The soil bacterial community and some inoculated bacteria were monitored to assess the microbial responses to prescribed fire in their microcosm. An acridine orange direct count of the bacteria in the unburned control soil were maintained at a relatively stable level $(2.0\~2.7\times10^9\;cells/g^{-1}{\cdot}soil)$ during the 180 day study period. The number of bacteria in the surface soil was decreased by fire, but was restored after 3 months. Inoculation of some bacteria increased the number of inoculated bacteria several times and these elevated levels lasted several months. The ratios of eubacteria detected by a fluorescent in situ hybridization (FISH) method to direct bacterial count were in the range of $60\~80\%$ during the study period, with the exception of some lower values at the beginning, but there were no definite differences between the burned and unburned soils or the inoculated and uninoculated soils. In the unburned control soil, the ratios of $\alpha-,\beta-\;and\;\gamma-subgroups$ of the proteobacteria, Cytophaga-Flavobacterium and other eubacteria groups to that of the entire eubacteria were 13.7, 31.7, 17.1, 16.8 and $20.8\%,$ respectively, at time 0. The overall change on the patterns of the ratios of the 5 subgroups of eubacteria in the uninoculated burned and inoculated soils were similar to those of the unburned control soil, with the exception of some minor variations during the initial period. The proportions of each group of eubacteria became similar in the different microcosms after 6 months, which may indicate the recovery of the original soil microbial community structure after fire or the inoculation of some bacteria. The populations of Azotobacter vinelandii, Bacillus megaterium and Pseudomonas fluorescens, which had been inoculated to enhance the microbial activities, and monitored by FISH method, showed similar changes in the microcosms, and maintained high levels for several months.
Bioaugmentation of bioreactors focuses on the removal of numerous organics, with little attention typically paid to the maintenance of high and stable nitrite accumulation in partial nitrification. In this study, a bioaugmented membrane bioreactor (MBR) inoculated with enriched ammonia-oxidizing bacteria (AOB) was developed, and the effects of dissolved oxygen (DO) and temperature on the stability of partial nitrification and microbial community structure, in particular on the nitrifying community, were evaluated. The results showed that DO and temperature played the most important roles in the stability of partial nitrification in the bioaugmented MBR. The optimal operation conditions were found at 2-3 mgDO/l and $30^{\circ}C$, achieving 95% ammonia oxidization efficiency and nitrite ratio ($NO_2^-/{NO_x}^-$) of 0.95. High DO (5-6 mg/l) and low temperature ($20^{\circ}C$) had negative impacts on nitrite accumulation, leading to nitrite ratio drop to 0.6. However, the nitrite ratio achieved in the bioaugmented MBR was higher than that in most previous literatures. Denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH) were used to provide an insight into the microbial community. It showed that Nitrosomonas-like species as the only detected AOB remained predominant in the bioaugmented MBR all the time, and coexisted with numerous heterotrophic bacteria. The heterotrophic bacteria responsible for mineralizing soluble microbial products (SMP) produced by nitrifiers belonged to the Cytophaga-Flavobacterium-Bacteroides (CFB) group, and $\alpha$-, $\beta$-, and $\gamma$- Proteobacteria. The fraction of AOB ranging from 77% to 54% was much higher than that of nitrite-oxidizing bacteria (0.4-0.9%), which might be the primary cause for the high and stable nitrite accumulation in the bioaugmented MBR.
USEPA 1605 방법을 이용하여 서울시 상수도계통에서 Aeromonas를 조사하였다. 2002년 7월부터 2003년 12월까지 매월 한강수계 하천수와 서울시 정수장에서 공급되는 정수, 수돗물에서 시료를 채취하였다. Aeromonas는 선택배지(ADA-V)를 사용하여 membrane 필터 표면에 성장한 노란색 집락을 계수하여 측정하였다. 하천수에서는 Aeromonas가 $1.0{\times}10^{0}-9.8{\times}10^{3}\;CFU/ml$의 농도로 검출되었으며, 정수에서는 검출되지 않았다. 수돗물에서는 141개 중 1개의 시료에서 1 CFU/500 ml의 농도로 검출되었다. 확인된 Aeromonas는 대부분 비병원성인 A. salmonicida(51%)였고, 이 외에도 A. caviae(4.7%), A. schubertti(3.4%), A. sobria(3.8%), A. hydrophila(2.1%), A ichithiosmia(0.4%)등이 동정되었다. A. salmonicida에 대한 염소 저항성을 평가한 결과, 0.2 mg/L 염소농도에서 30초 접촉 후, 99.99% (일부)가 제거되었다. USEPA 1605 방법에서 제시한 정도관리를 수행한 결과, 정도관리 허용기준을 만족하였다. 본 연구를 통해 서울시 상수도계통에서 Aeromonas에 대한 안전성이 확보되었다.
