녹각이 Galactosamine에 의해 유도된 간장해에 미치는 영향 (Effect of Old Antler Extracts on the Galactosamine-induced Hepatotoxicity in Rats)

  • 간장해에 미치는 녹각추출물(water-ext., neutral ext. 및 ether-ext.)의 효과를 구명할 목적으로 녹각을 추출 분리하여 급여하고 매주 일정시각에 galactosa-mine (400mg/kg of body weight)으로 간장해를 유도한 후 녹각추출물이 간기능 상태의 지표인 효소활성에 미치는 영향을 생화학적 측면에서 비교.고찰하였다 체중증가량, 식이섭취량 및 식이효율은 Control군에 비해 galactosamine 투여군에서 감소를 보였고, 각각의 녹각추출물 급여로 증가하는 경향을 보였다. 체중100g당 장기무게는 간장의 경우 Balactosamine 투여로 현저한 증가를 보였으나 각각의 녹각추출물 급여가 이들의 수치를 감소시켰고, 신장, 비장 및 심장에는 별다른 차이가 없었다. 혈청 aminotransferase 활성은 galactosamine투여로 인해 증가하였으며, 각각의 녹각추출물중 water-ext.가 galactosamine투여에 의한 효소 활성도의 변화를 현저히 완화시켰다. 혈청 LDH와 ALP의 활성은 galactosamine투여로 증가하였고, 녹각추출물을 병행 투여한 군에서 Water-CaIN군의 활성이 Control군 수준에 가장 가깝게 감소하였다.

  • 이간분수산(利肝分水散)의 galactosamine에 의해 야기된 백서의 간손상에 대한 해독능을 측정하기 위하여 이간분수산(利肝分水散)을 전처지한 후 시험관내에서 과산화지질의 생성 및 간장의 지질과산화물의 함량 및 간장에서 항산화방지제(SOD)의 활성에 미치는 영향, 시험관내에서 xanthine oxidase의 활성변화 및 간장조직의 xanthine oxidase의 형전환 변화를 측정하여 유의한 결과를 나타내었는데 이는 이간분수산(利肝分水散)의 이요작용(利尿作用)을 통한 독성물질의 배설을 촉진시킴으로서 세포나 조직의 손상 및 생체독성에 대하여 유의한 방어효능을 나타내는 것으로 이해할 수 있다.

  • 경골어류 4종(조피볼락, 용치놀래기, 송곳니베도라치 및 졸복) 장관의 복합당질 성상의 차이를 밝히기 위해 장을 네 부위로 나누워 아홉 종류의 biotinylated lectin들(DBA, SBA, PNA, BSL- I , RCA- I , sWGA, UEA- I 및 Con A)로 반응시켰다. 조피볼락을 제외하면 부위에 따른 차이는 없었다. 용치놀래기 장 상피는 모든 부위에서 D-glucose/mannose 잔기를 함유하고 있었다. 조피볼락, 송곳니베도라치 및 졸복 장 모든 부위에서는 정도의 차이는 있지만 ${\beta}$-N-acetyl-D-gal-actosamine가 함유되어 있었다. 또한 조피볼락의 근위장, 중간장 및 원위장에는 galactose-${\beta}$-1,3-N-acetyl-D-galactosamine 및 ${\alpha}$-D-galactose도 함유되어 있었으나 직장에는 이들 당 잔기들이 함유되어 있지 않았다. 송곳니베도라치에는 ${\beta}$-N- acetyl-D-galactosamine 외에 ${\beta}$-N-acetyl-D-glucosamine 및 D-glucose/mannose도 발견되었으며, 졸복에서는 Galactose-${\beta}$-1,3-N-acetyl-D-galactosamine, D-galactose 및 D-glucose/mannose도 발견되었다.

리포폴리사카라이드와 갈락토사민의 투여로 인한 생쥐 치사율에 미치는 글리시리진의 억제효과 (Inhibitory Effect of Glycyrrhizin on the Lethality Induced by Lippolysaccharide and Galactosamine)

  • This study was done to investigate the effect of glycyrrhizin on the lethality induced by galactosamine and lipopolysaccharide coadministration in mice. Glycyrrhizin was injecte d intravenously as a multiple dose at 20, 15, 10, 5, and O hr before galactosamine and lipopolysaccharide coadministration. Lethality and tumor necrosis factor (TNF${\alpha}$) level in serum were surveyed as markers of glycyrrhizin effect. Glycyrrhizin had no effect on the lethality induced by galactosamine and lipopolysaccharide when glycyrrhizin was administered as a single dose. Glycyrrhizin reduced the lethality induced by galactosamine and LPS in dose-dependent manner when glycyrrhizin was administered as a multiple dose at 20, 15, 10, 5 and O hr before galactosamine and lipopolysaccharide coadministration. Glycyrrhizin reduced the serum TNF${\alpha$ level.

