Gametophyte formation and Microspore Germination in Astragalus membranaceus Bunge (황기의 배우체 형성과 소포자 발아 특성)

  • This study was carried out to obtain the basic informations on the characteristics of gametophytes formation and microspore germination in Astragalus membranaceus Bunge. Pollen mother cells passed through meiosis when the flower bud length reaches around 3.5 mm, thus creating the tetrad when it is 4.0 mm long. Pollen attains full growth when the bud is about 10.0 mm long and the anther is found to dehisce when the length of tate bud reach around 12.0 mm. Embryo sac develops at a similar speed as pollen did and it attains its full growth when the bud is about 10-12 mm long. After being stored at 4$^{\circ}C$ or -4$^{\circ}C$, the pollen maintained its germination ability to almost full extent by the 30th day after store. However, the germination rate at room temperature (23~28$^{\circ}C$) decreased below 3% by the 3rd day of storage and so did the germination speed.

Cultivation Technique of Ecklonia cava Kjellman for Restoration of Natural Resources (자연자원 회복을 위한 감태(Ecklonia cava Kjellman)의 인공양식기법)

  • Outdoor cultivation experiment was conducted with artificial seeds using free-living gametophytes and zoospores of Ecklonia cava to develop FLG (free-living gametophyte) seeding technique. Growth of thalli between FLG and zoospore seeding groups was compared monthly from May to October 2009 at culture farm in Wando, Korea. In September 2009, thalli in the FLG seeding experimental group were $35.9{\pm}0.8$ cm in length, $24.1{\pm}2.62$ g in weight, $2.32{\pm}0.26$ kg $m^{-1}$ in biomass and $22.0{\pm}2.8$ individuals $m^{-1}$ in density. There were no significant differences in length, weight, biomass and density between FLG and zoospore seeding group. Therefore, the FLG seeding technique could replace the zoospore seeding method which would eventually contribute in restoration and conservation of natural resources.

  • This study aimed to develop a suitable method for inducing the proliferation of prothallus and producing sporophytes of rock polypody (Polypodium vulgare L.). The prothalli used in all experiments were obtained from spore germination and sub-cultured for 8-week intervals. The most appropriate media for prothallus propagation were investigated by culturing 300 mg of prothallus in MS ($1/4{\times}$, $1/2{\times}$, $1{\times}$, and $2{\times}$ strength) medium and in Knop medium for 8 weeks. Cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $30{\pm}1.0{\mu}mol-m-2{\cdot}s-1$, and a photoperiod of 16/8 h (light/dark). Fresh weight of prothalli was 4.8 g on $1{\times}$ MS, 4.5 g on $1/2{\times}$ MS and 4.3 g on 1/4 MS medium. To select a suitable soil combination for sporophyte formation, 1.0 g of prothallus was ground with distilled water, spread in five combinations onto different soil substrates (decomposed granite, horticultural substrates, peat moss, and perlite), and then cultivated for 13 weeks. The sporophyte cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $43{\pm}2.0{\mu}mol-m-2{\cdot}s-1$, humidity of $84{\pm}1.4%$, and a photoperiod of 16/8 h (light/dark). The results showed that a mixture containing a 2:1 (v:v) ratio of horticultural substrate and perlite, increased sporophyte formation to 462.5 sporophytes per pot (7.5 cm2). The other soil substrates produced from 314.5 to 405.3 sporophytes per pot. Therefore, our results will provide conditions suitable for mass production of Polypodium vulgare L.

  • Ferns have been consumed as food in many countries for centuries. As rich sources of protein, fiber, minerals, vitamins, essential amino acids, and fatty acids, ferns provide important nutrients to humans. Eastern bracken (Pteridium aquilinum var. latiusculum (Desv.) Underw. ex A. Heller) is the most popular edible fern in South Korea where, additionally, it has long been used as an edible wild leaf vegetable. Recently, the production of eastern brackens in South Korea (2018) has reached 14,032 tons, for an annual revenue of 83.5 billion won, and even more eastern brackens are marketed if imports are taken into account as well. Most of the common ferns can be propagated using spores. However, fern farmers cultivate seedlings through traditional propagation methods, such as root pruning or rhizome division. These propagation methods exhibit limitations in forming roots and growing-points and are labor intensive. Quality seedlings of eastern bracken can be obtained through spore propagation, but the spores are fine and difficult to handle in the field. In addition, it would require appropriate environmental control. The production of synthetic seeds using encapsulation technology is easy to establish and it can be used to achieve high productivity at low cost. Synthetic seeds contain explants embedded into a seed foam, and they overcome the limitations of micropropagation and offer the possibility of using plug seedlings. Synthetic seed matrix, such as sodium alginate, has the advantages of low cost, low toxicity, and gel stability. The present study aimed to develop and produce synthetic seeds for the commercial exploitation of eastern bracken. Furthermore, we verified spore germination and the extent of gametophyte and sporophyte development achieved with our new synthetic seeds, whose production was intended to solve current problems with the handling, storage, and transportation of eastern bracken.

