Simultaneous Determination of Albiflorin, Cinnamaldehyde, Cinnamic Acid, Daidzin, Glycyrrhizin, Liquiritin, Paeoniflorin and Puerarin in Galgeun-tang by HPLC-PDA

  • Objectives: We investigated to develop and validate HPLC-PDA methods for simultaneous determination of eight constituents in Galgeun-tang (GGT). Methods: Reverse-phase chromatography using a Gemini C18 column operating at $40^{\circ}C$, and photodiode array (PDA) detection at 230 nm, 254 nm, and 280 nm, were used for quantification of the eight marker components of GGT. The mobile phase using a gradient flow consisted of two solvent systems. Solvent A was 1.0% (v/v) aqueous acetic acid and solvent B was acetonitrile with 1.0% (v/v) acetic acid. Results: Calibration curves were acquired with $r^2$ > 0.9999, and the relative standard deviation (RSD) values (%) for intra- and inter-day precision were less than 3.0%. The recovery rate of each component was in the range of 87.33-101.38%, with an RSD less than 7.0%. The contents of the eight components in GGT were 1.98-12.17 mg/g. Conclusions: The established method will be applied for the quantification of marker components in GGT.

Simultaneous Determination of Four Bioactive Constituents in Galgeun Tang by HPLC/DAD. (HPLC-DAD를 이용한 갈근탕 중 4종 성분의 동시분석법 확립)

  • For the quality control of traditional herbal medicine, Galgeun tang, simultaneous determination of glycyrrhizin, paeoniflorin, puerarin, 6-gingerol was established by using a high performance liquid chromatographic (HPLC) method with diode array detector. To separate five four constituents, DIONEX $C_{18}$ column ($5{\mu}m$, $120{\AA}$, $4.6\;mm{\times}150\;mm$) was used with gradient elution system of water and methanol. Validation of the chromatography method was evaluated by linearity, recovery, and precision test. Calibration curve of standard components showed excellent linearity ($R^2$>0.9906). Limits of detection (LOD) and limits of quantification (LOQ) varied from 0.15 to $0.52{\mu}g$/ml and 0.27 to $0.80{\mu}mg$/ml, respectively. The relative standard deviations (RSDs) of data of the intra-day and inter-day experiments were less than 2.88% and 1.21%, respectively. The results of recovery test were ranged from 96.71 to 106.29% with RSD values 0.01-0.80%.

Influence of Galgeun-tang on Gene Expression in Diet-Induced Obese Mice (갈근탕이 고지방 식이 유발 비만 쥐에서 비만 관련 유전자 발현에 미치는 영향)

  • Objectives The purpose of this study was to evaluate anti-obesity effect of Galgeuntang (gegentang) and elucidate the effect of it on gene expression related to obesity. Methods The experiments were performed with the use of Diet-Induced Obese mice. They were grouped NC (normal control), HFD (high fat diet control), GGT (Galgeun-tang (gegentang), 700 mg/kg), ORL (Orlistat, 10 mg/kg). GGT was orally administered for 12 weeks. Body weight was measured every week. Real-time PCR was performed to investigate the effect of GGT on gene expression in liver tissue. Results GGT group and ORL group were reduced in body weight compared with HFD. HFD increased $PPAR{\gamma}$, SREBP-1, Leptin, aP2, FATP1, FAS gene expression compared with NC. GGT increased FATP1 gene expression. But GGT reduced $PPAR{\gamma}$ & FAS gene expression in liver tissue of diet-induced obese mice compared with HFD. Conclusions These results suggest that GGT is supposed to have a certain impact on the treatment of obesity. But more study is needed in the future.

The Study on The Origin, Changes, and Composition of Galgeulhaegi-tang (갈근해기탕(葛根解肌湯)의 기원(基源), 변천(變遷), 구성(構成)에 대한 고찰(考察))

  • 1. Objectives : This paper was written in order to understand the origin and changes of Galgeunhaegi-tang. 2. Methods : We analysis Galgeunhaegi-tang through pathology and new prescription in "Donguibogam(東醫寶鑑)", "Chobongwon(草本卷)", "Gabobon(甲午本)" and "Sinchukbon(辛丑本)" of "Dongyi Suse Bowon". 3. Results and Conclusions : We can find the first origin of Galgeunhaegi-tang in "Sinchukbon(辛丑本)" in "Donguibogam(東醫寶鑑)" which invelves Galgeunhaegi-tang in the categories of pestilence(瘟疫) and Yangmyeonggyeongbyeong(陽明經病). Also, we can find the origin in Jugoeng(朱肱)' Jojung-tang. Seungeumgalgeun-tang in the "Chobongwon(草本卷)" developed by a Galgeunhaegi-tang of "Gabobon(甲午本)", and Galgeunnabokja-tang in the "Chobongwon(草本卷)" developed by a Galgeunhaegi-tang of "Sinchukbon(辛丑本)". It is considered that Galgeunhaegi-tang of "Gabobon(甲午本)" solves heat depression(鬱熱), due to not throwing off, by using Galgeun(葛根) and Daehwang(大黃), and accompanied dryness(燥) by using Hangin(杏仁) and Sanjoin(酸棗仁) which moisten dryness(燥). It is considered that Galgeunhaegi-tang of "Sinchukbon(辛丑本)" solves heat depression(鬱熱) by using Galgeun(葛根) and accompanied dryness(燥) by using Gobon(藁本) which sets free dryness(燥).