삼두해정탕가미방이 $CCl_4$ 및 d-Galactosamine에 依하여 誘發된 흰쥐의 肝損傷에 미치는 影響 (The effects of Samduhaejungtang-gamibang on liver injury of rats induced by $CCl_4$ and d-Galactosamine)

  • In order to investigate the protective effect of Samduhaejungtang-gamibang on the liver injury of rats induced by $CCl_4$ and d-galactosamine, the serum transaminase(GOT& GPT) alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities and triglyceride, total bilirubin amounts for serum component were measured. All animals were divided into 4 groups, those were normal group(untreated), control group(treated with vehicle 0.9% Saline solution), sample Ⅰ group(1500mg/kg administrated), sample Ⅱ group(3000mg/kg administrated). The results were as follows: 1. The inhibitory effects of the serum GOT activities in rats induced by $CCl_4$ were noted in sample Ⅱ group(p<0.01). In serum GPT activities, sample Ⅱ group(p<0.05) only showed the inhibitory effects. 2. The inhibitory effects of the serum ALP activities in rats induced by $CCl_4$ were noted in sample Ⅱ group(p<0.01) 3. The inhibitory effects of the serum LPH activities in rats induced by $CCl_4$ were noted in sample Ⅱ group, but it is not recognized statistically. 4. The increases effects of the serum triglyceride content level in rats induced by $CCl_4$ were inhibited in both sample Ⅰ group(p<0.05) and sample Ⅱ group(p<0.01) 5. The increases effects of the serum total bilirubin content level in rats induced by $CCl_4$ were inhibited in sample Ⅱ group(p<0.05) 6. The inhibitory effects of the serum GOT, GPT activities in rats induced by d-galactosamine were noted in sample Ⅱ group (p<0.001), but sample Ⅰ group was not recognized. 7. The signiticantly inhibitory effects of in the serum LDH activities in rats induced by d-galactosamine were noted in both sample Ⅰ group(p<0.05) and sample Ⅱ group (p<0.001) 8. The increases of the serum ALP content level in rats induced by d-galactosamine were inhibited in sample Ⅱ group(p<0.05) 9. The increases of the serum total bilirubin content level in rats induced d-galactosamine were inhibited in sample Ⅱgroup(p<0.05) According to the above results, it is considered that Samduhaejungtang-gamibang has protective effect against liver injury in rats induced by $CCl_4$ and d-galactosamine. So it is required to study about the actions of mutual relation of medicines and patho-mechanism by experiment.

  • Hepatitis, liver cirrhosis and liver cancer caused by viral infection are among the most prevalent causes of death in Korea. Several medicines have been in use despite their nonsatisfactory effects on these disease. Some herbal medicines put to use recently have not shown beneficial effects, either. This paper evaluates the effects of extracts from 10 traditional Korean herbal medicines on rats with hepatic damage induced by galactosamine. Rubus coreanus showed an anti-inflammatory effect as shown on the data of activities of serum transaminases.

지구자(枳椇子) 산청목(山靑木) 배합(配合)이 D-galactosamine 유발 백서의 간손상에 미치는 영향(影響) (Effects of Mixture with Hovenia dulcis Thunb and Acer tegmentosum Maxim on Liver Failure Induced by D-galactosamine in Rats)

  • Objectives : This study was carried out to investigate the hepatoprotective effects of mixture with Hovenia dulcis Thunb (HDT) and Acer tegmentosum Maxim(ATM) on D-galactosamine-induced liver failure in rats. Methods : The animals were divided into 4 groups: control, with liver failure and no treatment; H1A1, with liver failure and oral treatment with HDT 1 and ATM 1; H1A2, with liver failure and oral treatment with HDT 1 and ATM 2; H1A4, with liver failure and oral treatment with HDT 1 and ATM 4. The animals were treated for 3 weeks and then examinations of change of body weight, aspartate aminotransferase (AST), alanine aminotransferase (ALT), gamma glutamyl transpeptidase ($\gamma$-GTP), total cholesterol, triglyceride, total bilirubin, superoxide dismutase (SOD), catalase, histopathologic change, and complete blood count (CBC) were performed. Results : All experimental groups had significantly decreased AST in serum and markedly increased activity of SOD as compared with the control group. H1A1, and H1A4 significantly decreased ALT in serum and H1A4 at 2 weeks was significantly higher on the change of body weight as compared with the control group. In histopathologic change of liver tissue by light microscopy, all experimental groups showed recovery effects of liver cells which were damaged by D-galactosamine. Conclusions : Based upon these results, it could be assumed that a mixture of HDT and ATM has hepatoprotective and antioxidative effects on D-galactosamine-induced liver failure. Therefore, a mixture of HDT and ATM might be utilized as a protective agent in therapy for liver diseases.

