Effects of Ginseng Total Saponin on [3H]DAGO Bindings of Opioid μ-Receptors

  • Crude synaptic membrane fractions from the frontal cortex, striatum, brain stem and whole brain of rat were prepared to assay the effects of ginseng total saponin (GTS) on [$^3$H]DAGO bindings of the opioid $\mu$-receptors. Scatchard plots analysis binding data demonstrated that GTS (0.1 mg/ml) decreased the affinity of specific [$^3$H]DAGO bindings without changes in B$\_$max/ in the frontal cortex and striatum. On the other hand, GTS did not affect the [$^3$H]DAGO bindings iii the brain stem and whole brain. These results suggest that the regulation of [$^3$H]DAGO bindings by GTS may play roles in the change of the pharmacological responses of $\mu$-opioids.

  • We have previously reported that the antagonism of U-50,488H-induced antinociception in mice pretreated with ginseng total saponins (GTS) Ivas abolished by pretreatment with a serotonin precursor, 5-hydroxytryptophan (5-HTP), but not by a noradrenaline precursor, L-dihydroxyphenylalanine (L-DOPA) in the tail flick test. In the present experiments, the effect of the same GTS on the development of tolerance to U-50,488H-induced antinociception was determined. GTS inhibited the development of tolerance to U-50,488H-induced antinociception. The inhibitory effect of GTS on the development of tolerance to U-50,488H-induced antinociception was reversed by 5-HTP, but not by L-DOPA. These findings suggest that the inhibitory effect of GTS on the development of tolerance to U-50,488H-induced antinociception is dependent on serotonergic mechanisms. Green Tobacco Sickness Among Tobacco Harvesters in a Korean Village

  • Background: Green tobacco sickness (GTS), an occupational disease in tobacco harvesters, is a form of acute nicotine intoxication by nicotine absorption through the skin from the wet green tobacco plant. We carried out a questionnaire survey and measured cotinine concentration, the metabolic product of nicotine, to determine the prevalence, incidence, and risk factors of GTS in Korean tobacco harvesters. Methods: We measured cotinine concentrations, and administered a questionnaire survey to tobacco harvesters in Cheongsong-gun, Gyeongsangbuk-do, Korea. We repeatedly measured urine cotinine concentration five times with a questionnaire survey. Results: Cotinine concentration at dawn was significantly higher than that at other times; it was significantly lower during the nonharvesting period than during the harvesting period. However, little change in cotinine concentration was detected in the daytime during the harvesting period. Study participants included 20 men and 20 women. The prevalence of GTS was 37.5% and was significantly higher in women than in men (55.0% vs. 20.0%, p < 0.01). GTS incidence according to number of workdays was 3.4 occurrences/100 person days. Conclusion: In this study, nicotine exposure and metabolism were experimentally determined from the time of cotinine exposure, and biological monitoring was performed in each season. In the future, this information may be valuable for medical decision-making in GTS prevention.

Effect of Ginseng Total Saponin on the Development of Psychic and Physical Dependence on Nalbuphine

  • This study was undertaken to estimate whether nalbuphine, a mixed agonist/antagonist opioid analgesic produced psychic dependence. Moreover, the physical dependence liability of nalbuphine was compared with that of morphine after 7 days administrations of the drugs in mice and rats, and the effects of ginseng total saponin (GTS) on the development of physical dependence on nalbuphine were also studied. Nalbuphine did not produce psychic dependence. However, various abstinence signs precipitated by naloxone were observed in nalbuphine-dependent mice and rats. As the nature of the dependence syndrome produced by nalbuphine 30 mg/kg under these conditions seems similar to that induced by morphine 10 mg/kg, it is clear that nalbuphine possesses the substantial abuse potential. Therefore, nalbuphine may be needed to initiate more stringent controls for the prevention of nalbuphine abuse. On the other hand, GTS inhibited the development of physical dependence on nalbuphine and reduced the contents of dopamine and its metabolite in the brains of mice. Accordingly, results of this study suggest that the inhibitory effects of GTS on the development of physical dependence on nalbuphine may involve dopaminergic mechanism. GTS may be useful for the therapy of physical dependence on nalbuphine.