Galgeun-tang, an Herbal Formula, Ameliorates Atopic Dermatitis Responses in Dust Mite Extract-treated NC/Nga Mice

  • Objectives: Galgeun-tang (GGT, gegen-tang, kakkon-to), an herbal formula, is used to treat the common cold, fevers, headaches, hangovers and neck and upper back stiffness. The drugs currently used to treat atopic dermatitis (AD) are limited by the significant adverse effects associated with their long-term usage. The need to efficiently manage the AD response while reducing side effects has led to the development of alternative remedies. Methods: To assess the effects of GGT on AD, the anti-inflammatory and anti-AD properties of GGT were evaluated in both in vitro and in vivo systems. Results: Nitric oxide (NO) and histamine production on lipopolysaccharide (LPS)-treated RAW264.7 cells and phorbol-12 myristate 13-acetate (PMA)/A23187-treated MC/9 cells, respectively, were inhibited by GGT. GGT reduced thymus and activation-regulated chemokine (TARC/CCL17) release on TNF-${\alpha}$/IFN-${\gamma}$ stimulated HaCaT cells in a dose-dependent manner. GGT reduced both plasma levels of IgE and histamine and the dermatitis score in house dust mite induced atopic dermatitis-like lesions on NC/Nga mice. However, there were no significant histopathological differences observed between the GGT group and the AD-induced group, such as AD-like lesions in the dorsal skin or ear or mast cell infiltration in the dorsal skin. Conclusions: These results indicate that GGT inhibits chemokine production by keratinocytes and the atopic dermatitis response in NC/Nga mice, suggesting that GGT may be useful as a therapeutic remedy for treating AD and allergic inflammation-related diseases.

Comparison of Ingredient and Efficacy of Galgeun-tang (Gegen-tang) Mix Extract Powder and Decoction (갈근탕 혼합단미엑스제 및 전탕액의 성분 및 생리활성 비교 연구)

  • Galgeun-tang (GGT) is a traditional herbal medicine generally used to treat cold, fever, headache, migraine and convulsion. To investigate the difference of ingredient and efficacy of GGT according to production method, mix extract powder and decoction, the quantities of index components, antioxidant and anti-inflammatory effects of two kinds of GGT formulation were compared. The contents of ten index components were simultaneously analyzed with HPLC. A HPLC method was developed and validated for the simultaneous determination. The GGT mix extract powder contained higher puerarin and daidzin content than the decoction, while the GGT decoction contained higher cinnamic acid and glycyrrhizic acid content than the mix extract powder. But ingredient of GGT mix extract powder and decoction was statistically very similar (r=0.8335). GGT decoction showed higher antioxidant effect than GGT mix extract powder, but the difference was low. The patterns of productions for NO, iNOS, COX-2, TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 were decreased along the increasing dose of GGT formulation in LPS-induced RAW 264.7 macrophages. The inhibitory effects on NO, iNOS, COX-2, IL-$1{\beta}$ and IL-6 in Raw 264.7 cell were very similar to each other. The results obtained in this study could be used as fundamental data for verifying the equivalence and effectiveness of the herbal medicines.

Protective Effect of Galgeun-Tang Against $CCl_4$ Induced Hepatotoxicity (갈근탕의 사염화탄소에 의한 간세포 독성 억제효과)

  • Galgeun-tang (GGT) has been a great source for treating cold diseases in the folk medicine recipe. Carbon tetrachloride ($CCl_4$) is one type of hepatotoxin that can eventually cause liver injury. During the experiment, we first studied the protective effects of GGT against $CC_4$-induced hepatotoxicity. GGT was pretreated for 3 h, and 1% $CCl_4$ was added to mouse primary liver cells. After 4 h, ROS generation and expression of antioxidant enzymes (catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx)) were analyzed by FACS and real time PCR. Also, the activities of ALT and LDH were measured using cultured medium. The hepatic levels of TNF-alpha and iNOS, which are related to inflammation and stress response gene, HSP72 and HO-1 were analyzed by PCR or real time PCR. Liver tissues were analyzed by HE stain. From the observation, we discovered that GGT treatment protects $CCl_4$-induced hepatotoxicity, and that GGT pretreatment decreases ROS generation, TNF-alpha and iNOS expression. However, gene expression of CAT, SOD, GPx, HSP72 and HO-1 were increased by GGT. These results lead to the conclusion that GGT has protective effects against $CCl_4$-induced hepatotoxicity.

