LncRNA CRNDE Promotes the Progression of B-cell Precursor Acute Lymphoblastic Leukemia by Targeting the miR-345-5p/CREB Axis

  • The imbalance between the proliferation and apoptosis of B-cell precursors is an important contributor to the pathogenesis of B-cell precursor acute lymphoblastic leukemia (BCP-ALL), while its specific regulatory mechanism remains perplexing. This study aimed to expound the underlying mechanism of the proliferation and apoptosis of BCP-ALL cells from the perspective of non-coding RNA. In this study, long non-coding RNA colorectal neoplasia differentially expressed (LncRNA CRNDE) was upregulated in the bone marrow of BCP-ALL patients and BCP-ALL cell lines (NALM-6 and RS4;11). Functionally, LncRNA CRNDE knockdown restrained cell proliferation and boosted cell apoptosis in NALM-6 and RS4;11 cells. The subsequent investigation confirmed that LncRNA CRNDE bound to miR-345-5p and negatively regulated miR-345-5p expression. The overexpression of miR-345-5p suppressed cell proliferation and boosted cell apoptosis in NALM-6 and RS4;11 cells. Further experiments revealed that miR-345-5p downregulated cyclic AMP response element-binding protein (CREB) expression by targeting its mRNA directly. CREB overexpression reversed the effect of miR-345-5p mimic on cell proliferation and apoptosis in NALM-6 and RS4;11 cells. Finally, in vivo experiments showed that LncRNA CRNDE knockdown prolonged the survival of mice xenotransplanted with NALM-6 cells. In conclusion, LncRNA CRNDE upregulated CREB expression by suppressing miR-345-5p, thus promoting cell proliferation and reducing cell apoptosis in BCP-ALL.

Classification of Microsatellite Alterations Detected in Endoscopic Biopsy Specimens of Gastric Cancers (단순반복염기서열의 변이 형태에 따른 위암 내시경 조직의 유전자형 분류)

  • Purpose: Individual gastric cancers demonstrate complicated genetic alterations. The PCR-based analysis of polymorphic microsatellite sequences on cancer-related chromosomes has been used to detect chromosomal loss and microsatellite instability. For the purpose of preoperative usage, we analyzed the correspondance rate of the microsatellite genotype between endoscopic biopsy and surgical specimens. Materials and Methods: Seventy-three pairs of biopsy and surgical specimens were examined for loss of heterozygosity and microsatellite instability by using 40 microsatellite markers on eight chromosomes. Microsatellite alterations in tumor DNAs were classified into a high-risk group (baselinelevel loss of heterozygosity: 1 chromosomal loss in diffuse type and high-level loss of heterozygosity: 4 or more chromosomal losses) and a low-risk group (microsatellite instability and low-level loss of heterozygosity: 2 or 3 chromosomal losses in diffuse type or $1\∼3$ chromosomal losses in intestinal type) based on the extent of chromosomal loss and microsatellite instability. Results: The chromosomal losses of the biopsy and the surgical specimens were found to be different in 21 of the 73 cases, 19 cases of which were categorized into a genotype group of similar extent. In 100 surgical specimens, the high-risk genotype group showed a high incidence of nodal involvement (19 of 23 cases: $\leq$5 cm; 23 of 24 cases: >5 cm) irrespective of tumor size while the incidence of nodal involvement for the low-risk genotype group depended on tumor size (5 of 26 cases: $\leq$5 cm; 18 of 27 cases: >5 cm). Extraserosal invasion was more frequent in large-sized tumor in both the high-risk genotype group ($\leq$5 cm: 12 of 23 cases; >5 cm: 23 of 24 cases) and the low-risk genotype group ($\leq$5 cm: 7 of 26 cases; >5 cm: 16 of 27 cases). The preoperative prediction of tumor invasion and nodal involvement based on tumor size and genotype corresponded closely to the pathologic tumor stage (ROC area >0.7). Conclusion: An endoscopic biopsy specimen of gastric cancer can be used to make a preoperative genetic diagnosis that accurately reflect the genotype of the corresponding surgical specimen.

In Vivo Dosimetry with MOSFET Detector during Radiotherapy (방사선 치료 중 MOSFET 검출기를 이용한 체표면 선량측정법)

  • In Vivo dosimetry is a method to evaluate the radiotherapy; it is used to find the dosimetric and mechanical errors of radiotherapy unit. In this study, on-line In Vivo dosimetry was enabled by measuring the skin dose with MOSFET detectors attached to patient's skin during treatment. MOSFET dosimeters were found to be reproducible and independent on beam directions. MOSFET detectors were positioned on patient's skin underneath of the dose build-up material which was used to minimize dosimetric error. Delivered dose calculated by the plan verification function embedded in the radiotherapy treatment planning system (RTPs), was compared with measured data point by point. The dependency of MOSFET detector used in this study for energy and dose rate agrees with the specification provided by manufacturer within 2% error. Comparing the measured and the calculated point doses of each patient, discrepancy was within 5%. It was enabled to verify the IMRT by using MOSFET detector. However, skin dosimetry using conventional ion chamber and diode detector is limited to the simple radiotherapy.

