폴리페놀 및 DPPH 라디칼 소거활성 관점에서의 복합갈근탕 및 단미혼합갈근탕의 동등성 (Equivalence of Mixed and Individual Preparation of Galgeun-tang in terms of Polyphenol Contents and DPPH Radical Scavenging Activity)

  • Traditional medicines usually use combinational formula that are prepared by mixing different varieties of medicinal herbs and boiling them in water to yield a decoction. In recent years a modified method has been proposed and practiced wherein the individual herbs are boiled with water separately and later these extracts are mixed together for use. We attempted to evaluate their equivalence in terms of its polyphenol contents and DPPH radical scavenging activity. The polyphenol contents as well as the DPPH radical scavenging activities were very similar to each other.

한약 처방 25종에 대한 간 보호 효과 비교 연구 (Hepatoprotective Effects of 25 Herbal Formulas in Primary Rat Hepatocytes)

  • The purpose of this study is to investigate the protective effects of 25 herbal formulas on acetaminophen (APAP) or D-galactosamine (D-GalN)-induced hepatotoxicity in primary rat hepatocytes. Cell viability was measured using by Cell Counting Kit-8. 15 kinds of herbal formulas significantly reversed the cell viabilities of D-GalN-treated rat hepatocytes compared with D-GalN alone (p<0.05). In particular, 9 herbal formulas (Bangpungtongseong-san, Bojungikgi-tang, Galgeun-tang, Gumiganghwal-tang, Guibi-tang, Sagunja-tang, Samsoeum, Pyeongwi-san and Yijin-tang) showed the potent protective effects. However, 8 herbal formula exerted weak protective effects and 2 herbal formula did not exert effects on hepatotoxicity by D-GalN. On APAP-induced hepatotoxicity, 7 kinds of herbal formulas increased the viabilities of hepatocytes compare with APAP alone (p<0.05). These results could be provide a valuable information for the future in vivo or clinical studies to predict the hepatoprotective effects of herbal formulas.

전통 한의약 처방 10종에 대한 혈관이완 효능 연구 (Vasorelaxant effects of 10 traditional Korean Herbal Prescriptions on isolated rat aortic rings)

  • Objectives : The objective of present study was to investigate the vasorelaxant effects of 10 traditional Korean Herbal Prescriptions (KHP) on isolated rat thoracic aorta precontracted with potassium chloride (KCl). Methods : An electric extractor was used to extract KHP in distilled water for 3h. Rat aorta rings were isolated and were precontracted using KCl in organ chambers containing 10 ml Krebs Henseleit (KH) buffer. KHP extracts were added in increasing concentrations (10-1000 ㎍/㎖) to investigate vasorelaxant effects. The vasorelaxant responses induced by KHP were expressed as a percentage in response to contraction generated by KCl. Results : Among the 10 KHP, Gamisoyo-san, Galgeun-tang, Gyeji-tang, Gwakhyangjeonggi-san, Daeyoung-jeon, and Socheongryong-tang showed significant vasorelaxant effect at high concentration. In contrast, Gyejibokryeong-hwan constricted more the aorta rings precontracted by KCl. And Gumiganghwal-tang, Guibi-tang, Saengmaek-san showed no significant effect. Also, rat aorta rings treated with Gyejibokryeong-hwan or Gyeji-tang after pre-relaxation by amlodipine did not cause any significant change. Conclusion : Thus, these results provide the experimental evidence as useful herbal prescriptions for the treatment of hypertension and suggest guidelines in conjunction with other western drugs, including amlodipine.