금강 하구에서 분리한 용혈독소를 생산하는 V. mimicus와 유사하지만 분류되지 않은 기수성 Vibrio속으로 추정되는 균주 Vibrio sp. E10 (V. kunsan)의 세균학적 특징은 다음과 같다. 기본적인 생화학적 시험과 전자현미경 상의 형태에서는 V. mimicus와 유사하였지만 TCBS 배지 상의 colony 형태는 V. vulnifirus와 유사하였으므로 V. mimicus와는 차이를 보였다. Human과 sheep blood agar에서의 뚜렷한 용혈환과 rat에 피부혈관투과항진 작용으로 용혈독성이 있었으며, mouse에 대한 치사독성이 있었으므로 병원성이 확인되었다. 생육 가능한 환경 조건은 염도 0$\~$$4.5\%$, pH 6.2$\~$9.2, 온도 $14\~42^{\circ}C$이 었다. Vibrio sp. E10의 16S rDNA 염기서열은 10여종의 V. cholerae와 1종의 Vibrio mimicus의 염기서열과 $99\%$의 상동성을 보여 유사 종으로 판명되었다. 그러나 세균 자동동정기를 이용한 동정 결과, genus는 Vibrio 속으로 동정되었으나 species는 일치하는 균종이 없었다. 알려진 Vi-brio속 중에서 유사성이 높았던 V. mimicus와 비교해 볼 때 18종류의 항목이, V. cholerae와는 11종류의 항목이 다른 반응을 나타내어 미분류 균종으로 확인되었다.
JB10과 $JB15^{T}$ 균주들은 양강도 삼지연군의 북서부에 위치한 백두산에 있는 장려 폭포의 폭포수에서 분리되었다. 그람 음성의 호기성 균주로서, 약$0.9-1.6{\times}1.5-2.5\;{\mu}m$의 짧은 간균이었다. JB10, $JB15^{T}$ 균주들과 표준 균주들은 생리 생화학 특성 실험에서 서로 차이점을 보였다. JB10과 $JB15^{T}$ 균주들의 16S rDNA 염기서열을 분식한 결과 ${\gamma}-proteobacteria$에 속하였으며, Acinetobacter tandoii 4N13T (97.3%), Acinetobacter haemolyticus $ATCC17906^{T}$ (97.2%), Acinetobacter johnsonii $DSM6963^{T}$ (97.2%), Acinetobaoter junii $DSM6964^{T}$ (96.7%), Acinetobacter schindleri $LUH5832^{T}$ (97.0%) 및 Acinetobacter ursingii $LUH3792^{T}$ (96.6%)와 높은 유사도치 염기서열 상동성을 보여주었다. 그리고 이 외의 표준 균주들과는 93-96%의 염기서열 상동성을 보였다. 균체 지장산 분석 결과주요 지방산으로 $C_{18:1}\;{\omega}9c$와 $C_{16:1}\;{\omega}7c/C_{15:0}\;iso\;2OH$를 함유하고 있는 것을 확인하였으며, 흥미롭게도 $JB15^{T}$ 균주에서 $C_{19:1}\;iso\;I$이 검출되었다. 이상과 같이 생리 생화학적 특성, 16S rDNA 염기서열 분석 결과 및 균체 지방산분석 결과에서 선별된 JB10과 $JB15^{T}$ 균주들이 표준 균주들과는 다른 특성을 나타내는 것으로 확인되어 JB10 (=KEMC 52-093)과 $JB15^{T}$ ($=KEMC\; 52-094^{T}$) 균주들을 Acinetobacter koreensis sp. nov.로 동정하였다.
