability range was pH $4.0{\sim}7.0$. The optimum temperature was $55^{circ}C$ and the enzyme retained 80% original activity after heated at $50^{\circ}C$ for 60 min. The activation energy of the enzyme for CMC degradation was caculated and found to be 6.2 Kcal/mole. The enzyme was activited by the addition of $Co^{++},\;Mn^{++}$, but slightly inactivated by $Hg^{++}$. Various enzyme inhibitors and chemical reagents did not affect the enzyme activity. The enzyme acted on native celluose as well as CMC. The Michaelis constant for CMC was calculated to be 2.4 mg glucose ep/ml.
