Grp78/BiP과 Grp94의 생화학적 분석 (Biochemical Characterization of Glucose-Regulated Proteins, Grp94 and Grp78/BiP)

  • Glucose-regulated proteins (grp's), srp94 3nd grp78/BiP, are a group of stress proteins which are highly synthesized in cells exposed to a variety of stressful agents including tunicamycin 3nd Ca2+ ionophore. Grp78/BiP is hon to function as a molecular chaperone which regulates the folding and assembly of secretory or membrane proteins, but the biological function of grp941 remains to be elucidated. In this study, we have examined the intracellular distribution of grV's and the function of srp94. Grp's are resident in the endoplasmic reticulum (ERI 3nd a specific sequence (Lys-Asp-Glu-Leu) at their C-terminus is known to be responsible for their retention within the ER. However, it has been unclear whether upon disturbance of cellular Caa+ homeostasis by the Ca2+ ionophore, grp94 is retained within the ER or secreted into the medium. In this study, we showed that in the presence of C3a+ ionophore, grp94 and gif78/BiP are present in the cells, mainly within the ER. We have also investigated whether grp94 might function as a molecular chaperone. Here we showed that in the immunoglobulin (Ig)-secreting hvbridom3 cells, grp94 transientlY interacts with fully glycosylated Is heavy chain, suggesting that grpg94 may be involved in facilitating the folding and assembly of Ig heavy chains.

Distribution, Content and Molecular Heterogeneity of Gastrin-Releasing Peptide in Rat Pancreas

  • Although importance of intrapancreatic neurons containing gastrin-releasing peptide (GRP) in control of exocrine secretion has been raised, the nature of GRP in the pancreas is unclear. Thus, the present study was undertaken to see distribution, content and molecular heterogeneity of immunoreactive GRP in the rat pancreas. Content of immunoreactive GRP in the rat pancreas was $2.99\;{\pm}\;0.66$ ng/g wet tissues determined by radioimmunoassay. Immunoreactive GRP was most abundantly expressed in the duodenal part among 3 parts of the pancreas; duodenal, body and splenic part. Vagotomy failed to change the content of immunoreactive GRP in the pancreas. Three distinct forms of immunoreactive GRP, very identical to GRP-27, bombesin-24 and neuromedin C, were observed in the rat pancreas by using reversed phase $C_{18}$ HPLC and Sephadex G-50 superfine column chromatography. Cell bodies of neurons containing immunoreactive GRP were scattered in pancreatic connective tissues and their nerve fibers innervated pancreatic acini and large ducts as determined by immunohistochemistry. The present results suggest that three distinct forms of GRP exist in intrapancreatic GRPergic neurons, which exert a stimulatory role in pancreatic exocrine secretion in rats.

GRP78 Secreted by Colon Cancer Cells Facilitates Cell Proliferation via PI3K/Akt Signaling

  • Glucose regulated protein 78 (GRP78) is usually recognized as a chaperone in the endoplasmic reticulum. However, increasing evidence indicates that GRP78 can be translocated to the cell surface, acting as a signaling receptor for a variety of ligands. Since little is known about the secretion of GRP78 and its role in the progression of colon cancer we here focused on GRP78 from colon cancer cells, and purified GRP78 protein mimicking the secreted GRP78 was able to utilize cell surface GRP78 as its receptor, activating downstream PI3K/Akt and Wnt/${\beta}$-catenin signaling and promote colon cancer cell proliferation. Our study revealed a new mode of action of autocrine GRP78 in cancer progression: secreted GRP78 binds to cell surface GRP78 as its receptor and activates intracellular proliferation signaling.

지중매설 유리섬유복합관의 관변형에 관한 안전성 평가 (A Safety Evaluation on the Ring Deflection of Buried GRP Pipes)

  • 최근 지중매설 유리섬유복합관(GRP관)은 유해한 환경에서 뛰어난 성능을 보유하고 있어 하수관거용으로서 사용이 증가 되고 있다. 또한 지중에 매설되어 있는 조건에서 기존의 콘크리트관 등 강성관에 비해 구조적 성능이 뛰어나다. 지중매설 GRP관은 주로 상부에 작용하는 상부토압과 활하중에 의한 압축응력에 의해 원주방향으로 변형이 일어나게 된다. GRP관의 구조적 거동은 매설토와 주변의 지반의 성질에 따라 다르게 설계되어야 한다. GRP관의 설계는 Spangler 의 변형량 계산식을 Watkins에 의해 수정되어 사용되고 있다. 이 연구에서는 Watkins의 관변형 추정식에 GRP관의 재료적 특성을 고려하여 관변형량을 예측하였다.

GRP 복합관의 열팽창계수 측정 (Measurements of Thermal Expansion Coefficients in GRP Pipe)

  • 본 연구에서는 스트레인게이지 회로를 이용하여 GRP 복합관의 열팽창계수를 측정하고자 하였다. 이를 위해 다양한 스트레인게이지 회로를 적용하여 알루미늄 보의 열팽창계수를 측정함으로써 측정방법의 타당성을 검증하였다. 또한 스트레인게이지의 부착위치와 반복횟수를 달리하며 또한 열팽창계수가 다른 스트레인게이지를 적용한 경우에 대해 GRP 복합관의 길이방향 및 원주방향 열팽창계수를 측정함으로써 열변형률 결과와 측정결과의 재현성에 미치는 영향을 조사하였다. 연구결과에 따르면 GRP 복합관의 경우 보강된 유리섬유에 의해 원주방향의 열변형률이 제한되어 원주방향 열팽창계수가 길이방향의 열팽창계수에 비해 낮게 나타났다. 또한 GRP 복합관의 후경화로 인해 측정횟수가 증가할수록 측정된 열팽창계수는 다소 증가하지만 증가폭은 점차 감소하였다. 아울러 열팽창계수가 다른 스트레인게이지를 적용하더라도 기준보상시편을 통해 스트레인게이지의 열변형률을 보상하면 동일한 열팽창계수가 얻어짐을 알 수 있었다.