주차환(舟車丸)이 $CCl_4$ 및 d-galactosamine으로 유발된 흰쥐의 간손상(肝損傷)에 미치는 영향 (Protective effects of Jucha-whan on liver injury of guinea pigs induced by $CCl_4$ and d-galactosamine)

  • Objective : In order to investigate the curative effect of Jucha-whan on the protective liver of guinea pigs induced by $CCl_4$ and d-galactosamine, serum transaminase(GOT, GPT), lactic dehydrogenase(LDH), alkaline phosphatase(ALP), glutathione S-transferase(GST), superoxide dismutase(SOD) were used to measure enzyme activities and lipid peroxide level. Method : The subject animals were divided into 5 groups; a control group(untreated), a subject group(administered with 0.9% Saline solution), a sample I (500mg/kg administered), sample II group (1000mg/kg administered), positive control group(administered with 200mg/kg silymarine). Result : The inhibitory effects on the serum GOT, LDH, ALP, SOD and Lipid peroxide level activities in protective liver of mice induced by $CCl_4$ were noted both in the sample I group and sample II. The inhibitory effects on the serum GPT activities in protective liver of guinea pigs induced by $CCl_4$ were noted in sample II group, but it was not noted in the sample I. The inhibitory effects on the GST activities in protective liver of guinea pigs induced by $CCl_4$ were not noted in both sample I and sample group II. The inhibitory effects on the serum GOT, GPT activities in protective liver of guinea pigs induced by d-galactosamine were noted in both sample I and sample II, but it was not recognizable statistically. The inhibitory effects on the serum LDH activities in protective liver of guinea pigs induced by d-galactosamine were noted in sample II, but it was not noted in sample I group. Conclusion : According to the above results, it is considered that Jucha-whan has treatment effects on liver injury in guinea pigs induced by $CCl_4$ and d-galactosamine. So it is required to study about the actions of mutual relation of medicines and patho-mechanism through experiment.

인진사령산이 흰쥐 간손상(肝損傷)에 미치는 영향(影響) (Effects of Injinsaryung-san on Experimental Liver Damage in Rats)

  • The following results were made by observation GOT, GPT, ALP, total cholesterol and higlyceride in serum to research the effects of medicines which are Injinsaryungsan(Sample- A) and another medicine(Sample-B) of which Injin(Artemisiae Capillaris Herba)was increased in quantity on liver damaged by $CCl_4$ and d-galactosamine in rats. 1. The high concentrated extracts of Sample A group and Sample- B group showed significant inhibltory effects on the increase of serum GPT, ALP, LDH, total cholesterol and triglyceride levels induced by $CCl_4$ and d-galactosamine. 2. The high concentrated extracts of sample A group and Sample B group showed more significant inhibitory effects(P〈0.001) than the low ones' effects(P〈0.01) on the increase of serum triglyceride level induced by $CCl_4$ 3. Sample-A group showed significant inhibitory effects on the increase of serum GOT, GPT. ALP, total cholesterol and triglyceride levels, but no significance on the increase of serum LDH level induced by d-galactosamine. 4. Sample-B group showed very significant inhibitory effects on the increase of serum GOP, GPT, ALP, LDH, total cholesterol and triglyceride levels induced by d-galactosamine. 5. As compared with Sample-A group, Sample-B group of which Injin was increased in quantity showed more significant inhibitory effects on all items of this experiment induced by $CCl_4$ and d-galactosamine. As mentioned above. it seemed that both Injinsaryungsan and another medicine of which Injin was increased in quantity had effects protecting liver and anti-fatty liver induced by $CCl_4$ and d-galactosamine in rats. Specially Sample-B group had very significant effects on liver damage as compared with Sample-B group. Therefore it seems that more researches on variation according to the increase of Injin dose must be continued for curing liver diseases.

The increases of the serum total bilirubin content level in rats induced by d-galactosamine were inhibited in sample Ⅱ group(p<0.05). According to the above results, it is considered that Samduhaejungtang has protective effect against liver injury in rats induced by $CCl_4$ and d-galactosamine.