방풍갈근탕(防風葛根湯)의 항알레르기효과에 관한 연구 (Study on Anti-allergic Effect and Safety of Bangpung-galgeun-tang)

  • The purpose of this study was to examine the anti allergic effect in vivo and in vitro, and to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated anti DNP IgE-mediated passive cutaneous anaphylaxis in rodents and compound 48/80-induced active systemic anaphylatic shock in mice after oral administration with BGT of 0.4 g/kg and 0.8 g/kg for 8 days, and also examined MTT assay, ${\beta}-hexosaminidase$ activity, IL-4 and $TNF-{\alpha}$ from RBL-2H3 and $TNF-{\alpha}$ from Raw264.7 after pre-treatment with BGT of 0.25 mg/ml, 0.5 mg/ml, 1 mg/ml and 2 mg/ml. To ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg /p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. BGT inhibited passive cutaneous anaphylaxis and active systemic anaphylatic shock by oral administration. All the concentrations of BGT from 0.25 to 2 mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosaminidase$ release, IL-4 and $TNF-{\alpha}$, and in Raw264.7, $TNF-{\alpha}$ were significantly reduced by treated all concentrations of BGT. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.

병원성 미생물에 대한 다빈도 51종 한약처방의 항균 활성 연구 (Antimicrobial Activities of 51 Herbal Formulae on Pathogenic Microorganisms)

  • Objectives: The purpose of this study was to establish experimental evidence for the antimicrobial effects of 51 herbal formulae commonly used in traditional Korean medical institutions. Methods: The antimicrobial activities of herbal formulae were screened using the disc diffusion method against 10 pathogenic microorganisms (Bacillus cereus, Staphylococcus aureus, Listeria monocytogenes, Vibrio parahaemolyticus, Escherichia coli DH5α, E. coli O157, Salmonella enteritidis, Yersinia enterocolitica, Shigella flexneri, and Helicobacter pylori). Of the 51 herbal formulae, 13 herbal formulae with antimicrobial activity were selected and their dose-dependency were confirmed. Results: Nine herbal formulae, including Gyeji-tang, Dangguisu-san, Saengmaek-san, Samul-tang, Ssanghwa-tang, Socheongryong-tang, Yukmijihwang-tang, Jakyakgamcho-tang, and Paljung-san, presented antibacterial activity against B. cereus. The effects of Saengmaek-san and Paljung-san was sustained for 48 hr. On L. monocytogenes, Dangguisu-san and Hyangsapyeongwi-san showed antimicrobial activity, but only Hyangsapyeongwi-san maintained the activity for 48 hr. Thirteen herbal formulae such as Galgeun-tang, Gyeji-tang, Dangguisu-san, Mahwang-tang, Banhasasim-tang, Saengmaek-san, Socheongryong-tang, Yukmijihwang-tang, Jakyakgamcho-tang, Cheonwangbosim-dan, Palmijihwang-tang, Paljung-san, and Hwanglyeonhaedok-tang showed antimicrobial activity against V. parahaemolyticus, and the activity was maintained for 48 hr. The 51 herbal formulae did not show any antimicrobial activity against seven strains such as E. coli DH5α, E. coli O157, S. aureus, S. enteritidis, Y. enterocolitica, S. flexneri, and H. pylori. Conclusions: Nine, two, and thirteen herbal formulae showed antimicrobial activities against B. cereus, L. monocytogenes, and V. parahaemolyticus in a dose-dependent manner, respectively. The results of antimicrobial activity of 51 herbal formulae against 10 microorganisms might be used as the basis for new application of herbal formulae.

전탕 전과 후의 중금속, 잔류농약 및 잔류이산화황의 농도변화 - 감기약을 중심으로 - (Concentration of Heavy Metals, Residual Pesticides and Sulfur Dioxide of before/after a Decoction)