Characterization of Fusarium udum Causing Fusarium Wilt of Sunn Hemp in Korea (클로탈라리아 시들음병을 일으키는 Fusarium udum의 특성)

  • Sunn hemp (Crotalaria juncea) is used as a nitrogen-fixing green manure in Korea to improve soil quality, reduce soil erosion, and suppress weeds and nematodes. In 2014, wilting sunn hemp plants were observed in green manure-cultivated fields in Wanju, Korea. Leaves of the infected plants began yellowing, starting with the lower leaves, eventually leading to their death. Moreover, a number of dark perithecia were observed on the wilting stems. Six isolates were obtained from these perithecia by single spore isolation. Based on their morphological characteristics, the isolates were identified as Fusarium udum (teleomorph: Gibberella indica). Macroconidia were slightly curved with almost hooked apical cell, and microconidia were formed on false heads by monophialides. Chlamydospores were produced abundantly in the hyphae, either singly or in clusters. To confirm the identification, multilocus sequence analysis was conducted using translation elongation factor 1 alpha (TEF), calmodulin (CAL), and histone 3 (HIS3). The sequences of TEF, CAL, and HIS3 showed 94.4~96.2%, 99.7%, and 99.6~99.8% similarity to the reference sequences of F. udum in NCBI GenBank, respectively. Pathogenicity was tested on sunn hemp and two soybean cultivars using the inoculation method of soil drenching with spore suspension. The wilting symptoms were observed only in sunn hemp and one cultivar of soybean (cv. Teagwang) after 14~21 days of inoculation. This is the first report of wilt disease in sunn hemp caused by Fusarium udum in Korea.

Ridge and field tile aerodynamics for a low-rise building: a full-scale study

  • Recent major post-hurricane damage assessments in the United States have reported that the most common damages result from the loss of building roof coverings and subsequent wind driven rain intrusion. In an effort to look further into this problem, this paper presents a full-scale (Wall of Wind --WoW--) investigation of external and underneath wind pressures on roof tiles installed on a low-rise building model with various gable roofs. The optimal dimensions for the low-rise building that was tested with the WOW are 2.74 m (9 ft) long, 2.13 m (7 ft) wide, and 2.13 m (7 ft) high. The building is tested with interchangeable gable roofs at three different slopes (2:12; 5:12 and 7:12). The field tiles of these gable roofs are considered with three different tile profiles namely high (HP), medium (MP), and low profiles (LP) in accordance with Florida practice. For the ridge, two different types namely rounded and three-sided tiles were considered. The effect of weather block on the "underneath" pressure that develops between the tiles and the roof deck was also examined. These tests revealed the following: high pressure coefficients for the ridge tile compared to the field tiles, including those located at the corners; considerably higher pressure on the gable end ridge tiles compared to ridge tiles at the middle of the ridge line; and marginally higher pressure on barrel type tiles compared to the three-sided ridge tiles. The weather blocking of clay tiles, while useful in preventing water intrusion, it doesn't have significant effect on the wind loads of the field tiles. The case with weather blocking produces positive mean underneath pressure on the field tiles on the windward side thus reducing the net pressures on the windward surface of the roof. On the leeward side, reductions in net pressure to a non-significant level were observed due to the opposite direction of the internal and external pressures. The effect of the weather blocking on the external pressure on the ridge tile was negligible.

Correlation between Visceral Adipose Tissue and Stagnation of the Liver Qi (gan-yu, 肝鬱) in Korean Perimenopausal Women (한국 갱년기 여성의 간울과 내장비만의 상관성)