  • As one of the fundamental studies for the breeding of marine algae, this paper deals wit_the effects of plant hormone on the growth of microscopic filamentous gametophyte of Undaria pinnatifida. The results obtained are summarized as follows: (1) All zoospores were settled on slide glass, germinated and developed into gametophytes, without the growth of germination tubes when treated with 0.1 mg/l of 2.4-D, or 0.1, 1.0, or 5.0 mg/1 of Keinetin. (2) The best growth effect in total average was observed at 0.1 mg/1 of Keinetin, when the growth-rate was $248.9\%$ in contrast with control, and was followed by 1.0 mg/1 of IAA ($243.3\%$), and 0.05 mg/1 of 2.4-D ($205.6\%$). (3) It was certain that the growth-effect by each plant hormone had some differences between male and female gametophytes. IAA was very effective in the growth of male gameto-phytes but Keinetin in that of females. Especially in females, the efficiency of Keinetin was recorded best at $239.0\%$ at 5.0 mg/l and $222.0\%$ at 0.1 mg/1. On the other hand, it was $195.1\%$ in its best at 0.5 mg/1 of IAA, and $146.6\%$ was recorded best in 2.4-D at 0.05 mg/1.

Spore Germination and Prothallium Development Conditions of Lygodium japonicum (Thunb.) Sw. (실고사리(Lygodium japonicum (Thunb.) Sw.) 포자발아와 전엽체 발달조건)

  • This study was conducted to determine the optimal conditions of growth medium, temperature, and light quality for efficient propagation of Lygodium japonicum spores. The rate of spore germination and prothalium development was high in Knop and 1/8MS and 1/4MS media, which had low mineral content; in particular, the germination rate exceeded 74%, and the germinated spores developed into heart-shaped prothallia. However, in Knop‘s medium with the lowest mineral content, a rapid prothallium senescence was observed; in 1/4MS medium, prothallium development was delayed. Germination rate increased with the increase in temperature and reached its maximum, 86.7%, at 30℃; however, at this temperature, the prothallia were thinner and abnormal development of rhizoids was observed compared to normally developed prothallia and rhizoids at 25℃. Therefore, the results suggested that the optimal temperature for L. japonicum spore germination was 25℃. The rate of germination was also measured under different light conditions, and the highest rate of 90.6% was observed under LED red light compared to fluorescent (77.2%) or LED blue (5.4%) lights. The germinated spores developed into heart-shaped prothallia under LED red light; however, 15 days after seeding, prothallium development decreased and the became elongated. In contrast, a normal and continuous development of heart-shaped prothallia was observed under fluorescent light.

Effect of Medium Components and Culture Methods on Prothallus Propagation of Pteridium aquilinum var. latiusculum (Desv.) Underw. ex Hell. (고사리 전엽체의 증식에 미치는 배지구성물질과 배양방법의 영향)

  • Present studies were conducted to evaluate the effects of medium strength(MS and Hyponex), carbon sources and their concentrations, agar concentrations, and inoculation amounts on prothallus propagation of Pterdium aquilinum var. latiusculum(Desv.) Underw. ex Hell in vitro. The optimum MS medium strength for prothallus propagation was 2MS concentration. Phosphate source was most effective for prothallus growth of P. aquilinum var. latisculum. The addition of 1% sucrose or glucose to MS medium promoted prothallus multiplication. Growth of prothallus was not affected by agar concentration. Propagation of homogenized prothallus was vigorous even in liquid medium. Chopped gametophytes(100 and 200 mg) were inoculated on 250 ml ${\Delta}$flask with 100 mL of 2MS concentration medium and suspension culture was done at 100 rpm for 22 days. After 20 days, prothallus multiplication slowed down, so 100 mg of chopped prothalli is recommended for initial inoculation, since initial amount of inoculum did not affect subsequent prothallus multiplication. Consequently after 20 days of suspension culture, prothallus should be subcultured or transplanted outside of growing vessels.

  • Growth and tetraspore release pattern of lithophyllum yessoense (Rhodophyta, Corallinaceae) were investigated from March 2000 to July 2001. Pinkish tetraspores were 40.2 $\pm$ 0.4 $\mu$m in diameter. After release, tetraspores attached on substrate shortly. Culture conditions were five temperatures (5, 10, 15, 20, 25^{\circ}C), four irradiances (0, 20, 50, 100 $\mu$mol${\cdot}m^{-2}$${\cdot}s^{-1}$) and six salinities (0, 9, 17, 25, 34, 43 ppt). Maximum growth of gametophyte was occurred at $20^{\circ}C$, $20{\mu}mol\;m^{-2}$${\cdot}s^{-1}$, 16: 8h (L:D) and 34 put. Maximum relative growth rate was 0.1232 at $20^{\circ}C$. The amount of tetraspore release showed maximum at September as 266 cells per crust area ($cm^2$), and tetraspores did not release from January to March.