  • Objectives: To compare the contents of heavy metals, residual pesticides and sulfur dioxide before/after a decoction. Methods: The heavy metal contents before/after a decoction were measured by Inductively Coupled Plasma Atomic Emission Spectrometer(ICP-AES) and mercury analyzer. In order to analyze pesticides in 5 samples we used simultaneous multi-residue analysis of pesticides by GC/ECD, which was followed by GC/MSD analysis to confirm the identity of the detected pesticide in each sample. In addition, the contents of sulfur dioxide($SO_2$) were performed by Monier-Williams distillation method. Results: 1. The mean values of heavy metal contents(mg/kg) for the samples were as follows: Galgeun-tang(before decoction-Pb; 0.793, Cd; 0.133, As; 0.016 and Hg; 0.005, after decoction-Pb; 0.033, Cd; 0.004, As; 0.002 and Hg; not detected), Gumiganghwal-tang(before decoction-Pb; 0.934, Cd; 0.197, As; 0.046 and Hg; 0.006, after decoction-Pb; 0.062, Cd; 0.007, As; 0.004 and Hg; 0.0001), Sosiho-tang(before decoction-Pb; 0.891, Cd; 0.134, As; 0.091 and Hg; 0.014, after decoction-Pb; 0.036, Cd; 0.002, As; 0.004 and Hg; not detected), Ojuck-san(before decoction-Pb; 0.907, Cd; 0.136, As; 0.084 and Hg; 0.007, after decoction-Pb; 0.074, Cd; 0.007, As; 0.011 and Hg; 0.0005) and Samsoeum(before decoction-Pb; 1.234, Cd; 0.154, As; 0.016 and Hg; 0.007, after decoction-Pb; 0.094, Cd; 0.006, As; 0.002 and Hg; 0.001). 2. Contents(mg/kg) of residual pesticides before/after a decoction in all samples were not detected. 3. Contents(mg/kg) of sulfur dioxide($SO_2$) before a decoction in Galgeun-tang, Gumiganghwal-tang, Sosiho-tang, Ojuck-san and Samsoeum exhibited 1.2, 3.4, 11.1, 12.0 and 5.7, respectively. However, contents of sulfur dioxide after a decoction in all samples were not detected. Conclusions: These results will be used to establish a criterion of heavy metals, residual pesticides and sulfur dioxide.

태음인(太陰人)의 표열증설사(表熱證泄瀉)에 관한 문헌연구(文獻硏究) (A Study on Tae-eumin Exterior Heat Syndrome Diarrhea)

  • 1. Backgrounds: There are a lack of comprehensive study on Taeeumin Exterior Heat syndrome diarrhea(表熱證泄瀉). Thus, The author examines the content of various literatures related to Tae-eumin Exterior Heat syndrome diarrhea. 2. Methods: Literature examination is used Lee, Je-ma's writings and Sasang Constitutional Medical doctor's writings. Syndrome examination is used Tae-eumin's study on Syndrome of DonguisusebowonGaboGu-bon("咸山沙村東醫壽世保元甲午舊本") and Donguisusebowon("東醫壽世保元") and treatises of the academic world. 3. Results: and Conclusions: 1) "Profuse sweating and reddish and difficult urination"(汗多而小便赤澁) is regard as Taeeumin Interior Febrile Disease(裏熱病). 2) There is no possibility of a misprint about Taeeumin Exterior Heat syndrome diarrhea. Taeeumin Exterior Heat syndrome diarrhea is engaged in both the Exterior and Interior(表裏俱病). 3) It is difficult to prescribe GalgeunHaegi-Tang(葛根解肌湯) instead of Galgeunnabokja-Tang(葛根蘿葍子湯). 4) Tae-eumin Exterior Heat syndrome diarrhea is formed in the early period and absorbed into Donguisusebowon ("東醫壽世保元") in the latter period. "Profuse sweating and reddish and difficult urination" is estimated to be added in the latter period.

교통사고 환자에 대한 상한론 처방의 유의성 연구 (A Study on the Significance of Sanghanron Prescription in Traffic Accident Patient)

  • Objectives : The aim of this study is to make proof of availability for Sanghanron prescription. Methods : The study group comprised 80 Koreans, male:female ratio of 31:49, with mean age of 40.78 years. The SF-MPQ and PDI were checked hospitalized and discharged patients. Results : 1. The most uesd prescription is Galgeun-Tang(Gegen-tang) and the other 30 prescriptions were uesd. 2. Hospitalized patients, there was significant difference in SF-MPQ and PDI scores(P<0.05). 3. The treatment was effective to 72 patients out of 80 who had been hospitalized for less than 14 days. 4. Experimental group compared with Lee's thesis was significantly difference in 7 days of treatment, but the other compared were not significant difference. Conclusions : This data suggested that the Sanghanron prescription could be effective treatment about traffic accidental patients without surgical injury. Further study is needed about the effectiveness of Sanghanron prescription.