Study on Safety of Bangpung-galgeun-tang (방풍갈근탕(防風葛根湯)의 안전성에 관한 연구)

  • The purpose of this study was to examine to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated to ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg/p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.

The Study on Clinical Characteristics of Taeeumin Dry-Heat Symptomatic Patten (태음인(太陰人) 조열병증(燥熱病證)의 임상적 특징에 대한 연구(硏究))

  • 1. Objective The object of this study was to understand the clinical characteristics of the Taeeumin Symptomatic Pattem. 2. Methods The patients visiting the department of Sasang Constitution of Traditional Korean Medicine clinic from June to November of 2008 were used in this study. Those who showed a favorable turn after over ten days of medication and who showed up for at least two follow-up sessions were chosen among first-time visitors. Of these, 75 patients identified and verified to be of the TE type were chosen for the study sample. The Taeeumin were verified into two groups according to clinical symptoms into Exterior Cold Disease and Interior Heat Disease groups, of which the Interior Heat Disease group was further subdivided into the Galgeun-medicament group and the Cheongshimyeonja-tang group, based on questionnaire results. The Exterior Cold Disease group was set as the control group in order to contrast and compare it with the study group, the Interior Heat Disease group. 3. Results and Conclusions The questionnaire items shown to differentiate the Exterior Cold Symptomatic Patterm and the Interior Heat Symptomaic Pattern (indicating Heat-affceted Liver Symptomatic Pattern) were related to "dryness of stool," "dark urine colorm," "dryness of nose," and "dryness of lips." The items shown to differentiate the Galgeun-medicament group and the Cheongshimyeonja-tang group (of the Dry - Heat Symptomatic Pattern) were related to "nausea," "stifling sensations and palptation," "disturbed sleep," and "dizziness." Therefore, the sleep pattern, palpitation, dizziness, and nausea can be said to be indicators of the Interior Heat Symptomatic Pattern, and Dry-Heat symptoms are better distinctions between the Chongshimyeonja-tang group and the Galgeun-medicament group than Heat-affected Liver symptoms.

The Study on the Formation of Pathology to which Jogakdaehwang-tang is Applicable and the Change of its Composition (조각대황탕(皂角大黃湯) 적용 병증의 형성과 처방구성의 변천에 관한 고찰)

  • Objectives The purpose of this study is to understand the formation of Jogakdaehwang-tang(JD-T)'s patholoy and the compositional change of it in Sasang Constitutional Medicine(SCM). Methods The pathologic condition in which JD-T was used and the change of its composition was studied in SCM Results and Conclusions 1. Jogak and Daehwang in "Dongmuyoogo Haedong" were presented with Ungdam because of the common purpose flowing the energy of lung. Therapeutic method for exogenous Ungibyeong in "Dongyisusebowon sangchobongwon" was subdivided into Hangwul's treated by Ungdam, and the heat pattern featuring contagious cold and constipation, treated by Jogakhwanggeumdaehwang-tang(JHD-T). 2. JD-T in "Dongyisusebowon Gabogubon" was used for Yangdok and Onbyeong which are exogenous disease causing by heat. The composition of JD-T has been completed as it included Mahwang, Gilgyeong, and Seungma derived from prescriptions relevant with Yangdok. 3. JD-T in "Dongyisusebowon Sinchukbon(Sinchukbon)" was included in pathology of Ganyeol, and it substituted Galgeunhaegi-tang(GH-T) for Onbyeong, not for Yangdok, so Galgeun and Seungma became the main drug in JD-T. Galgeunseung-gitang(GS-T) was created for treating heat pattern owing to Daehwang, its main drug, and moreover Jogak's effect is too strong. Therefore, Mahwang, Hwanggeum and Gilgyeong was excluded from the composition of JD-T. 4. The origins of JD-T could be Yiseonggugo-hwan, Daesiho-tang, Jowiseungchung-tang, Heukno-hwan, and GH-T. The change of JD-T went for them throughout JHD-T to JD-T. JD-T in "Sinchukbon" consists of Galgeun and Daehwang which directly gets rid of severe heat from small intestine, and Jogak and Seungma which help to eliminate mild heat existing from small intestine throughout nose to flesh.