Glucose regulated protein 78 promotes cell invasion via regulation of uPA production and secretion in colon cancer cells

  • Glucose regulated protein 78 (GRP78) is frequently highly expressed in tumor cells, contributing to the acquisition of several phenotypic cancer hallmarks. GRP78 expression is also positively correlated with tumor metastasis, and promotes hepatocellular carcinoma cell invasion via increasing cell motility, however, other mechanisms involving the prometastatic roles of GRP78 remain to be elucidated. Here we report that forced GRP78 expression promotes colon cancer cell migration and invasion through upregulating MMP-2, MMP-9 and especially uPA production. These effects of GRP78 are mediated by enhancing the activation of ${\beta}$-catenin signaling. Interestingly, we identify that GRP78 interacts with uPA both in the cells and in the culture medium, suggesting that GRP78 protein is likely to directly facilitate uPA secretion via protein-protein interaction. Taken together, our findings demonstrate for the first time that besides stimulation of cell motility, GRP78 can act by increasing proteases production to promote tumor cell invasion.

  • Microiniection of genes Into Xenopus laeuis oocvtes in highly useful in the annvsis of gene regulation, since a large number of oocvtes can be injected in a relatively short time. The GRP78 enhancer has been identified to a 291-bp fragment that spans a region of GRP78 promoter between -378 and -87 (Lin et at., 1986: Kim and Lee, 1989). We examined whether this GRP78 enhancer is effective in directing expression of heterologous gene in Xenopus laeuis oocytes. The chloramphenicol acetvltransferase (CAT) fusion constructs containing the GRP78 promoter and the SV4O early promoter were constructed and were injected into nuclei of Xencpus laeuis oocvtes. The recipient oocvtes were then assayed for CAT activity. The fusion constructs exhibited higher activity as compared to SV40 promoter tested here. The GRP78 enhancer showed 8.5- to 9.2-fold enhancement over that of the SV4O promoter. The orientation of GRP78 enhancer with respect to the direction of CAT transcription unit had no significant effect. Thus, the GRP78 enhancer is a viable candidate for the construction of expression system for use in Xenopus laevss oocvtes and will be important for the studY of a gene expression throughout development.

지중매설 GRP 관의 관변형 예측 (Prediction of Ring Deflection GRP Pipe Buried Underground)

  • Glass fiber reinforced plastic (GRP) pipes buried underground are attractive for use in harsh environments, such as for the collection and transmission of liquids which are abrasive and/or corrosive. In this paper, we present the result of investigation pertaining to the structural behavior of GRP flexible pipes buried underground. In the investigation of structural behavior such as a ring deflection, experimental and analytical studies are conducted. In addition, vertical ring deflection is measured by the field test and finite element analysis (FEA) is also conducted to simulate behavior of GRP pipe buried underground. Based on the results from the finite element analyses considering soil-pipe interaction the vertical ring deflection behavior of buried GRP pipe is predicted. In addition, analytical and experimental results are compared and discussed.

  • Endoplasmic reticulum (ER)내에 단백질의 folding과 안정화에 관여하는 단백질을 molecular cha-perone이라고 한다. GRP94 역시 ER내에 존재하는 molecular chaperone으로 알려지고 있지만 갑상선세포에서 단백질의 folding에 관여한다는 증거는 아직 불충분하다. 본 설험은 molecular chaperone을 세포내에서 overexpression시킬 수 있는 system을 확립하였다. 그 중에서 GRP94가 단백질의 folding에 직접적으로 관여한다는 증거를 얻기 위하여, endogenous GRP94를 code한 cDNA를 overexpression vector에 의해서 forced expression시킴으로 신생thyroglobulin의 folding에 직접적으로 관여하는 증거를 immun-oprecipitation으로 증명하였다.

Purification and characterization of a 1,3-β-D-glucan recognition protein from Antheraea pernyi larve that is regulated after a specific immune challenge

  • Pattern recognition receptors are known to participate in the activation of Prophenoloxidase system. In this study, a 1,3-${\beta}$-D-glucan recognition protein was detected for the first time in Antheraea pernyi larvae (Ap-${\beta}GRP$). Ap-${\beta}GRP$ was purified to 99.9% homogeneity from the hemolymph using traditional chromatographic methods. Ap-${\beta}GRP$ specifically bind 1,3-${\beta}$-D-glucan and yeast, but not E. coli or M. luteus. The 1,3-${\beta}$-D-glucan dependent phenoloxidase (PO) activity of the hemolymph inhibited by anti-Ap-${\beta}GRP$ antibody could be recovered by addition of purified Ap-${\beta}GRP$. These results demonstrate that Ap-${\beta}GRP$ acts as a biosensor of 1,3-${\beta}$-Dglucan to trigger the Prophenoloxidase system. A trace mount of 1,3-${\beta}$-D-glucan or Ap-${\beta}GRP$ alone was unable to trigger the proPO system, but they both did. Ap-${\beta}GRP$ was specifically degraded following the activation of proPO with 1,3-${\beta}$-Dglucan. These results indicate the variation in the amount of Ap-${\beta}GRP$ after specific immune challenge in A. pernyi hemolymph is an important regulation mechanism to immune response.