  • 목적 : 갱년기 여성은 내장비만의 위험이 높으며 또한 심리적 변화를 겪는 시기이다. 갱년기 여성의 복부지방, 내장지방, 피하지방 및 이와 관련된 단순비만지표, 간울증, 스트레스, 우울, 자존감 등을 조사하여 갱년기 건강의 위협이 되는 신체 심리 인자를 알아보고자 이 연구를 시행하였다. 방법 : 병원공고를 통해 체질량지수 $23kg/m^2$ 이상 비흡연자인 만 45세 이상 55세 이하의 폐경 전후의 갱년기 여성 환자를 모집하여 2007년 8월 20일부터 24일까지 47명이 모집되었으며 이중 체질량지수 $25kg/m^2$ 이상이면서 복부 전산화 단층촬영 상 내장지방이 $100cm^2$ 이상인 20명의 단순비만지표, 체성분검사, 혈액검사, 복부 전산화 단층촬영 및 한방비만변증 (간울) 및 스트레스, 우울, 자존감 설문검사 결과의 상관성을 분석하였다. 본 연구는 경희대학교 동서신의학병원의 임상연구심사위원회의 승인을 받았다. 결과 : 1. 갱년기 비만여성에서 복부지방 면적, 피하지방 면적은 체질량지수, 체지방률, 허리둘레/신장비 등의 단순비만지표와 유의한 상관성을 보인 반면 (p < 0.01), 내장지방 면적은 단순비만지표와 상관성을 보이지 않았다. 2. 내장지방 면적은 사회 재적응 평가척도 (${\gamma}=0.577$, p < 0.01)와 유의한 상관성이 있었고, 선형회귀분석에서 유의한 결과를 나타냈다. $VAT(cm^2)=116.1+0.101{\times}(SRRS\;score)$ (${\gamma}^2=0.332$) 3. 피하지방은 식이태도 점수와, 총복부지방은 식이태도 점수 및 갱년기지수 중 혈관운동증상과 상관성이 있었다. 4. 간울증은 스트레스 반응척도, 우울지수, 갱년기지수 및 그 하부항목 중 혈관운동, 정신, 운동, 소화, 전신증상과 양의 상관성을, 자존감척도와는 음의 상관성을 나타냈으며, 내장지방과 직접적인 상관성을 보이지 않았다. 결론 : 간울증은 높은 스트레스, 우울, 갱년기 증상 및 낮은 자존감과 관련되는 것으로 나타났다. 폐경전후 비만여성에서 내장지방 면적은 간울증과 유의한 상관성을 보이지 않았으나, 생활 스트레스 사건이 많을수록 높아지는 것으로 나타나 갱년기 여성 내장비만에서 스트레스에 대한 대처가 중요할 것으로 사료된다. 추후 임상적 연계성 및 설문 보완에 대한 연구가 필요하리라 생각된다.

Free-standing GaN 기판을 이용한 GaN 동종에피성장 및 높은 인듐 조성의 InGan/GaN 다층 양자우물구조의 성장

  • 이전 연구에서는 사파이어 기판 위에 이종에피성장 방법으로 성장한 높은 인듐 조성의 극박 InGaN/GaN 다층 양자우물 구조를 이용한 근 자외선 (near-UV) 영역의 광원에 대하여 보고하였다. 본 연구에서는 HVPE (Hydride Vapor Phase Epitaxy) 법을 이용하여 성장된 free-standing GaN 기판 위에 유기금속 화학증착법 (MOCVD) 을 이용하여 GaN 동종에피박막과 높은 인듐 조성의 InGaN/GaN 다층 양자우물을 성장하였고 그 특성을 분석하였다. Free-standing GaN 기판은 표면 조도가 0.2 nm 인 평탄한 표면을 가지며 $10^7/cm^2$ 이하의 낮은 관통전위밀도를 가진다. Freestanding GaN 기판 위에 성장 온도와 V/III 비율을 조절하여 GaN 동종에피박막을 성장하였다. 또한 100 nm 두께의 동종 GaN 박막을 성장한 후에 활성층으로 이용될 높은 인듐 조성의 InGaN/GaN 다층 양자우물구조를 성장하였다. Free-standing GaN 기판 위에 성장된 GaN 동종에피박막과 다층 양자우물구조의 표면 형상은 주사 탐침 현미경 (scanning probe microscopy, SPM) 을 이용하여 관찰하였고 photoluminescence (PL) 측정과 cathodoluminescence (CL) 측정을 통하여 광학적 특성을 확인하였다. 사파이어 기판 위에 성장된 2 um 의 GaN을 이용하여 성장된 높은 인듐 조성의 InGaN/GaN 다층 양자우물의 결함밀도는 $2.5 \times 10^9/cm^2$ 이지만 동일한 다층 양자우물구조가 free-standing GaN 기판 위에 성장되었을 경우 결함 밀도는 $2.5\;{\times}\;10^8/cm^2$로 감소하였다. Free-standing GaN 기판의 관통전위 밀도가 $10^7/cm^2$ 이하로 낮기 때문에 free-standing GaN 기판에 성장된 높은 인듐 조성의 다층 양자우물구조의 결함밀도가 GaN/sapphire 에 성장된 다층 양자우물의 결함밀도 보다 감소했음을 알 수 있다. Free-standing GaN 기판에 성장된 다층 양자 우물은 성장온도에 따라 380 nm 에서 420 nm 영역의 발광을 보이며 PL 강도도 GaN/sapphire 에 성장한 다층 양자우물의 PL 강도 보다 높은 것을 확인할 수 있다. 이것은 free-standing GaN 기판에 성장된 높은 인듐 조성의 InGaN/GaN 다층 양자우물구조의 낮은 결함밀도로 인하여 활성층의 발광 효율이 개선된 것임을 보여준다.