Effect of Several Cultivation Condition on Growth of Brachythecium rivulare and Myuroclada maximoviczii (몇 가지 재배조건이 물가양털이끼와 쥐꼬리이끼의 생육에 미치는 영향)

  • This study was carried out to develop the proper cultivation methods of Brachythecium rivulare and Myuroclada maximowiczii which showed high-value for the interior landscaping and potting. Growth of two moss species cultivated in the compost covered with cloth was vigorous compared to that grown in containers only using cloth or compost, and their harvesting processes were easier. The growth and harvest easiness of mosses cultivated in compost were great rather than in bark or peatmoss. Compared to division, the spray of crushed mosses using mixer was effective for both gametophyte generation and their harvesting processes. In addition, the optimum inoculum for each container ($27{\times}17{\times}3cm$) was 2.0 g in B. rivulare and 4.0 g in M. maximowiczii. Overall growth of B. rivulare treated with nutrient solution (N:P:K=20:20:20) was inhibited compared to control, fresh-weight gain was reduced toward the higher concentration. But fresh-weight gain of M. maximowiczii was the highest with $0.25g{\cdot}L^{-1}$ treatment. Therefore, adequate moisture supply, after spraying crushed mosses (2.0 and 4.0 g each) in the compost covered with cloth, were the appropriate cultivation methods for B. rivulare and M. maximowiczii. Nutrient solution treatment with low concentration, during the cultivation period, would be the proper way only for M. maximowiczii.

Cultural Conditions Affecting Gametophyte Propagation and Sporophyte Formation of Asplenium trichomanes L. (차꼬리고사리의 전엽체 증식 및 포자체 형성에 영향을 미치는 배양조건)

  • 차꼬리고사리(Asplenium trichomanes L.)는 남방계식물로 제주도에 자생하는 것으로 알려져 있으며, 잎에 광택이 있고 총생하는 식물이다. 주로 실내 외 조경 및 분화소재로 이용되며, 한방에서는 철각봉미초라하여 뿌리를 포함한 전초를 이질, 임병, 만성질염, 월경불순 및 요통의 약재로 사용한다. 또한 식물구계학적 특정식물종 IV급, 적색자료목록 준위협 (NT)에 분류된 식물로 보호가 필요하다. 본 연구는 차꼬리고사리의 대량생산을 위한 기내전엽체 증식 및 기외포자체 형성조건을 구명하고자 수행되었다. 실험재료는 포자를 기내 발아시켜 전엽체를 획득한 다음 8주 간격으로 계대하면서 확보하였다. 전엽체의 증식과 생육에 적합한 배지를 비교하고자, 1/4, 1/2, 1, 2MS와 Knop배지를 조성하여 배양하였다. 배양은 전엽체 300mg을 메스로 균일하게 다지는 방법을 이용하였으며, 배양환경은 온도 $25{\pm}1.0^{\circ}C$, 광도 $30{\pm}1.0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 광주기 16/8h(light/dark)로 조절되었다. 실험결과, 1MS배지에 배양된 전엽체의 생체중이 4.3g으로 가장 많이 증가하였다. 형태형성발달도 하트형의 전엽체로 정상적으로 유도되었으며, 생식기관도 관찰되었다. 전엽체로부터 포자체의 형성을 유도하고자, 원예상토, 피트모스, 펄라이트 및 마사토의 비율을 5종류로 달리하여 혼합된 토양을 사각분($7.5{\times}7.5{\times}7.5cm$)에 충진하였다. 전엽체 1g과 증류수 25mL를 핸드블랜더로 10초간 분쇄하여 토양표면에 분주하는 실험방법을 사용하였다. 재배환경은 온도 $25{\pm}1.0^{\circ}C$, 광도 $43{\pm}2.0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 광주기 16/8h(light/dark)로 유지하면서 12주간 재배되었다. 실험결과, 원예상토 단용, 원예상토와 펄라이트가 2:1(v:v)로 혼합된 토양, 원예상토와 마사토가 2:1(v:v)로 혼합된 토양에서 각 31.7, 24.3, 19.3개의 포자체가 생산되었다. 한편 포자체의 생육은 원예상토 단용 토양에서 엽수, 엽장, 엽폭 등의 수치가 비교적 우수하였다. 따라서 차꼬리고사리의 전엽체는 MS배지에 배양하고 증식된 전엽체를 원예상토에 분주하여 포자체의 형성을 유도하는 것이 가장 효과적이었다.