고방(古方)으로 치료한 두드러기 환자 39례의 임상보고(臨床報告) (A Clinical Report on 39 Urticaria Patients Treated With The Ko-Bang(古方))

  • Objectives : This study was designed to investigate the chronical features of urticaria and the effects of urticaria treatment with Ko-Bang(古方). Methods : We treated 39 patients for urticaria with Ko-Bang(古方), who visited to care urticaria at the Dept. of Dermatology Oriental medical hospital Dong-eui university from Jan, 2008 to Sep, 2009. This study was assessed using the chart analysis and the follow-up survey by telephone. Results & Conclusions : 1. 39 outpatients were surveyed, who were 20 males and 19 females. And patients who were ages 21-30 were the largest group, 11(28.21%). Among the 39 patients, acute urticaria patients were 8(20.5%), chronic urticaria patients were 31(79.5%), and contraction of a disease period between 6 weeks with 3 months was 17.9%. 2. The causes of urticaria were measured unknown 37.5%, foods 37.5%, drugs 25% in acute and unknown 35.5%, a change of temperature 29%, foods 16% in chronic. And suspected provocation factors were measured unknown and foods each 12 cases. The most of accompanied symptoms was itching sign(34 cases), the second most was self-conscious fever(10 cases). 3. Out of treat prescription(Ko-Bang,古方), Injinho-Tang(茵蔯蒿湯) was used most in 5 cases, Daehwanghwangryunsasim-Tang(大黃黃連瀉心湯), Hwangryun-Tang(黃連湯), Daesiho-Tang(大柴胡湯) were used in each 4 cases, Galgeun-Tang(葛根湯), Chijadaehwangsi-Tang(梔子大黃豉湯), Sosiho-Tang(小柴胡湯) were used in each 3 cases, Gaemagakban-Tang(桂麻各半湯), Chijasi-Tang(梔子豉湯),Oryeongsan(五笭散) were used in each 2 cases. 4. When the processes of treatment were classified by one poison(一毒), there were the 19 cases (48.7%) which was caused by Bun(煩), the 12 cases(30.8%) by Water(水) and the 4 cases(10.3%) by Gyur(結).

Effects of Korean traditional herbal formula for common cold on the activities of human CYP450 isozymes

  • Objectives: Most drug interactions are attributed to the inhibition or induction of the activity of cytochrome P450s (CYP450). Although the regulation of CYP450s by drugs has been widely reported, there have been few studies on influence of traditional herbal formulas on the drug-metabolizing enzymes. Because herbal formulas have been used traditionally to treat various diseases and because herb-drug interactions are crucial factors determining therapeutic efficacies, a systematic evaluation of the effects of herbal formulas is important. Methods: The effects of Galgeun-tang (GGT, gegen tang), Gumiganghwal-tang (GMGHT, jiuweiqianghuo tang), Insampaedok-san (ISPDS, renshenbaidu powder), Samsoeum (SSE, shensu drink), Socheongryong-tang (SCRT, xiaoqinglong-tang) and Sosiho-tang (SSHT, xiaochaihu tang) that are traditional herbal formulas used to treat common cold, on drug-metabolizing enzymes were evaluated through an in vitro CYP3A4, CYP2D6, CYP2C19 and CYP2E1 inhibition assay to assess its interaction potential with synthetic drugs. The inhibitory effects of herbal formulas were characterized with $IC_{50}$ values. Results: These six herbal formulas inhibited the activities of CYP3A4, 2C19, 2D6 and 2E1, in a concentration-dependent manner. Among the six herbal formulas, GGT critically inhibited CYP2C19, CYP2D6 and CYP2E1. GMGHT also inhibited CYP2D6 and CYP2E1 to a greater extent than the other CYP450 isozymes. Additionally, SSE and SSHT may change the effects of medicines that depend primarily on the CYP2C19 and CYP2E1 pathways. On the other hand, ISPDS and SCRT were not inhibited CYP3A4, CYP2C19, CYP2D6 and CYP2E1-mediated metabolism. Conclusions: These findings provide useful information regarding the safety and effectiveness of herbal formulas.