The Clinical Study of Tongue Change in Diabetes Mellitus Patients as Oriental Diagnostic Method (糖尿환자의 27例의 舌診에 關한 臨床 硏究)

  • Tongue examination is the most unique and important diagnostic method of oriental medicine. It reveals patients condition and give some information about direction of therapy and background of disease as well as produces clue of duration of disease changing that grasps it from outside. The aims of this study show that results of tongue examination is related to somewhat special pattern. we study 27 patients(age of means: 63.01 years old, male: 14, female: 13) who come to Wonkwang University Oriental-Medicine Hospital at Chonju with diabetes mellitus or diabetes mellitus complications. Tongue colors of this study result in pale-red 8 cases, pale 6 cases, red 6 cases, crimson 5 cases, blue-green-purple 2 cases and tongue fur of this study result in white-fur 16 cases, yellow-fur 7 cases, black-fur 1 case, none-fur 3 cases. And the condition of tongue fur with grimy and thin result in thin-white-fur 12 cases, white-grimy-fur 4 cases, thin-yellow-fur 5 cases, grimy-yellow-fur 2 cases, black-fur 1 case. Means level of fasting glucose during 7 days was $223.24{\pm}32.l7mg/dl$ and postprandial 2hours was $286.37{\pm}24.54mg/dl$ There were no changes in tongue body and tongue color but changes occur in tongue fur with 9cases in this period. 5 cases of patient make a difference between FBS(fasting blood glucose) and postprandial blood glucose level more than l00mg/dl (2 cases of patient with thin-white-fur gradually turned to slight-yellow-fur. There were no changes in 2 cases of patient with crimson-none-fur and pale-white-fur. 1 case of patient with slimy-yellow-fur turned to black fur.) 11 cases of patient had lesser than $10\%$ hemoglobin $A_lC$ and tongue color of these patient were pale-red 5cases, pale 2 cases, red 2 cases, crimson 2 cases. 16 cases of patient had more than $10\%$ Hemoglobin $A_1C$ and tongue color of these patients were pale-red 3 cases, pale 4 cases, red 4 cases, crimson 3 cases, blue 2 cases. This result shows that quantity of Hemoglobin $A_1C$ make a somewhat role in tongue color. The above results show that tongue color, fur color, condition and change of fur in diabetes mellitus patients is various in pale-red, pale, crimson, none-fur. So it is difficult to give an exact diagnosis on pathology of diabetes mellitus only with tongue examination because there are a little matches between blood glucose level, prevalence-period, short-term blood glucose regulation and tongue and fur colors.

  • A survey was conducted to investigate the chemical properties of soils such as pH, electrical conductivity, total organic matter content, soluble nitrate, available phosphate and major exchangeable canons, in plastic film houses at 513 sites. All the parameters surveyed in the plastic film house were much higher than those of open field soils. Particularly conspicuous was the accumulation of available P, exchangeable K and the occurrence of nitrate at relatively high concentration in both top soil(0-20 cm) and sub-soil(20-40 cm). In 70-80% of cases, the contents of available P and exchangeable K in top soils, were found to be higher than optimum levels. There was positive linear correlation between the content of exchangeable rations, and nitrate and EC of soils. The correlation coefficient was greater in the order of nitrate-EC > Mg-nitrate > K-nitrate > Ca-nitrate. The successive cultivation of horticultural crops in the plastic houses tended result in the accumulation of available P, exchangeable K and total organic matter in the soil.

Effects of Auricular Acupressure on Quality of Sleep and Depression (귀 지압이 수면의 질과 우울증에 미치는 영향)

  • This study was conducted to examine the effects of auricular acupressure on quality of sleep and depression. Adults who had insomnia for over 1 month were recruited. Participants were distributed randomly to the experimental group(n=22) and the control group(n=22). The experimental group received auricular acupressure on Shen men(TF2), insomnia 1(SF4), insomnia 2(SF1), and anti-depression(LO8) for 1 week. The contrast group received sleep hygiene education. The mean differences of the quality of sleep were 4.05±2.52 in the experimental group and 1.45±3.77 in the control group. Auricular acupressure significantly decreased insomnia in the experimental group compared to the control group(t=3.07, p=.004). Depression scores after acupressure were not different between the experimental group and the control group. After controlling the religion as a covariate, the mean difference of depression in the experimental group was higher than the control group(F=4.22, p=.046). In conclusion, auricular acupressure decreased insomnia and depression. Therefore, further studies on different acupressure points such as shen men, jiao gan, heart, pi zhi xia, and endocrine are recommended.