Effect of Panax ginseng on morphine- and steroids-induced immunosuppression

  • Effects of Panax ginseng were tested on morphine-and steroids-induced immunosuppression, focusing on mechanism and identification of active components. To investigate overall effects of morphine and ginseng total saponin (GTS) on immune system, body weight and lymphoid organ weight were measured. Morphine significantly reduced body weight, spleen/body weight, and thymus/body weight ratio. GTS, at 100mg/kg (oral), restored spleen/body weight ratio. Because morphine is known to increase corticosterone level, serum corticosterone level was measured by radioimmunoassay. Serum corticosterone was increased by morphine and it was restored to the control level by GTS 100mg/kg (oral). In vitro proliferation studies were also conducted to study the effects of ginseng on steroids-induced immunosuppression. While ginsenoside Rg$_1$ and ginseng alkaloids were effective on proliferation and dexamethasone-induced death of thymocytes, 50% ginseng ethanol extract and polysaccharides were effective on splenocytes. In vivo mprohine-induced apoptosis of thymus was partly protected by GTS.

A GTS Scheduling Algorithm for Voice Communication over IEEE 802.15.4 Multihop Sensor Networks

  • The recent increase in use of the IEEE 802.15.4 standard for wireless connectivity in personal area networks makes of it an important technology for low-cost low-power wireless personal area networks. Studies showed that voice communications over IEEE 802.15.4 networks is feasible by Guaranteed Time Slot (GTS) allocation; but there are some constraints to accommodate voice transmission beyond two hops due to the excessive transmission delay. In this paper, we propose a GTS allocation scheme for bidirectional voice traffic in IEEE 802.15.4 multihop networks with the goal of achieving fairness and optimization of resource allocation. The proposed scheme uses a greedy algorithm to allocate GTSs to devices for successful completion of voice transmission with efficient use of bandwidth while considering closest devices with another factor for starvation avoidance. We analyze and validate the proposed scheme in terms of fairness and resource optimization through numeral analysis.

  • This study examined the Influence of ginseng total saponins (GTS) on the analgestic action and tolerance development of pentazocine in mice. Pentazocine prolonged the latency to response in the tail flick rather than in the tail pinch test. The analgesic effect of pentaEocine was antagonized by naloxone and completely eliminated by pretreatment u·ith f-chlorophenylalanine (PCPA). GTS provented the pentasocine-incuced analgesia ann inhibited the development of tolerance to pentazocine. The antagonistic effect of GTS on the pentazocine-induced analgesia was abolished by 5-HTP, but not by L-DOPA. These results suggest that GTS inhibits the analgesic action of pentazocine by the interaction with serotonergic neuron.

Ginseng Total Saponin Attenuate Cardiac Hypertrophy Induced by Homocysteine in Rats

  • Recent studies have shown that Panax ginseng has a variety of beneficial effects on the cardiovascular system. Homocysteine (Hcy), which is derived from methionine, has been closely associated with the increased risk of cardiovascular diseases. In the present study, whether the in-vivo long-term co-administration of ginseng total saponins (GTS), active ingredients of Panax ginseng, with L-methionine (Met) inhibits methionine-induced hyperhomocysteine (HHcy) and H-Hcy-induced cardiovascular dysfunctions was investigated, and it was found that the plasma Hcy level, which was measured after 30 and 60 days, in the GTS+Met co-administration group was more significantly reduced than in the Metalone-treatment group. The left-ventricle (LV) wall thickness of the heart was likewise examined in each treatment group, and it was found that the co-administration of GTS with Met significantly reduced the Met-induced LV wall thickness. The results of the study indicate that the in-vivo long-term co-administration of GTS with Met not only inhibited H-Hcy induced by long-term Met-alone administration but also attenuated the H-Hcy-induced cardiovascular dysfunctions in rats.

  • 본 논문에서는 산업용 무선 제어네트워크의 네트워크 이용률과 가용 무선 I/O 노드 수를 동시에 증대시길 수 있는 동적 GTS할당 기법을 제안하였다. 모의실험을 통해, 제안된 동적 GTS할당 기법이 가용 노드수와 네트워크 이용률의 향상시킴을 살펴 볼 수 있다.

  • In this paper, a dynamic GTS allocation method of wireless control networks is proposed for the use of factory automation using IEEE 802.15.4 MAC protocol. A superframe of IEEE 802.15.4 is applied to the transmission method of real-time periodic I/O data of wireless control systems within the limited time in factory environment. The method is proposed for efficient transmission of real-time periodic I/O traffic. The simulation results show the average network utilization and available I/O node numbers could be increased by the proposed method.
